고속액체크로마토그래피에 의한 나프록센 광학이성질체의 분리 및 품질관리

Enantiomeric separation of naproxen is necessary in drug evaluation processes as the pharmacological activity of naproxen resides mainly in the (S)-enantiomer, The enantioseparation of naproxen was carried out with three different stationary phases. The methods were validated with the terms of linearity, intraday and interday precision, accuracy, selectivity and recovery. Among the selected stationary phases, ChiroSil DMB column with hexane : t-butylmethylether : acetic acid (60:40:0.1, v/v/v) as mobile phase showed the best enantioseparation of the naproxen in the commercial preparations. The wavelength for detection was 270 nm and flow rate was 2.0 mL/min. The contents of (S)-naproxen in the commercial preparations were 92.7 - 98.3%, indicating that all tested preparations were satisfactory to current regulation. However, the contents of (R)-naproxen as impurity in the commercial preparations were varied from 0.08% to 1.08%.

In this study, we analyzed and quantified the amounts of bioactive phenolic constituents, xanthotoxin (1), oxypeucedanin (2), and imperatorin (3) in each part of the root of Angelica dahurica by HPLC, which validated by ICH guide lines comparing the linearity, intra day precision, inter-day precision. As a result, the amount of imperatorin 2.96% in the rootlet was two fold higher than that of the main root 1.32%. On the other hand, the amounts of xanthotoxin and oxypeucedanin in the rootlet showed similar to those of main root. In addition, the cortex of root is more plentiful of three consitituents (0.66%,0.53%, and 1.85%) than those of xylem (0.29%, 0.05%, and 0.07%). These results show that the rootlet and cortex contain a large amount of bioactive phenolic constituent including xanthotoxin, oxypeucedanin, and imperatorin than other parts of the root.
A Cytotoxic Monoterpene-Neolignan from the Stem Bark of Magnolia officinalis

A monoterpene-neolignan, piperitylmagnolol (1), was isolated from the stem bark of Magnolia officinalis, together with syringaresinol (2), caffeic acid (3), and sinapaldehyde (4). The isolated compounds were established on the basis of spectroscopic and physicochemical analyses including 1D- and 2D- NMR techniques, as well as on comparing the spectral data with those in the literature and of authentic samples. Compounds 1 - 4 were tested for their cytotoxic activity against the HeLa, K562, A549, and HCT116 cancer cell lines in vitro. Of the isolates, piperitylmagnolol (1) exhibited cytotoxic activity against the tested cancer cell lines with IC50 values of 7.7 - 9.5 mg/mL.

Herbal drugs have been playing an important role in the medical therapy system in the oriental culture for thousands of years. Nowadays, the needs for the quality evaluations of these materials are very strong. In this research costunolide in Saussureae Radix was analyzed and the content of regulation was established for the quality evaluation of Saussureae Radix. Costunolide in the acetone extract of Saussureae Radix was successfully separated on Zorbax Eclipse XDB C_(18)(2.1×150 ㎜) with methanol-water(65:35, v/v) as mobile phase. The flow rate was 0.25 ㎖/min and detection was carried out at UV 215 ㎚. The HPLC method was validated by ICH guide lines comparing the linearity, precision, accuracy and detection limit. The average content of costunolide in Saussureae Radix was 1.04%. The highest costunolide content among the samples was 18 times higher than the lowest content. The content of costunolide for regulation in Saussureae Radix, set as the lowest costunolide content of analyzed sample which lied in the range of 95% confidence interval, was 0.68%. The appropriate number of samples to establish the costunolide content for regulation was proposed as 20, with the error level of not higher than 10%. These results provided the statistical back ground for the quality evaluation of Saussureae Radix.
