Nicotine exacerbates tacrolimus-induced renal injury by programmed cell death

Background/Aims: Cigarette smoking is an important modifiable risk factor in kidney disease progression. However, the underlying mechanisms for this are lacking. This study aimed to assess whether nicotine (NIC), a major toxic component of cigarette smoking, would exacerbates tacrolimus (TAC)-induced renal in-jury. Methods: Sprague-Dawley rats were treated daily with NIC, TAC, or both drugs for 4 weeks. The influence of NIC on TAC-caused renal injury was examined via renal function, histopathology, oxidative stress, mitochondria, endoplasmic reticulum (ER) stress, and programmed cell death (apoptosis and autophagy). Results: Both NIC and TAC significantly impaired renal function and histopathology, while combined NIC and TAC treatment aggravated these parameters beyond the effects of either alone. Increased oxidative stress, ER stress, mitochondrial dysfunction, proinflammatory and profibrotic cytokine expressions, and programmed cell death from either NIC or TAC were also aggravated by the two combined. Conclusions: Our observations suggest that NIC exacerbates chronic TAC nephrotoxicity, implying that smoking cessation may be beneficial for transplant smokers taking TAC.
Adaptive polymorphism of tetrameric alpha-amylase inhibitors in wild emmer wheat

α-Amylase inhibitors are attractive candidates for the control of seed weevils as these insects are highly dependent on starch as an energy source. Wheat tetrameric α-amylase inhibitor (WTAI) is a mixture (60 kDa) of 3 units: WTAI-CM2 plus 2 WTAI-CM3 plus WTAI-CM16, where none of the subunits is active on its own. A total of 334 gene sequences were obtained from 14 populations (131 accessions= genotypes) of wild emmer wheat. The frequencies of SNPs in WTAI-CM2,WTAI-CM3 and WTAI-CM16 were 1 out of 87.6, 101.4, and 108.0 bases, where 5, 5 and 4 SNPs were detected in the coding sequence, respectively. The nucleotide sequence of each unit of tetrameric α-amylase inhibitors were much more conserved than that of dimeric or monomeric inhibitors. The wild emmer wheat populations showed diversity on three WTAI loci,both between and within populations. It was revealed that WTAI were naturally selected for across populations by a ratio of dN/dS as expected. The results of purifying and positive selection hypothesis (p<0.05) also showed that the sequences of WTAI were contributed by natural selection, which ensures the protein function conservation as well as the inhibition diversity with insects amylase enzyme. Ecological factors, singly or in combination, explained a significant proportion of the variations in the SNPs. Ecological factors have an important evolutionary role in gene differentiation at these loci, and tetrameric α-amylase inhibitors are obviously adaptively selected under different environments.

Esophageal squamous cell carcinoma (ESCC) is the most common histologic subtype of esophageal cancer and is characterized by a poor prognosis. Determining gene changes in ESCCs should improve understanding of putative risk factors and provide potential targets for therapy. We sequenced about 55 million pair-end reads from a pair of adjacent normal and ESCC samples to identify the gene expression level and gene fusion. Sanger sequencing was used to verify the result. About 17 thousand genes were expressed in the tissues, of which approximately 2400 demonstrated significant differences between tumor and adjacent non tumor tissue. GO and KEGG pathway analysis revealed that many of these genes were associated with cellular adherence and movement, simulation responses and immune responses. Notably we identified and validated one fusion gene, HLA-E and HLA-B, located 1 MB apart. We also identified thousands of remarkably expressed transcripts. In conclusion, a novel fusion gene HLA-E and HLA-B was identified in ESCC via whole transcriptome sequencing, which would be a biomarker for ESCC diagnosis and target for therapy, shedding new light for better understanding of ESCC tumorigenesis.

On the basis of conducting a comprehensive investigation to agricultural sightseeing garden landscape resources, we established a evaluation index system of agricultural sightseeing garden landscape resources. From the perspective of experts, comparing the importance of all levels indicators, we obtained the weights of landscape resources evaluation index through using the Analytic Hierarchy Process. The result showed that the descending order of the weights of influence of agricultural sightseeing garden nineteen evaluation index is: reach ability (0.128), safety (0.083), location (0.078), participatory (0.076), cultural value (0.058), ecological conditions (0.057), scenic beauty (0.0505), environmental quality (0.051), featured properties (0.0501), environmental tolerance (0.048), reputation (0.047), environmental capacity (0.045), humanize (0.041), spots configuration (0.034), applicable travel period (0.033), scientific value (0.032), art value (0.031), holistic (0.03), suitability (0.027), it can provide a framework and basis for the planning, management, protection and exploitation of agricultural sightseeing garden landscape resources.

<P>Polyethyleneimine (PEI) has been described as a highly efficient gene carrier due to its efficient proton sponge effect within endosomes. However, many studies have demonstrated that PEI is toxic and associated with a lack of cell specificity despite high transfection efficiency. In order to minimize the toxicity of PEI, we prepared chitosan-graft-spermine (CHI-g-SPE) in a previous study. CHI-g-SPE showed low toxicity and high transfection efficiency. However, this compound also had limited target cell specificity. In the present study, we synthesized galactosylated CHI-g-SPE (GCS) because this modified GCS could be delivered specifically into the liver due to hepatocyte-specific galactose receptors. The DNA-binding properties of GCS at various copolymer/DNA weight ratios were evaluated by a gel retardation assay. The GCS copolymer exhibited significant DNA-binding ability and efficiently protected DNA from nuclease attack. Using energy-filtered transmission electron microscopy (EF-TEM), we observed dense spherical, nano-sized GCS/DNA complexes with a homogenous distribution. Most importantly, GCS was associated with remarkably low cytotoxicity compared to PEI in HepG2, HeLa, and A549 cells. Moreover, GCS carriers specifically delivered the gene-of-interest into hepatocytes in vitro as well as in vivo. Our results suggest that the novel GCS described here is a safe and highly efficient carrier for hepatocyte-targeted gene delivery.</P>
Aerobic Denitrification: A Review of Important Advances of the Last 30 Years

Understanding aerobic denitrification has become an important focus of environmental microbiology. Aerobic denitrification can be performed by various genera of microorganisms and describes the use of nitrate (NO3 −) as oxidizing agents under an aerobic atmosphere. Isolation of aerobic denitrifiers, enzymes involved in aerobic denitrifiers, phylogenetic distribution of aerobic denitrifiers, factors affecting the performance of aerobic denitrifiers, attempts of applications and possible future trends are depicted. The periplasmic nitrate reductase is vital for aerobic denitrifiers and NapA gene may be the proof of aerobic denitrification. Phylogenetic analysis revealed that aerobic denitrifiers mainly belong to α-, β- and γ-Proteobacteria. Aerobic denitrifiers tend to work efficiently at 25 ~ 37°C and pH 7 ~ 8, when dissolved oxygen concentration is 3 ~ 5 mg/L and C/N load ratio is 5 ~ 10. In addition, recent progresses and applications on aerobic denitrifiers are described, including single aerobic reactors, sequencing batch reactor and biofilm reactors. The review attempts to shed light on the fundamental understanding in aerobic denitrification.

Three cases of supertall connected structures are presented and each of them represents a quite style. The first case is a strong-connected structure. The coupling function of towers and connector contributes a lot to the structural stiffness and stability. Its special construction scheme had great impact on the construction quality and the structural safety, and must be accurately considered. For the second case which is a weak-connected structure, the influences of different connecting modes to the structural dynamic characteristic were explained. Then the combined bearings were proposed to achieve the design presume. In the third case which represents the multi-supported structures, the structural distinctive mechanical properties were discussed. For the structural state during construction process is quite different from that in final service condition, two construction procedures were simulated to get an optimal one. Although there are great challenges to designers, the advantages of the supertall connected buildings are obvious. Further work is needed in this area to adapt to the development of future cities.
A novel M2e-multiple antigenic peptide providing heterologous protection in mice

Swine influenza viruses (SwIVs) cause considerable morbidity and mortality in domestic pigs, resulting in a significant economic burden. Moreover, pigs have been considered to be a possible mixing vessel in which novel strains loom. Here, we developed and evaluated a novel M2e-multiple antigenic peptide (M2e-MAP) as a supplemental antigen for inactivated H3N2 vaccine to provide cross-protection against two main subtypes of SwIVs, H1N1 and H3N2. The novel tetra-branched MAP was constructed by fusing four copies of M2e to one copy of foreign T helper cell epitopes. A high-yield reassortant H3N2 virus was generated by plasmid based reverse genetics. The efficacy of the novel H3N2 inactivated vaccines with or without M2e-MAP supplementation was evaluated in a mouse model. M2e-MAP conjugated vaccine induced strong antibody responses in mice. Complete protection against the heterologous swine H1N1 virus was observed in mice vaccinated with M2e-MAP combined vaccine. Moreover, this novel peptide confers protection against lethal challenge of A/Puerto Rico/8/34 (H1N1). Taken together, our results suggest the combined immunization of reassortant inactivated H3N2 vaccine and the novel M2e-MAP provided cross-protection against swine and human viruses and may serve as a promising approach for influenza vaccine development.
