      • BubR1 acetylation at prometaphase is required for modulating APC/C activity and timing of mitosis

        <P>Regulation of BubR1 is central to the control of APC/C activity. We have found that BubR1 forms a complex with PCAF and is acetylated at lysine 250. Using mass spectrometry and acetylated BubR1-specific antibodies, we have confirmed that BubR1 acetylation occurs at prometaphase. Importantly, BubR1 acetylation was required for checkpoint function, through the inhibition of ubiquitin-dependent BubR1 degradation. BubR1 degradation began before the onset of anaphase. It was noted that the pre-anaphase degradation was regulated by BubR1 acetylation. Degradation of an acetylation-mimetic form, BubR1–K250Q, was inhibited and chromosome segregation in cells expressing BubR1–K250Q was markedly delayed. By contrast, the acetylation-deficient mutant, BubR1–K250R, was unstable, and mitosis was accelerated in BubR1–K250R-expressing cells. Furthermore, we found that APC/C–Cdc20 was responsible for BubR1 degradation during mitosis. On the basis of our collective results, we propose that the acetylation status of BubR1 is a molecular switch that converts BubR1 from an inhibitor to a substrate of the APC/C complex, thus providing an efficient way to modulate APC/C activity and mitotic timing.</P>

        통합탐구 단원에 대한 초등학교 교사들의 인식과 지도 실태

        이 연구는 2009 개정 교육과정 5∼6학년 과학 교과서에 새롭게 제시된 통합탐구 단원에 대한 초등학교 교사들의 인식과 지도 실태를 조사하여 통합탐구 단원 구성과 지도의 개선 방안을 모색하고자 하였다. 이를 위해 탐구지도와 관련된 기존 설문지를 수정, 보완하여 통합탐구 단원을 지도한 경험이 있는 교사들에게 설문을 실시해서 응답내용을 분석하였다. 그 결과, 교사들은 통합탐구과정 지도의 중요성에 대해 긍정적으로 인식하고 있었다. `통합탐구능력 습득 및 발달, 과학 학습의 전이, 자유탐구 실행` 측면에서 교사들의 통합탐구 단원의 적용 효과에 대한 인식은 긍정적이었으며 그 이유로 `통합탐구과정 학습을 위한 별도의 시간 확보, 후속 학습에서의 활용, 자유탐구 활동에서 통합탐구과정 적용의 용이함`을 들었다. 반면에 교사들은 `통합탐구과정의 분절적 학습, 통합탐구 단원과 다른 단원 간의 연계성 부족, 학생의 호기심 유발이 어려운 교과서 내용 구성`을 이유로 통합탐구 단원의 효과에 대해 부정적인 의견을 제시하기도 했다. 또한 통합탐구 단원의 지도 실태에 대한 조사를 통해 많은 교사들이 통합탐구과정 지도 시 교과서 내용을 그대로 따르는 것을 확인하였다. 교사들이 통합탐구 단원 지도 시 어려움을 가장 크게 겪는 과정은 문제인식이었으며 그 다음으로 가설설정, 변인통제, 자료변환 및 자료해석, 일반화, 결론도출 순이었다. 교사들이 어려움을 겪는 이유는 `학생들의 호기심·경험·이해력 부족, 교사의 전문성 부족, 교과서 구성의 한계 및 학습 시간 부족` 등으로 나타났다. 이러한 연구 결과는 통합탐구 단원의 개선 및 통합탐구과정 지도 관련 교사 전문성 제고를 위한 노력이 필요함을 시사한다. The `Integrated Inquiry` unit is a new addition in the 5th∼6th grade science textbooks, and was developed for the 2009 revised national curriculum. The unit deals with six integrated inquiry process skills: problem cognition, formulating hypotheses, controlling variables, transforming and interpreting data, drawing results, and generalization. The purpose of this study is to investigate the perceptions and the teaching practices of elementary school teachers on this unit in their school curriculum and where improvements can be made. Data was collected from questionnaires filled out by 92 elementary school teachers. The results are as follows: First, teachers do have a positive perception on the importance of teaching integrated process skills. Second, with that being true, this unit ended up receiving both positive and negative reviews by teachers. This research found that there were good and bad responses on the educational aspects of this unit in three particular areas: dealing with the development of integrated inquiry process skills, facilitating science learning in other units, and implementing open inquiry. Third, teachers have difficulties particularly in problem cognition, formulating hypotheses, controlling variables, transforming and interpreting data, generalization, and drawing results. There is a lack of student understanding as well as a lack of professionalism for teachers on this unit, and many issues related to the composition of the textbook. This study may have important implications for making improvements in this unit and teaching integrated process skills.

        수학활동자료 보유현황 및 활용정도에 관한 교사 인식

        본 연구는 수학활동자료 보유현황 및 활용정도에 대한 교사 인식을 알아보고자 하는데 그 목적이 있다. 본 연구는 부산시 소재 공ㆍ사립 유치원교사 386명을 대상으로 질문지를 사용하였으며, 수집된 자료는 x2(chi-square)검증으로 분석하였다. 본 연구의 결과는 다음과 같다. 첫째, 교사의 경력에 따라 분류하기와 순서짓기, 수의 기초개념 이해하기, 기초적인 측정과 관련된 경험하기, 공간과 도형의 기초개념 알기, 컴퓨터에서 보유현황 및 활용정도에 유의미한 차이가 있었다. 둘째, 교사의 학력에 따라 분류하기와 순서짓기는 보유현황에, 수의 기초개념 이해하기, 기초적인 측정과 관련된 경험하기, 공간과 도형의 기초개념 알기는 보유현황 및 활용정도에, 기초적인 통계와 관련된 경험하기는 활용정도에, 컴퓨터는 보유현황 및 활용정도에서 유의미한 차이가 있었다. 셋째, 교사의 담당유아연령에 따라 분류하기와 순서짓지는 보뉴현황에, 수의 기초개념 이해하기, 기초적인 측정과 관련된 경험하기느 보유현황 및 활용정도에, 시간에 대한 기초개념 알기는 활용정도에, 공간과 도형의 기초개념 알기, 기초적인 통계와 관련된 경험하기, 컴퓨터는 보뉴현황 및 활용정도에서 유의미한 차이가 있었다. 넷째, 기관의 설립유형에 따라 분류하기와 순서짓기는 보유현황에, 수의 기초개념 이해하기, 기초적인 측정과 관련된 경험하기, 공간과 도형의 기초개념 알기, 컴퓨터는 보유현황 및 활용정도에서 유의미한 차이가 있었다. The purpose of this study is to examine teachers" perceptions of possession and use of mathematical activity materials. For this purpose, the researchers utilized a questionnaire survey of 386 public and private kindergarten teachers located in Busan. Data obtained from the survey were statistically processed and analyzed through x2(chi-square) test. Results of the study are summarized below. First, there were differences in the possession and use of mathematical activity materials such as "classification and ordering", "understanding the basic concept of number", "experience related to basic measurement", "knowing basic concepts of space and figure" and "computers" based on the amount of teaching experience of those surveyed. Second, there were differences in the possession of such mathematical activity material as "classification and ordering, the possession and use of "understanding the basic concept of number", "experience related to basic measurement" and "knowing basic concepts of space and figure", use of "experience related to basic statistics" and in the possession and use of "computers" based on the educational background of the teachers surveyed. Third, there were differences in the possession of such mathematical materials as "classification and ordering", the possession and use of "understanding the basic concept of number" and "experience related to basic measurement", use of "knowing the basic concept of time", and in the possession and use of "knowing basic concepts of space and figure", "experience related to basic statistics" and "computers" based on the age of the students the surveyed teachers were in charge of educating. Last, there were differences in the possession of such materials as "classification and ordering" and the possession and use of "understanding the basic concept of number", "experience related to basic measurement", "knowing basic concepts of space and figure" and "computers" based on the type of pre-school educational institution.

        Comparison of on-Statin Lipid and Lipoprotein Levels for the Prediction of First Cardiovascular Event in Type 2 Diabetes Mellitus

        Background: A substantial cardiovascular disease risk remains even after optimal statin therapy. Comparative predictiveness of major lipid and lipoprotein parameters for cardiovascular events in patients with type 2 diabetes mellitus (T2DM) who are treated with statins is not well documented.Methods: From the Korean Nationwide Cohort, 11,900 patients with T2DM (≥40 years of age) without a history of cardiovascular disease and receiving moderate- or high-intensity statins were included. The primary outcome was the first occurrence of major adverse cardiovascular events (MACE) including ischemic heart disease, ischemic stroke, and cardiovascular death. The risk of MACE was estimated according to on-statin levels of low-density lipoprotein cholesterol (LDL-C), triglyceride (TG), highdensity lipoprotein cholesterol (HDL-C), and non-HDL-C.Results: MACE occurred in 712 patients during a median follow-up period of 37.9 months (interquartile range, 21.7 to 54.9). Among patients achieving LDL-C levels less than 100 mg/dL, the hazard ratios for MACE per 1-standard deviation change in ontreatment values were 1.25 (95% confidence interval [CI], 1.07 to 1.47) for LDL-C, 1.31 (95% CI, 1.09 to 1.57) for non-HDL-C, 1.05 (95% CI, 0.91 to 1.21) for TG, and 1.16 (95% CI, 0.98 to 1.37) for HDL-C, after adjusting for potential confounders and lipid parameters mutually. The predictive ability of on-statin LDL-C and non-HDL-C for MACE was prominent in patients at high cardiovascular risk or those with LDL-C ≥70 mg/dL.Conclusion: On-statin LDL-C and non-HDL-C levels are better predictors of the first cardiovascular event than TG or HDL-C in patients with T2DM.

        Many engineering polymer is suitable for Crack Layer theory (CL). According to this theory, polymer have different size and shape of process zone (PZ) and in the case of high density polyethylene (HDPE), crack propagates in a d discontinuous manner. In this article, slow crack growth was simulated by microsoft visual C++ 2010 on the grade of HDPE with single edge notched bar (SENT) specimen. In addition, the effects of the stress ratio (R-ratio) that is defined as the ratio of minimum and maximum stress of loading, fracture energy, and draw stress parameter were studied. These parameters have clear tendency and the program has view which appropriate for many other polymers just by turning some parameters into proper parameters.

        Statin Therapy and the Risk of Osteoporotic Fractures in Patients with Metabolic Syndrome: a Nested Case-Control Study

        Objective: We aimed to investigate the association between statin use and the risk of major osteoporotic fractures in patients with metabolic syndrome (MetS). Methods: A nested case-control study was performed in patients with MetS (≥50 years) who had no history of osteoporotic fracture using the Korean National Health Insurance Service- Health Screening Cohort. This study included 17,041 patients diagnosed with new-onset osteoporotic fractures and controls matched in a 1:1 ratio by age, sex, body mass index, cohort entry date, and follow-up duration. Conditional logistic regression analysis was used to evaluate covariate-adjusted odds ratios (ORs) and 95% confidence intervals (CIs). Results: During a 4-year follow-up period, the risk of major osteoporotic fractures was significantly reduced by 9% (OR, 0.91; 95% CI, 0.85–0.97) in statin users compared with that in non-users. Among subtypes of major osteoporotic fracture, a risk reduction with statin therapy was significant for vertebral fracture (OR, 0.86; 95% CI, 0.79–0.94) but not for non-vertebral fracture (OR, 0.97; 95% CI, 0.88–1.06). Longer duration (OR, 0.97; 95% CI, 0.96–0.99, per 1-year increase) and higher cumulative dose (OR, 0.97; 95% CI, 0.95–0.99, per 365 defined daily doses) of statins were negatively associated with the risk of major osteoporotic fracture. Conclusion: This study supports the hypothesis that statin therapy has a beneficial effect on major osteoporotic fractures, especially vertebral fractures, in patients with MetS.

        Gender differences in the presentation of chest pain in obstructive coronary artery disease: results from the Korean Women’s Chest Pain Registry

        Background/Aims: Chest pain in patients with obstructive coronary artery disease (OCAD) is affected by several social factors. The gender-based differences in chest pain among Koreans have yet to be investigated. Methods: The study consecutively enrolled 1,549 patients (male/female, 514/1,035; 61 ± 11 years old) with suspected angina. The predictive factors for OCAD based on gender were evaluated. Results: Men experienced more squeezing type pain on the left side of chest, while women demonstrated more dull quality pain in the retrosternal and epigastric area. After adjustment for risk factors, pain in the retrosternal area (odds ratio [OR], 1.491; 95% confidence interval [CI], 1.178 to 1.887) and aggravation by exercise (OR, 2.235; 95% CI, 1.745 to 2.861) were positively associated with OCAD. In men, shorter duration (OR, 1.581; 95% CI, 1.086 to 2.303) and dyspnea (OR, 1.610; 95% CI, 1.040 to 2.490) increased the probability for OCAD, while left-sided chest pain suggested a low probability for OCAD (OR, 0.590; 95% CI, 0.388 to 0.897). In women, aggravation by emotional stress (OR, 0.348; 95% CI, 0.162 to 0.746) and dizziness (OR, 0.457; 95% CI, 0.246 to 0.849) decreased the probability for OCAD. Conclusions: This is the first study to focus on gender differences in chest pain among Koreans with angina. Symptoms with high probability for OCAD were different between sexes. Our findings suggest that patient’s medical history in pretest assessment for OCAD should be individualized considering gender.

        Impact of carotid atherosclerosis in CHA<sub>2</sub>DS<sub>2</sub>- VASc-based risk score on predicting ischemic stroke in patients with atrial fibrillation

        Background/Aims: Vascular disease is an established risk factor for stroke in patients with atrial fibrillation (AF), which is included in CHA<sub>2</sub>DS<sub>2</sub>-VASc score. However, the role of carotid atherosclerosis remains to be determined. Methods: Three hundred-ten patients with AF who underwent carotid sonography were enrolled. Results: During a median follow-up of 31 months, 18 events (5.8%) of stroke were identified. Patients with stroke had higher carotid intima-media thickness (CIMT) (1.16 ± 0.33 mm vs. 0.98 ± 0.25 mm, p = 0.017). CIMT was significantly increased according to the CHA<sub>2</sub>DS<sub>2</sub>-VASc score (p < 0.001) and it was correlated with left ventricular mass index and early diastolic mitral annular velocity (e’), a ratio of early transmitral flow velocity to e’ (E/e’) and pulmonary artery systolic pressure (all p < 0.05). Cox regression using multivariate models showed that carotid plaque was associated with the risk of stroke (hazard ratio, 3.748; 95% confidence interval [CI], 1.107 to 12.688; p = 0.034). C-statistics increased from 0.648 (95% CI, 0.538 to 0.757) to 0.716 (95% CI, 0.628 to 0.804) in the CHA<sub>2</sub>DS<sub>2</sub>-VASc score model after the addition of CIMT and carotid plaque as a vascular component (p = 0.013). Conclusions: Increased CIMT and presence of carotid plaque are associated with a high risk of ischemic stroke, and CIMT is related to myocardial remodeling and diastolic dysfunction, suggesting that carotid atherosclerosis can improve risk prediction of stroke in patients with AF, when included under vascular disease in the CHA<sub>2</sub>DS<sub>2</sub>-VASc scoring system.
