Purpose: To investigate the efficacy and toxicity of a combination of gemcitabine with nedaplatin (GN) or cisplatin (GC) for patients with unresectable or recurrent esophagus squamous cell carcinoma. Methods: Gemcitabine was administered at 1 g/m2 intravenously on days 1 and 8; and nedaplatin or cisplatin were administered at 80 mg/m2 intravenously on day 1. We analyzed the response rate, overall survival time, progression-free survival time, and toxicity in 21 patients treated with GN and 27 patients treated with GC. Results: In patients treated with gemcitabine plus nedaplatin, the ORR was 47.6%, the median progression-free survival time was 4.1 months, and the median survival time was 9.3 months. In patients treated with gemcitabine plus cisplatin, the ORR was 48.2%, the median progression-free survival time was 3.9 months, and the median survival time was 9.1 months, respectively. There were no statistically significant differences in ORR, PFS and OS between the two groups. In both, the most commonly observed toxicities were thrombocytopenia and fatigue. Nausea and vomiting was more frequent in the GC group than in the GN group. Conclusion: Gemcitabine based chemotherapy was effective and tolerable for patients with unresectable or recurrent esophagus squamous cell carcinoma refractory to first line chemotherapy.

Matrix metalloproteinase (MMP)-2 and MMP-9 are important proteases involved in invasion and metastasis of various tumors. Extra-gastrointestinal stromal tumors (EGISTs) are rare neoplasms. This study was performed to assess MMP-2 and MMP-9 expression in EGIST tissue samples for association with clinicopathological data from the patients. Twenty-one surgical EGIST tissue specimens were collected for analysis of MMP-2 and MMP-9 expression using immunohistochemistry. MMP-2 and MMP-9 proteins were expressed in all of the epithelial cell types of EGISTs, whereas they were only expressed in 75% of the spindle cell type, although there was no statistically significant difference (p>0.05). Expression of MMP-2 and MMP-9 proteins was associated with tumor size, mitotic rate, tumor necrosis, and distant metastasis (p<0.05). MMP-2 expression was linked with MMP-9 levels (p<0.05). However, there was no correlation between MMP-9 expression and age, sex, primary site, or cell morphology in any of these 21 EGIST patients (p>0.05). Moreover, expression of MMP-2 and MMP-9 proteins increased with the degree of EGIST risk. This study provided evidence of an association of MMP-2 and MMP-9 expression with advanced EGIST behavior.

Objective: To make sure the feasibility with $^{18F}FDG$ PET/CT to guided dynamic intensity-modulated radiation therapy (IMRT) for nasopharyngeal carcinoma patients, by dosimetric verification before treatment. Methods: Chose 11 patients in III~IVA nasopharyngeal carcinoma treated with functional image-guided IMRT and absolute and relative dosimetric verification by Varian 23EX LA, ionization chamber, 2DICA of I'mRT Matrixx and IBA detachable phantom. Drawing outline and making treatment plan were by different imaging techniques (CT and $^{18F}FDG$ PET/CT). The dose distributions of the various regional were realized by SMART. Results: The absolute mean errors of interest area were $2.39%{\pm}0.66$ using 0.6cc ice chamber. Results using DTA method, the average relative dose measurements within our protocol (3%, 3 mm) were 87.64% at 300 MU/min in all filed. Conclusions: Dosimetric verification before IMRT is obligatory and necessary. Ionization chamber and 2DICA of I'mRT Matrixx was the effective dosimetric verification tool for primary focal hyper metabolism in functional image-guided dynamic IMRT for nasopharyngeal carcinoma. Our preliminary evidence indicates that functional image-guided dynamic IMRT is feasible.

Post-earthquake fire is a common disaster which causes serious safety issues to infrastructures. This study aims to investigate the residual loading capacities of circular concrete-filled steel tube (CFST) columns under post-earthquake fire experimentally and numerically. The experimental programme contains two loading steps - pre-damage cyclic loading at room temperature and transient state tests with constant compression loads. Three finite element models are developed and validated against the test results. Upon validation, a total of 48 numerical results were generated in the parametric study to investigate the effects of thickness and strengths of steel tube, axial compression ratio and damage degree on the fire resistance of circular CFST columns. Based on the analysis on experimental and numerical results, the loading mechanism of circular CFST columns is discussed. A design method is proposed for the prediction of fire resistance time under different seismic pre-damage and compression loads. The predictions by the new method is compared with the newly generated experimental and numerical results and is found to be accurate and consistent with the mean value close to the unity and a coefficient of variation around 1%.
ZNF424, a novel human KRAB/C2H2 zinc finger protein, suppresses NFAT and p21 pathway

Zinc finger-containing transcription factors are the largest single family of transcriptional regulators in mammals, which play an essential role in cell differentiation, cell proliferation, apoptosis, and neoplastic transformation. Here we have cloned a novel KRAB-related zinc finger gene, ZNF424, encoding a protein of 555aa. ZNF424 gene consisted of 4 exons and 3 introns, and mapped to chromosome 19p13.3. ZNF424 gene was ubiquitously expressed in human embryo tissues by Northern blot analysis. ZNF424 is conserved across species in evolution. Using a GFP-labeled ZNF424 protein, we demonstrate that ZNF424 localizes mostly in the nucleus. Transcriptional activity assays shows ZNF424 suppresses transcriptional activity of L8G5-luciferase. Overexpression of ZNF424 in HEK- 293 cells inhibited the transcriptional activity of NFAT and p21, which may be silenced by siRNA. The results suggest that ZNF424 protein may act as a transcriptional repressor that suppresses NFAT and p21 pathway to mediate cellular functions. [BMB reports 2010; 43(3): 212-218]
Association Between the Ku70-1310C/G Promoter Polymorphism and Cancer Risk: a Meta-analysis

Ku70 plays an important role in DNA double-strand break repair. Studies revealing conflicting results on the role of the Ku70-1310C/G promoter polymorphism on cancer risk led us to perform a meta-analysis to investigate this relationship. Ten case-control studies with 2566 cases and 3058 controls were identified. Odds ratios (ORs) and 95% confidence intervals (CIs) were used to assess the strength of associations. The overall results suggested no association between the Ku70-1310C/G promoter polymorphism and total cancer risk. However, on stratified analysis, significantly increased risks were observed among the Asian population (GG vs. CC: OR=1.50, 95%CI=1.10-2.06; GG vs. CC/CG: OR=1.47, 95%CI=1.07-2.01) and population-based case-control studies (GG vs. CC: OR=1.57, 95%CI=1.12-2.22; CG vs. CC: OR=1.35, 95%CI=1.11-1.64; CG/GG vs. CC: OR=1.37, 95%CI=1.14-1.65). Additionally, variant genotypes were associated with a significantly increased breast cancer risk (GG vs. CC: OR=1.80, 95%CI=1.26-2.56; GG vs. CC/CG: OR=1.40, 95%CI=1.01-1.95).
Cerium vanadate and reduced graphene oxide composites for lithium-ion batteries

<P><B>Abstract</B></P> <P>There has been a significant interest in the development of novel anode materials that can solve the problems of lithium plating and dendrite formation during the discharge-charge process, thus ensuring safety in Li-ion batteries. We synthesized tetragonal CeVO<SUB>4</SUB> as an alternative to graphite, the active material in commercial Li-ion batteries, via a hydrothermal reaction; CeVO<SUB>4</SUB> has lower lithium insertion potentials of 1.0 and 1.5 V versus Li<SUP>+</SUP>/Li compared to those of lithium titanium oxide. In order to overcome the drawbacks of the metal oxide, such as low electrical conductivity and volume change upon cycling, CeVO<SUB>4</SUB>/RGO composites were synthesized by mixing CeVO<SUB>4</SUB> uniformly with reduced graphene oxide (RGO) via a solid-state reaction. The CeVO<SUB>4</SUB>/RGO composites exhibited improved cycling performance and rate capability, with relatively low charging potential of 1.35 V and high power density of 235 W g<SUP>−1</SUP> at 10 wt% RGO, as compared to the pure CeVO<SUB>4</SUB>. Fangchinoline (Fan) inhibits cell proliferation and induces apoptosis in several cancer cell lines. The effects of Fan on cell growth and proliferation in breast cancer cells remain to be elucidated. Here, we show that Fan inhibited cell proliferation in the MDA-MB-231 breast cancer cell line through suppression of the AKT/Gsk-3beta/cyclin D1 signaling pathway. Furthermore, Fan induced apoptosis by increasing the expression of Bax (relative to Bcl-2), active caspase 3 and cytochrome-c. Fan significantly inhibited cell proliferation of MDA-MB-231 cells in a concentration and time dependent manner as determined by MTT assay. Flow cytometry analysis demonstrated that Fan treatment of MDA-MB-231 cells resulted in cell cycle arrest at the G1 phase, which correlated with apparent downregulation of both mRNA and protein levels of both PCNA and cyclin D1. Further analysis demonstrated that Fan decreased the phosphorylation of AKT and GSK-3beta. In addition, Fan up-regulated active caspase3, cytochrome-c protein levels and the ratio of Bax/Bcl-2, accompanied by apoptosis. Taken together, these results suggest that Fan is a potential natural product for the treatment of breast cancer.

Two new diterpenoid alkaloids, N(19)-en-denudatine (1) and N(4)-butanone-flavaconitine (2), were isolated from Aconitum brachypodum Diels.. Their structures were elucidated by comprehensive spectroscopic analyses including UV, IR, MS, 1D- and 2D-NMR.
UTrustDisk: An Efficient Data Protection Scheme for Building Trusted USB Flash Disk

Data protection of removable storage devices is an important issue in information security. Unfortunately, most existing data protection mechanisms are aimed at protecting computer platform which is not suitable for ultra-low-power devices. To protect the flash disk appropriately and efficiently, we propose a trust based USB flash disk, named UTrustDisk. The data protection technologies in UTrustDisk include data authentication protocol, data confidentiality protection and data leakage prevention. Usually, the data integrity protection scheme is the bottleneck in the whole system and we accelerate it by WH universal hash function and speculative caching. The speculative caching will cache the potential hot chunks for reducing the memory bandwidth pollution. We adopt the symmetric encryption algorithm to protect data confidentiality. Before mounting the UTrustDisk, we will run a trusted virtual domain based lightweight virtual machine for preventing information leakage. Besides, we prove formally that UTrustDisk can prevent sensitive data from leaking out. Experimental results show that our scheme`s average writing throughput is 44.8% higher than that of NH scheme, and 316% higher than that of SHA-1 scheme. And the success rate of speculative caching mechanism is up to 94.5% since the access pattern is usually sequential.
