        글리코사미노글칸 분해 효소와 핵자기공명을 이용한 오염된 헤파린 시료의 분석

        We examined the purity of six heparin samples by using heparinase, chondroitinase, 1 H-NMR, and polyacrylamide gel electrophoresis. To obtain high molecular weight contaminants from heparin samples, heparinase I - digested samples were subjected to the exhaustive microcon filtration. The filtration process removed heparin-derived di- and oligosaccharides effectively. By combining chondroitinase ABC treatment and strong anion exchange - high performance liquid chromatography, the result showed all six samples contained chondroitin sulfate as a contaminant ranging from 1.3 to 14.9%. Among them, sample S3 showed the highest content of 14.9%, which was further analyzed by chondroitinase AC treatment to confirm chondroitin sulfate B (dermatan sulfate). 1 H-NMR chemical shifts of N-acetyl groups clearly suggested the existence of chondroitin sulfate B (sample S3) and oversulfated chondroitin sulfate (samples S2 and S4) as contaminants. In addition, polyacrylamide gel electrophoresis was useful for qualitative detection on the sample's purity. These results suggest that the tools of heparinase I a nd chondroitinase ABC in combination with NMR spectroscopy would give very useful information for investigation of heparin contaminants such as oversulfated chondroitin sulfate and dermatan sulfate in heparin samples.

        Water Extract of Dolichos lablab Attenuates Hepatic Lipid Accumulation in a Cellular Nonalcoholic Fatty Liver Disease Model

        Nonalcoholic fatty liver disease (NAFLD) is a common chronic liver disease that is rising in prevalence worldwide. Therapeutic strategies for patients with NAFLD are limited by a lack of effective drugs. In this report, we show that Dolichos lablab water extract (DLL-Ex) protects against free fatty acid (FFA)-induced lipid accumulation and attenuates expression of genes involved in lipid droplet accumulation in cellular NAFLD models. The hepatoprotective effects and underlying mechanism of DLL-Ex were assessed using an in vitro cellular model in which NAFLD was simulated by inducing excessive FFA influx into hepatocytes. HepG2 cells were treated with DLL-Ex and FFAs for 24 h, after which intracellular lipid content was observed by using Nile Red and Oil Red O staining. Quantitative real-time polymerase chain reaction was used to measure expression levels of genes related to FFA-mediated cellular energy depletion. Western blotting was used to measure protein levels of phosphorylated c-Jun N-terminal kinase, AMP-activated protein kinase alpha (AMPKa), and peroxisome proliferator-activated receptor c coactivator 1 alpha. In HepG2 cells, DLL-Ex inhibited expression of CD36, which regulates fatty acid uptake, as well as BODIPY-labeled fatty acid uptake. Additionally, DLL-Ex significantly attenuated FFA-mediated cellular energy depletion and mitochondrial membrane depolarization. Furthermore, DLL-Ex enhanced phosphorylation of AMPK, indicating that AMPK is a critical regulator of DLL-Ex-mediated inhibition of hepatic lipid accumulation, possibly through its antioxidative effect. These results demonstrate that DLL-Ex exerts potent anti-NAFLD activity, suggesting that it could be a potential adjuvant treatment for patients with NAFLD.

        Skin Moisturizing and Antiphotodamage Effects of Tyndallized Lactobacillus acidophilus IDCC 3302

        Photoaging is generally the result of chronic exposure to the sun and ultraviolet (UV) radiation, which causes skin damage. In this study, we developed a UVB-induced hairless mouse model to determine whether Lactobacillus acidophilus IDCC 3302 tyndallizate (ACT3302) can enhance photodamaged skin repair. Mice (6 weeks old) were divided into six groups containing normal, UVB-treated vehicle, and UVB-treated ACT3302 (1 × 105, 1 × 106, 1 × 107, and 1 × 108 cells). Epidermal thickness was increased by UVB, but the thickening was lessened by ACT3302 as was the transepidermal water loss (TEWL). However, ACT3302 increased capacitance and decreased TEWL. Skin tissue staining to evaluate skin collagen increases in the number of skin collagen bundles in UVB-treated ACT3302 mice. UVB irradiation increased matrix metalloproteinase (MMP) and proinflammatory cytokine expression and activated mitogen-activated protein kinases in hairless mice; these changes were also attenuated by ACT3302. We conclude that ACT3302 effectively suppressed wrinkle formation induced by UVB irradiation through MMP downregulation. Therefore, ACT3302 potentially prevents skin photoaging and wrinkle formation.

        Dolichos lablab Protects Against Nonalcoholic Fatty Liver Disease in Mice Fed High-Fat Diets

        Hyacinth bean, Dolichos lablab or Lablab purpureus, has been used for centuries in India and China as an edible pod and animal forage, as well as to treat diarrhea and other gastrointestinal disease in traditional Korean medicine. Recently, we have demonstrated that D. lablab extract (DLL-Ex) prevented free fatty acid-induced lipid accumulation in an in vitro cellular nonalcoholic fatty liver disease (NAFLD) model. In this study, we, thus, aimed at clarifying the hepatoprotective effects of DLL-Ex in a high-fat diet-induced in vivo animal NAFLD model, as well as at elucidating underlying mechanisms of identified effects. Sixty, 6-week-old, male C57BL/6J mice were randomly divided into six groups: a control group fed a low-fat diet, four high-fat diet (HFD) groups, three receiving daily oral supplementation of DLL-Ex (25, 50, and 100 mg/kg/day), and one HFD group receiving daily oral supplementation of MILK (100 mg/kg/day). Effects of DLL-Ex supplementation were evaluated by histopathological and histochemical assessments. DLL-Ex supplementation inhibited HFD-induced increases in body weight and body fat mass and ameliorated increases in body weight, manifested as decreased liver function tests, lower serum triglycerides and cholesterol levels, and increased serum adiponectin levels. The expression of hepatic genes involved in lipid droplet accumulation and in fatty acid uptake was also decreased. We provide evidence of a protective effect of DLL-Ex against HFD-induced fatty liver disease in an animal model.

      • 빅데이터 분석을 이용한 보은대추 언론보도 동향 분석: 텍스트마이닝과 의미연결망 분석을 중심으로

        2017년 기준 단기소득임산물 생산액은 임업총생산액(8조 9,652억 원)의 30%(2조 9136억 원)를 차지하며, 우리나라 임가 중 91.1%가 단기소득임산물 재배업에 종사하고 있다. 현재 여러 지방 자치 단체들은 단기소득임산물을 이용하여 임가 소득향상과 지역 활성화를 위해 다양한 노력을 기울이고 있으며, 단기소득임산물로 관련 산업을 활성화시킨 사례분석을 통한 시사점 제시가 필요한 시점이다. 본 연구에서는 충북 보은군의 대추산업 동향과 성장 과정을 살펴보고 시사점을 도출하고자한다. 이를 위해 빅데이터 분석방법인 텍스트마이닝을 활용하여 2005년~2018년까지 보은대추라는 특정 이슈의 언론보도 동향을 온라인뉴스를 중심으로 분석하고, 도출된 키워드들을 대상으로 의미연결망분석을 실시하였다. 분석결과를 요약하면 다음과 같다. 첫째, 보은군에서 대추재배가구는 2006년 289호에서 2016년 1,186호로 410% 증가하였다. 같은 기간 대추 생산량은 278톤에서 4,075톤으로 1,465%, 생산액은 17억 원에서 379억 원으로 2,176% 증가하는 등 대추 산업은 보은군을 대표하는 산업으로 발돋움 하였다. 둘째, 텍스트마이닝 분석결과 1기(2005년~2008년)에서는 ‘보은대추축제’ 키워드가 출현빈도 21개, 빈도순위 37위로 낮게 나타났지만, 2기(2009년~2013년)에서는 출현빈도 341개, 빈도순위 3위, 3기(2014년~2018년)에서는 출현빈도 1,645개, 빈도순위 2위로 시간이 지남에 따라 분석 항목 대부분에서 비중이 높아졌다. 셋째, 의미연결망 분석결과도 텍스트마이닝 결과와 유사하게 기수에 따라 ‘대추축제’와 관련된 연결 관계 및 정도, 그룹이 중요하게 나타났다. 넷째, 보은대추산업이 발달하는 과정에서 보은군에서 추진하고 진행하는 보은대추와 관련된 정책 결과의 일부가 언론보도에 반영되어 의미화 되었고, 그 중 비중이 가장 높게 나타난 것은 ‘보은대추축제’였다. 본 연구는 보은대추 산업이 성장하는 과정에서 보은대추 관련 이슈가 어떻게 부각되고, 의미화 되는지 빅데이터 분석을 적용하여 구명하였다는 데에 의의가 있다. 이를 통해 지방정부에서 단기소득임산물과 관련된 정책 및 전략 수립 시 활용 가능한 기초자료로써의 시사점을 제공할 수 있을 것으로 기대한다.

        군집분석을 이용한 산촌경관 유형 구분 및 특성 분석

        Recently, public awareness regarding mountain villages’ landscapes is increasing. Thus, this study aimed to provide standards for conservation, management and creation of mountain village landscape by characterizing and classifying those exist. 286 mountain villages’ data were collected and 19 variables – extracted from GIS spatial information and statistic data of mountain villages, chosen as right sources according to former studies - were utilized to conduct factor and cluster analysis. As a result of the factor analysis, 7 characteristics of the mountain villages’ landscapes were defined - ‘Location’, ‘Cultivation’, ‘Ecology⋅Nature’, ‘Tourism’, ‘Residence’, ‘Recreation’. The K-means cluster analysis categorized the mountain villages’ landscapes into four types - ‘Residential’, ‘Touristic’, ‘General’, ‘Environmentally protected’. The classification was examined to be appropriate by field assessment, and basic guidelines of mountain village landscape management were set. The results of this study are expected to be utilized planning and implementing regarding mountain village landscape in the future.

        보은대추 산업 발달요인에 관한 연구: 보은군 추진정책과 빅데이터 분석을 중심으로

        The purposes of this study are to 1) investigate the jujube industry and its history of development in Boeun-gun, Chungbuk province and 2) suggest implications from the results. For the research, Boeun’s jujube industry trend was analyzed. Add to that, from each election period, articles related to Boeun Jujube were collected to be analyzed by big data methods - text mining and semantic network analysis. The two analyzed results were compared and analyzed with policies implemented in each election period. The result of this study has been summarized as follows. First, regarding Boeun Jujube, the indicators such as the number of its cultivating households, the acres of its cultivated area, its produced amount, and the revenue have generally increased. Second, Articles referring to Boeun Jujube differed depending on the election period. The each period has approximately doubled number of articles compared to the period immediately before that period, which also resulted in the increased number of used texts and their frequency in the articles. Third, after comparing and analyzing the policies and articles of each election period, it is revealed that each period has a different focus on developing the jujube industry in Boeun. Finally, the development of Boeun’s jujube industry is considered to be the outcome of these efforts: highlighting the historical significance of Boeun Jujube, changing concept to focus on unprocessed jujube rather than dried one to merchandise, building up education system to improve farming skill so as to maintain Boeun Jujube’s quality stable, and developing Boeun jujube festival. Furthermore, the success in implementing those policies is due to the long-term perspective approach by Boeun governers, which was also stable and focused. This research has found significant implications from the development process of Boeun’s jujube industry by comparing and analyzing Boeun’s policies and the big data analyses’ results from articles related to Boeun Jujube. It is hoped that this study can base local governments’policies and strategies related to non-timber forest products. 본 연구는 충북 보은군의 대추 산업 동향과 성장 과정을 살펴보고 시사점을 도출하기 위한 목적으로 수행되었다. 연구방법은 보은군의 보은대추 산업동향을 살펴보고, 민선시기별 언론기사를 수집해 빅데이터 분석 방법인 텍스트마이닝과 의미연결망 분석을 실시하여 도출된 결과를 민선시기별로 추진한 정책과 비교·분석하였다. 분석 결과를 요약하면 다음과 같다. 첫째, 보은군에서 대추는 시기가 지남에 따라 재배 가구 수, 재배면적, 생산량, 생산액 등 재배 관련 지표가 전반적으로 증가하였다. 둘째, 민선 시기에 따라 언론보도건수가 2배 이상으로 증가하였고 기사에 사용된 텍스트 수와 빈도 수도 증가하였다. 셋째, 민선 시기별 추진정책과 언론기사를 비교 분석한 결과, 민선 4기는 보은의 생대추를 알리기 위해 기반을 마련하는 시기, 민선 5기는 보은대추축제를 발달시키고 집중하는 시기, 민선 6기는 보은대추축제를 보다 더 많이 알리는 시기로 분석되었다. 넷째, 보은대추산업의 발달은 보은대추의 역사성 강조, 생대추로의 인식 전환, 균일한 품질을 위한 대추재배교육, 그리고 대추축제 활성화 등의 종합적인 요인들에 의한 것으로 판단되었으며, 정책을 추진한 보은군수의 일관되고 장기적인 관점의 접근이 있었기에 가능한 것으로 해석된다. 본 연구는 산업이 발달하는 과정에서 보은대추와 관련된 언론기사를 빅데이터 분석을 통해 살펴보았고, 이를 보은군에서 추진한 정책과 비교·분석하여 구명하였다는 데에 의의가 있다. 이를 통해 지방정부에서 단기소득임산물과 관련된 정책 및 전략 수립 시 활용 가능한 기초자료로써의 시사점을 제공할 수 있을 것으로 기대한다.

        Beneficial Effects of Insect Extracts on Nonalcoholic Fatty Liver Disease

        It is well known that nonalcoholic fatty liver disease (NAFLD) is a common disease worldwide because of unhealthy changes in dietary habits. In this study, we determined the effects of Tenebrio molitor Linnaeus, 1758 extract (TML) and Allomyrina dichotoma Linnaeus, 1771 larvae extract (ADL) in cellular and animal models. In vitro, TML and ADL treatments did not cause cytotoxicity, but attenuated the accumulation of lipid in HepG2 cells induced by free fatty acids. In vivo, mice were orally treated with TML and ADL for 10 weeks during high-fat diet feeding. TML and ADL administration significantly reduced the weight of body, liver tissue, and adipose tissue. Serum lipid profiles, hepatic functional parameters, and glucose levels were ameliorated by TML and ADL. Moreover, TML and ADL suppressed increased lipogenesis and inflammation-related makers, and improved antioxidant enzyme activity. In liver tissue, the decreased lipid accumulation by administration of TML and ADL was observed using Oil Red O and Hematoxylin and Eosin staining. Therefore, we suggest that TML and ADL may be having a therapeutic potential and is used to develop a therapeutic agent for NAFLD.
