        Wogonin inhibits Cytokine-induced TARC/CCL17 Expression by Suppression of NF-κB activation via p38 MAP kinase Signalning Pathways in HaCaT Keratinocytes

        Thymus and activation-regulated chemokine (TARC/CCL-17), produced by keratinocytes, is a CC chemokine known to selectively Th2 type T cells via CCR4+ and is implicated in the development of atopic dermatitis (AD). TARC/CCL17 expression was induced by cytokines such as tumor necrosis factor-α (TNF-α) and interferon-γ (IFN-γ). We recently found that the wogonin, a flavone isolated from Scutellaria baicalensis, suppressed TARC expression via heme oxygenase 1 (HO1) in human keratinocytes induced with mite antigen. However, little is known about the inhibitory mechanism of wogonin on TARC/CCL-17 expression stimulated with cytokines. To investigate the inhibitory mechanism, I determined the inhibitory effects of wogonin on the activation of nuclear factor-κB (NF-κB) and IκBα phosphorylation, and also examined the activation of p38 MAP kainase in HaCaT keratinocytes stimulated with TNF-α and IFN-γ. Wogonin inhibited NF-κB-DNA complex, NF-κB binding activity, and the phosphorylation of IκBα in a dose dependent manner. Wogonin also inhibited the translocation of NF-κB from cytosol to nucleus. Moreover, the phosphorylation of of p38 MAP kinase in the TNF-α and IFN-γ-stimulated HaCaT keratinocytes were suppressed by wogonin in a dose dependent manner. These results suggest that wogonin may inhibit cytokine-induced NF-κB activation by IκBα degradation via suppression of p38 MAP kinase signaling pathway in keratinocytes and modulation of wogonin signaling pathway may be beneficial for the treatment of AD.

        활성화된 설치류 RAW 264.7 대식세포에서 당귀 에탄올 추출물의 항염증 효과

        The inhibitory effects of ethanol extract of Radex Angelica uchiyamana, on LPS- or IFN- plus LPS-induced production of NO, and $TNF-{\alpha}$, and expression of iNOS and COX-2 were investigated in the activated RAW 264.7 cells. This extract significantly inhibited the production of NO and $TNF-{\alpha}$, and suppressed the expression of iNOS and COX-2 in a dose-dependent manner. These results show that ethanol extract of Radex Angelica uchiyamana may explain some known biological activities including anti-inflammatory effect, and is of considerable benefit in the treatment for NO and pro-inflammatory cytokine overproduction related immunological diseases.

        인간 혈관 내피세포에서 NF-κB 억제를 통한 엉겅퀴 추출물의 VCAM-1 및 ICAM-1 발현 억제효과

        Cirsium japonicum var. maackii is a traditional Korean wild perennial herb used to treat blood circulation, high blood pressure, inflammation, diabetes, and kidney damage. However, it is not known whether C. japonicum var. maackii directly improves endothelial dysfunction. In this study, the effect of C. japonicum var. maackii (CJE) on tumor necrosis factor (TNF)-α-induced vascular inflammation was investigated in vitro using human umbilical vein endothelial cells (HUVEC). As a result, CJE inhibited the production of VCAM-1, ICAM-1 and ROS increased by TNF-α in HUVECs. In addition, treatment with CJE attenuated IκB phosphorylation and translocation of NF-κB to the nucleus. These results suggest that CJE can suppress TNFα-induced adhesion molecule expression by blocking NF-κB signaling and inhibiting ROS generation. The results of this study show that CJE has the potential to be used to treat and prevent inflammation associated with endothelial cell damage.

        Effect of Aqueous Extract of Schizandra chinensis and Evodia rutaecarpa Fruits on Experimental Mouse Colitis Induced by Dextran Sulfate Sodium

        The aqueous extract of Schisandra chinensis, Evodia rutaecarpa and meal (SEM-Ex) has been traditionally used in the Oriental countries as an astringent. However, little is known about the effects of aqueous extract of SEM-Ex on dextran-sulfate sodium (DSS)-induced colitis in mice. In this study, we investigated the protective effects of SEM-Ex on DSS-induced colitis in mice. An experimental colitis was induced by daily treatment with 5% DSS. SEM-Ex was orally administered from day 2 of DSS treatment in the different dose (10-50 mg/kg body weight). SEM-Ex reduced significantly clinical sign of DSS-induced colitis, including body weight loss, shorten colon length, increased disease activity index (DAI), and histological colon injury. Moreover, SEM-Ex suppressed significantly not only the serum haptoglobin levels and the activities of myeloperoxidase (MPO), but also the colon tissue expression levels of monocyte chemoattractant protein-1 (MCP-1) in DSS-induced mice. In contrast, SEM-Ex increased significantly the colon tissue expression levels of granular colony stimulating factor (G-CSF) well known as anti-inflammatory cytokine. These results suggest that SEM-Ex administration could reduce significantly the clinical signs and regulate of chemokine and anti-inflammatory cytokine in DSS-induced model mice. Therefore, these properties may contribute to the strong anti-ulcerative colitis (UC) response effect of SEM-Ex.

        Dextran Sulfate Sodium 유도 마우스 대장염에 미치는 오미자와 매실의 상승효과

        The fruits of Schisandra chinensis and Prunus mume have been traditionally used in the Oriental countries as an astringent against diarrhea and abdominal pain, a protectant for liver disease, an antimicrobial, and a blood tonic. However, little is known about the extract of Schizandrae Fructus and Mume Fructus (SMF-Ex) on dextran-sulfate sodium (DSS)-induced colitis in mice. In this study, we investigated the protective effects of SMF-Ex on DSS-induced colitis in mice. An experimental colitis was induced by daily treatment with 5% DSS. SMF-Ex was orally administrated the single dose (80 mg/kg, body weight/day) for 7 days with one time per day. SMF-Ex reduced significantly clinical sign of DSS-induced colitis, including body weight loss, shorten colon length, increased disease activity index (DAI), and histological colon injury. SMF-Ex also inhibited significantly nitric oxide (NO) and prostaglandine $E_2$ ($PGE_2$) productions in DSS-induced colitis mice. Furthermore, SMF-Ex increased significantly an superoxide anion (SOD), catalase, and glutathione peroxidase (Gpx) activity of the colon tissue in DSS-induced colitis mice. These results suggest that SMF-Ex administration could reduce significantly the clinical signs and inflammatory mediators, and increase antioxidant activity in DSS-induced colitis model mice and is a good candidate for further evaluation as an effective anti-ulcerative agent.

        쏘가리 (Siniperca scherzeri) 말초혈액 백혈구의 미세구조 분석

        광학현미경과 전자현미경을 이용하여 쏘가리(Siniperca scherzeri0의 말초혈액 세포의 형태적 특징을 조사하였다. 쏘가리 말초 혈구는 크게 적혈구, 트롬보사이트, 백혈구 등 3가지로 구분되었다. 적혈구는 성숙 적혈구, 미성숙 적혈구, 적혈 아세포 등 3가지로 구분되었다. 트롬보사이트는 둥근형에서 방추난형으로 구분되었고, 핵은 이질염색질이 풍부한 난형이었으며, 세포질에는 소포, 미세간질이 관찰되었다. 림프구는 소형 림프구(type Ⅰ), 중형 림프구(type Ⅱ), 대형 림프구(type Ⅲ)로 구분되었다. 과립백혈구는 핵과 과립의 형태에 따라 이질핵 과립백 혈구와 호산성 백혈구로 구분되었는데, 이질핵 과립백혈구는 다형립 핵과 세포질내 크고 작은 과립이 풍부했고, 호산성 과립백혈구는 둥근 핵과 크고 작은 둥근 과립이 풍부했다. 과립백혈구의 과립의 형태는 크고 작은 방추형과 원형, 침형 또는 난형으로 구분되었다. 쏘가리의 단핵구/대식세포는 형태적으로 포유동물과 유사하였다. The morphological characteristics of peripheral blood cells in the freshwater mandarin fish (Siniperca scherzeri) were investigated by light and electron microscopy. Erythrocytes, thrombocytes, lymphocytes, granulocytes and monocytes were identified as the peripheral blood cells. Erthrocytes were divided into three form types: mature, immature, and erthroblasts. Thrombocytes were round to long oval (fusiform), each containing vesicular and microtubular structures and an oval nucleus with abundant heterochromatins. Lymphocytes were divided into three types in size: small, medium, and large. Granulocytes were distinguished into three types (type Ⅰ, type Ⅱ, and type Ⅲ) according to the morphology of the granules and nucleus. Heterophilic granulocytes possessed lobulated nuclei and a large number of cytoplasmic granules, each of which was oval and contained electron-dense materials and a crystalloid. Acidophilic granulocytes possessed large and small granules, and round nuclei. The granules were filled with regularly arranged fibriform materials, some needle-like structures, and oval forms. Monocytes and lymphocytes in this fish species were morphologically similar to those of mammals.

        가감보중익기탕의 LPS 유도 염증성 매개물에 대한 억제 효과

        Kagam-bojungikgitang is the water extracts prepared from Ginseng Radix, Astragali Radix, Angelicae gigantis Radix, Astractylodis Rhizoma alba, Aurantii nobilis Pericarpium, Glycyrrhizae Radix, Artemisiae iwayomogii Herba, and Scutellariae Radix. This is a modified prescription of Bojungikgitang, which has been used for the treatment of indigestion, and immunological disease in oriental countries. In this study, the effects of Kagam-bojungikgitang and Bojungikgitang on the production of prostaglandin E_2 (PGE_2) and the expression of cyclooxygenase-2 (COX-2) were examined using Raw 264.7 macrophages activated with lipopolysaccharide (LPS). Both prescriptions dose-dependently reduced the release of PGE_2 and expression of COX-2 caused by stimulation of LPS without cytotoxic effect. Kagam-bojungikgitang's inhibitory effects were better than Bojungikgitang in PGE_2 production and COX-2 expression. Moreover, Kagam-bojungikgitang also attenuated markedly the production of tumor necrosis factor (TNF)-α, and IL-6 than Bojungikgitang in LPS-stimulated RAW 264.7 macrophages. These results suggest that Kagam-bojungikgitang decrease PGE_2 and pro-inflammatory cytokine production in macrophages and these properties may contribute to the anti-inflammatory activity of Kagam-bojungikgitang.

        인진슬기의 간섬유화 억제 효과

        The inhibitory effects of Injinseulki including extracts of melanian snail plus phytomedicinal plants on $CCl_4$-induced fibrosis, $IFN-{\gamma}$ plus LPS-induced production of NO, PMA-stimulated production $O_2$, and tacrine-induced hepatic injury were investigated in rats, RAW 264.7 or HepG2 cells. The ethanol extracts of melanian snail significantly inhibited tarcrine-induced inury of HepG2 cells. Injinseulki inhibited the production of NO and $O_2$ in a dose-dependent manner in activated RAW 264.7 macrophages. Injinseulki significantly inhibited chemical-induced fibrosis in Rats. These results show that Injinseulki including extracts of melanian snail plus phytomedicinal plants may explain some known biological activities of Injinseulki including their anti-inflammatory and anti-fibrotic effect, and is of considerable benefit in the treatment for live diseases.
