Multiplex PCR을 이용한 유전자변형 카네이션의 검출법

유전자변형 카네이션(Moonvista, Moonaqua, Moonshadow, Moonlite, Moonshade, 및 Moodust) 개발에 이용된 형질전환 벡터(pCGP1470, pCGP1991)를 동시에 검출할 수 있는 multiplex PCR이 개발되었다. 유전자변형 카네이션을 스크리닝하기 위해 acetolactate synthase(SuRB/ALS) terminator의 염기 서열로부터 한 쌍의 primer를 제작하였다. Anthocyanidin synthase(ANS)를 카네이션의 내재유전자로 PCR 검사법에 이용하였다. 100bp의 PCR 산물을 얻을 수 있는 ANS-KF/KR primer가 ANS 유전자를 증폭시키는데 이용되었고, 이것을 이용하여 11개의 다른 식물에 대해서 PCR한 결과 카네이션에서만 특이적인 증폭산물이 확인되었다. C1991-KF/KR primer가 형질전환 벡터 pCGP1991에 삽입된 Dihydroflavonol reductase(DFR) 유전자와 DFR terminator 사이의 염기서열을 확인할 수 있도록 제작되었고, 형질전환 벡터 pCGP1470를 확인하기 위해 C1470-KF/KR primer가 Mac promoter와 DFR 유전자 사이의 염기서열을 확인할 수 있도록 제작되었다. 본 연구에서 개발된 multiplex PCR의 검정한계치는 약 1%이며, 이러한 multiplex PCR이 국내 미승인 유전자변형 카네이션을 모니터링 하는데 유용하게 사용될 수 있을 것이다. A multiplex PCR method was developed to simultaneously detect two transformation vectors (pCGP1470 and pCGP1991) used in the development of GM carnation plants (Moonvista, Moonaqua, Moonshadow, Moonlite, Moonshade, and Moodust). To screen for GM carnation plants, one specific primer was designed from acetolactate synthase (SuRB/ALS) terminator sequence. The anthocyanidin synthase (ANS) was also used as an endogenous reference gene of carnation in PCR detection. The primer pair ANS-KF/KR producing 100 bp amplicon was used to amplify the ANS gene and no amplified product was observed in any of the 10 different plants used as a template. The primer pair C1991-KF/KR was designed to amplify the junction sequence between dihydroflavonol reductase (DFR) gene and DFR terminator in pCGP1991. To confirm pCGP1470, the primer pair C1470-KF/KR was designed to cover the junction sequence between Mac promoter and DFR gene. The detection limit of the multiplex PCR method is approximately 1%. This result indicates that this multiplex PCR method could be a useful tool for monitoring unauthorized GM carnation in Korea.

Chloride ions are considered to be the major cause of steel corrosion in concrete structures exposed to seashore environments and also permeation of chloride is controlled by chloride diffusion. Therefore, the study on chloride diffusion of concrete have been done so far by many researchers. In this study, to examine the chloride penetration properties of concrete using fly-ash and blast-furnace slag powder as replacements of cement, it is compared and investigated the replacement ratio of fly-ash and blast-furnace slag powder with each 3 levels. Therefore, it is intended to grasp the chloride penetration properties of concrete including effects of W/B ratio and age, as other mix factors.

최근 거의 모든 산업에 걸쳐 보안과 서비스에 대한 이슈가 대두되고 있다. 이는 항만물류산업에서도 큰 영향을 미쳐 기존 항만 생산성 향상에만 초점을 두어 업무를 추진하던 방식에서 탈피하여 점차 항만 안전과 대고객 서비스 향상을 중심으로 업무 전환을 꾀하고 있다. 그 중 대표적인 방법이 기존의 컨테이너에 특정한 장치를 추가 장착하여 지능화시킨 GE(General Electronic)의 CSD(Container Security Device)와 Savi Networks의 eSeal 등을 들 수 있다. 하지만 이 장비들은 국제표준과 기술적인 구현 부분에만 치중하여 개발함으로써 화주, 선사 및 하역업체 등 실사용자들의 불편함을 야기하고 있으며, 이는 사용자 요구사항을 체계적으로 충분히 고려하지 못한 결과로 판단된다. 본 연구에서는 기 개발된 보안 장치가 갖는 문제점들을 해결하고, 사용자 요구사항이 충실히 반영되었을 뿐만 아니라 현실세계를 고려한 보안 시스템을 개발하기 위해 시스템 요구사항을 도출하는 과정에서 QFD(Quality Function Deployment, 품질 기능전개) 방법론을 적용하였으며 그 결과 시스템 품질특성(CTQ: Critical To Quality)이 5가지의 세부 과제별로 총 21건이 도출되었다. Recently security is being an important issue in almost every field of industry. This situation has affected port logistics industry deeply. Ports are now leaving operational methods that only focus on productivity, and shifting to new ones which focus on safety and custom services on the basis of it. Thus a lot of companies and institutions have offered various solutions as this issue becomes more and more intense. Among them, most typical solutions involve installing special devices to ordinary containers to improve its security, such as CSD (Container Security Device) of GE (General Electric) and eSeal of Savi Networks. On the other hand, these devices focus only on international standards or technical implementation, and this causes inconvenience to actual users like cargo owners, sea carriers, or stevedoring companies. This is considered to be due to lack of sufficient consideration on user demands. This research uses QFD (Quality Function Deployment) method for deducting system requirements in order to solve the problems of previous security devices and to develop a security system that can not only reflect the demands of the users but also considers real-world conditions. According to the QFD results, a total of 21 system CTO"s were deducted under 5 categories.

近年의 航空運送의 伸張은 눈부신것이며 增大하는 需要에 答하듯 연이은 Jumbo機의 導入과 空港施設의 擴張이 이루어졌다. 그러나 이것은 一部 主要 空港에 極限되며 幹線을 除外한 支線의 경우는 오히려 收容의 限界가 極限되어 退步一路에 있는 實情이다. 旣存의 작은 空港을 利用할수 있고 中近距離 輸送에 適格인 STOL機는 將次 航空輸送에 큰 比重을 차지할 것임에 輸送 system으로서의 STOL 機에 對하여 system 全般의 問題點과 機體의 技術的 問題 및 航空保安無線裝備의 改良方案에 對하여 硏究한 것이다. Recent expansion made in the air transportation has proven to be remarkable and in response to its ever-increasing demand Jumbo airliners have been introduced with airport facilities improved and enlarged. Yet the current situation is that such development seems to be limited entirely to the principal airports of the main lines, whereas the local lines appear to be deteriorating due to their insufficient overladen capacity. Taking it into consideration that STOL airplanes which, being applicable to middle-short-distance transportation, may utilize comparatively small-scale facilties of the existing airports and thus would play an important role in the prospective air transportation, this study is intended to deal with the over-all efficiency of STOL airplanes as a transportation system including general problems concerning technical know-how which would create the improvement of navigational aids for air security.
종합적 품질경영(TQM)의 보상요인이 성과에 미치는 영향에 대한 연구

According to the study of TQM implementations, the odds that an organization will successfully implement total quality management are two to one-against it. However, organizations armed with knowledge of the factors that have the biggest impact on their success can take steps to influence or change those factors. In this paper, we first are reviewed those rewards factors through literature study and gave the test for them in the Korean industry. This study also proposes that appropriate successful rewards factors are necessary for TQM implementations.
