      • 中學校 男學生의 家政敎科에 대한 態度 硏究

        The purpose of this study is to accompany efficient education of middle school boys' household management subjects. For the purpose an empirical study to the 6th household management subjects was carried out. In this study an questionnaire survey to middle school boys was accomplished. The target of the research was to analyze the pupils' attitude to household management subjects and their family factor variables. In the study, two boy schools in Pusan Metro City were selected as the research samples. The number of answers were 530 boys and collected data were analyzed by frequency analysis, The important findings of the research were as follows. Part I : The attitude to necessity of the subjects in boy middle school. The respondents expressed positive attitudes in the necessity of household management subjects and the utility of the subjects. But they expressed relatively negative attitudes in the preference of the subjects. The main reason of necessity of the subjects were the increment of men's role in home. The obstacles of studying the subjects were the conception of less important subjects than others and the difficulty of practice at the home. Part II : The attitude to contents of the subjects in boy middle school. The answers expressed positive attitudes in the understanding of the subjects, and they thought that all the range of the subjects very important. But they expressed negative attitudes in the interest of the contents. Part III : Family factor variables. It's appeared that parents' variables affected middle school boys attitude to the subjects. The boy whose father is younger and more educated recognized necessity of the subjects moil than others. And the boy whose mother is more educated or had job recognized necessity of the subjects more than others. From the findings of this study, following alternatives were presented. First, to increase middle school boys' preference and interest of the subjects, development of teaching method and revision of the contents are necessary. Second, Parents concerning to their boys is important. If their boys have more chance to the home affairs, they will have more interest of the subjects. To increase the efficiency of the subjects, parents have to offer their boys more opportunity to the home affairs. Third, Practicing is more important than theory teaching in the subjects. To improve teaching method, teachers must prepare the lecture more elaborately.

      • 6-(3,4-디클로로페닐)아미노-7-클로로-5,8-퀴놀린디온의 항진균작용 및 안전성 평가

        6-(3.4-Dichlorophenyl)amino-7-chloro-5.8-quinolinedione (RCK50) was tested for antifungal activities in mice systemically infected with Candida albicans. The therapeutic potential of RCK50 was also assessed in comparison with ketoconazole. RCK50 had ED_50 0.22±0.01 ㎎/㎏. Ketoconazole as a positive control had ED_50 6.00±1.70 ㎎/㎏. Intraperitoneally administered RCK50 at the ED_50 for 7 days and 14 days reduced Candida albicans colony count in the kidneys and liver. And administered RCK50 at the ED_50 for 14 days improved survival rates. The genotoxicities of RCK50 had been evaluated. RCK50 was negative in Ames test with Salmonella typhimurium and chromosomal aberration test in CHL cells. RCK50 did not show any clastogenic effect in mouse peripheral blood and was negative in mouse micronucleus assay. These results indicate that RCK50 has no genotoxic potential under these experimental conditions. Acute oral toxicity studies of RCK50 were carried out in ICR mice of both sexes. RCK50 did not show acute oral toxicities and LD_50 values were over 2.850 ㎎/㎏ in ICR mice.

      • 고등학교 가정 의복 만들기 실습의 적합성에 관한 조사연구

        This research was made to examine the compatability of the practice activities required in the textbook of Home Economics in the high school. The method of it was questionnaires of 18 questions to 78 teachers who are in charge of the subject in the District of Kyungsangnam. The results of them are as follows; (1) Most of teachers don't think of the practice activities of making clothes as an important job. (2) With directions of making 6 items of Korean traditional Clothes and 7 of Western clothes in the textbook, the classroom activities are restricted to the simplest items, for example, Busun(Korean traditional socks) or a skirt. Teachers' pointed out that the incompatibility of it comes from lack of sewing rooms, hours for activities, difficult requirement to follow to make clothes and obtain stuff for them as a high school student. In this study we analyzed the cause of the incompatibility of clothing construction in the high school and suggested the various ways to improve it.

        당뇨병환자에서 뇨증 Fibronectin농도의 증가에 관한 연구

        Plasma fibronectin is an α₂-glyoprotein, which is produced by vascular endothelial cells. Raised level of plasma fibronectin has been observed in diabetic patients particularly in the presence of microvascular complications. However, no available data exist about urine level of this glycoprotein in diabetic patients. We measured urine fibronectin level by ELISA methods in 54 diabetic patients who have microvascular comlications or not. The following results were obtained. 1) Urine fibronectin level(㎍/g creatinine) in diabetic group(1740.0±678.0) is increased compaired with those in normal control group(471.0±59.0). but this results were not significantly different among two groups(P>0.05). 2) Urine fibronectin level(㎍/g creatinine) was significantly increased in patients with nephropathy (6188.0±3144.0) compared with those in normal control group(471.0±59.0) and patients without nephropathy(645.0±251.0)(P<0.001). 3) There were a significant correlation between BUN, creatinine, creatinine clearance, 24hr urine total protein and urine fibronectin level in diabetic patients. Our data suggest that urine fibronectin excretion level might be used as a sensitive guide for diabectic nephropathy.

        환경친화성 계면활성제를 이용한 어병균의 살균효과

        The antibacterial effect amino acid-copper(Ⅱ) surfactant on fish pathogens was studied. Fish pathogens of Edwardsiella tarda, Vibrio anguillarum, Aeromonas hydrophila and Streptococcus sp. were selected, cultured in nutrient agar and adjusted at 2 × 10 exp (5)∼10 exp (6) CFU/㎖ in phosphate buffer saline before the addtion of amino acid-copper(Ⅱ) surfactant with different concentrations. All tested pathogens died within 1 hour with 1 ppm of amino acid-copper(Ⅱ) surfactant. In comparison with formalin and ET, amino acid-copper(Ⅱ) surfactnat was more effective in antibacterial capacity.

      • GG-14 : Alteration of PYCR1 in pelvic organ prolapse- in mice and humans

        Pelvic organ prolapse (POP) is a common, debilitating disorder affecting millions of women. The pathophysiology of POP remain poorly understood, however, recent studies focus on elastin homeostasis. Recently, mutation of PYCR1 associated with cutis laxa was reported, which showed decreased elastic fiber. We compared PYCR1 expression in the uterosacral ligament(USL) of women with POP. Since menupause and vaginal delivery are known risk factors of POP, we evaluated PYCR1 expression change after ovariectomy and vaginal dilatation in mouse vagina. The study group consisted of 43 women and the control group consisted of 18 women without POP. All patients were menopause. 6weeks old C57/BL6 mouse was used for ovariectomy and vaginal dilatation. We operated both ovariectomy or sham operation and sacrificed after 4weeks. We infused 450ul of water or no water on 3Fr silicon catheter for 40min and sacrificed at 0h, 3h, 9h and 1weeks. Expression levels of mRNA, and protein for PYCR1 were determined by real-time PCR and western blot. respectively, using USL samples from each patient and vaginal tissue from mouse. Expression of tropoelastin, elastic fiber are decreased in women with POP. Expression of PYCR1 mRNA and protein were lower in women with POP than in those without POP. PYCR1 level were decrased as POP stage increased. PYCR1 level was increased after ovariectomy in mouse. PYCR1 was decreased after vaginal delatation 3h, 9h, and those level was almost recovered after 1week. We have demonstrate that expression of PYCR1 altered by vaginal delivery and menopause, suggestinf a mechanism by which PYCR1 regulated the pelvic floor. A better understanding of the influence of PYCR1 on the homeostasis of the elastin may prove beneficial in defining the underlying etiologies of the development of POP.

        Annexin A5 as a New Potential Biomarker for Cisplatin-Induced Toxicity in Human Kidney Epithelial Cells

        Cisplatin is a member of platinum-containing anti-cancer drugs that causes cross-linking of DNA and ultimately cancer cell apoptosis. The therapeutic function of cisplatin on various types of cancers has been widely reported but the side effects have been discovered together and nephrotoxicity has been regarded as major side effect of cisplatin. To select candidates for new sensitive nephrotoxicity biomarker, we performed proteomic analysis using 2-DE/MALDI-TOF-MS followed by cisplatin treatment in human kidney cell line, HK-2 cells, and compared the results to the gene profi le from microarray composed of genes changed in expression by cisplatin from formerly reported article. Annexin A5 has been selected to be the most potential candidate and it has been identifi ed using Western blot, RT-PCR and cell viability assay whether annexin A5 is available to be a sensitive nephrotoxic biomarker. Treatment with cisplatin on HK-2 cells caused the increase of annexin A5 expression in protein and mRNA levels. Overexpression of annexin A5 blocked HK-2 cell proliferation, indicating correlation between annexin A5 and renal cell toxicity. Taken together, these results suggest the possibility of annexin A5 as a new biomarker for cisplatin-mediated nephrotoxicity.

      • OB-19 : The effect of hypoxia on the placental productions of soluble endoglin, soluble fms-like tyrosine kinase receptor-1 in villous explants from normal pregnancy and preeclampsia

        목적: Increased production of antiangiogentic factors, soluble endoglin(sEng) and soluble fms like tyrosine kinase receptor-1(sFlt-1) by placenta contributed to the pathophysiogy in preeclampsia. Our objective was to determine the differences in sEng and sFlt-1 prodution by villous explant( VE) between normal and preeclampsia placentas cultured under hypoxic conditions. 방법: The study was approved by the institutional review board. Human placental tissues were obtained from normal or preeclampsia pregnancies after cesarean section. Villous explants from normal(n=5) and preeclamptic pregnancies(n=6) were cultures under normal oxygen conditions of 21% O2 at 37°C in a humidified incubator for 48 hours and then divided by normal oxygen group or hypoxic conditions (1% O2) for 6 and 24hours. Villous explants productions of sEng, sFlt-1 were determined by ELISA. The ELISA kits for sEng and sFlt-1 were purchased from R&D systems, Inc. Plates were read at 450nm by autoplate reader. For each assay, all samples were assayed in duplicate in the same plate 결과: The productions of sEng and sFlt-1 by villous explants that were cultures under normoxia and hypoxia conditions are shown in Fig 1. Under normoxia(0h), villous explants from preeclampsia produced significantly more sEng and sFlt-1 than from normal. In preeclampstic explants, after hypoxia, the secretion of sEng were increased at 6 hour and 24hour, although in normal explants, after hypoxia, the secretion of sEng were not changed. In normal explants, the low level of sFlt-1 was increased after hypoxia. In preeclamptic explants, after hypoxia, the sFlt-1 were decreased dramatically at 6hours, and recovered almost same level of 0 hour at 24hour. 결론: Preeclampsia villous explants produce more sEng and sFlt-1 than normal villous explants. Hypoxia conditions promote sEng, but reduce sFlt-1 at early hypoxia and promote it at late hypoxia, productions by preeclampsia villous explants
