Synthesis and structure-activity relationship of N-benzyl-3-phenylpropanamides as transient receptor potential vanilloid 1 (TRPV1) antagonists are described. A variety of substituents such as halide, ester, nitro, and alkyl groups at 2 or 3-position of 4-(methylsulfonylamino) benzyl unit were examined. These compounds exhibited potent 45Ca2+ uptake inhibition in rat DRG neuron via TRPV1 blockade. Especially compound 28c, has been identified as a potent antagonist with IC50 of 38 nM.

Guided by bioisosterism and pharmacokinetic parameters, we designed and synthesized a series of novel benzamide derivatives. Preliminary in vitro studies indicated that compounds 10b and 10j show significant inhibitory bioactivity in HepG2 cells (IC50 values of 0.12 and 0.13 μM, respectively). Compounds 10b and 10j induced the expression of HIF-1α protein and downstream target gene p21, and upregulated the expression of cleaved caspase-3 to promote tumor cells apoptosis.

<P>The hepatoprotective effects of a diterpenoid acanthoic acid isolated from <I>Acanthopanax koreanum</I> N<SMALL>AKAI </SMALL>were evaluated in a <SMALL>D</SMALL>-galactosamine/lipopolysaccharide-induced fulminant hepatic failure mouse model. Mice were pretreated orally with acanthoic acid 12 and 1 h before intraperitoneal injection of <SMALL>D</SMALL>-galactosamine and lipopolysaccharide. Pretreatment with the compound markedly reduced lethal liver injury in experimental animals. The effects were likely associated with a significant decrease in serum tumor necrosis factor (TNF)-α levels, which are correlated not only with those of alanine aminotransferase and aspartate aminotransferase but also with the reduced number of apoptotic hepatocytes in the liver as confirmed using the terminal deoxynucleotidyl transferase-mediated dUTP nick end-labeling method and DNA fragmentation assay. These results suggest that acanthoic acid protects against <SMALL>D</SMALL>-galactosamine/lipopolysaccharide-induced fulminant liver failure at least in part by a mechanism associated with the down-regulation of TNF-α secretion.</P>
Complete Mitochondrial Genome of a Tongue Worm Armillifer agkistrodontis

Armillifer agkistrodontis (Ichthyostraca: Pantastomida) is a parasitic pathogen, only reported in China, which can cause a zoonotic disease, pentastomiasis. A complete mitochondrial (mt) genome was 16,521 bp comprising 13 protein-coding genes (PCGs), 22 tRNA genes, 2 rRNA genes, and 1 non-coding region (NCR). A phylogenetic tree drawn with the concatenated amino acid sequences of the 6 conserved PCGs (atp6, cox1-3, and nad2) showed that A. agkistrodontis and Armillifer armillatus constituted a clade Pentastomida which was a sister group of the Branchiura. The complete mt genome sequence of A. agkistrodontis provides important genetic markers for both phylogenetic and epidemiological studies of pentastomids.

A generator-grid coupling calculation model is established to study the leading-phase operational capability of a 770 MW jumbo hydro-generator in a Chinese ultra-mega hydropower station. The static and dynamic stability of the generator are analyzed and calculated to obtain stability limits under leading-phase operating conditions. Three-dimensional (3D) time-varying nonlinear moving electromagnetic and temperature field models of the generator end-region are also established and used to determine the magnetic field, loss, and temperature of the end-region under the leading-phase operating condition. The simulation results agree with data measured from the actual 770 MW hydro-generator. This paper provides reliable reference data for the leading-phase operation of a jumbo hydro-generator, which will help to improve in the design and manufacture of future hydro-generators.
A high-efficiency simulation method of wind field and its application on transmission line

Generally, the fluctuating wind is simplified as several independent one-dimensional multivariate stationary Gaussian processes in simulating a natural wind field. The correlation in the lateral, longitudinal and vertical directions should all be considered in the simulation of longitudinal wind field for the large-span spatial structures. In fact, this type of structure has lots of simulation points. The calculation amount of wind field simulation by the harmonic superposition method depends on the scale of cross-spectral density matrix, which is directly related to the number of simulated points, leading to a low efficiency when generating the time-varying wind speed. This paper innovatively proposes a high-efficiency simulation method for the longitudinal wind field based on Taylor’s hypothesis. Subsequently, the effectiveness of the proposed wind field method was verified by the numerical simulation. Finally, the dynamic responses of a transmission tower-line system under the wind loadings generated with the new method and traditional method are calculated and compared. The percentages difference of the mean and maximum axial force at the main tower members are less than 0.02% and 1%, respectively, indicating the effectiveness of the proposed time delay method. The results also show that the proposed simulation method of wind field can not only ensure the simulation accuracy, but also significantly improve the efficiency of wind speed generation, which is suitable for the wind load simulation of large-span spatial structures.

Experimental study on aerodynamic characteristics of conductors covered with crescent-shaped ice

Conductor galloping is a common disaster for the transmission lines. Among the existing analytical methods, the wind tunnel test is highlighted as the most effective approach to obtain the aerodynamic coefficients. In this paper, the aerodynamic coefficients of 12 conductor models covered with the crescent-shaped ice, which were fabricated considering the surface roughness of the iced conductor, were obtained based on the wind tunnel test. The influence of the Reynolds number and the shape parameter β, defined as the ratio of ice thickness to the diameter, were investigated. In addition, the effect of surface roughness of the iced conductor was discussed. Subsequently, unsteady areas of conductor galloping were calculated according to the Den Hartog criterion and the Nigol criterion. The results indicate that the aerodynamic coefficients of iced conductors change sharply at the attack angles of 20° and 170° with the increase of β. The surface roughness of iced conductors changed the range of attack angle, which was influenced by the increase of the Reynolds number. The experimental results can provide insights for preventing and controlling galloping.

Background: This retrospective study aimed to validate the safety and effectiveness of hepatectomy for huge hepatocellular carcinoma (HCC). Materials and Methods: Data of patients who underwent hepatectomy for HCC between January 2006 and December 2012 were reviewed. The patients were divided into three groups: huge HCC(${\geq}10cm$ in diameter), large HCC(${\geq}5$ but<10 cm in diameter) and small HCC(<5cm in diameter). Results: Characteristics of pre-operative patients in all three groups were homogeneously distributed except for alpha fetal protein (AFP)(p<0.001).The 30, 60, 90-day post-operative mortality rates were not different among the three groups (p=0.785, p=0.560, and p=0.549). Laboratory data at 1, 3, and 7 days after surgery also did not vary. The 5-year overall survival (OS) and 5-year disease-free survival (DFS) rates in the huge and large HCC groups were lower than that of the small HCC group (OS: 32.5% vs 36.3% vs 71.2%, p=0.000; DFS: 20.0% vs 24.8% vs 40.7%, p=0.039), but there was no difference between the huge and large HCC groups (OS: 32.5% vs 36.3%, p=0.667; DFS: 20.0% vs 24.8%, p=0.540). In multivariate analysis, five independent poor prognostic factors that affected OS were significantly associated with worse survival (p<0.05), namely, AFP level, macrovascular invasion, Edmondsone Steiner grade, surgical margin and Ishak score. AFP level, macrovascular invasion, microvascular invasion, and surgical margin influenced disease-free survival independently (p<0.05). Conclusions: The safety of hepatectomy for huge HCC is similar to that for large and small HCC; and this approach for huge HCC may achieve similar long-term survival and disease-free survival as for large HCC.

To identify novel therapeutic agents to treatcancer, we synthesized a series of diaryl ether derivatives. Structure–activity relationship studies revealed that thepresence of a chlorine or hydroxyl at the para-position onthe phenyl ring (5h or 5k) significantly enhanced antitumoractivity. Compound 5h had stronger growth inhibitory activityin HepG2, A549, and HT-29 cells than compound 5k,with IC50 values of 2.57, 5.48, and 30.04 lM, respectively. Compound 5h also inhibited the growth of other cells lines,including Hep3B, PLC/PRF5, SMMC-7721, HeLa, andA375, with IC50 values of 2.76, 4.26, 29.66, 18.86, and10.21 lM, respectively. The antitumor activity of compound5h was confirmed by a colony forming assay. Further,our results indicated that the antitumor activity ofcompound 5h may be mediated by enhancing expression ofp21 and cl-caspase3, and leading to apoptosis of cancercells.
