        In vivo study for the hemostatic efficacy and foreign body reaction of a new powder-type polysaccharide hemostatic agent

        Purpose: Various hemostatic agents have been introduced in therapy as postoperative bleeding is a poor prognostic factor for postoperative outcomes. These products can be divided into those that directly promote the hemostatic cascade and those that physically form a barrier by absorbing blood. The latter, powder-type hemostatic agents have the advantages of being inexpensive and more absorbable with less foreign body reactions (FBRs) and are applicable to a relatively wide area. This study was conducted to verify the safety and efficacy of a newly invented polysaccharide product (OOZFIX, Theracion Biomedical), which improves blood absorption and hemostatic effects. Methods: Two separate animal experiments were performed. The first evaluated FBRs histologically at 3 days, 2 weeks, and 4 weeks, after implantation of OOZFIX in rats, and the second compared hemostatic performance of OOZFIX and Arista AH (Bard) in the porcine liver punch biopsy model. Results: We found minimal FBRs in the 3-day group and no reactions in both the 2-week and 4-week groups after implantation of hemostatic agents. The time to hemostasis of OOZFIX was not significantly different from that of Arista AH (median [interquartile range]: 9 [6–10] minutes vs. 8 [6–10] minutes, respectively; P = 0.522). When comparing the serial bleeding grade tendency, there was no statistical difference between OOZFIX and Arista AH (P = 0.656). Conclusion: OOZFIX caused a minimal FBR that disappeared within 2 weeks in vivo, and its hemostatic performance was comparable with that of an existing agent, Arista AH. Further clinical studies are required in the future.

        Occupational hearing loss became the leading occupational disease by constioution 56% of all work-related diseases in Korea. However, the prevalance rates showed wide variations among the specific medical examination providers and were very low(0.8-1.4%) compared with those 8.4% in Japan. The sources of these variations was regarded in the preview study as 1) Employee audometric estings were primarily conducted by nurses and nurse-aids who had no formal training for audiometry; 2) althought audiometric testing equipment used by the spedific medical examination providers, it should be periodically calibrated, a significant number of them were not properly calibrated : 3) Initial audiometric testings were conducted at any available spaces in workplace or at the testing room without considering bachground noise level. One of solutions that can be done for sloving surrent noise related problems is to provide a noise related manual for the occupational health managers including doctors, nurses and hyginests. This study was done to provide such a manual. The main core of the manual was hearing conservation program. And the contents of the manual was the same as the main frame of hearing conservation program such as noise survey, provided of the diagram showing the core contents of each factor of the hearing conservation program. Hopefully, this manual will be the references of the standardized performances of the specific medical examination providers.

        방사선조사가 악하선 미세혈관과 내피세포에 미치는 영향에 관한 실험적 연구

        The purpose of this study was to investigate the irradiation effects on the capillary and endothelial cell in the submandibular gland. Sprague-Dawley strain male rats were singly irradiated to their neck region with the dose of 5Gy and 10Gy by 6MV X-irradiation and sacrificed on the 6 hours, 12 hours, 1,3,7, and 14 days after irradiation. The authors observed the histological changes of the capillary at H & E and PAS staining under a light microscope, and also observed the ultrastructural changes of the endothelial cell using a transmission electron microscope. The obtaining results were as follows: 1. In the light microscopic examination, the capillary density was slightly increased on the 1 day after irradiation, and increased until the 7 days after irradiation. After then, capillary density was apparently decreased. 2. The reaction to PAS staining at acinar cells was decreased on the 6 hours after irradiation, and recovered on the 7 days after irradiation. But reaction was decreased on the 14 days after irradiation again, after then, gradually recovered with days. 3. In the transmission electron microscopic examination, mild proliferation of cytoplasmic process of the endothelial cell and reduction in luminal size were observed just after irradiation. After then, nuclear degeneration, marked proliferation of cytoplasmic process, thickened basal lamina, and numerous cytoplasmic vesicles were observed on the 1 day after irradiation. These changes were recovered to normal on the 14 days after 5GY group, but not with 10GY irradiation group. And destruction of endothelial cell and loss of basal lamina were not observed in both groups. 4. From the above results, reduction in luminal size, proliferation of cytoplasmic process and thickening of basal lamina were observed as the irradiation effects on the capillary and endothelial cell of the submandibular gland. And also, these changes may induce increase in capillary number and endothelial permeability by means of increase of cytoplasmic vesicle formation. The changes appeared earlier and more prominent in 10Gy irradiated group than in 5Gy irradiated group.

        Requirement of EGF Receptor Kinase for Signaling by Calcium - Induced ERK Activation and Neurite Outgrowth in PC12 Cells

        Membrane depolarization in PC12 cells induces calcium influx via an L-type voltage-sensitive calcium channel (L-VSCC) and increases intracellular free calcium, which leads to tyrosine phosphorylation of epidermal growth factor (EGF) receptor and the associated adaptor protein, Shc. This activated EGF receptor complex then can activate mitogen-activated protein (MAP) kinase, as in nerve growth factor (NGF) receptor activation. In the present study, we investigated the role of EGF receptor in the signaling pathway initiated by membrane depolarization of PC12 cells. Prolonged membrane depolarization induced phosphorylation of extracellular signal-regulated kinase (ERK) within 1 min in undifferentiated PC12 cells. Pretreatment of PC12 cells with the calcium chelator EGTA abolished depolarization-stimulated ERK phosphorylation, but NGF-induced phosphorylation of ERK was not affected. The chronic treatment of phorbol ester, which down-regulated the activity of protein kinase C (PKC), did not affect the phosphorylation of ERK upon depolarization. In the presence of an inhibitor of EGF receptor, neither depolarization nor calcium ionophore increased the level of ERK phosphorylation. These data imply that the EGF receptor is functionally necessary to activate ERK and neurite outgrowth in response to the prolonged depolarization in PC12 cells, and also that PKC is apparently not involved in this signaling pathway.

      • N-terminal arginylation generates a bimodal degron that modulates autophagic proteolysis

        <P>The conjugation of amino acids to the protein N termini is universally observed in eukaryotes and prokaryotes, yet its functions remain poorly understood. In eukaryotes, the amino acid L-arginine (L-Arg) is conjugated to N-terminal Asp (Nt-Asp), Glu, Gln, Asn, and Cys, directly or associated with posttranslational modifications. Following Ntarginylation, the Nt-Arg is recognized by UBR boxes of N-recognins such as UBR1, UBR2, UBR4/p600, and UBR5/EDD, leading to substrate ubiquitination and proteasomal degradation via the N-end rule pathway. It has been a mystery, however, why studies for the past five decades identified only a handful of Nt-arginylated substrates in mammals, although five of 20 principal amino acids are eligible for arginylation. Here, we show that the Nt-Arg functions as a bimodal degron that directs substrates to either the ubiquitin (Ub)proteasome system (UPS) or macroautophagy depending on physiological states. In normal conditions, the arginylated forms of proteolytic cleavage products, D101-CDC6 and D1156-BRCA1, are targeted to UBR box-containing N-recognins and degraded by the proteasome. However, when proteostasis by the UPS is perturbed, their Nt-Arg redirects these otherwise cellularwastes tomacroautophagy through its binding to the ZZ domain of the autophagic adaptor p62/STQSM/Sequestosome-1. Upon binding to the Nt-Arg, p62 acts as an autophagic N-recognin that undergoes self-polymerization, facilitating cargo collection and lysosomal degradation of p62-cargo complexes. A chemical mimic of Nt-Arg redirects Ub-conjugated substrates from the UPS to macroautophagy and promotes their lysosomal degradation. Our results suggest that the Nt-Arg proteome of arginylated proteins contributes to reprogramming global proteolytic flux under stresses.</P>

        Can Proopiomelanocortin Methylation Be Used as an Early Predictor of Metabolic Syndrome?

        <P><B>OBJECTIVE</B></P><P>The objectives of this study were to compare early predictive marker of the metabolic syndrome with proopiomelanocortin (POMC) methylation status and to determine the association among birth weight, ponderal index, and cord blood methylation status.</P><P><B>RESEARCH DESIGN AND METHODS</B></P><P>We collected pregnancy outcome data from pregnant women, cord blood samples at delivery, and blood from children (7–9 years old; <I>n</I> = 90) through a prospective cohort study at Ewha Womans University, MokDong Hospital (Seoul, Korea), from 2003–2005. POMC methylation was assessed by pyrosequencing. We divided subjects into three groups according to cord blood POMC methylation: the low methylation (<10th percentile), mid-methylation, and high methylation (>90th percentile) groups. We analyzed the association of POMC methylation status at birth with adiposity and metabolic components using ANCOVA and multiple linear regression analysis.</P><P><B>RESULTS</B></P><P>Birth weights (<I>P</I> = 0.01) and ponderal indices (<I>P</I> = 0.01) in the high POMC methylation group were significantly lower than in the mid-POMC methylation group. In terms of metabolic components of childhood, blood triglycerides (57.97, 67.29 vs. 113.89 mg/dL; <I>P</I> = 0.03, 0.01) and insulin (7.10, 7.64 vs. 10.13 μIU/mL; <I>P</I> = 0.05, 0.02) at childhood were significantly higher in the high POMC methylation group than in the low and mid-POMC methylation group.</P><P><B>CONCLUSIONS</B></P><P>High POMC methylation in cord blood was associated with lower birth weight, and children with high POMC methylation in cord blood showed higher triglycerides and higher insulin concentrations in blood. Thus, POMC methylation status in cord blood may be an early predictive marker of future metabolic syndrome.</P>

        A Novel Chitinolytic Bacterial Strain: Characterization of an Extracellular Chitinase

        Based on chitinolytic enzyme production, the most potent isolate named as Enterobacter cowanii CH-02 was selected and identified as a novel chitinolytic enzyme producing bacterium. The effects of media composition on bacterial cell growth and chitinase production were also studied. The maximum conditions for chitinase production were obtained at 37°C and pH 8.0 in the presence of colloidal chitin in sea water. E. cowanii CH-02 produced 0.12 U/mg of protein in batch fermentation after 24 h of incubation at 37°C using colloidal chitin in culture medium. The molecular weight of chitinolytic enzyme was estimated to be 52-kDa by SDS-PAGE which was observed in silver staining. To the best of our knowledge, we first report the biochemical function of chitinolytic enzyme from E. cowanii strain.

        Stem cell analysis with intraductally treated mammary epithelial cells

        Purpose: The activity of mammary stem cells (MaSCs) is essential to mammary growth, differentiation and regeneration in cycles of pregnancy, lactation, and involution. The capability to recruit the mammary gland through the cycles is attributed to stem cells. It was shown that the intraductal (i.duc) injection of pegylated liposomal doxorubicin (PLD) to multiparous FVB/N mice was associated with a significantly reduced outgrowth potential of mammary gland cells. We have explored i.duc PLD’s effect on stem cell number and function in mouse mammary gland and aldehyde dehydrogenase (ALDH)’s availability as a mouse MaSC marker. Methods: The total mammary epithelium was purified from 6 to 8-month-old FVB/N control and i.duc PLD-administered mice treated twice and analyzed by flow cytometry and limiting dilution cleared mammary fat pad transplants. Results: There was no significant difference in the proportions of stem cell-enriched population (CD49fhighCD24med) between control and i.duc PLD-treated groups. However, we found a significant reduction in the outgrowth potential of CD49fhighCD24med and CD49fhighCD24medALDH(+) cells from i.duc PLD-treated mammary glands. We discovered that adding ALDH to CD49fhighCD24med had the possibility of better marker selection for MaSC of mice. Conclusion: We present i.duc administration of PLD to reduce MaSC function, but not the number; and ALDH activity may add further selection of MaSCs to CD49f CD24 in mouse mammary glands. Screening of chemotherapeutic drugs or other natural products by this method of stem cell analysis may provide safe i.duc treatment in breast cancer.
