Salmonella enteritidis와 S. typhimurium의 invasion 유전자 invH, invF, hilA의 클로닝과 유전적 분석

In this study, a gene sequence was determined after cloning genes invH, invF and hilA from Salmonella enterica subsp. enterica, serovar typhimurium and enteritidis. Genes were identified using electrophoresis after being PCR conducted. The result of comparison of determined gene sequence with Salmonella typhimurium LT2 and S. enterica U84273.1 (S. enterica subsp. enterica) showed highly correlated characteristics, i.e,. 99% for invH gene, and 100% for genes invF and hilA, respectively. Finally, in this study was formed a library of invasion, which is known as a toxicity factor of Salmonella, and determined the candidate genes of Salmonella epitope necessary for the production of the oral vaccine.
삼채․강황 복합추출물의 기능성 원료 표준화를 위한 HPLC 분석법 검증

This study was preformed to establish a simple and reliable HPLC/UV analytical method to determine p-coumaric acid contents for the standardization of water extracts of Allium hookeri and Curcuma longa complex as functional health food ingredients. The quantitative HPLC method was optimized using a reversed-phase C18 column at 35℃ with methanol and H2O (30:70, v/v) as the gradient mobile phase at a flow rate of 1.0 mL/min and along with detection wavelength of UV 300 nm. This HPLC/UV analytical method exhibited good specificity and high linearity in the tested range of 1.0–10.0 mg/mL with excellent coefficient of determination (R2) of 1.0. The limit of detection and limit of quantification were 0.08 and 0.24 μg/mL, respectively. In addition, the relative standard deviation values from the intra and inter-day precision measurements were 0.4 and 0.1%, respectively. These results indicate that the established HPLC/UV analytical method is simple, specific, precise, accurate, and reproducible and so could be employed in the quantitative analysis of p-coumaric acid as a functional compound in Allium hookeri Thwaites and Curcuma longa complex.

This study examined the laxative effects of hot-water extracts of Hovenia dulcis Thunb. (HD), Phyllostachys pubescens Mazel (PM), and a 2:8 mixture of both (HP) in two chronic constipation models. For the loperamide-induced constipation model, animals were divided into an untreated group, negative control group (loperamide 4 mg/kg), positive control group (bisacodyl 4 mg/kg) group, and six treatment groups (HP 100 or 400, HD 50 or 100, and PM 100 or 400 mg/kg). For the lowfiber diet-induced constipation model, animals were divided into an untreated group (normal diet), negative control group (low-fiber diet), positive control group (Agio granule, 620 mg/kg), and the same treatment groups. Fecal number, weight, fecal water content, and intestinal transit ratio were higher in the groups treated with HP, HD, and PM than in the groups treated with loperamide or lowfiber diet. Thickness of colon mucosa and muscle layers were increased in the treated groups. Colon tension increased in the HP groups, and [Ca2+]i measurements using fura-2 as an indicator showed that HP inhibits ATP-mediated Ca2+ influx in IEC-18 cells. These results showed that the HP mixture has laxative activity by increased mucin secretion and inducing contractile activity and relaxation. It may be a useful therapeutic strategy for ameliorating in chronic constipation.

Hypercholesterolemia is one of the well-established risk factors for cardiovascular mortality and morbidity in coronary heart disease. The present study was designed to investigate the effects of different types of extracts (aqueous, 20%-, and 80% ethanol) of Nelumbo nucifera leaf (NL) and Curcuma Longa L. (CL) on hypercholesterolemia in in vitro and in vivo models. Our results in in vitro experiments showed that treatment with various extracts of both NL and CL dose-dependently inhibited lipid accumulation and total cholesterol content. The 20% ethanol extract of NL (NL20E) and the aqueous extract of CL (CLW) showed the highest activity. The in vivo experimental group was fed a high cholesterol diet (HCD) with oral administration of NL20E, CLW, or a mixture of NL20E with CLW(NC; mixed equal ratio) for 8 weeks. The oral administration of NL20E, CLW, and NC (100 or 300 mg/kg/day) resulted in the suppression of the levels of serum total cholesterol (TC), low-density lipoprotein (LDL)-cholesterol, liver lipid level, glutamic oxaloacetic transaminase (GOT), glutamic pyruvic transaminase (GPT), atherogenic index (AI), cardiac risk factor (CRF), cholesteryl ester transfer protein (CETP), HMG-CoA reductase, and thiobarbituric acid-reactive substances (TBARS). Similarly, the NL20E, CLW, and NC extracts elevated high-density lipoprotein (HDL) cholesterol levels and lecithin cholesterol acyltransferase (LCAT) activity. Furthermore, NC showed a remarkable hypocholesterolemic effect by decreasing the levels of hepatic TC, AI, LDL cholesterol, HMG-CoA reductase, and CRE and by increasing the levels of HDL cholesterol. Thus, the results of this study indicate that the NC has combined hypocholesterolemic effects that block lipid accumulation, thus preventing hypercholesterolemia.
살모넬라의 Fimbrial Adhesion Group에 포함되는 Type 1 Fimbriae 유전자 분석

본 연구에서는 살모넬라 표준균주 4종과 분리균주 2종을 대상으로 type 1 fimbriae 유전자 fimC, fimD, fimI, fimZ를 각각 비교한 결과 많은 아미노산 변이가 관찰되었다. S. typhimurium ATCC 14028 균주에서 가장 많은 14개의 아미노산 변이가 관찰되었다. 반면 S. typhimurium ATCC 13311 균주에서는 가장 적은 2개의 아미노산 변이가 관찰되었다. 그중에서 유전자 fimC와 fimZ에서는 아미노산 변이가 관찰되지 않았고, fimD와 fimI 유전자에서 Leu527→Pro, Ile130→Val으로 각각 아미노산 변이가 관찰되었다. In this study, a number of amino acid mutations were observed in the type 1 fimbriae genes fimC, fimD, fimI, and fimZ in four Salmonella reference strains and two isolates. S. typhimurium ATCC 14028 strain showed most numerous 14 amino acid variations. On the other hand S. typhimurium ATCC 13311 strain, the least two amino acid variants were observed. Among them, amino acid mutations were not observed in the genes fimC and fimZ, and in the fimD and fimI genes, amino acid variations were observed as Leu527→Pro, Ile130→Val.

Skin aging is caused by structural changes in the extracellular matrix (ECM) due to internal and external factors. It is characterized by decreased skin moisture, increased skin roughness, increased skin wrinkles, and decreased skin elasticity. In this study, Elastase and Collagenase inhibitory activities and SOD, CAT, and GSH-Px activities were confirmed in HaCaT cells, and Collagen-I content and skin elasticity were also confirmed in response to UVB irradiation in HR-1 hairless mice. In addition, a double-blind, randomized, placebo-controlled study was conducted on the effects of oral administration of NP-2007 in 84 women. In the group that consumed NP-2007 or placebo, the amount of skin moisture, trans-epidermal water loss (TEWL), mass of skin keratin, skin asperity (Ra, Rz), eye wrinkle, and skin elasticity were checked. Compared to the placebo group, significant changes were confirmed at all-time points after 6 and 12 weeks of intake. This study was investigated the potential of oral administration of NP-2007 as a functional food material that improves skin.
