      • Benzotriazole계 유도체의 제초활성과 분자 설계

        Benzotriazole 유도체들의 정량적인 구조와 발아후 벼(Oryzae sativa L) 및 피(Echinochloa crus-galli)에 대한 생장저해 활성값(pI_50)사이의 관계들이 QSAR 방법으로 연구되었다. QSAR 분석에 의하면, 자유 라디칼 파라미터(E_R)가 매우 중요한 요소이었으며 pI_50 값은 파라치환기(X)의 E_R 파라미터에 대하여 포물선형의 관계를 나타내었다. (3)의 생장저해 활성은 (4)보다 우세하였으며 생장저해 활성이 가장 큰 화합물은 (3b)이었고 피의 생장저해 활성에 대한 E_R 파라미터의 적정값은 각각 E_R(3)=0.52와 E_R(4)=0.15이었다. 분자설계 결과, 치환기(X)가 전자 끌게이고 적정값 (0.52)와 E_R 파라미터를 가지는 분자가 가장 활성이 클 것으로 기대되었으며 Benzotriazole 유도체들의 생장저해 활성은 광합성반응(PS-II)에서 전자전달을 효과적으로 차단하는 결과로 믿어진다(1991년 9월 15일 접수, 1991년 9월 31일 수리). The relationships between the quantitative structure of benzotriazoles and their post-emergence growth inhibiting activity(pI_50) against Oryzae sativa L. and Echinochloa crus-galli were investigated using a generalized quantitative structure activity relationships(QSAR). According to the QSAR analysis, the free radical parameter (E_R) is a very important factor and the growth inhibiting activity values showed parabolic relation to E_R parameter of para-substituents(X). The activity of (3) was superior to those of (4) and (3b) is selected as the most highly effective compound. The optimal values of E_R parameter of the growth inhibiting activity aganist E. crus-galli are E_R(3)=0.52 and E_R(4)=0.15, respectively. From the result of molecular design, the substituents(X) of electron withdrawing properties and E_R parameter of optimal value(0.52) were most desirable for high activity of the benzotriazoles. And in view of this, benzotriazoles may also be effective in blocking the photosynthetic electron transfer.

      • 진행성 폐 종양 환자에서 종양 내 OK-432 주입술의 효과

        Background: Intratumoral injection of OK-432 produced significant antitumor effects. There are reports that OK-432 was injected directly into tumor tissue on gastrointestinal tracts and HCC tissue, producing decrease in serum alpha-fetoprotein level. We adopted this technique for treating locally advanced lung cancer. The object of this study is to evaluate the local therapeutic efficacy, side effects of injection for treating symptomatic mass. Methods: Twenty lung cancer patients (13 males and 7 females) underwent sonography guided OK-432 injection. We analyzed relationships between response of OK-432 and response of skin test, histology and age. Pain that patients feel was evaluated by VAS (Visual Analog scale). Results: A significant decrease of tumor size was observed. The mean pre-injection diameter of the lesions was 54.7±11.3mm, PR rate were 15.0% (3/20) and minimal change (50%<tumor size decreased.) patients were 35.0% (7/20). The VAS score before injecting OK-432 into the lesion was 5±3, this was decreased to 2.5±1.5 (p=0.04). Object response was significantly correlated with skin response, adenocarcinoma, and age. Conclusion: A significant decrease of tumor size and pain was observed. So OK-432 is a valuable treatment option for patients with symptomatic lesion from advanced lung cancer who are not amenable to further therapy.

        Evaluation of Hot Deformation and Dynamic Recrystallization Behaviors of Advanced Reduced-Activated Alloy (ARAA)

        The hot deformation behavior of advanced reduced-activation alloy (ARAA) was investigated using hot torsion tests. Thefl ow stress decreased as deformation temperature increased and as strain rate decreased. The fl ow behavior demonstratedthe typical dynamic recrystallization (DRX). Based on the constitutive analysis of peak stress, the activation energy for hotdeformation was found to be 330.3 kJ mol −1 . Peak stress was analyzed as a function of the Zener–Hollomon parameter, andcalculated and experimental values were in good agreement. A DRX kinetic model for ARAA was derived with deformationconditions based on the Avrami-type model. It was confi rmed that the volume fraction of dynamically recrystallized grainsincreased as deformation temperature increased and as strain rate decreased. The necklace structure and grain boundarybulging were observed in the deformed microstructure of ARAA. The suggested DRX mechanism for ARAA during hotworking is discontinuous DRX.

        Long-term alcohol consumption have significant harmful effects as fatty liver, hepatitis, and cirrhosis on the hepatic tissues. After voluntary administration of 8% alcohol(18.67㎖/kg/day) for 120 days, the hepatic tissues of ICR mice were daministered with Kunryeongtang extract(KRE;1.24g/kg/day) were observed to investigate the repaire effects of KRE for hepatic damages that caused by chronic alcohol administration. In chronic alcohol administered group, the vacuolation such as fat accumulation(steatosis) were increased in cytoplasm, the destructive configuration of liver plates were shown in necrotic lesion. These regions are aggregation of inflammatory component cell as lymphocytes, neutrophil leucocytes and Kuffer's cell. These hepatic damages were aggravated with time course. In KRE treated group, the decreased of vacuolation and normal construction of liver plates were appeared. These results indicated that the KRE work on the repair of hepatic damages induced chronic alcohol administration.

        In this paper, we conducted a study on implementing an urban air mobility scenario of takeoff and landing in an indoor environment. To achieve this, we utilized the Crazyflie drone, one of the nano drones, and the Lighthouse precision navigation system for indoor navigation. As a result of our research, we concluded that it is possible to implement an urban air mobility scenario in an indoor environment using nano drones.

        효소 단백 가수분해물의 항균 활성

        밀 단백질에 효소가수분해 할 때 생산되는 peptide의 항균활성과 천연항균제로서의 이용가능성을 검토하기 위하여 실험을 행하였다. 밀 단백질에 7종의 단백질가수분해효소를 작용시켜 생성된 가수분해물의 항균활성을 측정하고 한외여과, membrane filtration, HPLC를 이용하여 항균성peptide를 분리 정제한 후 분자량과 아미노산 결합 순서를 측정한 결과는 다음과 같다. 밀 단백질에 7종의 단백질 분해효소를 적용시켜 제조한 가수분해물중 Asp. saito protease를 적용시켜 얻어진 peptide 만이 항균활성을 나타내었다. Asp. saito protease는 37 ℃, pH 6.0에서 작용시킨 경우에 항균활성이 가장 높았으며, 50℃ 이상에서는 활성을 나타내지 않았다. 밀단백 효소가수분해물은 membrane filtration 에 의하여 분자량 1.000~3.000에서 항균활성이 나타났다. Membrane filtration으로 얻어진 항균활성분획을 HPLC로 분리한 결과 retention time 31.1~31.8 min에서 항균활성을 나타내었다. 밀단백 효소가수분해물은 121℃에서 15분간 가열하여도 효소활성이 유지되는 매우 안정한 화합물이었다. 항균활성분획을 MALDI-mass로 질량을 분석한 결과 1.633이었다. 항균성 peptide의 아미노산 결합순서는 cysteine, glycine, prolin, prolin, polin, valine, valine, alanine, alanine, arginine의 순서였다. This study was carried out to investigate whether peptide produced from wheat protein by enzyme hydrolysis can be used as a natural antimicrobial agent. Antimicrobial peptide was obtained form wheat protein by protease of 7 species. The produced antimicrobial peptide was purified through ultrafiltration membrane filtration and HPLC, and molecular weight and amino and sequence of the purified antimicrobial peptide were determined. Among hydrolysate produced from wheat protein by protease of 7 species, antimicrobial activity was observed for the peptide obtained from Asp. satio protease. The Asp. satio protease did production antimicrobial hydrolysate showing the highest antimicrobial activity at reaction condition of 37℃ and pH 6.0, but not at reaction condition above 50℃. Wheat protein hydrolysate was fractionated by membrane filtration and showed antimicrobial activity between molecular weight 1,000 - 3,000. The antimicrobial activity fraction obtained by membrane filtration was separated through HPLC and showed antimicrobial activity in the peak of retention time 31.1 - 31.8 min. Since after wheat protein protease hydrolysate was heated during 15 min at 121℃, antimicrobial activity was maintained we could be conviction as heat-stable peptide. Molecular weight of antimicrobial peptide identified by MALDI-mass was 1,633. Amino acid sequence of antimicrobial peptide was cysteine, glycine, prolin, prolin, prolin, valine, valine, alanine, alanine and arginine.

        표피 성장인자의 국소적용이 인체 표피화에 미치는 효과에 대한 연구

        An understanding of the mechanism of healing has increased dramatically during the past decade. The phenomenon of epithelization is important in the normal resurfacing of a donor site or a partial thickness burn wound. This study was conducted to clarify the effect of epidermal growth factor in wound healing of human. Paired donor sites were created in 18 patients who required skin grafting for either burns or reconstructive surgery. One donor site from each patient was treated topically with silver sulfadiazine cream, and the other was treated with silver sulfadiazine cream containing epidermal growth factor(10ug/ml) (Human recombinant epidermal growth factor ; Sigma, St.Louis, Mo). The donor sites were photographed daily, and healing was measured with the use of planimetric anlysis. The donor sites treated with silver sulfadiazine containing epidermal growth factor had an accelerated rate in all 18 patients as compared with that in the paired donor sited treated with silver sulfadiazine alone. Combining data from all patients demonstrated reduced the average time to 100% healing by approximately 2 to 3 days (p<0.01). Light and electron microscopical study of biopsy specimens taken from the centers of donor sites three days after the onset of healing supported these results. In conclusion, epidermal growth factor accelerates the rate of healing of partial thickness skin wound.
