        Experimental Simulation on Open-Ended Pipe Pile Penetration Using Transparent Granule

        In order to attain the soil movement and explore the penetration mechanism of open-ended pipe piles in saturated sand, the transparent granule, which could substitute the natural sand, was used to simulate the pile jacking with different pile sections. The granule displacement vector field, the plug neutral surface and the plug height were obtained. Test results showed that when the pile inner radius is larger, the squeezing displacement and the plug height are larger. Normalization of the horizontal and vertical components of granule movement employing the pile outer radius showed that the 4d range of the pile end was the area where the inner shaft resistance exerted. In addition, it was found that the linear relationship between incremental filling ratio (IFR) and plug length ratio (PLR).

        L-carnitine treatment attenuates renal tubulointerstitial fibrosis induced by unilateral ureteral obstruction

        Background/Aims: Accumulating evidence indicates that L-carnitine (LC) protects against multiorgan damage through its antioxidant properties and preservation of the mitochondria. Little information is available about the effects of LC on renal fibrosis. This study examined whether LC treatment would provide renoprotection in a rat model of unilateral ureteral obstruction (UUO) and in vitro. Methods: Sprague-Dawley rats that underwent UUO were treated daily with LC for 7 or 14 days. The influence of LC on renal injury caused by UUO was evaluated by histopathology, and analysis of gene expression, oxidative stress, mitochondrial function, programmed cell death, and phosphatidylinositol 3-kinase (PI3K)/ AKT/forkhead box protein O 1a (FoxO1a) signaling. In addition, H<sub>2</sub>O<sub>2</sub>-exposed human kidney cells (HK-2) were treated with LC. Results: LC treatment inhibited expression of proinflammatory and profibrotic cytokines, and was followed by a significant attenuation of tubulointerstitial inflammation and fibrosis. The increased oxidative stress caused by UUO was associated with mitochondrial dysfunction and excessive apoptosis and autophagy via PI3K/AKT/FoxO1a-dependent signaling, and this was abrogated by administration of LC. In H<sub>2</sub>O<sub>2</sub>-exposed HK-2 cells, LC decreased intracellular production of reactive oxygen species, and suppressed expression of profibrotic cytokines and reduced the number of apoptotic cells. Conclusions: LC protects against the progression of tubulointerstitial fibrosis in an obstructed kidney.

      • Effect of Tea Polyphenols on the Adhesion of Highly Metastatic Human Lung Carcinoma Cell Lines to Endothelial Cells in Vitro

        Aim: Tea polyphenols are known to play roles in critical steps of human lung carcinoma cell metastasis. For understanding the mechanisms whereby they inhibit tumor metastasis, the present study was conducted to investigate their effects on the adhesion of highly metastatic lung carcinoma cell lines (PG cells) to endothelial cells (EC cells) and adhesion molecule expression in vitro. Methods: The expression of CD44 or CD54 in the PG cells was detected by flow cytometry and adhesion of PG cells to EC cells was assessed by confocal microscopy double fluorescence staining. Results: The results showed that tea polyphenols: (1) inhibited the expression of CD44 and CD54, two important adhesion molecules in the PG cells in a dose-dependent manner; (2) significantly blocked the adhesion of PG cells to EC cells not only in a state of rest but also when active; and (3) influenced CD44 and CD54 expression during the adhesion process of PG cells to EC cells. Conclusions: The data indicated that the blocking role of tea polyphenols in the adhesion of PG cells to EC cells is related to CD44 and CD54. The mechanism of tea polyphenol prevention of human lung carcinoma metastasis might be through inhibiting adhesion molecule expression to block cancer cell adhesion.

        Nematicidal activity against Aphelenchoides besseyi and Ditylenchus destructor of three biflavonoids, isolated from roots of Stellera chamaejasme

        Aphelenchoides besseyi and Ditylenchus destructor can cause serious problems for a number of important agricultural crops and vegetables. In this study, the ethanol extract of Stellera chamaejasme L. roots showed strong nematicidal activity against Aphelenchoides besseyi and Ditylenchus destructor. By using a bioactivity-driven fractionation, three biflavonoids were isolated from the extract and their structures were identified by mass and nuclear magnetic resonance spectral data. Nematicidal activity bioassays revealed that isoneochamaejasmin A had the strongest nematicidal activity against A. besseyi and D. destructor with LC 50 values of 2.32 and 0.18 mM at 72 h, respectively. Chamaejasmenin B displayed weaker nematicidal activity against A. besseyi with an LC 50 value of 3.94 mM at 72 h. Neochamaejasmin B induced the lowest mortality against D. destructor with an LC 50 values of 15.6 mM at 72 h. These results suggested that the kind and position of substitutions and the relative configuration of 2-H/3-H and 2”-H/3”-H could be considered as important factors responsible for the nematicidal activity of these purified C-3/C-3″ biflavonoids.

        Repeated Administration of Newly Synthesized Aceclofenac Sustained Release Form Causes Agranulocytosis: Case Report of an Unforeseen Adverse Event during the Phase 1 Trial

        Aceclofenac is a non-steroidal anti-inflammatory drug (NSAIDs) for inflammatory diseases. In this report, we report a serious adverse event (AE) occurred during the phase I clinical trial for a new sustained-release (SR) formulation of aceclofenac. There was a serious adverse event (AE), agranulocytosis, induced by aceclofenac SR form. An open-labeled, repeated-doses, randomized, crossover study was conducted at Kyung Hee University Hospital and 26 Korean healthy male volunteers were enrolled. All subjects received both aceclofenac SR 200 mg once daily and aceclofenac IR 100 mg twice daily for 4 days with 11 days washout period. After 11 days washout period, one subject showed a serious decrease in the segment neutrophil (267/mm3) on a laboratory test prior to the reference drug administration in period 2. We first report a case of agranulocytosis, during a phase I clinical trial.

        Dimethylsulfoxide-soluble Smoking Particles and Nicotine Affect Vascular Contractibility

        The aim is to study the effect of dimethylsulfoxide-soluble smoking particles (DSP) and nicotine on the contractility of rat mesenteric artery. The superior mesenteric artery segments were cultured with DSP or nicotine for 24 h. The vascular contractibility was recorded with myograph system. DSP 0.4 mL/L and nicotine 0.48 and 0.96 mg/L shifted the concentrationcontractile curves induced by sarafotoxin 6c, a selective agonist for ETB receptor toward the left with increased Emax. DSP 0.4 mL/L and nicotine 0.96 mg/L shifted ETA receptor-mediated the concentration-contractile curves toward the left with increased Emax. However, nicotine 0.06 mg/L which is the equivalent concentration of nicotine in DSP 0.4 mL/L did not affect the curves and the Emax mediated with ETA receptor and ETB receptor. DSP 0.2 and 0.4 mL/L shifted the concentration-contractile curves induced by noradrenaline toward the right with decreased Emax. Neither did nicotine 0.06 and 0.96 mg/L. Both DSP and nicotine shifted the concentration-contractile curves induced by 5-hydroxytryptamine (5-HT) toward the right parallely. DSP changed the phenotypes towards an increased efficacy of ETA receptor and ETB receptor, and a reduced efficacy of 5-HT receptor and α-adrenocceptor. The effects of DSP on ETB receptor, ETA receptor and α-adrenocceptor were independent of nicotine. The effect on 5-HT receptor was responsible to nicotine.

        Heavy concrete shielding properties for carbon therapy

        As medical facilities are usually built at urban areas, special concrete aggregates and evaluation methods are needed to optimize the design of concrete walls by balancing density, thickness, material composition, cost, and other factors. Carbon treatment rooms require a high radiation shielding requirement, as the neutron yield from carbon therapy is much higher than the neutron yield of protons. In this case study, the maximum carbon energy is 430 MeV/u and the maximum current is 0.27 nA from a hybrid particle therapy system. Hospital or facility construction should consider this requirement to design a special heavy concrete. In this work, magnetite is adopted as the major aggregate. Density is determined mainly by the major aggregate content of magnetite, and a heavy concrete test block was constructed for structural tests. The compressive strength is 35.7 MPa. The density ranges from 3.65 g/cm<sup>3</sup> to 4.14 g/cm<sup>3</sup>, and the iron mass content ranges from 53.78% to 60.38% from the 12 cored sample measurements. It was found that there is a linear relationship between density and iron content, and mixing impurities should be the major reason leading to the nonuniform element and density distribution. The effect of this nonuniformity on radiation shielding properties for a carbon treatment room is investigated by three groups of Monte Carlo simulations. Higher density dominates to reduce shielding thickness. However, a higher content of high-Z elements will weaken the shielding strength, especially at a lower dose rate threshold and vice versa. The weakened side effect of a high iron content on the shielding property is obvious at 2.5 µSv=h. Therefore, we should not blindly pursue high Z content in engineering. If the thickness is constrained to 2 m, then the density can be reduced to 3.3 g/cm<sup>3</sup>, which will save cost by reducing the magnetite composition with 50.44% iron content. If a higher density of 3.9 g/cm<sup>3</sup> with 57.65% iron content is selected for construction, then the thickness of the wall can be reduced to 174.2 cm, which will save space for equipment installation.

        Histone deacetylase inhibitor trichostatin A affects porcine oocyte maturation in vitro

        <P> Previous studies show that porcine oocyte aging resulting from asynchronised IVM impairs embryo developmental competence. In the present study we investigated whether trichostatin A (TSA; an inhibitor of histone deacetylation) prolongs the maturation time and prevents the aging of oocytes. Porcine oocytes were cultured in medium containing increasing concentrations of TSA (300 nM) for 24, 44 or 64 h. The percentage of oocytes that underwent germinal vesicle breakdown was significantly lower in the TSA-treated group (300 nM) than in the control group. TSA did not affect oocyte quality at MII based on levels of maturation-promoting factor, the phosphorylation status of mitogen-activated protein kinase or histone H3K9 acetylation analysis. We also compared the preimplantation developmental competence and the viability of pathenogenetic embryos treated with 100 nM TSA for 24 h and then continuously cultured for another 24 h in TSA free condition. No significant differences were observed for either parameter between the TSA-treated and control groups. These results indicate that TSA prolongs the IVM of porcine oocytes but that oocyte quality and aging are not affected. These findings provide a feasible option by which to adjust the initiation time of downstream experiments based on porcine matured oocytes. </P>

        Expression pattern of meiosis associated SYCP family members during germline development in chickens.

        <P>Synaptonemal complexes (SCs) are associated with synapsis of homologous chromosomes, chiasmata distribution, recombination and segregation of chromosomes during the extended prophase of meiosis I. Three isoforms of SC proteins, SYCP1, SYCP2 and SYCP3, were identified as the structural proteins of SCs, and may be involved in the assembly and disassembly of SCs. The aim of this present study is to determine the pattern of expression of chicken homologues of SYCP family members during ovarian and testicular development. Protein sequence analysis using CLUSTAL X revealed that the sequences and potential phosphorylation sites of chicken SYCP family proteins were highly conserved with mammalian homologues of SYCP family proteins. Quantitative real-time-PCR and in situ hybridisation analysis revealed that chicken SYCP family members were differentially expressed during ovarian and testicular development. During ovarian development, all chicken SYCP family members were detected in primordial germ cells (PGCs) until embryonic day (E) 8.0; the expression continued in proliferating pre-meiotic oogonia until E15.5 and was upregulated in meiotic prophase I oocytes until hatching. After hatching, all chicken SYCP family members were detected at a low level until 24-weeks-old. During testicular development, all chicken SYCP family members were detected in PGCs until E13.0; the expression continued in pro-spermatogonia and proliferating spermatogonia for up to 8 weeks, and was upregulated in meiotic prophase I spermatocytes in adults. Our data demonstrate the expression pattern of meiosis associated SYCP family members during ovarian and testicular development in chickens.</P>

        Evidence for the medicinal value of Squama Manitis (pangolin scale): A systematic review

        Background: Squama Manitis (pangolin scale) has been used in traditional Chinese medicine for thousands of years. However, its efficacy has not been systematically reviewed. This review aims to fill the gap. Methods: We searched six electronic databases including PubMed, Embase, Cochrane Library, China National Knowledge Infrastructure Database (CNKI), WanFang Database and SinoMed from inception to May 1, 2020. Search terms included “pangolin”, “Squama Manitis”, “Manis crassicaudata”, “Manis javanica”, “Malayan pangolins”, “Manis pentadactyla”, “Ling Li”, “Chuan Shan Jia”, “Shan Jia”, “Pao Jia Zhu”, “Jia Pian” and “Pao Shan Jia”. The Cochrane Risk of Bias (RoB) assessment tool and Newcastle-Ottawa Scale (NOS) were used to evaluate the risk of bias of the included randomized controlled trials (RCTs) and case control studies (CCSs). Results: After screening, 15 articles that met the inclusion criteria were finally included. There were 4 randomized controlled trials, 1 case control study, 3 case series and 7 case reports. A total of 15 different diseases were reported in these studies, thus the data could not be merged to generate powerful results. Two RCTs suggested that Squama Manitis combined with herbal decoction or antibiotics could bring additional benifit for treating postpartum hypogalactia and mesenteric lymphadenitis. However, this result was not reliable due to low methodological quality and irrational outcomes. The other two RCTs generated negative results. All the non-RCTs did not add any valuable evidence to the efficacy of Squama Manitis beacause of small samples, incomplete records, non-standardized outcome detection. In general, currently available evidence cannot support the clinical use of Squama Manitis. Conclusion: There is no reliable evidence that Squama Manitis has special medicinal value. The removal of Squama Manitis from Pharmacopoeia is rational.
