        Transmission ability of Zika virus with artificially infected Aedes albopictus in Korea

        Zika virus (ZIKV), a virus mainly occurring in South America, is now globally distributed. Understanding the pathogen transmission cycle in a vector is exceptionally important in developing disease control strategies. In this study, we performed ZIKV microinjection in Aedes albopictus (Skuese), to estimate its vertical and horizontal transmission ability. The virus infection rate was confirmed by real-time qPCR in the F1 generation derived from the ZIKV-injected Ae. albopictus. The minimum infection rate (MIR) of the F1 generation was highest when they were injected with the virus at 6–12 h (MIR: 16.1) and lowest at 12–24 h (MIR: 4.7) after feeding. In the developmental stage of the F1 progeny, MIR values were 1.0, 1.3, and 6.7 in each egg, larval, and adult stage, respectively, but ZIKV was not detected in the pupa stage. Virus transmission ability was not significantly different between the collection areas (Tongyeong and Jeju). Ae. albopictus demonstrated a high venereal transmission rate of ZIKV and was detected in males (6 pool/7 pool) and females (6 pool/7 pool), confirming that ZIKV can be transmitted from infected mosquitoes to uninfected mosquitoes via mating. This is the first study of the administration of a ZIKV microinjection to Ae. albopictus in Korea and suggests a possibility of a potential mechanism for the virus to survive during adverse conditions via vertical transmission.

        Fungi are well suited for studying mechanism controlling development and cell differentiation in multicellular eukaryotes. The asexual and sexual reproductive cycles are tightly coordinated during the life cycle of Aspergillus nidulans. The asexual (mitotic) reproductive cycle involves the formation of a number of differentiated cell types. Successful formation of multicellular reproductive structures and spores (conidia) requires the establishment of spatiotemporal gradients in essential transcriptional regulators, such as brlA, abaA and wetA. Altered gene expression is coordinated with critical changes in the movement of nuclei and the regulation of the cell division cycle. Therefore, it has been proposed to define a central regulatory pathway. Several early regulatory genes involved in signal transduction pathway controlling asexual sporulation are also required for activating the central pathway. Sexual (meiotic) reproduction is more complex, requiring the differentiation of several specialized tissue types. Although technical difficulties have limited efforts to study the genetics of sexual fruiting-body formation directly, some recent progress has been made. With the recent availability of mutants blocked in sexual development, understanding the genetic interactions between genes that function primarily in determining sexual sporulation will be achieved in the near future.

      • 프리미돈의 한국인에서의 생체이용율시험

        A reversed phase HPLC method was developed and validated for the determination of primidone in human plasma. Propylthiouracil was used as an internal standard. Calibration curves were linear in the concentration range of 10-120 ng/㎖. The coefficient of variation of the intra- and inter-day precision were below 15%. The coefficient of variation of the accuracy were below 15% in the concentration range investigated. A bioavailability study was performed using the validated HPLC method. Twenty four healthy human male volunteers were orally administered 20 mg of primidone. The pharmacokinetic parameters were calculated using WinNonlin. The mean values of AUC_(LAST) was 85.0±17.7 ㎍ hr/㎖, C_(max) was 4.22±1.04 ㎍/㎖, T_(max) was 1.85±1.13 hr, t_(1/2) was 17.7±2.4 hr. The pharmacokinetic parameters and the HPLC method can be used for the desigm of bioequivalence study of primidone.

        전시를 위한 토기와 청자의 복원 : 토기기대, 노형기대, 청자사이호를 중심으로

        국립김해박물관 개관전시 유물인 가야시대 土器 器臺 2점과 중앙박물관의 특별전 “고대국가의 형성” 전시회 출품 유물인 靑磁四耳壺에 대한 보존처리를 수행하였다. 육안 및 실체현미경 관찰을 통하여 처리 전 상태, 文樣과 製作技法 등을 파악하였고 이를 토대로 각 유물의 특성에 맞는 보존처리를 실시하였다. 기대는 결실부분이 커 에폭시系 복원재료를 이용해 파손된 토기 面과 직접 접착시켜 형태를 복원하고 문양과 색, 질감을 類似하게 처리하였다. 청자는 토기와 같은 복원재를 이용했지만 파손된 청자 면과 分離시켜 형태를 복원하여 해체가 가능하도록 조치한 다음 색과 질감도 구분이 되도록 처리하였다. Conservation treatment was done for the pottery-stand of Gaya period which was to be exhibited in celebration of the opening of Gimhae National Museum and for the jar of Chinese celadon which was to be exhibited in Korea National Museums special exhibition "Formation of Ancient States". Through the examinations by naked eyes and under the microscope, condition before treatment, patterns and manufacturing techniques of the objects were observed. According to these examinations, conservation treatments, suitable for each object was finished. As the pottery-stand was damaged severely, the epoxy resin was pasted directly on the broken surface of the pottery to restore the original shape and pattern, color and feeling similar to original state were given to the restored area. Although same restoration material was applied on the celadon, it was not pasted directly on the broken surface of the celadon so that the restored area could be dismantled, and color and feeling were also treated somewhat differently.

      • 계룡산 구룡사지 당간지주의 기원암, 암석학적 및 보존과학적 진단

        구룡사지 당간지주의 암석학적 풍화와 훼손를 진단하였으며, 보존처리를 대비한 동일 종류의 기원암을 추적하였다. 이 당간지주는 홍장석 화강암으로 구성되어 있으며 주좌는 흑운모 화강암이다. 왼쪽 지주는 완전히 파괴되어 방치되어 있으며, 오른쪽 지주는 하나의 부재로 구성되어 있으나 계단식으로 절단되어 있어 도괴의 우려가 있다. 이 지주들은 균열 및 탈락과 훼손이 심하여 박리와 박락현상이 발생하였고 석재의 표면은 입상분해 작용이 진행 중이다. 이차적 오염현황도 심각하여, 황갈색 수산화철에 의한 암석의 변색과 강수의 유동흔적을 따라 나타나는 암회색 철과 망간의 침전물이 산재한다. 지주의 주변과 암석의 표면에는 지의류와 선태류의 오염이 심각하며, 기단부는 토양화의 진행에 의하여 잡초가 암석의 생물학적 풍화작용을 가중시키고 있다. 따라서 부지 주변의 나무와 잡초 및 불연속면에 서식하는 부착생물을 제거하기 위한 생화학적 처리도 필요하다. 이 당간지주는 석재자체의 성능이 저하되어 있으므로, 균열부의 장기적인 모니터링과 함께 석재의 표면을 강화할 수 있는 처리방법을 강구해야 할 것이다. 특히 왼쪽 지주는 보존처리도 어려운 상태여서 교체하여야 할 상황이다. 이 당간지주와 동일한 암종은 하신리 무지개 농장 부근의 도로변 노두에 있다. The Flag Pole Supporting Rocks (Dangganjiju) of the Guryongsa temple site, Korea was studied on the basis of petrological weathering, conservational scientific diagnosis and presumed to source area of the original supporting rocks. Constituting rocks of this supporting rocks are pink feldspar granite, and the rounded basement is biotite granite. However, the left supporting rock is remained at perfectly destruction state in situ. The right supporting rock showed also highly fractured as step-like cutting plane which is very dangerous state because of possible collapse. As a whole supporting rocks occurred with fracturing, exfoliation or deterioration, and rock surface appeared to grain dissolution. Host rock of the supporting rocks were highly secondary contaminations, and most rock surface was color changed to the dark black to dark brown due to the clay, iron and manganese hydroxides along the rain water flow. In the vicinity of the supporting rocks and rock surface show partly green and black patchs because of coated by algae, lichen and moss. The lower part of the basement rocks are transition to the some weeds. This biological problems are need for cleaning and chemical treatments. The main materials of the supporting rocks is highly degradation, therefore the supporting rocks need to be long term monitoring in the fracture planes and to prepare for synthetic conservation treatments. Especially, the left supporting rock is difficult to conservation owing to nearly perfect destruction. In the Hashinri Mujigae farm area, the same kind of pink feldspar granitic rocks with this supporting rocks are occurred.

      • 親環境農業을 위한 有機農業의 作付體系 確立에 관한 硏究

        This study was carried out to produce safe agricultural product and to improve the soil environment of crop cultivation using the reasonable cropping system in organic farming to the case study of organic farmhouse in Chungnam Hongdong area for sustainable agriculture. In the ratio of farming forms in these organic farmhouses, paddy field-upland field-greenhouse form, paddy field-upland field form, crop-livestock form and only greenhouse form were 23.8%, 23.8%, 23.8% and 19.0% respectively. In the greenhouse area of organic farmhouse, the 50% of total greenhouse number were 600~1,000 pyung(200~330a) and the 40% of total were 1,000~1,500 pyung(330~500a). In the area of upland field, the 80% of total upland field number were 500~1,000 pyung(167~330a). In the cropping systems of surveyed farmhouses, the number of crop-combination patterns were twenty-seven and the 96.3% of those was combined with vegetable. Vegetable-vegetable pattern, cereal crops-vegetable pattern and root and tuber crop-vegetable pattern were 44.4%, 18.5% and 14.8% respectively. Leguminous crop combined pattern was 7.4%, but patterns combined with barley or wheat did not exist. Among the vegetable combined patterns, leaf vegetables combined pattern, fruit vegetables combined pattern and root vegetables combined pattern were 66.7%, 31.0% and 22.2% respectively. Among the vegetable-vegetable patterns, the portion of fruit vegetables-leaf vegetables pattern was the highest and that of root and tuber vegetables-root and tuber vegetables pattern was the lowest. Among the twenty-seven crop-combination patterns, unreasonable crop rotation pattern was 50% over and especially the almost crop-combination pattern of greenhouse was unreasonable to crop rotation pattern. The farmhouse that control the amount of compost application by means of periodical soil analysis did not exist in this area.

      • 직장암환자에서 복회음병합절제술(Miles' operation) 후 발생한 지속적인 배뇨장애의 치료

        Bladder dysfunction is one of the serious postoperative complications after rectal cancer surgery. To get sufficient objective data and factors for adequate management of bladder dysfunction following rectal cancer surgery, we reviewed medical records of 122 patients who had abdominoperineal resection of rectum (Miles' operation) for rectal cancer. The incidence of bladder dysfunction in 122 rectal cancer patients was 18.9 percent (23 patients). Among 23 patients with bladder dysfunction, 10 patients were referred to our rehabilitation service and their bladder was areflexic type neurogenic bladder at referred time. In 10 patients referred to our rehabilitation service, 8 patients(80.0% got the balanced bladder and 2 patients(20.0% ) failed, and the interval between beginning of bladder training and establishment of balanced bladder in 8 patient was 13.4±6.8 days. In 13 patients did not referred to our service, only 3 patients(23.1% ) got the balanced bladder and 10 patients(76.9%) failed. To improve the life quality of the rectal cancer patients with neurogenic bladder following abdominoperineal resection of rectum, early proper rehabilitative managements including bladder training were should be given.

      • 배지의 종류가 식중독세균의 열손상으로 부터 회복에 미치는 영향

        10^6 - 10^7 cells/ml of pathogenic bacteria, vibrio parahaemolyticus 10135, Staphylococcus aureus 196E and Listeria monocytogenes were exposed at 45, 50 and 60℃ for 60 minutes in tryptic soy broth(TSB). Following thermal stress, cells were allowed to recover in three kinds of recovery media for 24 hour. TSB, Flounder homogenate and Oyster homegenate were used for heat injured Vibrio parahamolyticus and Staphylococcus aureus where as TSB, Listeria enrichment broth(LEB) and Oyster homogenate for Listeria monocytogenes as recovery media. Heat injury of Vibrio parahamolyticus was larger Staphylococcus aureus or Listeria monocytogenes at 50℃. Recovery of heat injured cells of Listeria monocytogenes was laster than other strains. TSB was a good media for recovery of heat injured cells but Oyster homogenate was poor.
