        중등학교 과학 수행평가의 평가 유형과 채점 방식 및 신뢰도 분석

        본 연구에서는 중등학교 과학 수행평가가 어떤 평가 유형과 채점 방식을 사용하고 있는지 분석하였으며, 이를 토대로 일반화가능도 이론을 이용하여 과학 수행평가 점수가 얼마나 신뢰로운지 분석하였다. 연구 결과, 과학 수행평가의 유형은 크게 지필형과 과제형으로 나눌 수 있었다. 지필형으로는 중등학교 모두 응답제한형 서술형검사만을 실시하고 있었다. 과제형은 과정 평가와 결과물 평가로 나눌 수 있었다 채점 방식은 1명의 교사가 모든 서술형문항과 수행과제를 채점하거나, 2명의 교사가 수행과제를 나누어 채점하고 있었다. 그러나 2명 이상의 교사가 서술형문항을 나누거나 교차하여 채점하는 경우는 없었다. 표집된 7개 중등학교 과학 수행평가의 신뢰도 분석 결과는 다음과 같다: (1) 서술형 문항의 특성이 수행과제의 특성보다 학생들의 수행평가 점수에 미치는 영향이 더 큰 것으로 나타났다 (2) 수행과제 채점에서 채점자가 피험자를 다르게 채점하는 정도는 학교에 따라 상당한 차이가 있었다. (3) 7개 중등학교 과학 수행평가의 일반화가능도 (신뢰도)는 대부분 적정 수준인 0.80에 미치지 못하는 것으로 나타났으며, 적정 수준의 일반화가능도를 얻기 위해서는 지금보다 더 많은 수의 서술형 문항과 수행과제 그리고 채점자가 필요한 것으로 분석되었다. In this study, we questioned what assessment types and scoring methods of science performance assessment(SPA) were being used in middle and high school, and how much these SPA scores were reliable(generalizable). To answer these questions, SPA data obtained from the seven schools were classified according to assessment type and scoring method. Based upon this classification, we analyzed the reliability by applying generalizability theory. The result, from the classification of assessment type and scoring method, showed that SPA types of the seven schools were divided into two types: paper-pencil type and task type. Paper-pencil type included answer(content)-restricted essay-type test solely. Task type has two parts: process and outcome assessment. As the results of analyzing scoring methods of the seven schools, there were two cases in the way of scoring methods: one case is scoring all essay-type items and performance tasks by one teacher, the other is scoring assigned performance tasks by two teachers. But the case of scoring assigned essay-type items or the case of cross scoring by two or more teachers were not found. The findings of the reliability analysis are as follows: (1) Effect of essay-type item to SPA score was larger than that of performance task. (2) There was remarkable difference among the seven schools' interaction effect of person and rater in scoring performance tasks. (3) Most of generalizability(reliability) coefficients of SPA for the seven schools were smaller than the acceptable generalizability coefficient(0.80). Therefore, the population of statistical parameters such as number of item, task and rater, should be increased for approaching the acceptable generalizability level.

        FISH법을 이용한 낙동강 상,중,하류의 세균군집 구조 분석

        For conducting the comparative analysis of the eubacterial community structure including nitrifying bacteria at 8 sampling sites throughout the N마-Dong River, FISH(fluorescence in situ hybridization) method was employed. The total ratio of each determined eubacterial group such as α·β·γ-subclasses protebacteria and Cytophaga-Flavobacterium(CF) group to total counts stained by DAPI at each site varied 9.3∼42.5% with the highest value at uppermost part. And each ratio of determined eubacterial groups reached mostly under 10% except those of CF group(23%) at uppermost part. Futhermore, compared to lower part, upper part represented higher proportions of γ-subclass proteobacteria comprised in most of fast growing bacteria on degradable organics. Also the variations of ammonia-oxidizing bacteria ranged from 2.7×10⁴to 18.3×10⁴ cells/㎖ with the lowest value in lower part and the highest value in mid part whereas those of nitrite-oxidizing bacteria varied 5.2∼7.7×10⁴cell/㎖ without noticeable differences throughout the sites. Additionally, the ratio of nitrifying bacteria to total counts ranged from 1.3% to 13.6?%, regardless of those of ammonia/nitrite-oxidizing bacteria. Overall, it implies that the upper part of Nak-Dong River represented comparatively better degradabilities of organic materials than their Lower part except at one site in upper part passed An-Dong City. By the contrast, the other part of Nak-Dong River, especially after merging the Gum-Ho River revealed lower detectability of FISH resulted from their cellular activity. So it could be said that the lower part of Nak-Dong River might be a ecosystem of which physico-chemical environment may have inhibited the activities of bacteria functioned as decomposer of organic matter.

      • Preparation and Characterization of Resorbable Bacterial Cellulose Membranes Treated by Electron Beam Irradiation for Guided Bone Regeneration

        <P>Bacterial cellulose (BC) is an excellent biomaterial with many medical applications. In this study, resorbable BC membranes were prepared for guided bone regeneration (GBR) using an irradiation technique for applications in the dental field. Electron beam irradiation (EI) increases biodegradation by severing the glucose bonds of BC. BC membranes irradiated at 100 kGy or 300 kGy were used to determine optimal electron beam doses. Electron beam irradiated BC membranes (EI-BCMs) were evaluated by scanning electron microscopy (SEM), attenuated total reflectance-Fourier transform infrared (ATR-FTIR) spectroscopy, thermal gravimetric analysis (TGA), and using wet tensile strength measurements. In addition, in vitro cell studies were conducted in order to confirm the cytocompatibility of EI-BCMs. Cell viabilities of NIH3T3 cells on 100k and 300k EI-BCMs (100 kGy and 300 kGy irradiated BC membranes) were significantly greater than on NI-BCMs after 3 and 7 days (<I>p</I> < 0.05). Bone regeneration by EI-BCMs and their biodegradabilities were also evaluated using in vivo rat calvarial defect models for 4 and 8 weeks. Histometric results showed 100k EI-BCMs exhibited significantly larger new bone area (NBA; %) than 300k EI-BCMs at 8 weeks after implantation (<I>p</I> < 0.05). Mechanical, chemical, and biological analyses showed EI-BCMs effectively interacted with cells and promoted bone regeneration.</P>

        인유두종 바이러스의 발암성 단백질의 발현을 저해하는 Trans Attack 항암성 유전자 백신의 개발

        목적 : 자궁경부암을 일으키는 것으로 알려진 인유두종 바이러스의 종양단백질 E6을 직접 저해하기 위하여 아데노 연관바이러스 (adeno-associated virus; AAV)의 Rep78유전자를 이용, E6의 작용을 차단함으로써 자궁경부암 세포성장의 억제에 이용하고자 하였다. 연구 방법 : 아데노 연관바이러스에서 유래된 Rep78유전자를 클로닝하여 pcDNA3.1/V5/His-Topo/Rep78벡터를 제작, 이를 자궁경부암 세포주에 이입시켰다. Rep78 유전자 이입 후 인유두종 바이러스에 의한 종양단백질 E6 mRNA의 발현을 보았으며 자궁경부암 세포성장 억제를 측정하였다. 결과 : CaSki, SiHa 세포주에서 Rep78 벡터에 의한 이입율은 30-60% 이었으며, 이입 24시간 후 제일 높은 발현이 나타났다. Rep78의 발현 후 세포 성장억제 실험 결과 자궁경부암 CaSki와 HeLa에서는 4일째에 세포성장을 억제하는 것을 보였으나, 그 외의 자궁경부암 세포주에서는 뚜렷한 차이를 보이지 않았다. 결론 : Rep78에 의한 세포내의 높은 이입율과 발현에도 불구하고 자궁경부암 세포주의 성장저해 효과는 나타나지 않았다. 아데노 연관바이러스를 이용한 자궁경부암의 치료에 있어서 Rep78의 발현을 오랜 시간 동안 지속시키는 전략이 필요하다. Objective : Adeno-associated virus Rep 78 protein is known to inhibit the promoter site of several onco-genes and viral gene, including the human papillomavirus type 16 E6 transforming genes. In this study, we investigated AAV Rep 78 mediated inhibition of HPV 16 E6 promotor activity. Methods : pcDNA3.1/V5/His-topo vector, cloned by AAV mediated Rep 78, is transfected into cervical cancer cell line (Caski). After that, we confirmed HPV 16 derived E6 expression and cell growth inhibition. Results : Transfection rate of Rep78 GFP-vector, approximately from 30 to 60 per-cent, is highly expressed at first day. But E6 expression is lower at this day. The growth of CaSki and HeLa cervical cancer cell lines was inhibited by Rep78 (p<0.05). But, the other cervical cancer cells lines was inhibited by Rep78 (p<0.05). But, the other cervical cancer cells were unaffected by Rep78 transfection. Conclusion : In spite of the high Rep78 transfection efficiency and expression rate, we could not show the cervical cancer cell growth inhibition. In our data, long term expression of Rep78 strategy is needed for cervical carcinoma gene therapy using adeno-associated virus vector.

      • A case of sarcoidosis associated with nontuberculous mycobacteria infection

        Sarcoidosis is a multisystem granulomatous disease of unknown cause. The lung is the most commonly affected organ, but the skin is frequently involved. Its etiology is unknown, though a range of factors including tuberculous infection, beryllium exposure and cold climate have been implicated in the pathogenesis of the condition. Both Mycobacterium tuberculosis and nontuberculous mycobacteria (NTM) have been implicated in sarcoidosis pathogenesis. Despite this, the role of mycobacteria in sarcoidosis remains unclear. A 45-year-old female presented with a 4-month history of an asymptomatic eye lid swelling and multiple erythematous papules on the neck. Computed tomography image showed a hypertrophy in bilateral lacrimal glands. Histological examination of the neck, orbital soft tissue and lacrimal gland revealed non-necrotizing granuloma consistent with sarcoidosis. Zielh-Neelsen stains and polymerase chain reactions (PCRs) for tuberculosis (TB) in skin biopsy specimens were negative. But NTM PCR was positive in the orbital soft tissue specimen. The patient was referred to rheumatologist and was finally diagnosed with sarcoidosis associated with NTM. Treatment with systemic steroid and hydroxychloroquine was started, resulting in an improvement of skin lesions. We herein report a case of sarcoidosis associated with NTM infection with review of the literature, as little is known regarding the role of mycobacteria in sarcoidosis.

      • A case of solitary mycosis fungoides on the scrotum

        Mycosis fungoides (MF) is the most common cutaneous T-cell lymphoma (CTCL), accounting for almost 50% of all primary cutaneous lymphomas. The occurrence of solitary lesions, which are clinically and histopathologically indistinguishable from classic MF has been described. Clinically these solitary lesions present as a single, erythematous scaly patch, plaque, or nodule that vary in size and location. The lesions are usually found in body regions typical for classic MF, mainly none exposed areas. To our knowledge, only few cases have been reported as solitary MF in the genitalia. A 54-year-old man visited our clinic with a 5-6mm sized skin colored nodule on the scrotum which had found 2 months ago. There were no other skin lesions. A punch biopsy specimen revealed a prominent infiltrate of atypical lymphocytes in dermis with some epidermotropism and lining up of atypical lymphocytes along the junctional zone. Molecular analysis revealed a monoclonal rearrangement of the T-cell receptor (TCR) genes. The patient was diagnosed with solitary MF and referred to another hospital. We herein report a case of solitary MF on the scrotum which is an unusual area with review of the literature.

        위암화과정에서 Helicobacter pylori 감염이 위점막 세포 역동학과 아포토시스 관련 단백질 발현에 미치는 영향

        Background/Aims: We investigated apoptosis, cell proliferation and the expression of apoptosis-related proteins in the gastric epithelial cells of Helicobacter pylori (H. pylori)-negative and positive patients. Methods: TUNEL staining, immunohistochemical staining for Ki-67, p53, Bcl-2, Bcl-XL and Bax were performed on paraffin-embedded specimens obtained from 11 H. pylori-negative controls, 20 H. pylori-positive chronic gastritis, 10 chronic gastritis with intestinal metaplasia (IM), 11 adenoma, and 7 adenocarcinoma patients. Apoptosis and proliferation were expressed as apoptosis index (AI) and proliferation index (PI). Results: In H. pylori-positive groups, apoptosis and proliferation were increased compared with controls. However, an AI/PI ratio was highest in chronic gastritis cases and then gradually reduced according to the progress from IM to cancer. p53 was expressed in 28.6% of adenocarcinoma cases. Expression of Bax was significantly increased in chronic gastritis and IM patients compared with controls. Conclusions: H. pylori infection induces marked increase in apoptosis and proliferation of gastric mucosa. In the premalignant lesions, the balance between apoptosis and proliferation is altered, and this imbalance may play a key role in the gastric carcinogenesis. (Korean J Gastroenterol 2003;42:12-19)
