        Lactobacillus helveticus CU 631 에 의한 Helicobacter pylon 의 Urease 및 공포 생성 독소 억제활성

        표준균주 혹은 유제품으로부터 분리된 Lactobacillus spp.와 Bifidobacterium spp. 32균주를 사용하여 H. pylori 생장을 현저하게 억제하는 L. helveticus CU631을 선발하고, urease와 공포생성 독소의 활성을 억제하는 효과를 측정하여 다음의 결과를 얻었다. L. helveticus CU631의 억제대의 직경이 10.0±1.5㎜ 나타내어 가장 강력한 생장 억제 능력을 보였으며 L. plantarum과 L. fermentum은 직경 4.0㎜ 내외의 억제대를 나타내어 비교적 약한 억제 활성을 보였으며 Bifidobacterium spp.에서 억제 활성을 보이지 않았다. L. helveticus CU631의 배양액과 배양 상층액 모두, H. pylori NCTC11637의 urese 억제 활성을 나타내었다. L. helveticus CU631를 H. pylori G88016를 같이 배양했을시 공포생성 독소의 역가가 50%로 감소하였으며 L. helvesticus CU631의 배양 상층액과 H. pylori G88016의 배양 상층액을 5:5와 6:4 비율로 혼합하였을 때 억제 활성이 나타났다. The inhibitory effects of 32 strains of lactobacilli against Helicobacter. pylori were determined and Lactobacillus. helveticus CU631 has been selected as the strain which possessed the strongest inhibitory effect against H. pylori NCTC11637 in inhibition zone test showing inhibition zone with the average diameter of 10±1.5㎜, whereas Lactobacillus. plantarum and L. fermentum made inhibition zone with the average diameter of 4.0㎜, H. pylori G88016 revealed the highest vacuolating toxin activity among the 8 strains of H. pylori, which showed positive reaction of vacuolating toxin gene in PCR amplification test. Both L. helveticus CU631 and cell free culture supernatant had a strong inhibitory activity on the urease activity of H. pylori NCTC11637. The inhibitory activity of L. helveticus CU631 on the vacuolating toxin activity of H. pylori manifested in the co-culture of two strains and in the 5:5 mixture of supernatant of the two strains.

        한우 어미소와 송아지의 행동 특성

        This study was performed to investigate the behavioral characteristics of Hanwoo(Bos taurus coreanae) cows and their calves. Four cows and their calves were placed in pens. The behaviors of each cow and calf were recorded using a time lapse VCR from 07:00 h to 18:00 h for 6 days and were analyzed using the scan point sampling method. The cow's behaviors in order of frequency were LD(lying down, 25%), ST(standing, 23%), EA(eating, 13%), LR(lying down rumination, 10%) and SR(standing rumination, 7%). The cows spent less than 3% of their time PG(pairwise grooming) and SF(sniffing). The calves mainly showed LD(52%), ST(28%), LF(lying flat, 4%), EA(3%) and WA(walking, 3%). The calves spent less than 2% of their time SF(sniffing) and SUK(sucking). The behavioral patterns of cows and calves at 07:00 h, 12:00 h and 18:00 h were different, especially in ST and LD.

      • A noncanonical E-box enhancer drives mouse Period2 circadian oscillations in vivo

        <P>The mouse Period2 (mPer2) locus is an essential negative-feedback element of the mammalian circadian-clock mechanism. Recent work has shown that mPer2 circadian gene expression persists in both central and peripheral tissues. Here, we analyze the mouse mPer2 promoter and identify a circadian enhancer (E2) with a noncanonical 5'-CACGTT-3' E-box located 20 bp upstream of the mPer2 transcription start site. The E2 enhancer accounts for most circadian transcriptional drive of the mPer2 locus by CLOCK:BMAL1, is a major site of DNaseI hypersensitivity in this region, and is constitutively bound by a transcriptional complex containing the CLOCK protein. Importantly, the E2 enhancer is sufficient to drive self-sustained circadian rhythms of luciferase activity in central and peripheral tissues from mPer2-E2::Luciferase transgenic mice with tissue-specific phase and period characteristics. Last, genetic analysis with mutations in Clock and Bmal1 shows that the E2 enhancer is a target of CLOCK and BMAL1 in vivo.</P>

        Obesity phenotype and cardiovascular changes

        OBJECTIVE: Healthy obese phenotype with favorable metabolic profiles is proposed. However, whether healthy obesity leads to target organ changes is controversial. We investigated the impact of a healthy obesity on cardiovascular structure and function. METHODS: A total of 2540 participants without known cardiovascular disease were enrolled. According to BMI and the metabolic syndrome (MetS) component, the participants were divided into six groups: healthy (none of five MetS components) normal weight (BMI <23 kg/m), unhealthy (one or more of five MetS components) normal weight, healthy overweight (BMI 23–24.9 kg/m), unhealthy overweight, healthy obesity (BMI ≥25 kg/m), and unhealthy obesity. The cardiovascular changes were assessed by echocardiography, tissue Doppler imaging (TDI), carotid ultrasonography, and pulse wave velocity (PWV). RESULTS: In a multivariate analysis after adjusting for age, sex, heart rate, high-sensitivity C-reactive protein, and medication for hypertension and diabetes mellitus, the unhealthy overweight and obese groups showed statistically significant changes in the left ventricular mass index, mitral E/A ratio, E/Ea ratio, TDI Ea velocity, common carotid artery intima–media thickness (CCA-IMT), and brachial-ankle PWV (P < 0.001), compared with the healthy normal weight individuals. In the healthy overweight and obese groups, CCA-IMT and brachial-ankle PWV values were similar, but left-ventricular mass index and TDI Ea velocity were significantly different (P < 0.001). CONCLUSION: Healthy obesity was associated with subtle changes in left ventricular structure and function. These data provide evidence that metabolically healthy phenotypes with excess weight may not be a benign condition.

        Search for lepton flavor and lepton number violating <i>τ</i> decays into a lepton and two charged mesons

        <P><B>Abstract</B></P><P>We search for lepton flavor and lepton number violating <I>τ</I> decays into a lepton (ℓ=electron or muon) and two charged mesons (h,<SUP>h′</SUP>=<SUP>π±</SUP> or <SUP>K±</SUP>), <SUP>τ−</SUP>→<SUP>ℓ−</SUP><SUP>h+</SUP><SUP>h′−</SUP> and <SUP>τ−</SUP>→<SUP>ℓ+</SUP><SUP>h−</SUP><SUP>h′−</SUP>, using 671 fb<SUP>−1</SUP> of data collected with the Belle detector at the KEKB asymmetric-energy <SUP>e+</SUP><SUP>e−</SUP> collider. We obtain 90% C.L. upper limits on the branching fractions in the range (4.4–8.8)×<SUP>10−8</SUP> for τ→eh<SUP>h′</SUP>, and (3.3–16)×<SUP>10−8</SUP> for τ→μh<SUP>h′</SUP> processes. These results improve upon previously published upper limits by factors between 1.6 to 8.8.</P>

        Characterization of Interface States in MOS Systems by Using Photonic High-Frequency Capacitance-Voltage Responses

        Based on the photonic high-frequency capacitance-voltage response of Metal-Oxide-Semiconductor capacitors, we report an improved characterization method for the analyzing of interface states in MOS systems. An optical source with a photonic energy $E_{ph}$ = 0.943 eV ($\lambda$ = 1314.5 nm) is employed for photonic deep-depletion (fast sweep rate) high-frequency Capacitance-Voltage (photonic DD HF-CV) characterization of interface states distributed in the photo-responsive energy band. Using the photonic DD HF-CV characterization, we obtained a U-shaped distribution of $D_{it}$ over ($E_V + E_g/2 - q\phi_f) < E_t < (E_V + E_g/2 - q\phi_f + q\phi_S$) for N-type Metal-Oxide-Semiconductor capacitors and ($E_C - E_g/2 - q\phi_f + q\phi_S) < E_t < (E_C - E_g/2 - q\phi_f$) for P-type Metal-Oxide-Semiconductor capacitors.

        Association and functional relevance of E237G, a polymorphism of the high-affinity immunoglobulin E-receptor β chain gene, to airway hyper-responsiveness

        <P>Summary</P><P>Background</P><P>The hyper-sensitivity reaction of IgE, with its high-affinity receptors (FcϵRI), is central to the phenomenon of atopic diseases.</P><P>Objective</P><P>To evaluate the genetic effects of non-synonymous single-nucleotide polymorphisms (SNPs) of FcϵRI on intermediate phenotypes of asthma, i.e. atopy and airway hyper-responsiveness (AHR), in the Korean general population.</P><P>Subjects and methods</P><P>Atopy and AHR were evaluated in a cohort of 2055 subjects, aged 10–18 years, using skin prick tests (SPTs) for common aeroallergens and total serum IgE and methacholine bronchial provocation tests. All FcϵRI-α, FcϵRI-β, and FcϵRI-γ gene exons of 24 healthy subjects were sequenced to locate informative non-synonymous SNPs (minor allele frequency >2%). Informative SNPs were then scored, using the high-throughput single base extension method. Relative risk (RR) was determined by multiple logistic regression analysis, after adjusting for confounding factors. The functional relevance of non-synonymous SNPs was analysed using the sorting intolerant from tolerant (SIFT) program.</P><P>Results</P><P>The SNP search found only one informative non-synonymous SNP in FcϵRI-β: E237G (minor allele frequency=0.21). The positive rate of AHR was lower among subjects with the 237<SUP>*</SUP>E allele than among those with 237<SUP>*</SUP>G [RR (95% confidence interval)=0.41 (0.19–0.89); <I>P</I>=0.01]. However, the E237G substitution was not associated with either a positive SPT response or total serum IgE levels. Sequence evolution analysis predicted that the E237G variation is an intolerant amino acid substitution, with functional importance.</P><P>Conclusion</P><P>In the Korean general population, AHR is significantly associated with the E237G polymorphism of FcϵRI-β, which results in an intolerant amino acid substitution.</P>

        Genotoxicity evaluation of electromagnetic fields generated by 835-MHz mobile phone frequency band

        It is still unclear whether the exposure to electromagnetic fields (EMFs) generated by mobile phone radiation is directly linked to cancer. We examined the biological effects of an EMF at 835 MHz, the most widely used communication frequency band in Korean CDMA mobile phone networks, on bacterial reverse mutation (Ames assay) and DNA stability (in vitro DNA degradation). In the Ames assay, tester strains alone or combined with positive mutagen were applied in an artificial mobile phone frequency EMF generator with continuous waveform at a specific absorption rate (SAR) of 4 W/kg for 48 h. In the presence of the 835-MHz EMF radiation, incubation with positive mutagen 4-nitroquinoline-1-oxide and cumene hydroxide further increased the mutation rate in Escherichia coli WP2 and TA102, respectively, while the contrary results in Salmonella typhimurium TA98 and TA1535 treated with 4-nitroquinoline-1-oxide and sodium azide, respectively, were shown as antimutagenic. However, these mutagenic or co-mutagenic effects of 835-MHz radiation were not significantly repeated in other relevant strains with same mutation type. In the DNA degradation test, the exposure to 835-MHz EMF did not change the rate of degradation observed using plasmid pBluescript SK(+) as an indicator. Thus, we suggest that 835-MHz EMF under the conditions of our study neither affected the reverse mutation frequency nor accelerated DNA degradation in vitro.

      • Improvement of the phosphoenolpyruvate carboxylase activity of Phaeodactylum tricornutum PEPCase 1 through protein engineering

        In order to mitigate CO<SUB>2</SUB> accumulation and decrease the rate of global warming and climate change, we previously presented a strategy for the development of an efficient CO<SUB>2</SUB> capture and utilization system. The system employs two recombinant enzymes, carbonic anhydrase and phosphoenolpyruvate carboxylase, which were originated from microalgae. Although utilization of this integrated system would require a large quantity of high quality PEPCase protein, such quantities could be produced by increasing the solubility of the Phaeodactylum tricornutum PEPCase 1 (PtPEPCase 1) protein in the Escherichia coli heterologous expression system. We first expressed the putative mitochondria targeting peptide- and chloroplast transit peptide-truncated proteins of PtPEPCase 1, mPtPEPCase 1 and cPtPEPCase 1, respectively, in E. coli. After affinity chromatography, the amount of purified PEPCase protein from 500mL of E. coli culture was greatest for cPtPEPCase 1 (1.99mg), followed by mPtPEPCase 1 (0.82mg) and PtPEPCase 1 (0.61mg). Furthermore, the enzymatic activity of mPtPEPCase 1 and cPtPEPCase 1 showed approximately 1.6-fold (32.19 units/mg) and 3-fold (59.48 units/mg) increases, respectively. Therefore, cPtPEPCase 1 purified using the E. coli heterogeneous expression system could be a strong candidate for a platform technology to capture CO<SUB>2</SUB> and produce value-added four-carbon platform chemicals.

        For the measurement on the fat content in University Students, an intensive examination was conducted on a total of 3,017 Students (male 2,566, female 451), from 16 to 25 years of age. The subjects were all students who had passed the written entrance examination for Seoul National University. Total Body Fat amount was measurement by skin-fold thickness with a skin-fold caliper (Manufactured by Cambridge Scientific Industrial Inc. U.S.A.) at four sites of the body, and it was calculated by Kim' s5,11, 25) and Lees1) regression equation. As a result of this survey, the following conclusions were obtained; 1. Average body height by Anthropometric was male 167.66cm and female 156.02cm. Average body weight by Anthropometric was male 54.601kg and female 48.23kg. 2. The mean -skin-fold thickness measured with a lange skin-fold caliper was; at the Sub-scapular: male 10.33mm and female 14.15mm, Abdominal: male 8.68mm and female 14.14mm, Lumbar: male 8.83mm and 13.09mm, Upper arm: male 5.83mm and female 11.94mm, Total average: male 8.43mm and female 13.33mm, in all cases the female measurements were higher than male. 3. Data on body fat amounts according to body fat weight regression equation were; In male: Fat Weight=9.41kg, Fat Free Weight=45.19kg, Fat Weight/Fat Free Weight(%)=20.82%, Fat Weight/Total Body Mass(%)=23.28%(Body-fat amount %). In female: Fat Weight=11.23kg, Fat Free Weight=37.00kg, Fat Weight/Fat Free Weight(%).=30.351, Fat Weight/Total Body Mass(%)=23.28%. The female's value was higher than the males.
