        단일클론 항 γ-Glutamyltrasferase(GCT) 항체를 이용한 혈뇌장벽 내 GGT의 분포에 관한 연구

        γ-Glutamyltransferase(GGT: E.C. 2.3.2.2.) is a glycoprotein enzyme which is involved in glutathione metabolism and amino acid transport through the plasma membrane. It is distributed widely in several organs including liver, kidney, pancrease and brain. GGTs derived from the brain of Wister rats and BALB/c mice were biochemically purified to a specific activity of 4246.2, 862.1 units per mg of protein, a purification folds 93.7, 43.8 and the final yield 65.8, 44.0% respectively. Electrophoretic pattern of purified GGTs from rats and mice brain shows very similar protein fraction each other. We have produced six monoclonal antibodies(GGT-MAb 1-6) against 2-acetamidofluorene treated rat liver GGT. Using these GGT-MAb 1-6 we performed immunohistochemistry(IHC) to study the distribution of GGY isozymes in normal tissues of rat brain and in ncoplastic tissues of human brain. The results indicated that human brain GGT was localized in pericytes of blood-brain barrier, especially in the blood-rich portion of the brain(e.g. cerebellum of rat, meningioma and craniopharyngioma of human). Therefore these MAbs may be used to evaluate the distribution of GGT isozymes in different tissues.

      • Ethephon의 浸漬處理 및 圃場處理가 고추의 着色 및 主要成分에 미치는 影響

        Color enhancement, contents of red pigment and capsaicin were studied with the dipping and field treatment of various levels of ethephon concentrations; 250, 500 and 1000ppms.The results were summarized as followings: 1. Dipping treatment effectively enhanced the coloring of the unmatured stages of fruit, especially, under younger fruit age. As the ethephon concentration increased, the remarkable effects were observed in Saegochu while Saemaul-Gumjang and Cheonan were less affected. 2. The optimum ethephon concentration for color enhancement were 500 and 250ppm in dipping and field treatments, respectively. 3. Red pigment content was slightly increased in 500ppm concentration level. 4. As the fruit age increased, capsaicin contents were increased in the ethephon concentration level from 500 to 1000ppm. In younger fruit slight increment was observed in the 1000ppm treatment.

        An Ac-type Plasma Display Panel (PDP) operating with Ne-Ar(0.1%) Penning mixture gas is fabicated. The characterics of the panel with electrodes covered with thin and thick dielectric layers are studied. The brightness of the Neon-orange light emitted by the panel measured as function of applied voltage and frequency. As an application, a graphic display system equipped with PDP showing still and moving pictures is made.

      • 프로폴리스 농도가 항균활성에 미치는 영향

        The objetives of this study are to set up optimum extraction temperature, time and organic solvent for propolis extraction, to investigate chemical properties, and to develop health foods from propolis preparation. In this study, ethanol and ultrasonic extracts method performed to optimum extraction temperature was at 60, 2012, optimum extraction time was at 12, 4 hours and optimum extraction amount of solvent was at 20, 15 times of propolis weight. When various ethanol solutions were used, whereas flavonoid content was highest in 70, 80% aqueous ethanol, respectively. So the ultrasonic extracts method used gave better results than the ethanol extracts method in this work. Extraction of propolis with etanol and ultrasonic extracts method was performed by using the water and various concentrations of aqueous ethanol as solvent. Sensidvity of propolis samples to Staphylococcus aureus was invesdgated and the results were shown. Samples of water extract did not inhibit microbial growth, where as 50% aqueous ethanol extract the largest inhibitory zone for Staphylococcus aureus, then decreased inhibition with increasing ethanol concentrations.

        In this paper, a fuzzy logic controller is designed for speed control of a hydraulic inverter elevator. Mathematical modeling of an elevator actuated with hydraulic system is presented and the friction characteristics of a cylinder is examined, which may cause the abrupt increase of the acceleration in the zero-crossing speed region. Simulation results show that the proposed fuzzy logic speed controller yields a better control performance than conventional PID controller.

      • 공기 제습용 무기막의 제조 및 특성

        Recently, the humidity adjustment of indoor air is of great importance in air conditioning system as a applied, in buildings, vehicles, and containers for storage and transport of perishable products. Membrane dehumidification or air is potentially attractive because it offers low capital and operating costs, along with low energy consumption. And membrane dehumidification process attracted the attention of the public instead of the other dehumidification processes, such as adsorption, absorption, and refrigeration cycles and so on. In this study, the prepared hydrophilic inorganic membrane-based dehumidifiers (membrane air dehumidification) examined the performance of dehumidification. The surface-modified inorganic membrane prepared in this study showed high dehumidification efficiency (over 80%). Thc membrane might be very useful for dehumidification industries.

      • 베체트씨병에 동반된 복부 대동맥 동맥류의 스텐트 삽입술 1 예 : Treatment with Endovascular Stent Graft

        Behcet's disease is an inflammatory disorder of unknown cause, characterized by recurrent oral aphthous ulcer, genital ulcers, ocular manifestations, and skin lesions. Large-vessel involvement of both arterial and venous system appears in 7-29% of patients, and it is major cause of morbidity and mortality. Venous involvement, including superficial thrombophlebitis and deep vein thrombosis, is more frequent. Arterial manifestations occurs infrequently and presents with aortitis or arterial aneurysm and arterial thrombosis. Arterial aneurysms or occlusions which occur in the systemic or the pulmonary arterial bed are present in approximately 2 to 3% of patients with Behcet's disease and accounts for only 12% of vascular complication. Aneurysms are more frequent than occlusions and are mostly present in the pulmonary arteries and abdominal aorta. They have poor prognosis because of rupture with fatal outcome and surgical treatment is necessary. But, Surgical treatment often difficult and may lead to formation of further false aneurysms at the site of vascular anastomosis. Endovascular stent graft placement emerged as an alternative treatment that is less invasive with a lower risk. We experienced a case of abdominal aortic aneurysm in 36-year-old man with Behcet's disease. The patient was diagnosed by CT scan and treated with percutaneous endovascular stent graft.

      • Next-generation sequencing reveals novel resistance mechanisms and molecular heterogeneity in EGFR-mutant non-small cell lung cancer with acquired resistance to EGFR-TKIs

        <P><B>Abstract</B></P> <P><B>Objectives</B></P> <P>Despite initial responses to epidermal growth factor receptor tyrosine kinase inhibitors (EGFR-TKIs) in EGFR mutant non-small cell lung cancer, patients invariably develop acquired resistance. In this study, we performed next-generation sequencing in pre- and post-EGFR-TKI tumor samples to identify novel resistance mechanisms to EGFR-TKIs.</P> <P><B>Material and methods</B></P> <P>We collected tumor tissues before EGFR-TKI treatment and after progression from 19 NSCLC patients to analyze genomic alterations in 409 cancer related genes. Bioinformatics analyses were used to identify mutations in which the allele frequencies are significantly changed, or newly appeared after progression.</P> <P><B>Results</B></P> <P>Overall, mutation rates and compositions were similar between pre- and post-EGFR-TKI tumors. We identified EGFR T790M as the most common mechanism of acquired resistance (63.2%). No pre-EGFR-TKI tumor had a preexisting T790M mutation, suggesting that tumors acquired T790M mutations following progression on EGFR-TKIs. Compared to T790M-positive tumors, T790M-negative tumors showed relatively high tumor mutation burden and shorter survival, suggesting T790M-negative patients as a potential candidate for immune checkpoint inhibitors. TP53 mutation was also significantly enriched in the T790M-negative tumors. Finally, we described here for the first time a novel missense mutation (T263P), which occurred concurrently with an activating G719A mutation, in the extracellular domain II of EGFR in a patient with poor response to erlotinib. Ba/F3 cells harboring EGFR T263P/G719A mutation showed higher sensitivity to afatinib, compared to gefitinib due to inhibition of EGFR/HER2 heterodimerization.</P> <P><B>Conclusion</B></P> <P>Comprehensive genomic analysis of post-EGFR-TKI tumors can provide novel insight into the complex molecular mechanisms of acquired resistance to EGFR-TKIs.</P> <P><B>Highlights</B></P> <P> <UL> <LI> NGS in paired NSCLC before and after EGFR-TKIs was performed. </LI> <LI> EGFR-TKI resistant T790M (+) NSCLC patients had significantly longer survival. </LI> <LI> Hypermutation/TP53 mutation seem enriched in EGFR-TKI resistant T790M (−) patients. </LI> <LI> Tumor mutation burden seem to be higher in EGFR-TKI resistant T790M (−) patients. </LI> <LI> T263P of EGFR was identified as the de novo mechanism of resistance to EGFR-TKIs. </LI> </UL> </P>

        Bacterial Composition of Respiratory Organs and Intestine in Mice and Rats

        The change of microbial flora in intestine and respiratory organs make some effects on animal health, which can cause some aberrations to experiment data. Therefore, it is an important factor to maintain the homogeneity of the bacterial composition of each organ. In this study, the bacterial distribution of respiratory organs and intestine were analyzed from 247 mice and 200 rats by analyzing biochemical properties and 16S rDNA sequencing. The total of 1,075 isolates were classified to 35 species of 17 genera. In case of rats, intestinal microbial flora were composed of 5 major bacteria including E. coli, P. mirabilis, M. morganii, C. freundii, and P. vulgaris, and 5 bacteria such as E. coli, P. mirabilis, S. aureus, S. cohnii, and M. morganii also mainly consisted of the microbial flora of respiratory organs. In case of mice, intestinal microbial flora were composed of 4 major bacteria including E. coli, P. mirabilis, A. lwoffi, and L. adecarboxylata, and 5 bacteria such as E. coli, B. sphaericus, S. auricularis, A. lwoffi, and L. adecarboxylata also mainly consisted of microbial flora of respiratory organs. This study suggested that B. sphaericus, G. morbillorum, S. paucimobilis, Staphylococcus spp. were specific in respiratory organs, and that M. morganii and Citrobacter spp. were almost specific in intestine. This study also can give some basic information to understand the relationship between animal care environment and microbial composition of animal organs.
