Study on Method of Sperm Aspiration and Injection into an Oocyte in Intracytoplasmic Sperm Injection(ICSI) Immobilization of spermatozoa prior to intracytoplasmic sperm iniection(ICSI) sometimes results in crooked tail and this makes it difficult to aspirate sperm into an injection pipette tail first. Head-first sperm aspiration into an injection pipette avoid this problem due to the bigger size of the sperm head. The effect of head or tail-first sperm injection into an oocyte on fertilization cleavage, percentage of grade I embryos and development to blastocyst stage in ICSI program has been studied. A single living immobilized spermatozoa from oligoasthenozoospermic patient was injected into an oocyte head-first or tail-first according to the treatment. Eighteen hours after microinjection, oocytes ware inspected for survival and fertilization Fertilized oocytes with two pronuclei were cultured in 30μl drop of mHTF supplemented with 10% heat-inactivated follicular fluid(FF) at 37℃. On day 2. embryo transfer was performed with cleaved embryos. The remaining 2-8 cell stage embryos were co-cultured with BRL cells in mHTF + 10% FF for 72 hours and the developmental stage was observed. The data were analyzed by Analysis of Variance. A total of 164 oocytes from 36 cycles were assigned to earth treatment and ICSI was performed(88 head-first, tail-first). The rates of normal fertilization were 81.8% and 76.3% for head-first and tail-first, respectively. Of the fertilized oocytes, the percentage of cleaved embryos and the percentage of grade 1 embryo among cleaved embryos were 88.9% and 68.8%, 93.1% and 74.1% for head-first and tail-first, respectively. Of the 2-8 cell embryos cultured, 44.4%(16/36) and 50.0%(10/20) for head first and tail first, respectively developed to blastocyst stage. There were no differences in fertilization, cleavage, rates of grade 1 embryos, and development to blastocyst stage. In conclusion, head-first or tail-first sperm injection into an oocyte in ICSI program does not affect fertilization and subsequent embryo development to blastocyst stage in vitro.

<P>Neurofibromatosis type 1 (NF1) caused by <I>NF1</I> gene mutation is a commonly inherited autosomal dominant disorder. Malignant peripheral nerve sheath tumors (MPNSTs), a type of aggressive sarcoma, are a major cause of mortality in NF1 patients. The malignant transformation of benign plexiform neurofibromas (PNs) to MPNSTs is a marked peculiarity in NF1 patients, yet the pathogenesis remains poorly understood. We found that an actin-associated protein transgelin (SM22) was highly expressed in <I>NF1</I>-deficient MPNST tissues compared to <I>NF1</I>-deficient PN tissues using immunohistological staining and primary cultured MPNST cells in western blot analysis. We further found that this transgelin upregulation was caused by increased transcriptional expression of the <I>TAGLN</I> gene encoding transgelin. Comparison of DNA methylation values in the promoter and subpromoter regions of the <I>TAGLN</I> gene in three types of <I>NF1</I>-deficient primary-cultured cells, derived from an NF1 patient’s normal phenotype, a benign PN and MPNST tissues, revealed that the <I>TAGLN</I> gene was hypomethylated in the MPNST cells. Next, to determine the functional role of transgelin in MPNST pathogenesis, we manipulated the <I>TAGLN</I> gene expression and investigated the alteration of the RAS-mitogen-activated protein kinase (MAPK) signaling pathway in the normal-phenotypic and malignant tumor cells. The downregulation of <I>TAGLN</I> expression in <I>NF1</I>-deficient MPNST tumor cells through the treatment of the small interfering RNA resulted in a decrease in the RAS activation (GTP-RAS) and the downstream ERK1/2 activation (phosphorylated ERK1/2), while the overexpression of <I>TAGLN</I> in normal-phenotypic <I>NF1</I>-deficient cells caused an increase in RAS and ERK1/2 activation. These results indicate that upregulation of transgelin caused by hypomethylation of the <I>TAGLN</I> gene is closely involved in tumor progression in NF1.</P>

AlCrNO cermet films were prepared on aluminum substrates using a DC-reactive magnetron sputtering method and a water-cooled Al:Cr target. The Al<sub>2</sub>O<sub>3</sub>/AlCrNO (LMVF)/AlCrNO (MMVF)/AlCrNO (HMVF)/Al/substrate of the 5 multi-layers was prepared according to the Ar and (N<sub>2</sub> + O<sub>2</sub>) gas-mixture rates. The Al<sub>2</sub>O<sub>3</sub> of the top layer is the anti-reflection layer of triple AlCrNO (LMVF)/AlCrNO (MMVF)/AlCrNO (HMVF) layers, and an Al metal forms the infrared reflection layer. In this study, the crystallinity and surface properties of the AlCrNO thin films were estimated using X-ray diffraction (XRD) and field-emission scanning electron microscopy (FESEM), while the composition of the thin films was systematically investigated using Auger electron spectroscopy (AES). The optical properties of the wavelength spectrum were recorded using UH4150 spectrophotometry (UV-Vis-NIR) at a range of 0.3 μm to 2.5 μm.

Lymphangioma is a benign tumor of lymphatic origin. Lymphangioma can occur anywhere in the body and only rarely affects the intestinal tract. Most intestinal lymphangiomas are asymptomatic and detected incidentally at autopsy or surgery. Occasionally, they may be large enough to present as a mass to cause obstruction or intussusception. Recently, we examined a case of a 27-year-old woman who complained right upper quadrant abdominal pain, increased bowel sound and weight loss. By surgical resection after the double contrast barium enema and colonoscopy, we could confirm cystic lymphangioma of the transverse colon. So we report this case with brief review of relevant literature.

석탄가스화 복합발전(IGCC)에서 석탄 가스화 기술이 전 공정의 성능에 큰 영향을 미치는 중요한 요소이다. 연료 및 산화제의 공급방식, 가스화기의 기본 구조, 벽면의 구성 방식, 용융 슬랙 및 생산되는 합성가스 배출 방식 등에 따라 가스화의 성능이 영향을 받는다. IGCC plant의 정확한 성능 해석을 위해서는 석탄가스화기 공정 모델의 정밀도를 높일 필요성이 있다. 기존의 열병합 발전 사이클 해석에서 적용되었던 열 및 물질정산과 평형계산 방식을 통하여 석탄가스화기 공정을 해석하는 방법을 확인, 정리하고 이를 개선하기 위한 절차 및 방안을 제시하고자 한다. 가스화기 내부 공정을 크게 탈휘발과 가스화의 단계로 구분하여 가스화기 출구조건을 예측하였으며, ASPEN PLUS를 이용한 공정해석을 실시하였다. 가스화기 출구에서의 합성가스는 주생성가스인 CO, H₂를 위주로 하여 조성을 얻을 수 있고, 그 결과들을 선행연구들과의 비교를 통하여 가스화기 모델의 분석을 실시한다. 그리고 가스화기 해석의 정밀도를 높이기 위한 향후 고려될 가스화기 모델에 관하여 논의한다.
