Development of Rapid Detection Technique for Canine Distemper Virus

Recently cases of canine distemper have occurred in Korea despite vaccination was carried out nationwidely. This study was performed to establish rapid diagnosis of canine distemper by RT-PCR, nested PCR, and serological test. A total of 30 dogs, which were suspected canine distemper clinically, was examined. RT-PCR and nested PCR were specific for the amplification of CDV H gene and sensitive to detect 7 TCID50 of Onderstepoort strain. By RT-PCR, H gene was detected in 6(20%) of 30 peripheral bloods from dogs. And H gene was detected in 10(33.3%) of 30 samples by nested PCR. H gene was detected from 1 brain of 6 years-old Beagle dog and 1 lung of 2 months-old Shihtzu dog, in which peripheral blood H gene was not detected. Serum neutralizing antibody titer against Onderstepoort strain ranged from 4 to 1,024 in 30 patients. No correlation was observed between the results of nested PCR and titiers of neutralizing antibody.

Antimicrobial susceptibility testing was conducted with 6 different spirochetal strains (4 strains of Leptospira spp. and 2 strains of Borrelia burgdorferi) against 3 antimicrobial agents, commonly used in equine and bovine practice. The ranges of MIC and MBC of amoxicillin against Leptospira spp. were 0.05-6.25 μg/ml and 6.25-25.0 μg/ml, respectively. And the ranges of minimal inhibitory concentration (MIC) and minimal bactericidal concentration (MBC) of amoxicillin against B. burgdorferi were 0.05-0.39 μg/ml and 0.20-0.78 μg/ml, respectively. The ranges of MIC and MBC of enrofloxacin against Leptospira spp. were 0.05- 0.39 μg/ml and 0.05-0.39 μg/ml, respectively. Two strains of B. burgdorferi were resistant to enrofloxacin at the highest concentration tested for MBC (≥100 μg/ml). Therefore, the potential role of tilmicosin in the treatment of leptospirosis and borreliosis should be further evaluated in animal models to understand whether the in vivo studies will confirm in vitro results. All spirochetal isolates were inhibited (MIC) and were killed (MBC) by tilmicosin at concentrations below the limit of testing (≤0.01 μg/ml).

We examined the responses of PBMCs to house dust mite (HDM) allergen in atopic and healthy, non-atopicdogs to identify differences in lymphocyte reactivity that might reflect the immunologic status of atopic dermatitis. Thirteenwas significantly higher in the atopic dogs than that in healthy, non-atopic dogs insensitive to the allergen (P = 0.007).The proliferative responses were positively correlated with the level of HDM-specific IgE in serum (P = 0.035), and werethereby confirmed to reflect the activity of lymphocytes competent to promote IgE production. These results suggestthat HDM-specific lymphocytes were present in peripheral blood and played a role in the pathogenesis of canine atopicdermatitis.
