        SOD2와 NDPK2 유전자 집적에 의한 페튜니아의 아황산가스 저항성 증진

        유전자 집적에 의해 아황산가스 저항성 증진 페튜니아를 개발하고자 비생물적스트레스 저항성 유전자로 널리 알려진 SOD2 와 NDPK2유전자가 각각 도입된 SOD2 형질전환 계통 SOD2-2-1-1-35(T₄)[S(T₄)]와 NDPK2 형질전환 계통 NDPK2-7-1(T₂)[N7-1(T₂)]간 상호교잡을 실시하여 교잡 제1세대를 획득하였다. 교잡 제1세대 중 SOD2와 NDPK2유전자가 모두 집적된 개체 획득율은 32.1-73.0%이었고, SOD2와 NDPK2유전자가 모두 집적된 개체는 SOD2 또는 NDPK2유전자가 단독 도입된 개체에 비해 아황산가스 30ppm처리 피해율이 2.6-5.1배 낮아 아황산가스 저항성이 증진되었음을 확인하였다. 또한 아황산가스 저항성이 증진된 교잡 제1세대를 자가수분에 의해 세대진전 시켜 획득한 교잡 제2세대의 아황산가스 저항성 검정을 통하여 교잡 제1세대에서의 아황산가스 저항성이 후대에서도 안정적으로 발현함을 확인하였다. 또한, 광합성 효율 증진 조사를 통해 아황산가스 저항성이 증진된 SOD2와 NDPK2유전자 집적 후대가 아황산 가스에 대한 스트레스를 덜 받고 있음을 확인하였고, RT-qPCR분석을 통해 SOD2와 NDPK2유전자 집적 후대의 아황산가스 저항성 증진이 SOD2와 NDPK2 유전자 집적에 의한 효과임을 확인하였다. The aim of this study was to develop a transgenic petunia with enhanced resistance to sulfur dioxide (SO₂) gas by stacking two genes, SOD2 and NDPK2, which are both known to confer resistance to abiotic stresses. The first-generation hybrids (TF1) were obtained through reciprocal crosses between an SOD2-transgenic line SOD2-2-1-1-35(T₄)[S(T₄)] and an NDPK2- transgenic line NDPK2-7-1(T₂)[N7-1(T₂)]. Approximately 32.1-73.0% of the first-generation hybrids (TF₁) carried both SOD2 and NDPK2 genes. These hybrids showed 2.6 and 5.1 times less damage than hybrids carrying only SOD2 or NDPK2 genes, respectively, when they were treated with SO₂ gas at 30 ppm. This confirmed that the heterozygous hybrids were more resistant to SO₂ than the hybrids carrying either one of the resistance genes. Second-generation hybrids (TF₂) were obtained by selfing the TF₁ individuals. We confirmed the expression of the stacked genes in the TF₂ hybrids by phenotypic observation of their response to SO₂ gas at 30 ppm as well as using RT-qPCR and photosynthetic efficiency.

        마이크로그로브 형태에 따른 평활근 세포 운동 측정

        Purpose: The spreading, orientation, and chemotaxis the gradient of a chemoattractant of smooth muscle cells (SMCs) were studied on the micro-grooved substrata by the light, fluorescence and scanning electron microscopy. Method: Vertical-walled grooves were produced in silicon wafers by the micromachining technique. All grooves were 4~20㎛ deep and 10~80㎛ wide, SMCs were cultured on each microgroove and examined under stereo-microscope. Result: Cell clusters were markedly oriented by all the grooved substrata examined. Tim-lapse images acquired from CCD (Charge Coupled Device) showed that the grooves directed the migration of SMCs. There was no prominent difference in the migration speed of SMCs according to the grooves. All the cytoskeletal fibers were reoragnized in the same direction with grooves. Especially the alignments of microtubule and intermediate filaments were distinguished in the SMCs on the micro grooves. Conclusion: These results could be applied to the analysis of vascular restenosis and the development of artificial blood vessels.

        정보교육과 학교도서관에 관한 연구

        Creative thinking in education is a common assumption to be accomplish in this information age. Information education can contribute to build the ability to think creatively. The Author explored how information education conduces the creative thinking ability that is necessary to the development of independent and competent study for students themselves. The writer also expressed the integrated education makes students think synthetically and synthetic educational experience derives creative thinking. She based her arguments upon the theory of the psychology of memory and the Piaget's cognitive structure. To increase the effects of information education, it is necessary to integrate the curriculums and learning method of the information education and those of other areas of learning, i.e., languages, literatures, social sciences, sciences, Mathematics, etc. Here, author asserted that the teaching of information skill within classroom curriculums for all subject areas can make the integrated effects on carious classroom curriculums. On the basis of the findings of this study, the author recommended that every school needs to prepare enough books and other media for the students to drill information skill. Consequently, to build creative thinking ability for the students, librarians, classroom teachers and school principals who have influence on the information education, have to cooperate to initiate integrated information education for the student

        There exists a major impediment to learning electricity which may illuminate why direct presentation of scientists' ideas is too aggressive a strategy. This impediment lies in thinking that electricity is used up and in envisaging the light bulb as a consumer. This view is based on a powerful idea which summarises much of our experience; it declares that 'things get used up', 'you cannot get something for nothing', and 'intuitive thinking'. To replace this view with scientific view, we made the analogy model. It is suggested that the analogy model is more effective in teaching electricity circuits than conventional teaching method.

        Microparticles for oral drug delivery were prepared by freeze drying the micelle solutions of amphiphilic copolymers comprising water soluble drugs such as 5-fluorouracil and rose bengal in the aqueous phase. It was found the release behavior from thus prepared microparticles followed the typical diffusion controlled mechanism. However, the microparticles exhibited a significant initial burst effect due to the high solubility of drug molecule in the aqueous phase. The incorporation of hydrophilic colloids including gelatin or xanthan gum in the aqueous phase has incorporation of hydrophilic colloids including gelatin or xanthan gum in the aqueous phase has effectively suppressed the instant release by gelling hydrophilic segments of the matrix.

        일 병원 간호사의 임파워먼트, 업무성과 및 직무만족의 관계

        Purpose: The purpose of this study was to investigate the extent of empowerment, task performance and job satisfaction perceived by hospital nurses and the relationship among those variables. Methods: The sample consisted of 261 nurses from a university affiliated hospital. Data were collected with self-administrated questionnaires including empowerment, performance and job satisfaction and analyzed using mean, standard deviation, ANOVA and Scheffe' test. Results: The mean scores of variables were as follows. Empowerment was 4.6472, task performance was 3.7814, and job satisfaction was 3.1240. The older, more educated and more experienced nurses showed more empowered and higher job performed than the younger, less educated and less experienced nurses. Empowerment had a positive correlation with task performance and job satisfaction. Conclusions: The results imply that hospital should develop the empowerment program for nurses to be more job performed and satisfied.

        The female consumer whose ages are between 20 and 29 is segmented according to benefit, similarity table is composed of the brand image in consumer perceptual dimension and the advertising image of that brand, and the purpose of this study is to make positive brand image, to require efficient advertising expression approaching brand perception, brand preference, brand loyalty. This study is a purpose like this ; First The female consumer whose ages are between 20 and 29 is segmented according to benefit. Second, Similarity table is composed of underwear brand image and advertising image by the perception of consumers. Third, Segmentation group is composed, analyzed according to composing of brand image and avdertising image, it is to require avdertising expression. The subject of this study wear gathered into 202 female in 20’s As measurement - implement, questionaire written by investigator wear designed for literature and precedence study. The analysis of data used fator analysis, principal component analysis, cluster analysis, ANOVA, T-test, F-test by using SAS. The result are as follows ; 1. Benefit sought by consumer wear found to included three different factor - brand value, fashionability and practicality on underwear. As a result of subdividing the consumers, three distinctive groups wear found on the basis of benefit factor - fashion oriented group, brand value oriented group, practicality oriented group. 2 Similarity table of brand image and advertising image by the perception of consumers was shown the distinction as each segmented group.

        평생학습사회의 기반구축과 학교도서관매체 센터에 관한 연구

        학교도서관매체센터가 학교교육에 긍정적 영향을 줄 뿐만 아니라 평생학습사회의 하부구조임을 밝히는 것이 본 논고의 목적이다. 평생학습사회는 사회구성원의 학습욕구와 이를 지원할 수 있는 학습환경이 구비됨으로써 형성될 수 있다. 그리고 학교도서관매체센터는 사회구성원들의 학습욕구를 형성시켜주는 産室로 규명된다. 민주주의 사회에서 사회구성원들이 학습욕구를 강하게 가질 때 사회환경이 학습하기에 적절하고 편리하도록 변형된다는 것은 자연스러운 논리이다. 결과적으로 학교도서관매체센터는 사회구성원의 학습욕구를 만들어 내고 또 이에 응하는 사회환경을 조성하는 원동력을 조성해 낸다고 할 수 있다. 그러므로 필자는 학교도서관매체센터가 평생학습사회의 중요한 하부구조라고 밝히고 있다 The purpose of this study is to assert the fact that School Library Media Center(SLMC) is the most important body which influences the whole process of learning in school and at the same time it functions as a very essential infrastructure of acquiring learning in the society. The writer analysed the studies on learning outcomes across a range of educational conditions and settings. She found that learning outcomes are influenced positively by the educational conditions and settings that accomodate the student diversity and individualization, and learning materials for their individual needs. It means that the outcomes of effective learning are influenced by using learning materials of individual student guided by teachers and teacher librarians. In case, school education furnishes desirable programs of SLMC and use it properly, students can have enough experiences what and how they learn by using library materials during their school days. As school days are in their early days of their lifetime, they are easy to carry their way of learning with them even after they finish school. The members of society who are accustomed to use library materials during their school days will have learning needs for their better life. The writer wants to call it the learning needs of society that school produces. A learning society is composed of two factors. One is the learning needs of the people and the other is the environment to meet them. SLMC produces learning needs, and it meets to the needs of student learning. Consequently it can be said SLMC is the infrastructure for learning society. The author pointed out that the Educational Reformation Draft of current Government does not mention about learning materials prerequisite to enhance student learning outcomes, especially in relation to their creativity. She concluded educators of every level of school including government officials in charge of education have to think and do something about reformation of School Library Media Center for the fulfillment of the goal of Korean Educational Reformation Draft announced by the Educational Reformation Committee May 31, 1995

      • 그레이브스병에서 방사성 요오드 치료에 따른 갑상선 자극 호르몬 수용체 결합억제 면역글로불린(Thyrotropin Binding Inhibitory Immunoglobulin, TBII)의 변화

        Prediction of thrapeutic response to radioiodine in Graves' disease is poorly understood. Although thyrotropin binding inhibitor immunoglobulin(TBII) level is a strong index for relapse after antithyroid drug medication, conflicting results are reported regarding its prognostic significance in Graves' disease treated with RAI. This study is dengned to evaluate possible relationship between post-treatment hypothyroidism and TBII in Graves' disease treated with RAI. Fourty two patient with Graves' disease after radioiodine treatment were studied retrospectively. The subject were divided into hypothyroid group and euthyroid or hyperthyroid group. We evaluated the association of hypothyroidism and TBII with radioiodine treatment dose. The mean age of hypothyroid group was 48±11 years and euthyroid or hyperthyroid group was 47±12 years. There was no difference in two groups. And there was no significant difference in post- treatment TBII between two groups(49.9±28.5%, 29.9±14.3%, p-value >0.05). The treatment-dose had no influence on post-treatment thyroid state. Euthyroid or hyperthyroid group was done with 13.6±6.9mCi and hypothyroid group was 17.0±10.4mCi(p-value > 0.05). TBII had no prognostic significance on long-term hypothyrodism in Graves' disease treated with radioiodine. And, treatment-dose had no influence on post-treatment thyroid state.
