        Evidence for the medicinal value of Squama Manitis (pangolin scale): A systematic review

        Background: Squama Manitis (pangolin scale) has been used in traditional Chinese medicine for thousands of years. However, its efficacy has not been systematically reviewed. This review aims to fill the gap. Methods: We searched six electronic databases including PubMed, Embase, Cochrane Library, China National Knowledge Infrastructure Database (CNKI), WanFang Database and SinoMed from inception to May 1, 2020. Search terms included “pangolin”, “Squama Manitis”, “Manis crassicaudata”, “Manis javanica”, “Malayan pangolins”, “Manis pentadactyla”, “Ling Li”, “Chuan Shan Jia”, “Shan Jia”, “Pao Jia Zhu”, “Jia Pian” and “Pao Shan Jia”. The Cochrane Risk of Bias (RoB) assessment tool and Newcastle-Ottawa Scale (NOS) were used to evaluate the risk of bias of the included randomized controlled trials (RCTs) and case control studies (CCSs). Results: After screening, 15 articles that met the inclusion criteria were finally included. There were 4 randomized controlled trials, 1 case control study, 3 case series and 7 case reports. A total of 15 different diseases were reported in these studies, thus the data could not be merged to generate powerful results. Two RCTs suggested that Squama Manitis combined with herbal decoction or antibiotics could bring additional benifit for treating postpartum hypogalactia and mesenteric lymphadenitis. However, this result was not reliable due to low methodological quality and irrational outcomes. The other two RCTs generated negative results. All the non-RCTs did not add any valuable evidence to the efficacy of Squama Manitis beacause of small samples, incomplete records, non-standardized outcome detection. In general, currently available evidence cannot support the clinical use of Squama Manitis. Conclusion: There is no reliable evidence that Squama Manitis has special medicinal value. The removal of Squama Manitis from Pharmacopoeia is rational.

        Immobilization of Acetobacter sp. CGMCC 8142 for Efficient Biocatalysis of 1, 3-propanediol to 3-hydroxypropionic Acid

        3-hydroxypropionic acid (3-HP) is an important chemical platform organic in material industry, daily chemical industry and biomedicine field due to its numerous valuable derivatives. However, no mature methods have been established in the synthesis industry for direct large scale production. The bacterium Acetobacter sp. CGMCC 8142 with high efficiency of alcohols oxidation property was immobilized for biosynthesis of 3-HP from 1, 3- propanediol (1, 3-PDO). Parameter values in mass transfer modeling indicated that mass transfer of immobilized biocatalysts affected the oxidation reaction (the internal effectiveness factor ηi < 1) but was not the rate-limiting step if Thiele modulus 1 > ϕ > 0.3. The optimal immobilization conditions for 3-HP biocatalysis was sodium alginate 40 g/L, gel beads diameter 1 mm, cross-linkage time 2 h and 0.1mM FeCl2. Immobilized cells showed promising substrate tolerance, pH stability, thermal stability and storability. After 5 cycles of reaction, 3-HP molar yield of immobilized beads was retained to 80.26%, and 66.95 g/L 3-HP were produced from 70 g/L 1, 3-PDO. The biocatalysis process of immobilized cells introduced in this study may provide an economical and efficiency alternative route for practical production of 3-HP.

        Downregulation of lysyl oxidase and lysyl oxidase-like protein 2 suppressed the migration and invasion of trophoblasts by activating the TGF-β/collagen athway in preeclampsia

        Preeclampsia is a pregnancy-specific disorder that is a major cause of maternal and fetal morbidity and mortality with a prevalence of 6–8% of pregnancies. Although impaired trophoblast invasion in early pregnancy is known to be closely associated with preeclampsia, the underlying mechanisms remain elusive. Here we revealed that lysyl oxidase (LOX) and LOX-like protein 2 (LOXL2) play a critical role in preeclampsia. Our results demonstrated that LOX and LOXL2 expression decreased in preeclamptic placentas. Moreover, knockdown of LOX or LOXL2 suppressed trophoblast cell migration and invasion. Mechanistically, collagen production was induced in LOX- or LOXL2-downregulated trophoblast cells through activation of the TGF-β1/Smad3 pathway. Notably, inhibition of the TGF-β1/Smad3 pathway could rescue the defects caused by LOX or LOXL2 knockdown, thereby underlining the significance of the TGF-β1/ Smad3 pathway downstream of LOX and LOXL2 in trophoblast cells. Additionally, induced collagen production and activated TGF-β1/Smad3 were observed in clinical samples from preeclamptic placentas. Collectively, our study suggests that the downregulation of LOX and LOXL2 leading to reduced trophoblast cell migration and invasion through activation of the TGF-β1/Smad3/collagen pathway is relevant to preeclampsia. Thus, we proposed that LOX, LOXL2, and the TGF-β1/Smad3/collagen pathway can serve as potential markers and targets for clinical diagnosis and therapy for preeclampsia.
