      • The Influence of Relative Humidity, Temperature, Insect Developmental Stages on the Susceptibility of Sweetpotato Whitefly to Conidia of Isaria javanica

        Bemisia tabaci, sweetpotato whitefly, has been recognized one of the most destructive insect pests worldwide because of increased resistance to some insecticide groups requiring alternative strategies for its control. We conducted a study of the influence of relative humidity, temperature and different developmental stages on the susceptibility of sweetpotato whitefly to conidia of Isaria javanica isolate, which had been reported high virulence against Q biotype of B. tabaci. The mortality of tobacco whitefly was low at low constant relative humidities, but was high when kept high humidity for first 24 hours and transferred to low humidity. The Isaria isolate had wide range of temperature (15℃ to 35℃) to control sweetpotato whitefly. The isolate has virulence to the egg and all developmental stages of nymph of B. tabaci. These results indicated that the isolate had good control effects at various environmental conditions and is an excellent candidate to develop a microbial pesticide to control sweetpotato whitefly.

        Steel Bridge Construction of Hong Kong–Zhuhai–Macao Bridge

        The 55-km-long Hong Kong–Zhuhai–Macao Bridge (HZMB) is the world’s longest sea-crossing bridge, connecting Hong Kong with Zhuhai and Macao at the mouth of the Pearl River Estuary in China, comprising 22.9-km-long steel bridges. HZMB is the leading steel bridge in China, with top-level manufacturing and installation technology. This paper outlines the steel bridge construction experiences of HZMB to provide comparisons for the construction of other long sea-crossing steel bridges at home or abroad. The main considerations of construction constraints, scheme selection, structural and aesthetic design of HZMB are presented, and the following points related to new strategies in the steel bridge construction of HZMB are elaborated: (1) construction quality assurance, (2) automatic manufacturing technology, (3) large segment off shore installation, (4) eco-friendly paint (content limitation of volatile organic compounds) and new multifunctional inspection gantry, and (5) Guss Mastic Asphalt steel deck pavement system. The successful implementation of those strategies shows that the steel bridge construction of HZMB promotes improvement in the overall construction and management level of the Chinese bridge industry. The advanced experience of HZMB has opened up broad prospects for the design and construction of off shore bridge engineering in China.

        Genome-wide analyses reveal the extent of opportunistic STAT5 binding that does not yield transcriptional activation of neighboring genes

        <P>Signal Transducers and Activators of Transcription (STAT) 5A/B regulate cytokine-inducible genes upon binding to GAS motifs. It is not known what percentage of genes with GAS motifs bind to and are regulated by STAT5. Moreover, it is not clear whether genome-wide STAT5 binding is modulated by its concentration. To clarify these issues we established genome-wide STAT5 binding upon growth hormone (GH) stimulation of wild-type (WT) mouse embryonic fibroblasts (MEFs) and MEFs overexpressing STAT5A more than 20-fold. Upon GH stimulation, 23 827 and 111 939 STAT5A binding sites were detected in WT and STAT5A overexpressing MEFs, respectively. 13 278 and 71 561 peaks contained at least one GAS motif. 1586 and 8613 binding sites were located within 2.5 kb of promoter sequences, respectively. Stringent filtering revealed 78 genes in which the promoter/upstream region (−10 kb to +0.5 kb) was recognized by STAT5 both in WT and STAT5 overexpressing MEFs and 347 genes that bound STAT5 only in overexpressing cells. Genome-wide expression analyses identified that the majority of STAT5-bound genes was not under GH control. Up to 40% of STAT5-bound genes were not expressed. For the first time we demonstrate the magnitude of opportunistic genomic STAT5 binding that does not translate into transcriptional activation of neighboring genes.</P>

      • Alkaloids from Beach Spider Lily (Hymenocallis littoralis) Induce Apoptosis of HepG-2 Cells by the Fas-signaling Pathway

        Alkaloids are the most extensively featured compounds of natural anti-tumor herbs, which have attracted much attention in pharmaceutical research. In our previous studies, a mixture of major three alkaloid components (5, 6-dihydrobicolorine, 7-deoxy-trans-dihydronarciclasine, littoraline) from Hymenocallis littoralis were extracted, analyzed and designated as AHL. In this paper, AHL extracts were added to human liver hepatocellular cells HepG-2, human gastric cancer cell SGC-7901, human breast adenocarcinoma cell MCF-7 and human umbilical vein endothelial cell EVC-304, to screen one or more AHL-sensitive tumor cell. Among these cells, HepG-2 was the most sensitive to AHL treatment, a very low dose ($0.8{\mu}g/ml$) significantly inhibiting proliferation. The non-tumor cell EVC-304, however, was not apparently affected. Effect of AHL on HepG-2 cells was then explored. We found that the AHL could cause HepG-2 cycle arrest at G2/M checkpoint, induce apoptosis, and interrupt polymerization of microtubules. In addition, expression of two cell cycle-regulated proteins, CyclinB1 and CDK1, was up-regulated upon AHL treatment. Up-regulation of the Fas, Fas ligand, Caspase-8 and Caspase-3 was observed as well, which might imply roles for the Fas/FsaL signaling pathway in the AHL-induced apoptosis of HepG-2 cells.

      • XPD Lys751Gln and Asp312Asn Polymorphisms and Susceptibility to Skin Cancer: A Meta-Analysis of 17 Case-control Studies

        Background: Numerous studies have explored the influence of XPD Lys751Gln and/or Asp312Asn polymorphisms on skin cancer susceptibility. However, the results remain inconclusive. To derive a more precise estimation, we conducted a comprehensive search to identify all available published studies and performed a meta-analysis. Materials and Methods: Electronic literature searches of the PubMed, CBM and CNKI databases were performed up to March 2014. Odds ratios (ORs) with 95% confidence intervals (CIs) were applied to assess the strength of associations. Results: Seventeen case-control studies were included with a total sample size of 6, 113 cases and 11, 074 controls for the XPD Lys751Gln polymorphism, and 10 studies (3, 840cases and 7, 637 controls) for the XPD Asp312Asn polymorphism were pooled for analysis. Overall, no significant associations were found between the XPD Lys751Gln polymorphism and skin cancer risk in any genetic model. On stratified analysis by tumor type, XPD Lys751Gln polymorphism was not associated with increased risk of non-melanoma skin cancer, but was significantly related with increased risk of cutaneous melanoma (Gln/Gln vs Lys/Lys: OR=1.15, 95%CI=1.02-1.29, p=0.023; dominant model: OR=1.09, 95%CI=1.01-1.18, p=0.036). For the XPD Asp312Asn polymorphism, no significant association with skin cancer risk was observed in overall or subgroup analyses. Conclusions: The present meta-analysis suggests that the XPD Lys751Gln polymorphism may contribute to the risk of cutaneous melanoma from currently available evidence. Further investigations are needed to obtain more insight into possible roles of these two polymorphisms in skin carcinogenesis.

      • Efficient Promotion of Autophagy and Angiogenesis Using Mesenchymal Stem Cell Therapy Enhanced by the Low-Energy Shock Waves in the Treatment of Erectile Dysfunction

        <P><B>Background</B></P><P> Mesenchymal stem cell therapy (MSCT) and defocused low-energy shock wave therapy (ESWT) has been shown to ameliorate erectile dysfunction (ED). However, the interactions and effects of action between MSCT and ESWT remain poorly understood. In this study, we investigated the mechanisms of combination therapy with MSCT and ESWT in a rat model of diabetic ED. </P><P><B>Materials and Methods</B></P><P> Eight-week-old male Sprague-Dawley rats were randomly divided into 2 parts. Diabetic rats induced by streptozotocin (65 mg/kg) were randomly divided into 4 groups: (1) DM control group, (2) DM + ESWT group, (3) DM + MSCT group, and (4) DM + ESWT + MSCT group. The sham group was a normal control group (without streptozotocin). MSCT and (or) ESWT were, respectively, administered to each group according to the proposal for 8 weeks. Immediately after recording of intracavernous pressure (ICP), the penis was then harvested for histologic analysis, ELISA, and Western blotting. </P><P><B>Results</B></P><P> The ratio of ICP/MAP was significantly higher in the DM + ESWT + MSCT group than in ESWT or MSCT treated group (<I>P</I> < 0.05). Also, the treatment stimulated angiogenesis and vasodilatation in the corpus cavernosum (<I>P</I> < 0.05). ESWT increased the quantity of MSCs in the corpus cavernosum and also induced MSCs to express more VEGF in vitro and vivo (<I>P</I> < 0.05) which activated the PI3K/AKT/mTOR and NO/cGMP signaling pathways in the corpus cavernosum. The combination approach stimulated autophagy and decreased apoptosis in the corpus cavernosum. NGF and BDNF expressions were higher in the DM + ESWT + MSCT group than in the DM control group (<I>P</I> < 0.01). Furthermore, the treatment promoted the MSC recruitment by inducing penile tissues to express more PECAM and SDF-1. </P><P><B>Conclusions</B></P><P> Combination of LI-ESWT and MSCT can get a better result than a single treatment by expressing more VEGF which can take part in autophagy by triggering the PI3K/AKT/mTOR signaling pathway. This cooperative therapy would provide a new research direction in ED treatment for the future.</P>

        고객참여 행동에 대한 이중경로 동기요인에 관한 연구 -모바일 SNS 위챗(WeChat)을 중심으로-

        본 연구는 높은 빈도의 상호작용을 특징으로 하는 온라인 “친분 사회”인 위챗(WeChat)을 연구대상으로, 모바일 SNS 플랫폼 상에서 고객참여 행동에 영향을 미치는 요인들을 탐색하였다. 이를 통해 모바일 SNS를 사용하는 고객들의 참여를 향상시키는 방안을 모색하여, 기업이 모바일 SNS 플랫폼에서 마케팅 활동을 효과적으로 수행하고 이를 개발 및 사용할 수 있도록 하는 관리적 시사점을 제공하고자 한다. 본 연구는 선행연구를 검토하여 연구 문제와 관련된 중요변수 및 측정항목을 선정하고, 342개의 데이터를 수집하여 구조방정식 모형 분석을 실시하였다. 분석 결과, 온라인 환경에서, 상황적 요인은 감정에 정(+)적인 영향을 미치고, 감정은 고객 참여의 적극적 및 소극적 행동에 정(+)적인 영향을 미치는 것으로 나타났다. 또한 자기효능감 역시 고객참여의 적극적 및 소극적 행동에 정(+)적인 영향을 미치는 것으로 나타났다. 하지만 브랜드 개성인식은 적극적 행동에만 정(+)의 영향을 미치는 것으로 나타났다. 다음으로 오프라인 환경에서, 정보 일관성은 감정과 고객참여의 적극적 행동에만 정(+)적인 영향을 미치는 것으로 나타났다. 또한 공동체 영향은 고객참여의 수동적 행동에만 정(+)적인 영향을 미치고, 적극적 행동에는 부(-)적인 영향을 미치는 것으로 나타났다. This study takes the online “acquaintance society” with high frequency interaction―WeChat as the research object, to create a research framework through the driving factors of customer participation behavior on the mobile social network platform, deeply understand the rules of improving customer participation in enterprises’ marketing activities on the mobile social network platform, and provide management enlightenment for enterprisers to better develop and use mobile social network. From a review of the literature, this study selects important variables and measurement items with reference to the research problem, conducting a structural equation model analysis to acquire data on 342 valid random samples to creat a modifed conceptual model. Finally, empirical analysis shows that in the online environmental, contextual factor had positve(+) effect upon emotion; emotion exercised positive(+) influence upon active and passive behaviours of customer paticipation; self-efficacy also exercised positive(+) effect on active and passive behaviours of customer paticipation; however, brand personality perception had positive(+) effect upon only active behaviours of customer paticipation; in the offline environmental, information consistency exercised positive(+) influence upon emotion and active behaviours of customer paticipation only; circle influence had positive(+) effect upon passive behaviours and negative(-) effect upon active behaviours of customer paticipation.

        Redundancy of Single-layer Dome under Earthquake Action Based on Response Sensitivity

        Structural redundancy is an important component of structural robustness that reflects the security of the structure under the influence of damage. In this paper, a method based on the response sensitivity is proposed for the assessment of the redundancy of a structure under earthquake excitation, and the response sensitivity is defined as the derivation of element strain to the elastic modulus of the structural materials. The redundancy analysis is carried out for three models of Kiewitt-6 single-layer reticulated dome built at a scale of 1:10 and subjected to earthquake action. The calculated redundancies and the results from the shaking table tests of the three models show that the members damaged by a severe earthquake are characterized by a small redundancy. The redundancy is able to reveal the weak links in the structure, and it represents the importance of the member in the structure. Members with small redundancy are identified as structural weaknesses that are easily damaged under loads, and their failure may cause catastrophic consequences to the dome. Such members are the key components of the structures.

        B3GNT3 acts as a carcinogenic factor in endometrial cancer via facilitating cell growth, invasion and migration through regulating RhoA/RAC1 pathway‑associated markers

        Background Aberrant expression of beta-1,3-N-acetylglucosaminyltransferase-3 (B3GNT3) has been frequently clarifed in various cancers, however, its role in endometrial cancer (EC) has not been assessed in detail. Purpose This study aimed to investigate the biological role of B3GNT3 in EC and simply explored the detailed mechanism. Methods The EC RNA-Seq dataset from TCGA database was applied to evaluate the expression of B3GNT3 and assess its role on prognostic value. HEC-1-A and KLE cell lines of EC were used to perform loss- and gain-of-function B3GNT3 assays respectively. Quantitative real-time PCR (qRT-PCR) and western blot were used to measure the mRNA and protein levels of indicated molecules respectively. Cell counting kit-8, clone formation tests, and Transwell assay served to determine the changes of proliferative, invasive and migratory abilities of EC cells after altering the expression of B3GNT3. Results B3GNT3 was found to be highly expressed in EC tissues compared to normal tissues according to the online public databases, which confrmed by the following qRT-PCR in 3 EC cell lines. Besides, high B3GNT3 expression presented a worse overall survival in EC patients as compared with low B3GNT3 expression group. Furthermore, functional experiments in vitro indicated that B3GNT3 could facilitate the cell growth, invasion and migration. Moreover, we found that downregulation of B3GNT3 signifcantly reduced the expression level of GTP-RhoA and GTP-RAC1, whereas upregulation of B3GNT3 presented the opposite results. Conclusion The results of current study demonstrate that B3GNT3 acts as an oncogene that promotes EC cells growth, invasion and migration possibly through regulating the RhoA/RAC1 signaling pathway-related markers, suggesting that B3GNT3 may be a candidate biomarker for EC therapeutic intervention.

      • XPC 939A>C and 499C>T Polymorphisms and Skin Cancer Risk: a Meta-analysis

        The xeroderma pigmentosum complementation group C gene (XPC) has been identified as important for repairing UV-related DNA damage. Some subtle changes in this gene may impair repair efficiency and influence susceptibility to human cancers, including skin cancer. Two polymorphisms in XPC, 939A>C (rs2228001) and 499C>T (rs2228000), are considered to have possible associations with the risk of skin cancer, but the reported results have been inconsistent. Here we performed a meta-analysis of the available evidence regarding the relationship between these two polymorphisms and the risk of skin cancer. All relevant studies were searched using PubMed, Embase and Web of Science before February 2012. A total of 8 case-control studies were included in this analysis, and no convincing associations between the two polymorphisms and risk of skin cancer were observed in any of the genetic models. Stratified analyses by skin cancer type also did not detect significant associations in any subgroup. This meta-analysis suggested that the XPC 939A>C and 499C>T polymorphisms may have little involvement in susceptibility to skin cancer.
