        In recent years, gonorrhea has been pandemic and remains one of the most common STDs in the world, especially in developing countries. For the detection of a more effective therapeutic regimen and assessing the prevalence of Penicillinase Producing Neisseria gonorrhoeae(PPNG), we have been trying to study the patients who have visited the Venereal Disease Clinic of Choong-Ku Public Health Center in Seoul since 1980 by menas of the chromogenic cephalosporin method. In 1998, 93 strians of N. genorrhoeae were isolated, among which 60(64.5%) were PPNG. The prevalence of PPNG in Seoul, which had been decreased to 39% in 1996 after a peak of 74.3% in 1993, is increased to 64.5% in 1998.

      • 백서 해마에서 카이닌산 유도 경련에 의한 JNK 경유 신호전달경로 활성화의 발달 단계에 따른 변화

        연구목적 : 어린 백서의 해마에서 전기경련충격(electroconvulsive shock, ECS) 후 활성화되는 것으로 알려져 있는 MAPK계 신호전달경로가 카이닌산(Kainic acid, KA)에 의한 경련 후 어떤 양상으로 활성화되는 지를 초기 발달 단계에 따라서 관찰하고자 하였다. 방 법 : 생후 7일, 14일, 21일 된 어린 백서와 성년 백서의 해마에서 면역블롯 방법으로 MAPK계 효소의 인산화 효소 중 하나인 SAPK/ERK kinase-1(SEK-1), MAPK계 효소인 c-Jun N terminal protein kinase(JNK), 전사인자 c-Jun 및 조기유전자 단백질 Fos의 단백질 양과 기저 인산화를 측정하였고, 백서에게 KA를 주사하여 경련을 유발한 후 위 단백질들의 인산화 변화의 양상을 같은 방법으로 관찰하였다. 결 과 : SEK-1, JNK, c-Jun 모두 연령 증가에 따른 단백질 양 자체에는 변화가 없었다. JNK와 c-Jun은 기저 인산화의 양적 변화도 없었으나 SEK-1의 경우 기저 인산화 양이 생후 7일에 가장 높고 이후 연령이 높아짐에 따라 감소하였다. JNK의 경우 KA 주사 후 인산화 변화 양상이 관찰되지 않았으나 SEK-1과 c-Jun은 생후 14일부터 연령의 증가와 함께 인산화 양도 증가하였다. Fos 역시 생후 7일부터 발현되어 연령의 증가와 함께 발현양이 증가하였다. 결 론 : 백서 해마에서의 MAPK계 신호전달계의 성숙은 연령의 증가에 따라, 또 신호전달 물질에 따라 차이를 보였다. KA 유도 경련 후 JNK는 생후 21일까지도 활성화가 되지 않는 것으로 보아, 세포주에서 알려진 SEK-1 - JNK-c-Jun - Fos로 연결되는 신호전달 경로가 어린 백서의 해마에서는 적용될 수 없음을 시사한다. Objective : We observed the developmental pattern of activation of MAPK signal transduction pathways known to be activated by eletroconvulsive shock(ECS) in young rat hippocampus after kainic acid(KA)-induced seizure. Methods : We used the method of immunoblotting for examining the basal protein amount and basal level of phosphorylation of MAPK kinase(SAPK/ERK kinase -1, SEK-1), MAPK(c-Jun N terminal protein kinase, JNK), transcription factor(c-Jun) and immediate early gene proteins(Fos) in rat hippocampus at postnatal day 7, 14 and 21, respectively. We also examined the changes of phosphorylation of those proteins after kainic acid-induced seizure in the same way. Results : The basal protein amounts of SEK-1, JNK, and c-Jun did not show age-dependent changes and basal level of phosphorylation of JNK and c-Jun remains unchanged throughout the early developmental period. The basal level of phosphorylation of SEK-1 was peaked at postnatal 7 days and then decreased with aging. After kainic acid-induced seizure, the change of phosphorylation of JNK was not observed but those of SEK-1 and c-Jun increased after postnatal day 14. The expression of Fos was observed at postnatal day 7 and also increased with aging. Conclusion : These results show that the MAPK signal transduction system in rat hippocampus matures in accordance with aging, but the process of maturation differs depending specific proteins. This study suggests the signal transduction cascade(SEK-1 - JNK - c-Jun - Fos) which is well established in cell line studies may not be applied to rat hipposcampus because we could not observe the activation of JNK after KA-induced seizure in young rat hippocampus.

        외분비선세포의 막전압에 관한 연구

        This experiment was performed to elucidate the effects of some drugs controlling the concentration of cyclic nucleotide on membrane potential in isolated salivary gland and pancreas of rats. Rats of both sexes were anesthetized by intraperitoneal injection of 25% urethan (0.5 ml/100gm). The segments of submaxillary gland and pancreas were isolated and placed in a Perspex tissue bath (20 ml) perfused with 37℃ Krebs-Henseleit solution, aerated with 5% CO_2-O_2 mixture, flowed at about 10 ml/min. Membrane potentials were measured with glass microelectrodes filled with 3 M KCl, having a tip resistance of 10-30 MΩ. The microelectrode was connected to a pre-amplifier and the membrane potential was recorded on a pen recorder. The tissues were stimulated by injecting 100 ㎍ acetylcholine directly into the tissue bath using a constant infusion pump for one minute. Theophylline and nicotinic acid were added to control Krebs-Henseleit solution. The mean resting membrane potential of submaxillary gland cells during perfusion with Krebs-Henseleit solution was -29.9 mV and 10 mM nicotinic acid increased the membrane potential to -33.3 mV. During exposure to a control Krebs-Henseleit solution, acetylcholine evoked hyperpolization of cell membrane of submaxillary gland by 16.0 mV. Acetylcholine-induced hyperpolarization was slightly increased by theophylline. But acetylcholine depolarized the membrane by 7.4 mV during exposure to 10 mM nicotinic acid. In isolated pancreas, mean resting membrane potential was -19.0 mV during exposure to a control solution. 10 mM theophylline increased the membrane potential to -23.4 mV and 10 mM nicotinic acid decreased to -17.1 mV. Acetylcholine always decrease the membrane potential during exposure to all solutions in pancreas. In control Krebs-Henseleit solution, the amplitude of the acetylcholine-induced depolarization was similar to that of theophylline solution but nicotinic acid reduced the acetylcholine-induced depolarization.

        經通草 煎湯液이 家兎의 腎臟機能 및 血漿 Aldosterone 濃度에 미치는 影響

        To evaluate effect of Lignum Aeschynomeni water extract on the renal functions and plasma aldosterone concentration, experiments performed in the rabbit The results obtained were followed : 1. Intravenous administration of Lignum Aeschynomeni water extract at the doses of 0.08㎖/㎏ significantly increased urine volume, glomerular filtration rate and urinary excretion of electrolytes. 2. Intravenous administration of Lignum Aeschynomeni water extract at the doses of 0.8㎖/㎏ remarkably decreased urine volume, free water clearance and glomerular filtration rate. 3. In travenous administratio of Lignum Aeschynomeni water extract at the doses of 0.08㎖/㎏ and 0.8㎖/㎏ significantly decreased plasma aldosterone concentration. These results suggest that Lignum Aeschynomeni water extract at the doses of 0.08㎖/㎏ has a diuretic effect which may be caused by renal hemodynamics and the decrement of aldosterone concentration, and that Anti diuretic effect of large dose may be relateo with Anti diuretic hormone.

      • 정상인과 혈액질환 환자에서의 혈소판 평균용적치에 관한 연구

        Molphologic study of platelet have been done by many authors for the past years, they investigated that the heterogeneity of platelet size was shown even in normal person and various hematologic disorder. In the past the platelet size was not utilized in clinical medicine because of its technical difficulties of measuring. Recently, and automated blood cell counter, Coulter counter Model Splus Ⅱ®, was introduced and platelet size could be easily gained, which expressed by mean platelet volume(MPV). Therefore, this study was planned to evaluated the clinical significance of MPV in health person and various hematologic disease. The following results were obtained: 1. In healthy person, MPV was decreased along with increasing the platelet number. 2. In thrombocytopenic patients, MPV was decreased more than normal. 3. In various hematologic disease, MPV was decreased in acute leukemia, marrow failure and aplastic anemia, normal in iron deficiency. anemia, increased in reactive marrow, MPV was varied by platelet number in various hematologic disease. This disclosed that MPV was related with marrow function and production of megakaryocytes. From these results, MPV could be used as a valuable parameter in the evaluation of platelet together with measuring the platelet number. Particular relation-ship between MPV and various hematologic disease were not observed. We should be more study for MPV.

        It is so far suggested from the results of investigation on the changes of quality during the freezing storage of shrimp and swell fish that increasing of VB-N, TMA rapidly break down, increasing of Coliform MPN and decreasing of concentration of pigment mainly in the early stage of storage. In this paper, the raw material used for experimental sample after soaking during 1hr. in 5% ascorbic acid in 2% NaCl solution and 5% EDTA solution. The results of this study was that the sea food of freezing storage for domestic use and expore had better soaking during 1hr. in 5% EDTA solution.

        한국인의 치아우식 조절에 관한 연구 : Ⅳ. 고추수침엑스가 Streptococcus mutans B-13의 성장에 미치는 영향 Ⅳ. THE EFFECT OF WATER-EXTRACT OF CAPSICUM ON THE GROWTH OF STREPTOCOCCUS MUTANS B-13

        This experiment was performed to elucidate the reported anti-cariogenic effect of capsicum in rats. Streptococcus mutans strain B-13, which was drived from human oral cavity and known to be cariogenic, was cultured in TYG with water-extract of capsicum. Glucose content of water-extract of capsicum was also determined by glucose oxidase method. The results are as follows. 1. Glucose content of water-extract of capsicum was 25.6 (±0.5) %. 2. In logarithmic phase, water-extract of capsicum group showed less steepness in growth curve than control group. 3. In early stationary phase, size of bacterial cell mass was in order of control, 0.5% and 1% water-extract of capsicum group. 4. It appears that water-extract capsicum might contain inhibitory substance to growth of Streptococcus mutans B-13, in spite of its high glucose content.
