      • GIS를 이용한 부여 서부지역의 산사태 연구

        지구정보시스템(GIS)을 이용하여 충청남도 부여군 서부지역의 산사태 발생 가능성을 분석하였다. 산사태 취약성 분석을 위하여 산사태 발생에 영향을 미치는 사면경사, 토양침식율, 지질, 토지이용도 등과 같은 환경지질학적 요인들을 선정한 후 산사태 발생지역에 대한 통계분석을 행하였다. 분석 결과를 이용하여 database를 구축하고 각각의 자료들을 중첩시켜 산사태 취약성 지수(LSI)를 산출한 다음 이 지수와 분포 면적에 대한 누적곡선을 작성하여 산사태 취약성도를 작성하였다. 산사태 취약성은 연구지역 남쪽과 북쪽 경계부, 그리고 은산면 중앙부 지역에 매우 높게 나타났으며, 은산면 북서쪽과 내산면 일대는 비교적 높은 산사태 취약성을 보였다. 그러나 규암면 일대와 구룡면, 옥산면을 잇는 수계하천을 따라서는 산사태 취약성이 매우 낮게 나타났다. This study used GIS to analyze the possibility of landslide occurrence in the western part of Puyeo-kun, Chung Nam Province. For the regional analysis of landslide, environmental and geological factors such as topographic slope, soil erodibility, geology, landuse which have influence on the landslide occurrence were extracted, and then analyzed statistically for the area of landslide occurrence. With the result obtained by statistical analysis, databases were constructed. LSI were calculated by overlapping each factor. Landslide susceptibility map was obtained by use of LSI and cumulative curve of area. The results of this study show that each border of the south and north regions and the center of Eunsan-myeon have the very high susceptibility and northwest of Eunsan-myeon and side area of drainage between Gurong-myeon and Oksan-myeon have the very low landslide susceptibility.

      • 한약제형 선호도에 관한 설문조사

        In order to investigate to develop the new dosage form of the traditional herbal medicine which have been used for the treatment of disease in oriental medicine, we conducted a survey on 217 patients who visited oriental hospital in Kyung Hee University Medical Centre during one month since October 2003. They were given questionnaires to answer and results of this study were as the following: 1. About 45% patients visited in oriental hospital believed that oriental medicine was better remedy than modern medicine. 2. Most of patients felt that the oriental medical expenses were expensive. 3. About 60% patients preferred the modernized dosage form such as extract powder and granule, etc. rather than medicinal herbs in package. 4. Most of patients hope that decoction of medicinal herbs in package would change the advanced dosage forms such as tablet, capsule and extract powder, Because patients suffered from inconvenience of making a decoction, unhandy, unwieldy and keeping. 5. About 60% patients pointed out that merits of advanced dosage forms of medicinal herbs were convience to take and handy, etc.. However, they had an Intension that there were some difference of efficacy between decoction and modernized dosage forms of oriental medicine. Because of several defects such as unusual flavor and smell, incompatible food, large volume with amount, inconvenience of carrying and decocting oriental herbal medicines, most of patients would expect the development of the advanced dosage forms of them. In summary, this study show that it was needed to develop the new dosage forms of oriental herbal medicine on the basis of scientific research and economical cost of medical expenses.

      • 만성조현병 환자가 지각한 가족의미에 대한 현상학적 연구 : 부산지역 정신의료기관 입원환자를 중심으로

        This study aimed to understand the meaning of the family as a subjective voice of the patients with chronic schizophrenia through the phenomenological method. In this study, 13 patients (7 males and 6 females), who were hospitalized in a mental institution in busan, were selected as participants in terms of a purposive sampling. The analysis method of Colaizzi was used for the study. The research process was conducted 1-2 times by indepth interviews and focus group interviews from August to October, 2017. Through the analysis of the research data, the main meanings of family perceived by the partcipants were integrated into 10 major semantic units, and fimallu categorized into 4 components. The 4 components were identified as an 'emotional supporter', a 'disconnected being', a 'beloved being', and an 'economic supporter'. Based on the results of this study, this study discussed the social support for family members of parients with chronic schizophrenia and rehabilitation of the patients themselves. 이 연구는 만성조현병 환자 당사자의 주체적인 목소리로 가족에 대한 의미와 본질을 이해하고 그 경험적 구조를 탐색하고자 하는 것이 목적이다. 연구참여자는 부산 지역 정신의료기관에서 입원 중인 만성조현병 환자 13명(남 7명 여 6명)을 의도적 표집으로 선정하였으며, 2017년 8월부터 10월까지 심층면담과 포커스면담을 병행하여 자료를 수집하여다. 결과분석은 Colaizzi 분석방법으로 이루어졌으며, 만성조현병 환자가 지각한 가족의 의미에 대한 주요 의미단위는 10개로 통합되었고, 최종적으로 '정서적 지지자', '단절된 존재', '경제적 지지자' 등 4개의 구성요소로 분석되었다. 이 연구를 통해 만성조현병 환자의 사회복귀와 그 가족에 대한 지원 대책을 논의하였다.

        The ultimate goal of periodontal therapy is to promote the regeneration of lost periodontal tissue, there have been many attempts to develop a method to achieve this goal, but none of them was completely successful. The purpose of this study was to evaluate the effects of BBP?? on alkaline phosphatase(ALP) activity in human fetal osteoblast (hFOB1). The results of this study were as follows, BBP?? didn't show any significant increased cell proliferation and cell activity in the human fetal osteoblast as compared to non-supplemented controls at 2 and 4 day. In ALP activity, 100 ㎍/㎖ BBP?? treated group showed significantly increased value than negative control group.

        Gintonin facilitates catecholamine secretion from the perfused adrenal medulla

        The present study was designed to investigate the characteristics of gintonin, one of components isolated from Korean Ginseng on secretion of catecholamines (CA) from the isolated perfused model of rat adrenal gland and to clarify its mechanism of action. Gintonin (1 to 30 μg/ml), perfused into an adrenal vein, markedly increased the CA secretion from the perfused rat adrenal medulla in a dose-dependent fashion. The gintonin-evoked CA secretion was greatly inhibited in the presence of chlorisondamine (1 μM, an autonomic ganglionic bloker), pirenzepine (2 μM, a muscarinic M<sub>1</sub> receptor antagonist), Ki14625 (10 μM, an LPA<sub>1/3</sub> receptor antagonist), amiloride (1 mM, an inhibitor of Na<sup>+</sup>/Ca<sup>2+</sup> exchanger), a nicardipine (1 μM, a voltage-dependent Ca<sup>2+</sup> channel blocker), TMB-8 (1 μM, an intracellular Ca<sup>2+ </sup>antagonist), and perfusion of Ca<sup>2+</sup>-free Krebs solution with 5mM EGTA (a Ca<sup>2+</sup>chelater), while was not affected by sodium nitroprusside (100 μM, a nitrosovasodialtor). Interestingly, LPA (0.3~3 μM, an LPA receptor agonist) also dose-dependently enhanced the CA secretion from the adrenal medulla, but this facilitatory effect of LPA was greatly inhibited in the presence of Ki 14625 (10 μM). Moreover, acetylcholine (AC)-evoked CA secretion was greatly potentiated during the perfusion of gintonin (3 μg/ml). Taken together, these results demonstrate the first evidence that gintonin increases the CA secretion from the perfused rat adrenal medulla in a dose-dependent fashion. This facilitatory effect of gintonin seems to be associated with activation of LPA- and cholinergic-receptors, which are relevant to the cytoplasmic Ca<sup>2+</sup> increase by stimulation of the Ca<sup>2+</sup> influx as well as by the inhibition of Ca<sup>2+</sup> uptake into the cytoplasmic Ca<sup>2+</sup> stores, without the increased nitric oxide (NO). Based on these results, it is thought that gintonin, one of ginseng components, can elevate the CA secretion from adrenal medulla by regulating the Ca<sup>2+</sup> mobilization for exocytosis, suggesting facilitation of cardiovascular system. Also, these findings show that gintonin might be at least one of ginseng-induced hypertensive components.

        영구치 치수 기질세포를 이용한 연골 분화 및 분화 시기에 따른 형태학적 변화

        Objectives: The aim was to confirm the stem cell-like properties of the dental pulp stromal cells and to evaluate the morphologic changes during in vitro chondrogenesis. Materials and Methods: Stromal cells were outgrown from the dental pulp tissue of the premolars. Surface markers were investigated and cell proliferation rate was compared to other mesenchymal stem cells. Multipotency of the pulp cells was confirmed by inducing osteogenesis, adipogenesis and chondrogenesis. The morphologic changes in the chondrogenic pellet during the 21 day of induction were evaluated under light microscope and transmission electron microscope. TUNEL assay was used to evaluate apoptosis within the chondrogenic pellets. Results: Pulp cells were CD90, 105 positive and CD31, 34 negative. They showed similar proliferation rate to other stem cells. Pulp cells differentiated to osteogenic, adipogenic and chondrogenic tissues. During chondrogenesis, 3-dimensional pellet was created with multi-layers, hypertrophic chondrocyte-like cells and cartilage-like extracellular matrix. However, cell morphology became irregular and apoptotic cells were increased after 7 day of chondrogenic induction. Conclusions: Pulp cells indicated mesenchymal stem cell-like characteristics. During the in vitro chondrogenesis, cellular activity was superior during the earlier phase (within 7 day) of differentiation.

        This study proposes an optimized diffuser strake design that can improve driving performance for cars in high-speed driving mode. The speed of the vehicle is set to 160km/h, and the number of strake is set to 4, 6 and 8, and the angle of the diffuser is set to 4, 5 and 6 degrees. The arrangement of the strake is centered, edged and equally arranged. The optimized diffuser design induced with the Ansys Fluent analysis shows a drag coefficient of 16% less than the basic diffuser-free model at a diffuser angle of 6 degree, 8 strakes, and equal arrangement.

        Detection of Antibodies against Orientia tsutsugamushi Sca Proteins in Scrub Typhus Patients and Genetic Variation of<i>sca</i>Genes of Different Strains

        <B>ABSTRACT</B><P>Scrub typhus, caused byOrientia tsutsugamushiinfection, is one of the main causes of acute febrile illness in the Asian-Pacific region. Although early diagnosis and immediate antibiotic treatment are critical for reducing disease severity and mortality, current diagnostic methods using serological and molecular approaches have some limitations in sensitivity and applicability in clinical laboratories. In this study, we identified and characterizedO. tsutsugamushisurface cell antigen (<I>sca</I>) family genes encoding autotransporter proteins in order to test them as novel diagnostic targets. We evaluated antibody responses against the Sca proteins in scrub typhus patient sera and examined the genetic diversity of these genes in different strains after PCR amplification. Specific antibody responses against ScaA and ScaC were observed in patients with high indirect immunofluorescence assay titers (≥1:640), whereas specific responses against ScaB and ScaE were relatively low. Genetic analysis using genomic DNAs revealed the<I>sca</I>genes to be quite variable among the different strains. In contrast to<I>scaA</I>,<I>scaC</I>, and<I>scaD</I>, which were detected in all of the tested strains,<I>scaB</I>and<I>scaE</I>were amplified differentially from the different strains, suggesting a differential presence of the genes in the genomes. Among the members of the gene family, the sequence of<I>scaC</I>is the most highly conserved between the different strains, and the size of<I>scaD</I>is the most variable due to the presence of different numbers of internal repeat sequences. These results suggest that the<I>sca</I>genes ofO. tsutsugamushimay be valuable targets for use in combination with classical assay methods for scrub typhus diagnosis.</P>

      • Production of an antibody for treatment of rheumatoid arthritis using plants

        Rheumatoid arthritis, which is known as an autoimmune phenomenon as the main mechanism, is a chronic inflammatory disease that causes inflammation in several joints, such as the hands, wrists, feet, and ankles. Humira, a monoclonal antibody, recognizes tumor necrosis factor-alpha (TNF-α) and is used to treat rheumatoid arthritis. Humira is mostly produced in animal cell-based production systems that have high investment costs, high production costs, and risk of contamination. In this study, Humira was produced using plants to overcome the problems of the existing production system. In order to improve the efficacy of the antibody and reduce side effects, glycoengineered plants without plant-specific residues developed in previous studies were used. In addition, in order to solve the low expression level of the recombinant protein, a vector containing the antibody gene was constructed using a virus-based expression system in which expression is regulated by an inducible promoter. The expression vector was introduced into Agrobacterium to transform wild-type plants and glycoengineered plants, and the transformed plants were selected using an antibiotic-containing medium and a reporter gene. The expression of the antibody expressed in the plant was confirmed using an antibody specific to the protein extracted from the leaf. It was identified at a size of approximately 50 kDa and 27 kDa, respectively, representing the heavy and light chains of the antibody. Single copy plants were selected through the segregation test, and homozygous lines were established through the selection process. The expression level of the recombinant protein by the inducible promoter was compared according to the concentration and time of the inducer. These results show the potential for lower production costs and safer production of rheumatoid arthritis drugs using plants.
