        다중 중합효소 연쇄반응을 이용한 소의 Johne병 진단 기법 확립

        반추수에서 발생하는 Johne병의 조기 진단 방법을 제시하고 이 질병의 원인체와 미생물학적 특징이 유사한 M. bovis, M. avium 등의 mycobacteria감염증을 감별 진단하는 방법을 개발하기 위하여 Mycobacterium 균속의 표준균주를 사용하여 중합효소 연쇄반응을 확립하였다. Johne병으로 의심되는 소의 혈액과 유즙을 채취하여 분리한 단핵구 및 거식세포로부터 genomic DNA를 추출하였다. 각 시료로부터 추출한 DNA를 template로 이용하여 Mycobacterium spp.에 특이적인 16S rDNA primer set를 이용한 PCR을 수행하여 시료내의 mycobacterial DNA 보유 여부를 확인하였다. 한편 mycobacteria 양성으로 확인된 시료는 M. avium complex 균종에 특이한 16S rDNA 염기서열을 기초로하여 제작한 primer set와 M. paratuberculosis 의 IS900 sequence에 특이한 primer set를 이용하여 duplex PCR을 수행하여 Johne병 원인체의 보균 여부를 조사하였으며, 이 결과를 oligonucleotide probe를 사용한 Southern blot hybridization을 통하여 다시 확인하였다. 이와 같은 duplex PCR 기법을 실제 축산 현장에서 수집한 유즙과 말초혈액으로부터 분리한 단핵구 및 거식세포 시료에 적용한 결과 본 연구에서 확립한 duplex PCR기법 유용성을 확인할 수 있었다. Diagnosis of Bovine Johne's Disease Using Multiplex Polymerase Chain Reactions In order to improve the early diagnosis of Johne's disease in ruminants, duplex polymerase chain reaction system for the detection of the etiologic agent of M. paratuberculosis and for the differentiation of other mycobacterial animal pathogens, such as M. bovis and M. avium, was applied. Genomic DNAs were purified from peripheral blood monocytes or milk macrophages and were used as templates in the duplex PCR. Detection of Mycobacterium spp. in the specimen was carried out by PCR using primer set specific to the mycobacterial 16S rDNA. And then, mycobacterial DNA-positive specimens were further differentiated with duplex PCR system which was composed of primer sets specific to 16S rDNA of M. avium complex and IS900 gene of M. paratuberculosis. The results were re-confirmed by Southern blot hybridization with oligonucleotide specific to the internal sequence of IS900 PCR amplicons. The applicability of this duplex PCR system was evaluated with DNAs extracted from clinical specimens of peripheral blood monocytes and milk macrophages. In summary, the duplex PCR amplification system described in this experiment is promising molecular technique for the early diagnosis of Johne's disease in ruminants.

      • 단기배양을 통한 말초혈액 CD34 양성세포의 체외증폭

        Background: It is suggested that clinical practice in the areas of bone marrow transplantation and gene therapy might rely on the ex vivo expansion of hematopoietic stem/progenitor cells. However, the condition for ex vivo expansion of hematopoietic progenitor cells is not well established. The authors pursued a series of experiments to define the proper conditions for the expansion of hematopoietic cells in the short-term liquid suspension culture of mobilized peripheral blood CD34+ cells. Methods: 1.0ml cultures were initiated with 9×10^3 PB CD34+ cells, which were isolated from PB mononuclear cells (MNCs) by high-gradient cell sorting, in 12 well plates with the various combinations of hematopoietic growth factors(HGF). The following recombinant human HGFs were used: stem cell factor(SCF) 100ng/ml, granulocyte colony-stimulating factor(G-CSF) 100ng/ml, GM-CSF(granulocyte, macrophage colony-stimulating factor) 100ng/ml, interleukin-1 beta(IL-1B) 1ng/ml, interleukin-3(IL-3) 20ng/ml, interleukin-6 (IL-6) 100ng/ml. At the end of culture, colony-forming cells were evaluated by semisolid clonogenic assay. Results: 1) Using the high-gradient magnetic sorting system, CD34^+ cells were isolated with a yield of 40 3% 2) In 7 day culture of PB CD34^+ cells(9×10^3 cells), nucleated cells expanded mean 10×10^3(range, 9 to 20×10^3) with the addition of SCF alone, 35×10^3(range, 10 to 60×10^3) with SCF plus G-CSF plus GM-CSF, and 130×10^3(range, 40 to 300×10^3) with the combination of SCF, G-CSF, IL-1, IL-3, IL-6, GM-CSF. In 14 day culture, nucleated cells expanded 10×10^3 to 1,860×10^3 with combination of human hematopoietic growth factors. 3) In 10 day culture without medium change of PB CD34^+ cells, CFU-GM numbers expanded 16. 5 fold(range, 7 to 59 fold) with the addition of SCF plus G-CSF plus Il-1 plus IL-3, 31.3 fold(range, 20.5 to 101.1 fold) with the combination of SCF, G-CSF, IL-1, IL-3, GM-CSF. In 14 day culture with or without medium change of PB CD34^+ cells was inferior to 10 day culture for CFU-GM expansion. 4) There was no significant difference for CFU-GM expansion between five growth factors(SCF,G-CSF,IL-1,IL-3,GM-CSF) and six growth factors(five growth factors plus IL-6). Conclusion: The authors could confirm that short-term suspension culture of peripheral blood CD34+ cells could expand hematopoietic progenitor cells. Ten-day culture with medium change of CD34+ cells with the addition of five growth factors, i.e. SCF, G-CSF, IL-1B, IL-3, and GM-CSF, might be the most efficient in this system.

      • 항암화학요법을 받는 암 환자에 대한 Nucare^R의 영양지지효과

        Purpose : The purpose of this study was to evaluate the effect of nutritional support with enteral liquid supplement in cancer patients receving chemotherapy for possible benefit in nutritional, immunologic and golbal function of patients. Patients and Methods : From October 1995 to February 1997, 30 advanced cancer patients receving chemotherapy were divided two roups. The Nycare group, in addition to normal diet, Nucare^R enteral supplement was given for 1week right after chemotherapy for the duration of 2 chemotherapy cycles. Control group received only normal diet without parenteral fluid supplement for 2 chemotherapy cycles. Results : Median ages were 53 end 56 years for Nucare group and Contrl group respectively. Performance scores was less than 2 by ECOG scale. All patients were stage Ⅲ or Ⅳ. The physical parameters, such as weight, arm muscle circumference(AMC) and triceps skin fold(TSF) were decreased in both groups after 2 cycles of anticancer therapy. but it was less severe in Nucare group(p<0.05). Serum transferrin was maintained in mild deficit state in Nucare group, whereas it aggravated form mild to moderate deficit in Control group(p<0.05). Serum albumin level increased in Nucare group without statistical significance. but it decreased from normal to mild deficit in Control group. Serum total protein did not change significantly in Nucare group. but in Control group, serum total protein was decreased from 7.24±0.9 to 6.52±0.5(P<0.05). Total lymphocyte count did not change significantly in both groups. Conclusion : This study shows that the nutritional support with Nucare^R was effective in the prevention of nutritional deficit status in patients receving a nticancer chemotherapy.

        임대주택법 일부개정법률안의 문제

        The bill, a partial amendment to the Housing Act, is submitted to the Land, Transport and Maritime Affairs Committee of the National Assembly at Congressman Kim Tae-ho’s proposal, with major support of tenants in Kim-hae area. It is argued that the amendment is written for the tenants. The amendment includes some new provisions and other partially amended provisions. The main contents are comprised of provisions regarding the recovery of the full amount of the National Housing Fund, the criteria for appraisal prices relating to the conversion of rental housing for sale and the upper limit, retrospective application of the criteria, approval by the head of a Si, the head of Gun, or the head of Gu of rental conditions, and relevant penal provisions. Those provisions are for lowering the appraisal prices relating to the conversion of rental housing for sale by changing the criteria and ultimately for promoting tenants who reside in public rental housing to own a home. The main point is to retrospectively apply the same criteria for appraisal prices relating to the conversion of rental housing for sale to ten-year public rental housing units, as applied to five-year public rental housing units. Because the appraisal prices relating to the conversion of rental housing for sale directly affects interests between a landlord and a tenant after the tenancy is terminated, it is necessary to analyze the criteria for appraisal prices relating to the conversion of rental housing units for sale, the upper limit, and the timing for the application of the appraisal prices, in terms of the objectives of policies for rental housing and impartiality between landlords and tenants. Especially, after the simulation of the upper limit of appraisal prices relating to the conversion of rental housing for sale in an area such as Pan-gyo where housing prices had dramatically risen, the result turned out that the real prices relating to the conversion of rental housing for sale, if the conversion is made after the termination of ten year rental period, is projected to range from 160 percent to 200 percent of the housing prices (construction cost) at the time of the recruitment of occupants. In cases of ten year rental housing units, since the profits of rental business operators are guaranteed for a rental period by law, the housing prices seems to rise higher. The more housing prices rise in the relevant area, the prices for the conversion of rental housing for sale seems to rise higher. Thus, it turned out that such increase deprived tenants of the opportunity to own a home. Also, the deprivation was against the objectives of the policies for rental housing. 이 논문의 분석대상이 된 임대주택법의 일부개정법률안은 김해지역의 임차인들이 주축이 되어 김태호의원의 대표발의로 국회의 국토해양위원회에 제출되었던 법률안이다. 그래서 임차인들을 위한 개정안이라고 주장하며, 그 주요내용을 보면, 국민주택기금의 전액 환수, 분양전환가격의 산정기준과 상한선 및 소급적 적용, 시장ㆍ군수ㆍ구청장의 임대조건 승인, 임대조건의 승인 및 그 벌칙에 관한 것을 일부 신설ㆍ개정하는 것이다. 주요내용들 중에서 분양전환가격을 산정하는 방식의 변경을 통해 분양전환가격의 인하를 유도하고, 궁극적으로 공공임대주택에서 거주하는 임차인들의 내집마련을 도모하고자하는 것이라고 볼 수 있다. 그 핵심적인 내용은 현재 5년 임대주택에 적용되는 분양전환가격의 산정기준과 상한선을 10년 임대주택에 대하여 동일하게 적용하되, 소급적으로 적용하려는 것이 특징이다. 임대기간 만료 후 분양전환을 할 때 산정되는 분양전환가격의 크기는 임차인과 임대인간의 이해관계에 직접적으로 영향을 미치는 사항이기 때문에 분양전환가격의 산정기준과상한선 및 그 적용시기를 임대주택정책의 목적 및 임차인과 임대인 간의 형평성 등의 관점에서 분석할 필요가 있다. 특히 주택가격이 크게 상승한 (판교)지역에 대한 분양전환가격의 상한선을 시뮬레이션을 해본 결과, 10년 임대기간 만료 후 분양전환이 될 경우 예상되는 실질 분양전환가격은 최초 입주자 모집당시의 주택가격(건설원가)의 160%~200%에 달하는 것으로 나타났다. 10년 임대주택의 경우, 임대기간 중 임대사업자의 이윤까지 보전해주면서 해당 지역에서 주택가격이 상승하면 상승할수록 실질 분양전환가격의 크기는 더욱더 크게 증가함으로써 사실상 임차인들의 내집마련 기회가 박탈되고, 이것은 결국 임대주택정책의 목적에 반하는 것으로 나타났다.

      • 경연 대회를 통한 초ㆍ중고생의 창의력 신장 효과에 관한 연구

        미래 사회를 대비하기 위한 개인의 개성을 존중과 창의적인 능력을 지닌 인간을 양성을 목적으로 충청남도 지역의 초 ㆍ 중고등학생들을 대상으로 한 창의력 경연대회가 공주대학교 과학교육연구소에서 개최되었다. 충청남도 초 ㆍ중 ㆍ 고등학생들을 대상으로 실시한 창의력 경연대회는 창조적 아이디어를 내어 스스로 문제를 해결해 나갈 수 있는 교육 프로그램을 제공하는 좋은 기회였음이 확인되었으나, 일부의 팀에서만 참신하고 재미있는 아이디어들이 제안되었다. 여학생의 참가자 수가 상당히 증가하였으며, 대체적으로 입상권의 학생들의 상당 부분이 여학생이라는 것이 특이할만하다. 학년이 올라감에 따라서 창의적인 아이디어를 내는 양이나 질에 있어서 뒤떨어지는 것으로 분석되었으며, 토론 학습의 적응력이 미흡하고 발표력 및 청취력에 있어서도 떨어지는 것으로 나타났다. 따라서 중 ㆍ고등학교의 학교교육에서도 학생들에게 프로젝트형 탐구학습 프로그램의 적용은 물론 토론식 수습을 통한 학생들의 사고의 전환의 기회를 제공해야 할 것으로 사료된다. The creativity competition was held to foster the creativity of elementary and secondary school students in the Institute of Science Education, Kongju national University. It was found that the creativity competition effected on the improvement of inventing creative ideas and problem solving activities. But, only some competitors participated in semifinal and final contests, presented original and interesting ideas. More female students participated in competition than last year. More female participants were awarded a prize than males. Totally, participants were defective in discussion and communication, and presentation and listening ideas. Increasing grades correlated inversely with creativity and originality. There is need of inquiry project teaming programs and presenting opportunities of conversion of thinking by discussing instructions in school

      • 수소취화형 응력부식균열에서 수소함유량 측정법의 개발

        본 연구의 목적은 수소취화형 응력부식균열기구에서 균열선단부의 수소집적량을 측정하기 위한것이다. 실험에 이용한 시편은 CT시험편을 개량해서 실제 균열을 전파시키면서 시험편내에 가공한 4mm 직경의 수소집적량 측정용구멍을 통해서 수소투과전류를 측정하였다. The purpose of this study is measurement of hydrogen content in crack tip. A new method for measureing the hydrogen content at the wall in modified CT type specimen was proposed. Two holes, 4mm in diameter and 37mm in length, were drilled closely to crack propagation path. The anodic current in the holes due to ionization of hydrogen which diffused through the specimen from the inside wall of crack was measureed.

      • 한국 전력산업구조개편 추진방향 및 개선방안에 대한 연구 : Driving Directions and Improvement Suggestions

        1961년부터 한국의 전력산업은 한국전력공사(KEPCO)가 발전, 송전, 배전분야를 독점하여 왔다. 그러나 외환위기 이후에 시행된 금융, 기업, 노동, 공공부문의 4대 구조개혁과 맞물려 기존의 KEPCO 독점체제에 대한 전력산업구조개편이 진행되고 있다. 정부는 1999년 전력산업구조개편 기본계획에서 (1) 시장경쟁 원리 도입에 따른 효율성의 증대, (2) 저렴하고 안정적인 전력공급의 지속적 보장, (3) 소비자 선택권의 확보에 의한 편의성 증가 등을 구조개혁의 목표로 제시하였다. 구조개혁과 연관하여 발전분야의 민영화도 진행되고 있다. 그러나, 전력산업구조개편이 국가경제에 미칠 영향과 부작용을 고려할 때, 구조개혁이 성공적으로 구현되기 위해서는 당면과제와 정책대안을 철저하게 검토하고 연구해야 할 것이다. 이 논문은 이런 연구의 연장에서 전력산업구조개혁의 추진방향과 주요한 이슈들을 논의하며 몇 가지 개선방안을 다루고 있다. In January 1999, the Korean government finalized and announced the restructuring plan of the electric power industry which has been state-monopolized by KEPCO for forty years. KEPCO has an integrated operation monopoly over power generation, transmission and distribution of electricity. The Korean electric power industry is now being undertaken fundamental restructuring toward the following goals. - To increase efficiency through market competition - To continuously guarantee a long-term, inexpensive and stable electric power supply - To promote consumer convenience through expansion of consumer choice in the electric power provider Also as a link in the restructuring, privatization of the power generation sector is underway. Considering the pervasive side effects and influences that restructuring have on the national economy, such extensive restructuring requires that a number of initial policy questions and challenges being thoroughly examined so as to ensure successful implementation of the plan.       • 하수배제방식이 하수처리 효율에 미치는 영향

        본 연구는 하수배제방식이 하수처리장의 효율에 미치는 영향을 조사하였다 이를 위하여 표준활성 슬러지공법을 이용하여 하수를 처리하는 두 모델처리장을 대상으로 , BOD, COD, SS, T-N, 그리고 T-P의 제거율을 비교하였다. 모델처리장 K1은 합류식 관거로 이송한 하수를 분뇨와 연계하여 처리하고 있으며, K2 처리장은 분류식 관거를 통하여 이송된 하수만을 처리하고 있다. 하수처리장 유입유량은 하절기에 증가하는 경향을 보였고, 유기물과 SS 농도는 봄철에 증가하는 경향을 보였다. K1 처리장의 유입유량과 농도가 K2 처리장에 비해 변화가 심하였는데, 이는 하수배제방식의 차이 때문인 것으로 판단된다. K1 처리장은 설계유량보다 처리량이 더 많았음에도 불구하고 방류수 수질기준을 만족하였는데, 이는 활성슬러지공법이 충분한 안전율로 설계되기 때문인 것으로 판단된다. K2 처리장의 유입유량의 설계유량보다 적어 두 모델처리장의 효율을 정확하게 비교하기 어려웠지만, 분류식 하수관거가 활성슬러지공법의 효율을 향상시키는데 더 유리한 것으로 보인다. The activated sludge process has been the most popular for the treatment of domestic wastewater. In this study, the effect of sewage collection system on the treatment efficiency of wastewater treatment plant (WTP) was investigated. For this purpose, two WTPs operated using the activated sludge process were selected as model plants and the removal efficiency of each contaminant, such as BOD, COD, SS, T-N, and T-P, was compared. Plant K1 treats the domestic wastewater transported through the combined collection system and the nightsoil which is intermittently added into aeration tank. On the other hand. Plant K1 treats only the domestic wastewater transported through the separated collection system. Influent flowrate increased during summer season, but organic content and SS concentration increased during spring. Plant K1 showed higher range of fluctuation with respect to both wastewater quantity and quality compared to plant K2 due to the difference in sewage collection system. Even though Plant K1 treated much more wastewater than the designed capacity, the concentration of effluent from Plant K1 was less than the discharge limit. This is due probably to the safety factor which was taken into consideration when the activated sludge process was designed capacity, it was impossible to compare exactly the efficiencies of two model plants. But, it seems that the separated collection system is more beneficial in improving the efficiency of activated sludge process.

      • 항공기 구조용 Ti-6Al-4V의 수소화처리를 통한 초소성특성 향상

        The Alloys can be hydrogenated with large amount of hydrogen and dehydrogenated invaceume at elevated temperature. Because hydrogen is β-Ti stabilizer and produces hydride(TiH₂) into microstructure optimum phase ratio is maintained at readly low temperautre and hydrides introduce localized high densitydislocation which provides with nucheation site for rerystallization. after hydrogenation treatment was performed at 830℃ for 10 minutes Pre-Treatment was performed at 760℃ and 830℃ with Water Quenching and Air Cooling. Agter pre-treatment Aging Treatement was carried out at 500℃ for 6 hours and Dehydrogenation Treatment was carried out at 760℃ for 24 hours. Specimen exhibits fine equiaxed granins of about 2㎛.
