        Synthesis and DNA-binding Properties of Trehalose-tethered Monomeric and Dimeric Berberines

        Trehalose-tethered monomeric and dimeric berberines were synthesized in 50% and 30% from the reaction of berberrubine with 6-tosyl-${\alpha}$,${\alpha}^{\prime}$-trehalose and 6,6'-ditosyl-${\alpha}$,${\alpha}^{\prime}$-trehalose, respectively, and fully characterized by MS (HR and ESI) and NMR ($^1H$, $^{13}C$, COSY and HSQC). Spectrophotometric and spectrofluorimetric titrations indicated that compared with berberine, trehalose-tethered monomeric berberine had comparable DNA-binding affinity toward calf-thymus DNA, whereas trehalose-spaced dimeric berberine exhibited higher DNA-binding affinity. The potential application of these conjugates is also briefly discussed.

        Composition Dependence of Structural and Magnetic Properties of Mn<SUB>3+x</SUB>Ga<SUB>1-x</SUB>C (0 ≤ x ≤ 1)

        Both Mn₄C (=Mn₃MnC) and Mn₃GaC have been studied previously. However, the reports on Mn<SUB>3+x</SUB>Ga<SUB>1-x</SUB>C (0 ≤ x ≤ 1) with intermediate compositions are very rare. In this work, the structure and magnetic properties of Mn<SUB>3+x</SUB>Ga<SUB>1-x</SUB>C prepared by using solid state reaction were studied systematically. High purity anti-perovskitetype Mn<SUB>3+x</SUB>Ga<SUB>1-x</SUB>C were obtained in the composition range of 0 ≤ x ≤ 0.5, above which Mn₂₃C₆ precipitates and the fraction of Mn₂₃C₆ in the samples increases with increasing x. The structural stability, lattice parameters, and room temperature saturation magnetization of ferromagnetic Mn<SUB>3+x</SUB>Ga<SUB>1-x</SUB>C (0 ≤ x ≤ 1) decreases with increasing x. The Curie temperature of Mn<SUB>3+x</SUB>Ga<SUB>1-x</SUB>C (0 ≤ x ≤ 1) increases with increasing x. Most Mn<SUB>3+x</SUB>Ga<SUB>1-x</SUB>C with varied x exhibit near-zero coercivity and zero remanent magnetization. This work indicates that the temperature coefficient of magnetization of Mn<SUB>3+x</SUB>Ga<SUB>1-x</SUB>C may be tuned by tuning the fraction of the Ga atoms.

        Depth-enhanced integral imaging system based on spatial filtering

        An integral imaging system enabling an enhanced depth of field is proposed based on spatial filtering. In the proposed system, spatial filtering is used on each elemental image to obtain a depth-enhanced elemental image in the pickup stage, and the obtained elemental images are composed to form an elemental image array for the computational reconstruction in the display stage. A preliminary experiment was carried out, and the experiment results verified the feasibility of the system.

        Association of Lipoprotein-Associated Phospholipase A_2 with Characteristics of Vulnerable Coronary Atherosclerotic Plaques

        Purpose: Lipoprotein-associated phospholipase A2 (Lp-PLA_2) is an inflammatory enzyme expressed in atherosclerotic plaques. We investigated the association of circulating Lp-PLA_2 with characteristics of vulnerable coronary atherosclerotic plaques. Materials and Methods: We recruited 113 patients with either unstable angina (UA, n=59) and stable angina (SA, n=54) by coronary angiography. Thirty-six healthy subjects served as controls. Intravascular ultrasound (IVUS) was used to evaluate the characteristics of coronary atherosclerotic plaque, and serum Lp-PLA2 concentration was measured as well. Results: Lp-PLA_2 concentration was significantly higher in both UA and SA patients [(396±36) μg/L and (321±39) μg/L, respectively]compared with the controls [(127±49) μg/L, p<0.01], and higher in UA than SA group. IVUS findings showed that remodeling index (RI) (0.91±0.15 vs. 0.85±0.11, p=0.005) and eccentricity index (EI) (0.73±0.16 vs. 0.65±0.22, p=0.039) were larger in UA than in SA group, and fibrous caps were thicker in SA than UA group [(0.91±0.23) mm vs. (0.63±0.21) mm, p=0.032]. Moreover, Lp-PLA_2 correlated positively with EI (r=0.439, p<0.01) and RI (r=0.592, p<0.05) in UA group. There was an inverse relationship between Lp-PLA_2 and fibrous cap thickness in both UA (r=-0.587, p<0.001) and SA (r=-0.318, p<0.05) groups. The independent risk factors in UA group were Lp-PLA_2 (OR=1.055, 95% CI: 1.03-1.08, p=0.013), LDL-cholesterol (OR=0.032, 95% CI: 0.00-0.05, p=0.041) and fibrous cap thickness (OR=0.008, 95% CI: 0.00-0.45, p=0.019). Lp-PLA_2 was strongly associated with both EI and fibrous cap thickness in both groups. Conclusion: Serum level of Lp-PLA_2 is associated with both eccentricity index and fibrous cap thickness in both UA and SA groups. Elevated levels of circulating Lp-PLA_2 might to be a strong risk factor and more serious for unstable angina than stable angina.

        Polymorphisms in TYMS for Prediction of Capecitabine-Induced Hand-Foot Syndrome in Chinese Patients with Colorectal Cancer

        Purpose Capecitabine is an extensively used oral prodrug of 5-fluorouracil in treatment of colon cancer and is known to cause hand-foot syndrome (HFS). As the target enzyme for capecitabine, thymidylate synthase (TYMS) plays a key role for 5-fluorouracil metabolism and has been associated with some side effects caused by capecitabine. The aim of our study is to identify the possible genetic predictors of capecitabine-induced HFS (CAP-HFS) in Chinese colorectal cancer patients.Materials and Methods Whole exons of TYMS were sequenced for 288 extreme phenotype HFS patients, including 144 severe or early-onset (first 2 cycles) moderate HFS extreme cases and 144 extreme controls with no reported HFS. The associations between polymorphisms and CAP-HFS were analyzed using logistic regression under an additive model.Results We identified a novel risk mutation (c.1A>G, chr18:657743), was associated with severe HFS in an extreme case who was affected during the first cycle of treatment. Moreover, we identified three new variants, rs3786362, rs699517, rs2790, and two previously reported variants, 5’VNTR 2R/3R and 3′-untranslated region 6-bp ins-del, which were significantly associated with CAP-HFS (p < 0.05). In silico analysis revealed that the effect of these polymorphisms in the TYMS region on the development of HFS might not be restricted solely to the regulation of TYMS expression, but also the TYMS catalytic activity through the indirect effect on ENOSF1 expression.Conclusion This study identified new polymorphisms in TYMS gene significantly associated with CAP-HFS, which may serve as useful genetic predictors for CAP-HFS and help to elucidate the underlying mechanism of HFS.

        Optical Switchable Electrowetting Lens

        <P>The authors report an optical switchable lens using an electrowetting liquid lens. The optical switchable lens is obtained through two different paths, either the annular aperture or the central aperture of the liquid lens. Three polarizers and a liquid crystal cell are used as a light shutter for the two-path transition. A prototype is demonstrated, and the authors analyzed the properties of the switchable electrowetting liquid lens. In contrast to the conventional liquid lenses, our lens system can present high optical power with high resolution for the path of annular aperture, while it can reduce the aberrations and extend the depth of field for the path of central clear aperture. Our lens system has potential applications in target tracking, biomedical science, imaging security, and portable electronic devices.</P>

      • An adaptive liquid lens with a reciprocating movement in a cylindrical hole.

        <P>We demonstrate a liquid droplet which can do a reciprocating movement in a cylindrical hole. The droplet in the hole exhibits a lens character. By applying a voltage, the border of the droplet is stretched to expand by the generated dielectric force. Due to the fixed volume, the dome of the droplet in the hole has to move toward the substrate without changing its surface profile. Therefore, the focal length of the droplet remains unchanged although the focal point is shifted. Once the voltage is removed, the droplet can return to its original state. The droplet with such a movement functions as an adaptive lens. Our lens can provide a high resolution (~114 lp/mm) whether or not it is actuated. The dynamic response time is reasonably fast. Integrating with a solid lens, the compound lens can provide a variable focal length.</P>

        Annular folded electrowetting liquid lens

        <P>We report an annular folded electrowetting liquid lens. The front surface of the lens is coated with a circular reflection film, while the back surface of the lens is coated with a ring-shaped reflection film. This approach allows the lens to get optical power from the liquid-liquid interface three times so that the optical power is tripled. An analysis of the properties of the annular folded electrowetting liquid lens is presented along with the design, fabrication, and testing of a prototype. Our results show that the optical power of the proposed liquid lens can be enhanced from 20.1 to 50.2??m(-1) in comparison with that of the conventional liquid lens (aperture 3.9??mm). It can reduce the operating voltage by 10??V to reach the same diopter as a conventional liquid lens. Our liquid lens has the advantages of compact structure, light weight, and improved optical resolution.</P>
