      • Efficacy Analysis of Simplified Intensity-modulated Radiotherapy with High or Conventional Dose and Concurrent Chemotherapy for Patients with Neck and Upper Thoracic Esophageal Carcinoma

        For patients with neck and upper thoracic esophageal carcinoma, it is difficult to control lymph node metastases with conventional dose therapy. In this study, we assessed the feasibility of simplified intensity-modulated radiotherapy (sIMRT) and concurrent chemotherapy for 44 patients and boosted high-dose to metastatic lymph nodes. Three radiation treatment volumes were defined: PGTVnd, with which 68.1Gy was delivered in high dose group (hsIMRT group), and 60Gy in the conventional dose group (csIMRT group); PTV1, featuring 63.9Gy in the hsIMRT group and 60Gy in the csIMRT group; PTV2, with 54Gy given to both groups. The sIMRT plan included 5 equi-angular coplanar beams. All patients received the cisplatin and 5-FU regimen concurrently with radiotherapy. The treatment was completed within six weeks and one case with grade three acute bronchitis was observed in hsIMRT group. For esophageal lesions, 80% complete response (CR) and 20% partial response (PR) rates were found in the hsIMRT group, and 79.2% CR, with 20.8% PR, in the csIMRT group; for lymph node lesions, 75% CR and 25% PR rates were observed in the hsIMRT group, with 45.8% and 37.5% respectively in the csIMRT group (P<0.05). The differences in 1-, 2- and 3-year relapse-free survival rates were all statistically significant (P<0.05). The major toxicity observed in both groups was Grade I~II leucopenia. sIMRT can generate a desirable dose distribution in treatment of neck and upper thoracic esophageal carcinoma with a better short-term efficacy. Boosted high dosing to metastatic lymph nodes can increase the relapse-free survival rate.

        Molecular cloning, chromosomal localization and expression profiling of porcine selenoprotein M gene

        Selenoprotein M may regulate a myriad of biological processes through its redox function. In pigs, neither the nucleotide sequence nor the amino acid sequence is known. Furthermore,patterns of tissue expression and regulation by dietary selenium (Se) have not been examined. We determined the full coding sequence (CDS) and the chromosomal location of the porcine gene, SELM, and described its expression profile in vivo under different dietary Se concentrations. The cDNA sequence of porcine SELM from the start codon to the poly(A) tail was cloned by reverse transcription PCR. The CDS contained 429bases with a typical mammalian selenocysteine insertion sequence of form 2 (F2) located in the 3′-untranslated region. The gene was mapped to chromosome 14q21, where porcine SELM and its neighboring genes exhibited a similar organization to human homologues on chromosome 22q12.2. The expression pattern of SELM mRNA in muscle, thyroid, cerebral cortex, pituitary, testis, liver, and kidney was analyzed with real-time quantitative PCR in young male pigs fed a Se-deficient corn-soybean meal basal diet supplemented with 0.0, 0.3,or 3.0 mg Se/kg in the form of Se-rich yeast. Though the SELM mRNA abundance in each of the 7 tissues was not affected by the dietary Se concentrations, it was significantly higher in thyroid (P < 0.01) than in cerebral cortex, pituitary,testis, liver, and kidney at all of the 3 dietary Se concentrations.

        Optimal magnetoelectric coupling performance of Terfenol-D/PZT composites in resonance state under bias magnetic field

        Laminated magnetoelectric (ME) composites with various thickness ratios were optimized, fabricated and experimentally investigated in this work. The Terfenal-D/PZT specimens with optimal thickness ratio between the magnetostrictive phase and piezoelectric phase, and two other values were tested for their ME coupling performance. The coupling voltage output increases linearly with the increase of DC bias magnetic field. The ME voltage coefficient increases more than 100 times in the resonance state for the optimal laminate. The DC bias magnetic field affects the ME voltage coefficient significantly, and also has little effect on the resonant frequency. The strength of AC magnetic field also slightly affects the ME voltage coefficient in resonance state, but does not affect the resonant state under which the same DC magnetic field is required. The experimental results can help understand the coupling performance of ME composite under bias magnetic field and prompt the application of ME devices.

        Molecular Cloning and Function Analysis of an Anthocyanidin Synthase Gene from Ginkgo biloba, and Its Expression in Abiotic Stress Responses

        Anthocyanidin synthase (ANS, leucoanthocyanidin oxygenase), a 2-oxoglutarate iron-dependent oxygenase, catalyzed the penultimate step in the biosynthesis of the anthocyanin class of flavonoids, from the colorless leucoanthocyanidins to the colored anthocyanidins. The full-length cDNA and genomic DNA sequences of ANS gene (designated as GbANS) were isolated from Ginkgo biloba for the first time. The full-length cDNA of GbANS contained a 1062-bp open reading frame (ORF) encoding a 354-amino-acid protein. The genomic DNA analysis showed that GbANS gene had three exons and two introns. The deduced GbANS protein showed high identities to other plant ANSs. The conserved amino acids (H-X-D) ligating ferrous iron and residues (R-X-S) participating in 2-oxoglutarate binding were found in GbANS at the similar positions like other ANSs. Southern blot analysis indicated that GbANS belonged to a multi-gene family. The expression analysis by real-time PCR showed that GbANS expressed in a tissue-specific manner in G. biloba. GbANS was also found to be up-regulated by all of the six tested abiotic stresses, UV-B, abscisic acid, sucrose, salicylic acid, cold and ethylene, consistent with the promoter region analysis of GbANS. The recombinant protein was successfully expressed in E. coli strain with pET-28a vector. The in vitro enzyme activity assay by HPLC indicated that recombinant GbANS protein could catalyze the formation the cyanidin from leucocyanidin and conversion of dihydroquercetin to quercetin, suggesting GbANS is a bifunctional enzyme within the anthocyanidin and flavonol biosynthetic pathway.

        A High Performance Permanent Magnet Synchronous Motor Servo System Using Predictive Functional Control and Kalman Filter

        A predictive functional control (PFC) scheme for permanent magnet synchronous motor (PMSM) servo systems is proposed in this paper. The PFC-based method is first introduced in the control design of speed loop. Since the accuracy of the PFC model is influenced by external disturbances and speed detection quantization errors of the low distinguishability optical encoder in servo systems, it is noted that the standard PFC method does not achieve satisfactory results in the presence of strong disturbances. This paper adopted the Kalman filter to observe the load torque, the rotor position and the rotor angular velocity under the condition of a limited precision encoder. The observations are then fed back into PFC model to rebuild it when considering the influence of perturbation. Therefore, an improved PFC method, called the PFC+Kalman filter method, is presented, and a high performance PMSM servo system was achieved. The validity of the proposed controller was tested via experiments. Excellent results were obtained with respect to the speed trajectory tracking, stability, and disturbance rejection.

        Disruption of Phaffia rhodozyma cells and preparation of microencapsulated astaxanthin with high water solubility

        A novel process was developed for encapsulationof astaxanthin from Phaffia rhodozyma. The yeast cellswere disrupted by glass beads and the high shearing forcepartially emulsified the astaxanthin in aqueous phase. Theenzymolysis method was then adopted to prepare the yeastextract for a full use of the cells. The gelatin and porousstarch were used to microencapsulate the emulsifiedastaxanthin. Under optimized conditions, the recovery ofamino nitrogen and solid reached 3.68 ± 0.32% and49.22 ± 2.34%, respectively. The microencapsulationconditions were optimized through orthogonal experimentand the encapsulation efficiency, loading astaxanthin, andamino-nitrogen reached 88.56%, 1.55 mg/g, and1.35 ± 0.14%, respectively. The water solubility ofmicrocapsules reached 81.5 ± 0.35%. Color and storagestability analysis showed that microencapsulation ofastaxanthin possessed higher thermal stability. The resultsdemonstrated that the established process was effective andpractical.

      • A Wireless Sensor Network Clustering Algorithm based on Hypergraph

        The paper proposes a clustering algorithm for wireless sensor network based on hypergraph. Under the hypergraph model, a wireless sensor network is mapped to a hypergraph. Then a hierarchical iterative clustering algorithm is applied to the hypergraph, thus dividing the hypergraph into multiple parts. A cluster head is selected from each part. In order to improve the partitioning process, a new modularity function is proposed. Compared with the classic clustering algorithm LEACH, simulation shows that our algorithm performs better in energy consuming and network lifetime.

        Inhibition of Poly(I:C)-Induced Inflammation by Salvianolic Acid A in Skin Keratinocytes

        Background: Skin keratinocytes participate actively in inducing immune responses when external pathogens are introduced, thereby contributing to elimination of pathogens. However, in condition where the excessive inflammation is occurred, chronic skin disease such as psoriasis can be provoked. Objective: We tried to screen the putative therapeutics for inflammatory skin disease, and found that salvianolic acid A (SAA) has an inhibitory effect on keratinocyte inflammatory reaction. The aim of this study is to demonstrate the effects of SAA in poly(I:C)-induced inflammatory reaction in skin keratinocytes. Methods: We pre-treated keratinocytes with SAA then stimulated with poly(I:C). Inflammatory reaction of keratinocytes was verified using real-time polymerase chain reaction, enzyme-linked immunosorbent assay and Western blot. Results: When skin keratinocytes were pre-treated with SAA, it significantly inhibited poly (I:C)-induced expression of inflammatory cytokines including interleukin (IL)-1β, IL-6, IL-8, tumor necrosis factor-α, and CCL20. SAA inhibited poly(I:C)-induced activation of nuclear factor-κB signaling. And SAA also inhibited inflammasome activation, evidenced by decrease of IL-1β secretion. Finally, SAA markedly inhibited poly(I:C)-induced NLRP3 expression. Conclusion: These results demonstrate that SAA has an inhibitory effect on poly(I:C)-induced inflammatory reaction of keratinocytes, suggesting that SAA can be developed for the treatment of inflammatory skin diseases such as psoriasis.
