        EC 격자의 자체 결상 분석

        We characterized theoretically and experimentally the self-images of an elongated circular (EC) grating which could be employed for visual measurement of linear displacements in Talbot Moire deflectometry. Based on the Fresnel diffraction integral, we estimated an shape of the self-image formed at a specific distance from an EC grating in which both parallel lines and semicircles were illuminated by a coherent plane wave. For a square--type transmittance of the grating, we compared the theoretical estimate with its self-image captured by using an array of charge-coupled devices (CCD). We show that the self-images of the EC grating are substantially aberrated near the center of the interface between the linear and the semicircular parts, but not in the off-central region. The Moir$\dot{e}$e patterns formed by using two self-imaged EC gratings were found not to be affected very much by the central aberration. 무아레 간섭법에서 선형 변위의 정밀 측정을 위해 고안된 EC격자의 자체 결상 특성을 조사하였다. 직선과 반원으로 구성된 EC격자에 가간섭성 평면파가 입사할 때, 일정 거리에 형성되는 자체 결상의 특성을 프렌넬 회절 적분식에 근거하여 분석하였다. 사각형 형태의 투과 함수를 갖는 EC격자의 자체 결상을 CCD 사진기로 기록하고 그 결과를 이론과 비교 평가하였다. 반원 부분의 자체 결상은 중심 근처에서 큰 수차를 보여 주었으나 중심에서 멀어질수록 수차가 없어짐을 발견하였다. 자체 결상된 EC 격자쌍이 만드는 무아레 무늬는 중심부 수차에 의해서 큰 영향을 받지 않음을 알 수 있었다.

      • FIA를 이용한 Collagenase 활성의 간편정량법 개발

        The parameters that play an important role in the measurements of the collagenolytic activity by flow injection analysis (FIA) are optimized. Borate buffer was used as sample carrier and OPA (o-phthalaldehyde) as reaction reagent, and the flow rates were 0.5mn/min and 0.05㎖/min, respectively. The collagenolytic activity was determined by florescence intensity at λ_ex 338nm and λ_em 446nm. The reaction of OPA with substrate was carried out in a reaction coil for 20 sec at room temperature. FALGFA (2-furanacryloyl-L-leucylglycyl-L-prolyl-L-alanin) was used as substrate and reacted with collagenase for 1 hour at room temperature. Calibration curve of collagenase was acquired in the range of 0.5-20㎍/㎖ and the correlation coefficient was 0.9997. This method can also be useful for the screening of enzyme inhibitor.

        HPLC-ECD를 이용한 흰쥐의 뇌 중 감마 아미노부티르산 및 글루탐산의 정량

        A sensitive and efficient assay method was applied to determine the level of glutamic acid (GA) and ${\gamma}-aminobutyric$ acid (GABA) in frontal cortex and hippocampus of rat administrated with ethanol and drugs. The compounds were derivatized with ο-phthalaldehyde (OPA) and 2-mercaptoethanof for precolunm analysis. The condition for the simultaneous determination of GA, GABA and $\beta-aminobutyric$ acid (BABA) by high performance liquid chromatography with electrochemical detection was reverse phase $C_{18}$ column as stationary phase, 0.1 M phosphate buffer containing 0.1 mM $Na_4EDTA$ : methanol = 55:45 (v+v) pH 3.8 as mobile phase and 0.7V electrode voltage. The stability of reaction product of GA, GABA and BABA with OPA could be increased by adding the same volume of polyethylene glycol 400 to reaction mixture. The GABA level in frotal cortex of rat was significantly decreased by the administration of picrotoxin and diazepam, but it was significantly increased by the administration of red ginseng total saponin, N-methyl-D-glucamine and (-)-deprenyl.

      • 크로마토그래프법을 이용한 사삼(沙蔘) 및 유사생약의 패턴분석 비교 연구

        Adenophora Radix is the dried root of Adenophora triphylla var. japonica Hara or the other crude drugs belong to genus Adenophora. However, other crude drug like Codonopsis lanceolata or C. pilosula were often misused as Adenophora radix. A pattern analysis method to evaluate the Adenophora Radix and similar crude drugs were carried out with the methanol and ethylacetate extract by HPLC and TLC. The components were separated by HPLC on ODS column with gradient elution of solvent A (10% acetonitrile) and B (70% acetonitrile). The multivariate peak data of the chromatograms of extracts of samples were used for hierarchical clustering analysis, principal components analysis and similarity calculation. The similarities like Euclidean distance, cosine and correlation for the methanol extract of A. tetraphylla and A. remitiflora showed good correlation with A. triphylla var. japonica, indicating that the similarity could be used as a chemical taxonomy. The genus Adenophora and genus Codonopsis were well classified by the hierarchical clustering method for the cluster analysis. The developed pattern analysis method was specific and could be readily utilized for comprehensive evaluation of Adenophora Radix.

        패턴분석법에 의한 대황의 비교 연구

        Three species, such as Rheum palmatum L., R. tanguticum Maxim. and R. officinale Baillon are recognized as the source plants of Rhei Rhizoma in Korean Pharmacopeia. However, other herbal sources such as R. undulatum L. and Rumex crispus L. have been often misused as Rhei Rhizoma. A pattern analysis method to discriminate Rhei Rhizoma in Korean Pharmacopeia from other herbal plants using HPLC and TLC chromatograms was developed. The multivariate peak data of the chromatograms of methanol extracts of Rhei Rhizoma were used for hierarchical clustering analysis, principal components analysis and similarity calculation. Besides of the statistic analysis, TLC patterns of samples could be used as criteria of the discrimination. The developed pattern analysis method was specific and could be readily utilized for comprehensive evaluation of Rhei Rhizoma.

      • TLC/HPTLC에서 측정된 자외/가시부 스펙트럼의 표준화 및 검색

        To improve the identification power of TLC/HPTLC the in situ reflectance spectra obtained directly from plates with commercial scanner are used. The spectrum normalization should be carried out prior to comparing and searching the spectra from library for the identification of compounds. Because the reflectance does not obey the Lambert-Beer's law, there arise some problems in normalization. These problems could be solved to some extent by normalizing the spectra with regression methods. The spectra are manipulated with the regression function of a curve obtained from the correlation plot. When the parabola was used as the manipulating function, the spectra were identified with the accuracy of 97% and this result was better than that of conventionally used the point and area normalization method.
