        Characterization of a novel variant HMW‐glutenin gene from Elymus canadensis

        High molecular weight (HMW) glutenin subunits (GS) play a key role in the determination of end‐use quality of wheat and other cereal crops. In this study, we report the isolation and characterization of both promoter region and ORF of novel HMW‐GS allele 1St1.3 from a perennial Triticeae species,Elymus canadensis. The amino acid (AA) sequences of E. canadensis 1St1.3 were deduced as 434 aa. Its protein primary structure comprises a signal peptide with a conserved N‐terminal domain, a central repetitive domain and a C‐terminal domain. E. canadensis 1St 1.3 possesses several distinct characteristics which are different from those of wheat HMW‐GSs. The N‐terminal domains of E. canadensis 1St 1.3 resemble that of y‐type subunits, while their C‐terminal domains are more similar to x‐type subunits. The deletion of 85 bp fragment has been observed in promoter region of 1St 1.3, however which has not interrupted the expression of this gene. Our results indicate that 1St 1.3 is novel HMW‐GS variants which will be valuable for enhancing our understanding of structural differentiation and the evolutionary relationship among HMW‐GSs in Triticeae species.

        Piezo2: A Candidate Biomarker for Visceral Hypersensitivity in Irritable Bowel Syndrome?

        Background/Aims Currently, there exists no biomarker for visceral hypersensitivity in irritable bowel syndrome (IBS). Piezo proteins have been proven to play an important role in the mechanical stimulation to induce visceral pain in other tissues and may also be a biomarker candidate. The aim of this study was to test the expressions of Piezo1 and Piezo2 proteins in the intestinal epithelial cells from different intestinal segments and to explore the correlation between Piezo proteins expression and visceral pain threshold. Methods Post-infectious IBS was induced in mice via a Trichinella spiralis infection. Visceral sensitivity was measured with abdominal withdrawal reflex to colorectal distention. Inflammation in the small intestine and colon was scored with H&E staining. Expression location of Piezo proteins was confirmed by immunohistochemistry. Abundance of Piezo proteins were measured with real-time reverse transcriptase polymerase chain reaction. Results Piezo1 and Piezo2 proteins were expressed in the intestinal epithelial cells. The expression levels of Piezo1 and Piezo2 were abundant in the colon than the small intestine (P < 0.001 for Piezo1, P = 0.003 for Piezo2). Expression of Piezo2 in the colon significantly correlated to the visceral sensitivity (r = -0.718, P = 0.001) rather than the mucosal inflammation. Conclusion Piezo2 is a candidate biomarker for visceral hypersensitivity in IBS. (J Neurogastroenterol Motil 2017;23:453-463)

        Yeast Extract: Characteristics, Production, Applications and Future Perspectives

        Yeast extract is a product prepared mainly from waste brewer’s yeast, which is rich in nucleotides, proteins, amino acids, sugars and a variety of trace elements, and has the advantages of low production cost and abundant supply of raw material. Consequently, yeast extracts are widely used in various fields as animal feed additives, food flavoring agents and additives, cosmetic supplements, and microbial fermentation media; however, their full potential has not yet been realized. To improve understanding of current research knowledge, this review summarizes the ingredients, production technology, and applications of yeast extracts, and discusses the relationship between their properties and applications. Developmental trends and future prospects of yeast extract are also previewed, with the aim of providing a theoretical basis for the development and expansion of future applications.

        Dynamic Surface Active Fault Tolerant Control Design for the Attitude Control Systems of UAV with Actuator Fault

        In this paper, an active fault tolerant control (FTC) approach based on transient performance index isproposed for the attitude control systems of unmanned aerial vehicle (UAV) with actuator fault. The nonlinearattitude control system model for UAV with actuator faults is given, which represents the dynamic characteristicsof UAV. A fault diagnosis component is used for fault detection and estimation. According to the fault estimationinformation obtained during the fault diagnosis, the fault tolerant control scheme is developed by adopting theadaptive dynamic surface control technique, which guarantees the asymptotic output tracking and ultimate uniformboundedness of the closed-loop attitude control systems of UAV in actuator faulty case. Further, a prescribedtransient performance of the FTC attitude control systems is considered which characterizes the convergence rateand maximum overshoot of the attitude tracking error. Finally, simulation results are shown that the attitude controlsystem states remain bounded and the output tracking errors converge to a neighborhood of zero.

        An Improved Mathematical Model for Estimating Polarization Parameters of Transformer Oil-Paper Insulation

        The parameters of dielectric response based on extended Debye circuit model can refl ect oil-paper insulation state. The model using the initial slop of recovery voltage to estimate circuit parameters, serve as good quality consistency with the measured data. However, the model can be applicable for large power transformers but not for distribution transformers. This paper proposed an improved mathematical model using recovery voltage peak, peak time and initial slope characteristics to solve dielectric response equivalent circuit parameters. The identifi cation of equivalent circuit parameters is converted into a mathematical optimization problem. And then particle swarm optimization algorithm is used for solving the problem. The improved mathematical model can decrease sampling data of recovery voltage. To check the validity of the estimated parameters, the on-site measured data of RVM experiments on actual transformers in various capacities is applied. The calculated result shows that the recovery voltage curve calculated and measured recovery voltage curve have good consistency, which can be advantageous to diagnose the oil-paper insulation status of transformer. It illustrates that the improved method proposed in this paper is feasible and eff ective.

      • Oxaliplatin Sensitizes OS Cells to TRAIL-induced Apoptosis Via Down-regulation of Mcl1

        Purpose: To investigate the killing effect on OS cells of a combination of oxaliplatin and TRAIL and related molecular mechanisms. Methods: TRAIL and oxaliplatin were applied to OS732 cells singly or jointly and survival inhibition rates were measured by MTT assay, changes of cellular shape being assessed with inverted phase contrast and fluorescence microscopy. Apoptotic rates were analyzed by flow cytometry (FCM) and immunocytochemistry was used to examine Mcl1 expression of OS732 cells. Results: The survival inhibition rate of combined application of $100{\mu}g/ml$ TRAIL and $1{\mu}g/ml$ oxaliplatin on OS-732 cells was significantly higher than that of either agent singly (p<0.01). Changes of cellular shape and apoptotic rates also indicated apoptosis-inducing effects of combined application to be much stronger than those of individual application. Oxaliplatin had the effect of down-regulating Mcl1 expression and sensitizing OS cells to TRAIL-induced apoptosis. Conclusion: A combination of TRAIL and oxaliplatin exerts strong killing effects on OS-732 cells which might be related to down-regulation of Mcl1 expression.

      • Transparent and Self-Powered Multistage Sensation Matrix for Mechanosensation Application

        <P>Electronic skin based on a multimodal sensing array is ready to detect various stimuli in different categories by utilizing highly sensitive materials, sophisticated geometry designs, and integration of multifunctional sensors. However, it is still difficult to distinguish multiple and complex mechanical stimuli in a local position by conventional multimodal E-skin, which is significantly important in the signals’ feedback of robotic fine motions and human–machine interactions. Here, we present a transparent, flexible, and self-powered multistage sensation matrix based on piezoelectric nanogenerators constructed in a crossbar design. Each sensor cell in the matrix comprises a layer of piezoelectric polymer sandwiched between two graphene electrodes. The simple lamination design allows sequential multistage sensation in one sensing cell, including compressive/tensile strain and detaching/releasing area. Further structure engineering on PDMS substrate allows the sensor cell to be highly sensitive to the applied pressures, representing the minimum sensing pressure below 800 Pa. As the basic combinations of compressive/tensile strains or detaching/releasing represent individual output signals, the proposed multistage sensors are capable of decoding to distinguish external complex motions.         Psychosocial Factors Associated With Irritable Bowel Syndrome Development in Chinese College Freshmen

        Background/Aims The role of psychosocial factors on irritable bowel syndrome (IBS) in adolescents is incompletely understood. The aim of this study is to investigate the association between the psychosocial factors and the risk of developing IBS in college freshmen. Methods A cross-sectional survey was conducted in a randomly selected freshmen population in Wuhan China (n = 2449). Questionnaire evaluated demographics and psychosocial risks. The population was divided into 3 groups: non-discomfort, chronic abdominal discomfort and IBS. The association between the development of IBS and psychosocial factors was analyzed by ordinal and multiple logistic regression analysis. Results A total of 2053 (83.8%) completed this survey (mean age, 18.2 ± 0.9 years; female, 35.6%). Among them, 82 (4.0%) fulfilled the Rome III criteria for IBS. Female (odds ratio [OR], 3.31; 95% confidence interval [CI], 2.47–4.45), experience of abuse (OR, 2.44; 95% CI, 1.10–5.56), and suicidal intention (OR, 2.17; 95% CI, 1.15–4.17) were more likely to have IBS. Compared with chronic abdominal discomfort, however, depression (OR, 5.55; 95% CI, 1.36–22.71) was the only dependent risk factor for IBS. Conclusion The prevalence of IBS in college freshmen is 4.0%, and to the freshmen, psychosocial factors such as experience of abuse, depression, and suicidal intention were associated with high risk of developing IBS.

      • Efficient Killing Effect of Osteosarcoma Cells by Cinobufacini and Cisplatin in Combination

Methods: Cinobufacini and cisplatin were applied to OS732 cells singly or jointly and survival rates were measured by MTT assay. Changes in cellular shape were observed with inverted phase contrast and fluorescence microscopy and apoptosis rates were analyzed with flow cytometry (FCM). Immunocytochemistry were used to examine the Fas expression of OS732 cells. Results: The combination of cinobufacini and cisplatin had the effect of up-regulating Fas expression and inducing apoptosis. The survival rate of combined application of 100 ${\mu}g/ml$ cinobufacini and 1 ${\mu}g/ml$ cisplatin on OS-732 cells was significantly lower than with either of the agents alone (p<0.01). Changes in cellular shape and apoptotic rates also indicated the apoptosis-inducing effects of combined application were much enhanced. Conclusion: The combination of cinobufacini and cisplatin demonstrated strong killing effects on OS-732 cells which might be related to up-regulation of Fas expression.

        Activating transcription factor 4 aggravates angiotensin II-induced cell dysfunction in human vascular aortic smooth muscle cells via transcriptionally activating fibroblast growth factor 21

        Abdominal aortic aneurysm (AAA) is a life-threatening disorder worldwide. Fibroblast growth factor 21 (FGF21) was shown to display a high level in the plasma of patients with AAA; however, its detailed functions underlying AAA pathogenesis are unclear. An in vitro AAA model was established in human aortic vascular smooth muscle cells (HASMCs) by angiotensin II (Ang-II) stimulation. Cell counting kit-8, wound healing, and Transwell assays were utilized for measuring cell proliferation and migration. RT-qPCR was used for detecting mRNA expression of FGF21 and activating transcription factor 4 (ATF4). Western blotting was utilized for assessing protein levels of FGF21, ATF4, and markers for the contractile phenotype of HASMCs. ChIP and luciferase reporter assays were implemented for identifying the binding relation between AFT4 and FGF21 promoters. FGF21 and ATF4 were both upregulated in Ang-II-treated HASMCs. Knocking down FGF21 attenuated Ang-IIinduced proliferation, migration, and phenotype switch of HASMCs. ATF4 activated FGF21 transcription by binding to its promoter. FGF21 overexpression reversed AFT4 silencing-mediated inhibition of cell proliferation, migration, and phenotype switch. ATF4 transcriptionally upregulates FGF21 to promote the proliferation, migration, and phenotype switch of Ang-II-treated HASMCs.
