The agarase gene gaa16a was identified from a draft genome sequence of Gilvimarinus agarilyticus JEA5, an agar-utilizing marine bacterium. Recently, three agarase-producing bacteria, G. chinensis, G. polysaccharolyticus, and G. agarilyticus, in the genus Gilvimarinus were reported. However, there have been no reports of the molecular characteristics and biochemical properties of these agarases. In this study, we analyzed the molecular characteristics and biochemical properties of agarases in Gilvimarinus. Gaa16A comprised a 1,323-bp open reading frame encoding 441 amino acids. The predicted molecular mass and isoelectric point were 49 kDa and 4.9, respectively. The amino acid sequence of Gaa16A showed features typical of glycosyl hydrolase family 16 (GH16) β-agarases, including a GH16 domain, carbohydrate-binding region (RICIN domain), and signal peptide. Recombinant Gaa16A (excluding the signal peptide and carbohydrate-binding region, rGaa16A) was expressed as a fused protein with maltose-binding protein at its N-terminus in Escherichia coli. rGaa16A had maximum activity at 55°C and pH 7.0 and 103 U/mg of specific activity in the presence of 2.5 mM CaCl2. The enzyme hydrolyzed agarose to yield neoagarotetraose as the main product. This enzyme may be useful for industrial production of functional neoagaro-oligosaccharides.

Leptolyngbya sp. KIOST-1 (LK1) is a newly isolated cyanobacterium that shows no obvious cytotoxicity and contains high protein content for both human and animal diets. However, only limited information is available on its toxic effects. The purpose of this study was to validate the safety of LK1 powder. Following Organisation for Economic Co-operation and Development (OECD) guidelines, a single-dose oral toxicity test in Sprague Dawley rats was performed. Genotoxicity was assessed using a bacterial reverse mutation test with Salmonella typhimurium (strains TA98, TA100, TA1535, and TA1537) and Escherichia coli WP2 uvrA, an in vitro mammalian chromosome aberration test using Chinese hamster lung cells, and an in vivo mammalian erythrocyte micronucleus test using Hsd:ICR (CD-1) SPF mouse bone marrow. After LK1 administration (2,500 mg/kg), there were no LK1-related body weight changes or necropsy findings. The reverse mutation test showed no increased reverse mutation upon exposure to 5,000 μg/plate of the LK1 powder, the maximum tested amount. The chromosome aberration test and micronucleus assay demonstrated no chromosomal abnormalities and genotoxicity, respectively, in the presence of the LK1 powder. The absence of physiological findings and genetic abnormalities suggests that LK1 powder is appropriate as a candidate biomass to be used as a safe food ingredient.
혁신역량과 혁신성과에 영향을 주는 요인에 관한 연구: 해외시장진출 중소기업을 대상으로

본 연구에서는 해외에 진출한 중견기업을 대상으로 혁신성공요인과 혁신성과를 알아보고자 하였다. 보다 구체적으로는 경영자, 조직, 종업원 차원의 요인과 관련하여 리더십, 운영시스템, 변화주도행위를 혁신역량에의 영향 변수로 설정하였으며 혁신역량이 혁신성과에 미치는 영향을 실증 연구를 통해 파악해 보고자 하였다. 더불어 혁신역량에의 영향요인과 혁신성과에 미치는 영향이 본국과 해외진출국가 에서 어떠한 차이가 나는지를 알아보았다. 실증 분석 결과 혁신영향요인 모두 혁신역량에 긍정적 영향을 미치고 있었으며, 혁신역량은 혁신성과에 긍정적 영향을 미치고 있었다. 영향력의 정도에 있어서는 운영시스템, 리더십, 변화주도행위의 순으로 나타났다. 국가간 차이에 있어서는 중국의 경우 운영시스템, 리더십, 변화주도행위의 순이나 큰 차이는 없는 것으로 나타났다. 한국의 경우는 운영시스템과 리더십은 긍정적 영향을 미치나, 종업원의 개별적 변화주도행위는 혁신역량에 영향을 미치지 않는 것으로 나타났다. 본 연구 결과는 해외 진출 기업에게 있어서는 현지 특성에 맞는 혁신역량 강화 노력이 필요하다는 점과 한국의 경우 보다 고도화된 혁신역량 및 혁신성과의 창출을 위해서 종업원 개인의 변화주도행위를 이끌어 내도록 하는 추가적인 활동이 요구됨을 보여주고 있다. The purpose of this study is to investigate the factors affecting innovation capability and innovation performance of midsize company entering overseas market. More specifically, in terms of managerial, organizational, and employee - level factors, we set leadership, operating system, and taking charge at work as variables influencing innovation capability and tried to understand the effect of innovation capability on innovation performance through empirical studies. In addition, we examined the differences in the influence factors on innovation capability and the effect on innovation performance in the home country and overseas countries. As a result of the empirical analysis, all of the influence factors had a positive effect on the innovation capability and the innovation capability had a positive effect on the innovation performance. The degree of influence was in the order of operating system, leadership and taking charge at work. In China, there is no significant difference in the order of operating system, leadership and taking charge at work. In the case of Korea, operating system and leadership have a positive effect, but the employee`s taking charge at work does not affect innovation capability. The results of this study are as follows. First, it is necessary to enhance innovation capability in accordance with local characteristics. In Korea, there are additional activities to induce employee`s taking charge at work is required for improved innovation capability and innovation performance.

L-Asparaginase (E.C. 3.5.1.1) is an enzyme involved in asparagine hydrolysis and has the potential to effect leukemic cells and various other cancer cells. We identified the Lasparaginase gene (L-ASPG86) in the genus Mesoflavibacter, which consists of a 1,035 bp open reading frame encoding 344 amino acids. Following phylogenetic analysis, the deduced amino acid sequence of L-ASPG86 (L-ASPG86) was grouped as a type I asparaginase with respective homologs in Escherichia coli and Yersinia pseudotuberculosis. The L-ASPG86 gene was cloned into the pET-16b vector to express the respective protein in E. coli BL21 (DE3) cells. Recombinant L-asparaginase (r-L-ASPG86) showed optimum conditions at 37-40oC, pH 9. Moreover, r-L-ASPG86 did not exhibit glutaminase activity. In the metal ions test, its enzymatic activity was highly improved upon addition of 5 mM manganese (3.97-fold) and magnesium (3.35-fold) compared with the untreated control. The specific activity of r-LASPG86 was 687.1 units/mg under optimum conditions (37°C, pH 9, and 5 mM MnSO4).

<P><B>Abstract</B></P><P>The regulation of transcriptional activation is an essential and critical point in gene expression. In this study, we describe a novel transcription factor activator protein-1 (Ap-1) gene from disk abalone <I>Haliotis discus discus</I> (AbAp-1) for the first time in mollusk. It was identified by homology screening of an abalone normalized cDNA library. The cloned AbAp-1 consists of a 945 bp coding region that encodes a putative protein containing 315 amino acids. The AbAp-1 gene is composed of a characteristic Jun transcription factor domain and a highly conserved basic leucine zipper (bZIP) signature similar to known Ap-1 genes. The AbAp-1 shares 46, 43 and, 40% amino acid identities with fish (<I>Takifugu rubripes</I>), human and insect (<I>Ixodes scapularis</I>) Ap-1, respectively.</P><P>Quantitative real time RT-PCR analysis confirmed that AbAp-1 gene expression is constitutive in all selected tissues. AbAp-1 was upregulated in gills after bacteria (<I>Vibrio alginolyticus, Vibrio parahemolyticus</I> and <I>Lysteria monocytogenes</I>) challenge; and, upregulated in hemocytes and gills by viral hemorrhagic septicemia virus (VHSV) challenge. Shell damage and tissue injury also increased the transcriptional level of Ap-1 in mantle together with other transcription factors (NF-kB, LITAF) and pro-inflammatory TNF-α. All results considered, identification and gene expression data demonstrate that abalone Ap-1 is an important regulator in innate immune response against bacteria and virus, as well as in the inflammatory response during tissue injury. In addition, stimulation of Ap-1 under different external stimuli could be useful to understand the Ap-1 biology and its downstream target genes, especially in abalone-like mollusks.</P>
제7회 광역단체장 투표선택 결정요인 분석: 보수의 분화와 중앙정치의 영향력을 중심으로

본 논문은 2018년 제7회 전국동시지방선거 유권자 설문조사를 분석하여 광역단체장 선거에서 투표결정 요인을 분석하였다. 분석의 초점은 크게 중앙정치의 영향력, 특히 대통령 국정지지가 투표선택에 미친 영향과 광역지방자치단체장의 직무수행 평가가 투표선택에 미친 영향력 비교와 보수유권자의 분화를 살펴보는 것에 맞추어졌다. 분석결과, 제7회 지방선거에서 대통령의 직무평가는 민주당 후보에 대한 투표에 긍정적인 영향을 끼친 반면, 지방자치단체장의 직무수행 평가는 유의미한 영향력을 보이지 않았다. 자유한국당에 투표한 유권자와 바른미래당에 투표한 유권자를 비교한 결과, 바른미래당 투표에 영향을 미친 것은 주로 정책적 요인보다는 정당 호감도와 같은 감정적인 요인이었다. This paper analyzes the voting decision in the 7th gubernatorial elections. The focus of analysis is largely on the influence of national politics on voting choice and on the comparison between conservative voters. As a result of the analysis, the evaluation of the president’s performance shows a positive effect on the support for the ruling party, whereas the evaluation of the job performance of the local governor shows no significant impact. As a result of comparing the voters who voted in the Liberty Korea Party and the voters who voted in the Bareunmirae Party, an emotional factor has more influence than policy preferences on the support for the Bareunmirae Party.
