      • Expression of mosquitocidal crystal protein genes in non-insecticidal Bacillus thuringiensis subsp. israelensis

Bacillus thuringiensis NTB-1 isolated from soil samples in Korea produces ovoidal parasporal inclusions with proteins of approximately 24-40kDa in size. Although serological study indicated that the isolate has a flagella (H) antigen identical with subsp. israelensis, it seemed to be non-insecticidal against Lepidoptera and Coleoptera as well as Diptera. To investigate the activity of non-insecticidal B.thuringiensis transformed with insecticidal crystal protein genes, cryIVD and cytA genes of B. thuringiensis subsp. morrisoni PG-14, highly toxic to mosquito larvae, were introduced into the isolate NTB-1. The expression of mosquitocidal crystal protein genes in NTB-1 was characterized by SDS-PAGE analysis and electron microscopy. The results showed that crystalline inclusions of host, CryIVD and CytA were stably expressed in the transformant. However, the mosquitocidal activity of transformant was similar to that of B. thuringiensis subsp. kurstaki Cry-B harbouring cryIVD and cytA genes, demonstrating that a synergistic effect by an interaction of both introduced insecticidal and resident non-insecticidal crystal proteins was not observed.

        In this study, we investigated how much job-satisfaction and self leadership nurses have. And making use of self leadership, we hope to enhance job- satisfaction and develop nurse organization. To find out the relationship between nurse's self leadership and Job-satisfaction, we carried out a sample survey : The subjects of our study are 150 nurses being in offices(hospitals) at the two university hospitals and three general hospitals in Seoul or Keong-gi province. Nurses agreed with and understood the purpose of our study. For analyzing the result, we used ANOVA of SPSS 14.0. The main results of our study are as follows : According to characteristic of subjects, level of self leadership has a meaningful difference statistically according to marriage, religion, department career, position. As for the marriage, the level of self leadership of the married are higher((F=19.95, p= .000) than the unmarried. . Level of self leadership perceived by subjects is an average 3.50 point. An average points of each factors are as follows : Self-compensation is 3.75 point, rehearsal is 3.57 point, self-expectation is 3.55 point, goal-setting is 3.53 point, self-criticism is 3.31 point, constructive thinking is 3.27 point. The total average is 62.9 point to 90 full marks. Level of self job-satisfaction perceived by subjects is an average 2.98 point to 5 full marks. Level of self job-satisfaction perceived by subjects is an average 2.98 point to 5 full marks. An average points of each factors are as follows : Interrelation is 3.42 point, interrelation between doctors and nurses is 3.35 point, job demand is 3.10 point, administration is 3.02 point, a professional position is 2.94 point, self control is 2.84 point, payment is 2.44 point. A coefficient of co-relation of self leadership and self job-satisfaction is .406 point, a meaning probability is .000 point and they seem to have a close interrelation. Rehearsal among factors of self leadership has the most interrelation with payment of self job-satisfaction. A professional position of self job-satisfaction has the most interrelation with self expectation(r= .314, p= .000). A job demand has the most interrelation with constructive thinking(r= .272, p= .001). A interaction has the most interrelation with self expectation(r= .272. p= .000). A administration has the most interrelation with constructive thinking(r= .411, p= .000). An interrelation between doctors and nurses has the most interrelation with self expectation(r= .385, p= .000). Our study shows there is a close relation between self leadership and job-satisfaction. Especially, self expectation and constructive thinking, the factor of self leadership, appear to affect the nurse's job-satisfaction. It appears that nurses feel to have a intensive job-satisfaction, having positive self leadership, a sense of purpose, and constructive thought. Thus, we need to steadily study self leadership, self control, and internal motivation as a new paradigm focused on self-control and internal motivation for their self-developments and advancement of nurses' organizations.

        Identifying the fault location has to deal with multiple possible candidates. It presents a new technique to diagnosis a fault region. The proposed diagnosis scheme identifies accurately the location based on the protective device operations and load current change at the substation. An Expert system is presented using these facts.

        Purpose: The study aimed to investigate the factors influencing consciousness of sexual violence in female high school students, focusing on sexual attitude and parental attitude. ethods: Data were collected from 248 female high school students in South Korea. A structured questionnaire was used for data collection through online survey. The collected data were analyzed by descriptive statistics, t-test, ANOVA, Tukey’s test, Pearson correlation coefficient, and multiple linear regression using the SPSS 25.0 program. Results: The mean score of consciousness of sexual violence in female high school students was 43.54 out of 48. The findings indicate that the mean score of consciousness of sexual violence differed y number of years in high school and academic achievement. Among sexual attitude, parental ttitude, and consciousness of sexual violence, there were meaningful correlations between onsciousness of sexual violence and sexual attitude (r=.132, p=.038,), love-hostility (r=.172, p=.006),and autonomous-control (r=.132, p=.038). Love-hostility aspect of parental attitude (=.159, p=.049) and academic achievement (=.208, p=.001) were found to be the important influencing factors on female high school student’s consciousness of sexual violence (F=5.720, p<.001). Conclusion: The findings of the study suggest that the love-hostility aspect of parental attitude and academic achievement influence consciousness of sexual violence of female high school students. Therefore, in order to improve consciousness of sexual violence in female high school students, sex education program and parent-education program should be developed with further research.

        The Possibility of TBC1D21 as a Candidate Gene for Teat Numbers in Pigs

        Based on a quantitative traits locus (QTL) study using a $F_2$ intercross between Landrace and Korean native pigs, a significant QTL affecting teat numbers in SSC7 was identified. The strong positional candidate gene, TBC1D21, was selected due to its biological function for epithelial mesenchymal cell development. Sequence analysis revealed six single nucleotide polymorphisms (SNPs) in the TBC1D21 gene. Among these, two SNP markers, one silent mutation (SNP01) for g.13,050A>G and one missense mutation (SNP04) for c.829A>T (S277C), were genotyped and they showed significant associations with teat number traits (p value = 6.38E-05 for SNP01 and p value = 1.06E-07 for SNP04 with total teat numbers). Further functional validation of these SNPs could give valuable information for understanding the teat number variation in pigs.

        Performance Analysis of Passive Magnetic Pointing Method for Implementation of Directional Antennas on CubeSats

        As CubeSats have become hugely popular in the recent years, the demand on their capability has also increased. As the missions and functionalities become more complex, coupled with the popular VHF and UHF bands becoming less accessible, CubeSats have been getting pushed into using higher frequency bands. For more capable CubeSats such as NASA and military satellites, utilization of S- and X-band communication have become a common place. While S- and X-band communication provide much higher data rate for the mission, the power and attitude control demand has also increased. Meeting this increased cost and demand on resources may not be feasible for university-level projects as attitude control systems can often cost more than the rest of the CubeSat. This research revisits passive magnetic pointing as an option for providing antenna pointing capability for CubeSats, enabling S- and X-band communication without requiring an active attitude control system. CubeSats can make contact with ground stations located at higher latitude to establish data link with directional antennas. The analyses shows that a higher data throughput can be achieved using S- and X-band communication over the traditional 9600 bps link with the proposed setup, without requiring active attitude control or deployable solar panels. The study characterizes the expected communication link performance for a typical 3U CubeSat when implementing the proposed communication architecture using a S- or X-band patch antenna taking into consideration the expected pointing performance and power generation capability of a 3U CubeSat.

        This paper presents a mew fault location algorithm using iterative method at unbalance condition for distribution feeder systems. Distribution feeders include single phase and three phase laterals. The proposed algorithm achieves a high accuracy by continuously updating voltage and current phasor using the phase components and admittance load model.

        Ultra-fast Detection and Differentiation of Mycoplasma haemofelis and Candidatus M. Haemominutum in Korean Feral Cats by Microchip Electrophoresis with Programmed Field Strength Gradients

        A microchip-based capillary gel electrophoresis (MCGE) technique was developed for the ultra-fast detection and differentiation of Candidatus Mycoplasma haemominutum (Candidatus M. haemominutum, California strain) and Mycoplasma haemofelis (M. haemofelis, Ohio strain) in Korean feral cats through the application of programmed field strength gradients (PFSG) in a conventional glass double-T microchip. The effects of the poly (ethyleneoxide) (PEO) concentration and electric field strength on the separation of DNA fragments were investigated. The PCR-amplified products of Candidatus M. haemominutum (202-bp) and M. haemofelis (273-bp) were analyzed by MCGE within 75 s under a constant applied electric field of 117.6 V/cm and a sieving matrix of 0.3% PEO (Mr 8 000 000). When the PFSG was applied, MCGE analysis generated results 6.8-times faster without any loss of resolution or reproducibility. The MCGE-PFSG technique was also applied to eleven samples selected randomly from 33 positive samples. The samples were detected and differentiated within 11 s. The analysis time of the MCGE-PFSG technique was approximately 980-times faster than that using conventional slab gel electrophoresis.

        For somatic cell nuclear transfer in Hanwoo, fetal fibroblast cell lines were established from 35, 50, 70 and 90-day fetuses of Korean native cattle. The sex of these fetal fibroblast cells were analyzed by PCR using Y-specific primers and confirmed that two cell lines were female and the other two cell lines were male. Karyotyping of these cell lines indicates that the chromosome numbers of fetal fibroblast cells were not affected by passage number and more than 80% of fetal fibroblast cells have normal chromosome number. To evaluate G_(0) stage in cell cycle of fetal fibroblast cells, Western blotting was performed to detect the expression level of PCNA which is known to be expressed in all cell cycle stages except G_(0) stage. Following serum starvation or confluent culture for 7 days, fetal fibroblast cells were effectively reached to G_(0) stage. The cell cycle was resumed after culture of these G_(0) stage-fetal fibroblast cells with normal medium. These results indicates that fetal fibroblast cells originated from Hanwoo were successfully isolated and culture system and induction of cell cycle of these cells were established for somatic cell nuclear transfer in Hanwoo.

        Molecular cloning and expression of fungal cellobiose transporters and β-glucosidases conferring efficient cellobiose fermentation in Saccharomyces cerevisiae

        Cellobiose was once regarded as a byproduct that should be removed from biomass hydrolysates because of its inhibitory activity to cellulases. It was revealed, however, that cellobiose could serve as a co-substrate for xylose fermentation by engineered Saccharomyces cerevisiae. Despite its advantages, to date, little is known about cellodextrin transporters that endow S. cerevisiae with cellobiose transporting ability. In this study, engineered S. cerevisiae strains capable of fermenting cellobiose were constructed by expressing various fungal cellobiose transporters and intracellular β-glucosidases. Among them, the strain expressing a putative sugar transporter from Penicillium chrysogenum (Pc_ST) and β-glucosidase from Thielavia terrestris (Tt_BG) showed an improved cellobiose fermentation performance compared to the strain expressing a cellodextrin transporter from Neurospora crassa (Nc_CDT-1) and β-glucosidase from N. crassa (Nc_GH1-1). Cellobiose fermentation by S. cerevisiae Pc_ST/Tt_BG under microaerobic conditions resulted in 14.5+/-0.5g/L of final ethanol concentration with a yield of 0.37+/-0.01g ethanol/g cellobiose, which are 22% and 26% higher than the corresponding values of S. cerevisiae Nc_CDT-1/Nc_GH1-1. These results suggest that the yield and rate of cellobiose fermentation can be improved by adopting optimal pairs of cellobiose transporters and β-glucosidase.
