        Disrupted Pathways Associated with Neonatal Sepsis: Combination of Protein-protein Interactions and Pathway Data

        To identify disrupted pathways associated with neonatal sepsis, we performed a research based on the combination of protein-protein interactions (PPIs) and pathway data. Firstly, a total of 23,292 genes, 787,896 PPIs and 1,675 human pathways were obtained, respectively. Then, under the threshold value of false discovery rate (FDR)<0.05 and a delta cut-off value >4.36, a total of 986 differentially expressed genes (DEGs) were identified. In the following, by degree centrality for the objective PPI network, 20 hub genes were obtained. Finally, pathway enrichment analysis and randomization tests indicated that pathways of gene expression, immune system and innate immune system were with remarkable significance in neonatal sepsis. Therefore, in the present study, we presented a novel pathway method, and we successfully identified several pathways in neonatal sepsis, which might be underlying indicators in the detection and treatment of neonatal sepsis.

        Biocatalysis and Fermentation Technology : Thermostable Sites and Catalytic Characterization of Xylanase XYNB of Aspergillus niger SCTCC 400264

        In order to improve the expression of heat-resistant xylanase XYNB from Aspergillus niger SCTCC 400264, XynB has been cloned into Pichia pastoris secretary vector pPIC9K. The XynB production of recombinant P. pastoris was four times that of E. coli, and the Vmax and specific activity of XynB reached 2,547.7 umol/mg and 4,757 U/mg, respectively. XynB still had 74% residual enzyme activity after 30 min of heat treatment at 80°C. From the van der Waals force analysis of XYNB (ACN89393 and AAS67299), there is one more oxygen radical in AAS67299 in their catalytic site, indicating that the local cavity is much more free, and it is more optimal for substrate binding, affinity reaction, and proton transfer, etc, and e ventually i ncreasing enzyme activity. The H-bonds analysis of XYNB indicated that there are two more H-bonds in the 33rd Ser of XYNB (AAS67299) than in the 33rd Ala(ACN89393 ), and two H-bonds between Ser70 and Asp67.

        The Evaluation of Chinese Railway Companies’ Efficiency

        철도는 국가경제발전에 반드시 필요한 물류인프라 시설이다. 중국에서의 철도 설비와 관련 산업은 최근 빠르게 성장하였으나, 여전히 운송에 대한 경제적 및 사회적 개발의 수요를 모두 충족하지는 못하고 있다. 선행연구를 살펴보면 중국 각 지역의 철도 서비스를 운영하는 각 철도 기업에 대한 최근 효율성 연구가 부족한 실정이다. 이에 따라 본 연구는 자료포락분석 (DEA)과 초효율성 (Super-efficiency) DEA 방법론을 사용하여 2018년 중국 철도 기업 17곳의 기술 및 순수 기술 효율성, 규모 효율성을 측정하고자 하였다. 분석 결과, Taiyuan의 경우 기술 효율성 측면에서 가장 높은 점수를 보이고, Harbin이 가장 낮은 점수를 보였다. 또한, 전반적으로 이전의 연구와 유사하게 중국 철도 기업들의 기술 효율성은 낮은 수준으로 도출되었다. 이를 개선하여 효율성을 높이기 위해서는 지속가능한 성장을 유지하고 철도 설비에 대한 과도한 투자를 조절하기 위해서 중국 철도 기업들은 철도 화물 운송과 여객 운송에 있어 균형적인 개발 조절을 해야 한다. 결과적으로 본 연구는 철도 기업에게 운영 개선점과 시사점을 제시하고자 하였다. The railway is an essential transportation infrastructure in a country. In China, the railway transportation industry has developed rapidly. However, it still cannot meet the needs for economic and social development of transportation. This study uses the data envelopment analysis (DEA) and super-efficiency DEA methods to evaluate the efficiency of Chinese railway companies in 2018. Results reveal that Chinese railway companies have different input redundancy and output deficiency. For instance, in 2018, Taiyuan Railway Company had the highest technical efficiency among all Chinese railway companies. Meanwhile, Harbin Railway Company had the lowest technical efficiency. Therefore, to maintain a sustainable development of railway companies and control excessive investment in infrastructure, Chinese railway companies must coordinate a balanced development of railway freight transport and passenger transport, which will also improve the railway companies’ efficiency. Several implications can be drawn from this study for railway companies.

        Correlation between fat accumulation and fecal microbiota in crossbred pigs

        Backfat thickness (BF) is an important indicator of fat deposition capacity and lean meat rate in pigs and is very important in porcine genetics and breeding. Intestinal microbiota plays a key role in nutrient digestion and utilization with a profound impact on fat deposition of livestock animals. To investigate the relationship between the pig gut microbiome and BF, 20 low-BF (L-BF) and 20 high-BF (H-BF) pigs were selected as two groups from Yunong Black pigs in the present study. Fecal samples from pigs were analyzed for microbial diversity, composition, and predicted functionality using 16S rRNA gene sequencing. The results showed that there were significant differences in microbial β diversity between the two groups. LEfSe analysis revealed a number of bacterial features being differentially enriched in either L-BF or H-BF pigs. Spearman correlation analysis identified the abundance of Oscillospira, Peptococcus, and Bulleidia were significantly positive correlations with BF (P < 0.05), while Sutterella and Bifidobacterium were significantly negatively correlated with BF (P < 0.05). Importantly, the bacteria significantly positively correlated with BF mainly belong to Clostridium, which can ferment host-indigestible plant polysaccharides into shortchain fatty acid (SCFA) and promote fat synthesis and deposition. Predictive functional analysis indicated that the pathway abundance of cell motility and glycan biosynthesis were significantly widespread in the microbiota of the H-BF group. The results of this study will be useful for the development of microbial biomarkers for predicting and improving porcine BF, as well as for the investigation of targets for dietary strategies.

      • Long Non-coding RNA GAS5 Functions as a Tumor Suppressor in Renal Cell Carcinoma

        Background: Renal cell carcinoma (RCC) is a malignancy with a poor prognosis. We aimed to explore whether the expression of Long Non-Coding RNA (LncRNA) growth arrest-specific transcript 5 (GAS5) is associated with RCC genesis. Methods: We selected twelve clinical samples diagnosed for renal clear cell carcinoma and found that the LncRNA GAS5 transcript levels were significantly reduced relative to those in adjacent unaffected normal renal tissues. Results: In addition, expression of GAS5 was lower in the RCC cell line A498 than that in normal renal cell line HK-2. Furthermore, using functional expression cloning, we found that overexpression of GAS5 in A498 cells inhibited cell proliferation, induced cell apoptosis and arrested cell cycling. At the same time, the migration and invasion potential of A498 cells were inhibited compared to control groups. Conclusion: Our study provided the first evidence that a decrease in GAS5 expression is associated with RCC genesis and progression and overexpression of GAS5 can act as a tumor suppressor for RCC, providing a potential attractive therapeutic approach for this malignancy.

        Research on Oscillation Character of Six-Phase Fractional-Slot Concentrated-Winding Permanent Motor with Different Slot-Pole Match

        The oscillation character of permanent magnetic motor is highly related to its slot-pole match. By calculating air-gap magnet field and radial electromagnetic force of 6-phase fractional-slot concentrated-winding permanent magnetic motor with slot–pole match of 48/44,48/46,48/50 and 48/52 under no load and load status, oscillation character of permanent magnetic motor is analyzed. A 20kW prototype with 48 slots and 44 poles is designed. With many sensors attaching to the corresponding parts, oscillatory acceleration is measured, and spectrum of oscillation frequency is recorded as well. The experiment results give proof to the analysis method for permanent magnetic motor oscillation in this paper.

        Albumin-to-Fibrinogen Ratio as an Independent Prognostic Parameter in Untreated Chronic Lymphocytic Leukemia: A Retrospective Study of 191 Cases

        Purpose Chronic lymphocytic leukemia (CLL) is one of the most frequent type of B-cell chronic lymphoproliferative disorders and chronic inflammation takes part in the development of CLL. However, there has been no valid immune biomarker to predict the prognosis of untreated CLL patients. Materials and Methods In this retrospective study, we analyzed the clinical correlations and prognostic value of albumin-to-fibrinogen ratio (AFR) detected at diagnosis in 191 CLL patients. Results The cut-off value of AFR was 9.7 calculated by X-tile. Patients who were more than 65 years old were often accompanied by low level of AFR (p < 0.001). Survival analysis showed that patients with low level of AFR had shorter overall survival (OS) than patients with high level of AFR (p < 0.001). Multivariate analysis illustrated that AFR had a negative impact on OS (p=0.003) and was independent of parameters involved in CLL international prognostic index and other prognostic markers such as CD38 and ZAP-70. Conclusion These data provide a comprehensive view of AFR and shows that AFR at diagnosis is an adverse prognostic factor in untreated CLL patients.

      • Role of Nucleation-Promoting Factors in Mouse Early Embryo Development

        <B>Abstract</B><P>During mitosis nucleation-promoting factors (NPFs) bind to the Arp2/3 complex and activate actin assembly. JMY and WAVE2 are two critical members of the NPFs. Previous studies have demonstrated that NPFs promote multiple processes such as cell migration and cytokinesis. However, the role of NPFs in development of mammalian embryos is still unknown. Results of the present study show that the NPFs JMY and WAVE2 are critical for cytokinesis during development of mouse embryos. Both JMY and WAVE2 are expressed in mouse embryos. After injection of JMY or WAVE2 siRNA, all embryos failed to develop to the morula or blastocyst stages. Moreover, using fluorescence intensity analysis, we found that the expression of actin decreased, and multiple nuclei were observed within a single cell indicating that NPFs-induced actin reduction caused the failure of cell division. In addition, injection of JMY and WAVE2 siRNA also caused ARP2 degradation, indicating that involvement of NPFs in development of mouse embryos is mainly through regulation of ARP2/3-induced actin assembly. Taken together, these data suggested that WAVE2 and JMY are involved in development of mouse embryos, and their regulation may be through a NPFs-Arp2/3-actin pathway.</P>

        New degradation feature extraction method of planetary gearbox based on alpha stable distribution

        The planetary transmission system has been widely used in industry because of its various advantages. And the study on degradation feature extraction method of planetary gearbox is of major significance for mechanical system prognostics and health management (PHM). In this paper, the alpha stable distribution characteristics of planetary gearbox vibration signals in performance degradation process are verified. By observing the change of alpha stable distribution for planetary gearbox degradation experiment data, a new degradation feature extraction method based on alpha stable distribution is proposed, which is called the height of probability distribution (HPD). Through comparative analysis, it is determined that HPD has better linearity and less fluctuation compared with conventional degradation features in planetary gearbox accelerated degradation stage. Moreover, in the accelerated degradation stage, the degradation trend prediction result based on HPD is closer to the actual data than conventional degradation features no matter using Wiener-based or LSSVM-based prediction method. These conclusions indicate that the newly proposed HPD works well and gives accurate estimates for condition monitoring and degradation trend prediction of planetary gearbox.

        Robust Experimental Study of Data-driven Optimal Control for an Underactuated Rotary Flexible Joint

        As an important component of industrial robot, the motion control of rotary flexible joint (RFJ) system is of great significance, especially when the system has unmodeled dynamics or is seriously disturbed. This paper presents an experimental robustness study on a kind data-driven optimal control approach based on an underactuated rotary flexible joint system. The data-driven approach combines the off-policy optimal control algorithm and the popular integral reinforcement learning technique. Through literature review, we find that the key step of the control design lies in that it learns the optimal value function and control policy simultaneously from the input and output (I/O) data. However, the I/O data are often disturbed by the system uncertainty or environmental noise, and then it will indirectly affect the optimal control performance. To investigate the robustness of the data-driven optimal control approach, we artificially set different experimental scenarios and take numerous control experiments on a RFJ experimental setup. The experimental results show that the data-driven optimal control method is quite robust against the system uncertainties in terms of maintaining the stability and delivering satisfactory tracking performance, even when the uncertainty is not a small quantity. In addition, the disturbance originating from environmental noise has certain impact on the controlling of RFJ system, but as long as the noise power is not too large, the control algorithm can converge to a satisfactory result. Finally, we find that the probing signal up has strong influence to this control algorithm, which reminds us to be cautious when selecting the probing signal.
