Site Preference of Alloying Elements in DO22-Ni3V Phase: Phase-Field and First-Principles Study

        Ding-Ni Zhang,Qian-Qian Shangguan,Fu Liu,Ming-Yi Zhang 대한금속·재료학회 2015 METALS AND MATERIALS International Vol.21 No.4

Site preference of alloying elements in DO22-Ni3V phase was investigated using phase-field and first-principles method. The concentrations of alloying elements on sublattices of DO22-Ni3V phase were quantitatively studied using phase-field model based on microscopic diffusion equations. The phase-field computation results demonstrate that the concentration differences of alloying elements on the NiI and NiII site are attributed to the coordination environment difference. Host atoms Ni and substitutional ternary additions Al prefer to occupy NiI site. Antisite atoms V show site preference on the NiII site. Further reason of site preference of alloying elements on the two different Ni sites were studied using first-principles method to calculate the electronic structure of DO22-Ni3V phase. Calculation of density of states, orbitals population and charge population of the optimized Ni3V structure found that the electronic structures of NiI and NiII sites are different. Electronic structure difference, which is caused by coordination environment difference, is the essential reason for site selectivity behaviors of alloying elements on NiI and NiII sites.

Suppressing Erwinia carotovora Pathogenicity by Projecting N-Acyl Homoserine Lactonase onto the Surface of Pseudomonas putida Cells

( Qian Qian Li ),( Hong Ni ),( Shan Meng ),( Yan He ),( Zi Niu Yu ),( Lin Li ) 한국미생물 · 생명공학회 2011 Journal of microbiology and biotechnology Vol.21 No.12

N-Acyl homoserine lactones (AHLs) serve as the vital quorum-sensing signals that regulate the virulence of the pathogenic bacterium Erwinia carotovora. In the present study, an approach to efficiently restrain the pathogenicity of E. carotovora-induced soft rot disease is described. Bacillus thuringiensis-derived N-acyl homoserine lactonase (AiiA) was projected onto the surface of Pseudomonas putida cells, and inoculation with both strains was challenged. The previously identified N-terminal moiety of the ice nucleation protein, InaQ-N, was applied as the anchoring motif. A surface display cassette with inaQ-N/ aiiA was constructed and expressed under the control of a constitutive promoter in P. putida AB92019. Surface localization of the fusion protein was confirmed by Western blot analysis, flow cytometry, and immunofluorescence microscopy. The antagonistic activity of P. putida MB116 expressing InaQ-N/AiiA toward E. carotovora ATCC25270 was evaluated by challenge inoculation in potato slices at different ratios. The results revealed a remarkable suppressing effect on E. carotovora infection. The active component was further analyzed using different cell fractions, and the cell surface-projected fusion protein was found to correspond to the suppressing effect.

Phase separation phenomena and thermodynamics of octylphenol polyoxyethylene ether in micellar solutions: Effect of high concentration of inorganic salts and linear correlation

Dan Ni Li,Jing Huang,Zhao Hua Ren,Rui Sheng,Zheng Bo Qian,Bei Bei Li,Xiao Feng Quan,Ye Xi Zhang,Jun Ru Wang,Huan Tian 한국공업화학회 2021 Journal of Industrial and Engineering Chemistry Vol.99 No.-

The phase separationphenomena or cloud points (CP) behaviorof nonionic octylphenol polyoxyethylene etherwith oxyethylene glycol ether unit of 15 (OP-15) were investigated in aqueous micellar solutions containinghigh concentrations of inorganic salts including NaCl, KCl, MgCl2, CaCl2, BaCl2, AlCl3, FeCl3, Na2SO4and Na2SiO3. The change in CP induced by salts and its mechanism are discussed theoretically. In aqueous solutionscontaining high concentration of salt, for thefirst time the linear relationship between CP and saltconcentrationiswellestablishedtocharacterizethedepressionofCP.Basedonpseudophaseseparationmodel, thermodynamic parameters are estimated and adopted to further describe the phase separation process andthe interactions between OP-15 molecules or the OP-15/water interactions. Thermodynamic parametersincluding standard Gibbs energy change (DG0CP), enthalpy change (DH0CP) and entropy change (DS0CP)indicate that the additions of different salts result in the change in entropy-driven nonspontaneousprocess of phase separation. These data, which are from the compensation temperatures and the changeof molar heat capacity, also prove that different ions induce the change in the interactions betweensurfactant molecules or the surfactant–water interaction and thus affect the depression of CP. Thesefindings help with understanding the effect of inorganic salts, especially the high concentration of salt, onCP of nonionic surfactant and then broadening the application of nonionic surfactant based on CP such asthe extraction of biomaterials in biological samples derived from animals or plants, etc.

Experimental investigation on penetration performance of larger volume needle-free injection device

        Dongping Zeng,Ni Wu,Lei Qian,Hanqing Shi,Yong Kang 대한기계학회 2020 JOURNAL OF MECHANICAL SCIENCE AND TECHNOLOGY Vol.34 No.9

The injection performance of a small volume of needle-free injection (up to 0.3 mL) has proven to be controllable and satisfactory in transdermal drug delivery. However, no comprehensive research on the behavior of larger volume injections with different requirements for clinical applications exists. This study aims to present the penetration characteristics of larger volume injections, including dynamic properties, dispersion pattern, and percent delivery. The researchers conduct impact and injection experiments with injection volumes from 0.1-1.0 mL, driving pressures of 0.5-1.75 MPa, and orifice diameters of 0.17-0.5 mm. This study uses high-speed photography and impact experiments to capture the dynamic properties of the liquid jet. The researchers observe the dispersion patterns of liquid penetration into the gels in the gel injection experiments and investigate the percentages of liquid delivered to the skin tissues in porcine tissues injection experiments. Moreover, this study uses the response surface methodology (RSM) to analyze the interactive effect between various injection parameters on the injection performance. Results describe the differences in penetration performance between larger volume injections and small volume injections, and the critical volume of the mentioned injections is approximately 0.6 mL.

CardioSentinal: A 24-hour Heart Care and Monitoring System

        Min Gao,Qian Zhang,Lionel Ni,Yunhuai Liu,Xiaoxi Tang 한국정보과학회 2012 Journal of Computing Science and Engineering Vol.6 No.1

People are willing to spend more for their health. Traditional medical services are hospital-centric and patients obtain their treatments mainly at the clinics or hospitals. As people age, more medical services are needed to exceed the potentials of this hospital-centric service model. In this paper, we present the design and implementation of CardioSentinal, a 24-hour heart care and monitoring system. CardioSentinal is designed for in-home and daily medical services. It mainly focuses on the outpatients and elderly. CardioSentinal is an interdisciplinary system that integrates recent advances in many fields such as bio-sensors, small-range wireless communications, pervasive computing, cellular networks and modern data centers. We conducted numerous clinic trials for CardioSentinal. Experimental results show that the sensitivity and accuracy are quite high. It is not as good as the professional measurements in hospital due to harsh environments but the system provides valuable information for heart diseases with low-cost and extreme convenience. Some early experiences and lessons in the work will also be reported.

Integration of single-cell transcriptomics and epigenetic analysis reveals enhancer-controlled TIMP1 as a regulator of ferroptosis in colorectal cancer

        Li Meng,Ni Qian-yang,Yu Su-yang 한국유전학회 2024 Genes & Genomics Vol.46 No.1

Background Ferroptosis is an iron-dependent non-apoptotic programmed cell death. However, the regulatory mechanism of ferroptosis in colorectal cancer (CRC) is still unclear. Objective The aim of this study was to investigate the role and mechanism of enhancer-controlled genes in ferroptosis in CRC. Methods Dimensionality reduction and differentially expressed genes (DEGs) identification were conducted using Seurat algorithm based on single-cell RNA sequencing (scRNA-seq) data from the GSE200997 dataset. Ferroptosis-related pathway enrichment analysis was performed using the FerrDb V2 database. Enhancers were identified using HOMER algorithm based on H3K27ac ChIP-seq data from the GSE166254 dataset. Kaplan-Meier Plotter online tool was used to analyze prognosis and gene expression correlation. Transcription factors were predicted using the transcription factor affinity prediction web tool. The binding of enhancer to transcription factor and H3K27ac enrichment were detected by ChIP-qPCR. RSL3 was used to induce ferroptosis in CRC cells. Gene transcription was detected by qRT-PCR. Cell proliferation was detected by CCK8 assay. Results Nine cell clusters including T cells, natural killer cells, macrophages, mast cells, epithelial cells, fibroblasts, goblet cells, B cells and dendritic cells were identified in CRC and normal colonic tissue samples. Compared to normal colonic tissue-derived epithelial cells, 1075 DEGs were screened in CRC tissue-derived epithelial cells. Ferroptosis-related pathway enrichment suggested that DEGs were associated with the regulation of ferroptosis. DPEP1, ETV4, CEBPG, TIMP1, DUOX2 and LCN2 were identified as the significantly upregulated genes enriched in the “ferroptosis regulator” term, and their H3K27ac signals were significantly higher in CRC tissues than in normal colonic tissues. Of these, only the expression of TIMP1 predicted a poor prognosis of CRC patients. Transcription factor SPI1 drove TIMP1 transcription by binding to its enhancer. Overexpression of TIMP1 significantly promoted the resistance to ferroptosis induced by RSL3 in CRC cells, which was partially restored by SPI1 knockdown. Conclusion Transcription of TIMP1 was driven by transcription factor SPI1 in combination with its enhancer, consequently promoting CRC cells against ferroptosis. The SPI1/TIMP1 axis confers ferroptosis resistance in CRC, and thus has the potential to be the molecular targets for CRC treatment. Sequential Prediction of the TBM Tunnelling Attitude Based on Long-Short Term Memory with Mechanical Movement Principle

        Ruirui Wang,Yuhang Xiao,Qian Guo,Hai Wang,Lingli Zhang,Yaodong Ni 대한토목학회 2024 KSCE Journal of Civil Engineering Vol.28 No.2

TBM tunnelling attitude controlling is a significant issue for guaranteeing the tunnel fitting the expected tunnel axis, with directly influence the tunnel quality. The key to solve the problem is to establish the relationship between the tunnelling attitude and the controlling parameters and to predict the tunnelling attitude accordingly. For this, this paper introduced a TBM tunnelling attitude predicting method. In detail, using Long-Short Term Memory (LSTM), the initial tunnelling attitude and the controlling parameters of each later ring are taken as input, while the tunnelling attitude of each later rings are regarded as the output, and the relationship between the input and output is established. Meanwhile, for avoid the over-fitting and error accumulation risk of LSTM, the theoretical relationship between the input and output is also built based on the TBM mechanical movement principle, and it is also involved into the LSTM-based relationship as constraints. The proposed method is verified by the field data collected from the 6th Section of the Qingdao Metro Project, and the results reveal that the proposed LSTM-based method is accurate and acceptable.
