Quantitative Assay for the Binding of Jun-Fos Dimer and Activator Protein-1 Site

The Jun and Fos families of eukaryotic transcription factors form heterodimers capable of binding to their cognate DNA enhancer elements. We are interested in searching for inhibitors or antagonists of the binding of the Jun-Fos heterodimer to the activator protein-1 (AP-1) site. The basic-region leucine zipper (bZIP) domain of c-Fos was expressed as a fusion protein with glutathione S-transferase, and allowed to form a heterodimer with the bZIP domain of c-Jun. The heterodimer was bound to glutathione-agarose, to which were added radiolabeled AP-1 nucleotides. After thorough washing, the gel-bound radioactivity was counted. The assay is faster than the coventional electrophoretic mobility shift assay because the gel electrophoresis step and the autoradiography step are eliminated. Moreover, the assay is very sensitive, allowing the detection of picomolar quantities of nucleotides, and is not affected by up to 50% dimethylsulfoxide, a solvent for hydrophobic inhibitors. Curcumin and dihydroguaiaretic acid, recently known inhibitors of Jun-Fos-DNA complex formation, were applied to this Jun-GST-fused Fos system and revealed to decrease the dimer-DNA binding.

This paper discusses System Identification using observer in time domain. It is carried out for a experimental Three story structure With Active Mass Damper(AMD). Applying gradually stable method. Observer Kalman Filter Identification(OKID) determines Maskov paramaters. System eigen value is presumed utilizing this variables. Simulation system is composed of this fluents's compounding. It is verticified to compare numerical simulation results with experimental measurements.

We have been proposed model equation which is able to predict the trihalomethane producing concentration formation, that is one of byproduct, in the water treatment processes. In proposed model, the effects of trihalomethane factors like chlorine contact time, pH, temperature, TOC and UV-254 are considered. The concentration of the trihalomethane produced is proportion to the contact with chlorine, pH of water, temperature of water TOC and UV-254, respectively. This proposed model could be predicted the formed concentration of trihalomethanes by trihalomethane factors.

This paper presents virtual machining system in order to realize turning process in virtual space. A reliable virtual turning process simulation was developed based on the surface shaping system which is capable of considering geometric model, thermal error model, and vibration model. Accuracy of surface shape resulting from proposed machining simulator was verified experimentally. This paper also developed the watchdog agent that continuously assessed, diagnosed, and predicted performance of products and machines in machining. The Watchdog agent extracted feature signal using time-frequency analysis among various signals from multi-sensor and evaluated machining condition using performance confidence value.

Background : Nontyphoid Salmonella (NTS) is a leading cause of human food-borne enteritis, It has been known that integron, a naturally occurring gene capture and expression element, Plays an important role in the development and dissemination of multidrug-resistance. In this study, we investigated the prevalences and molecular characteristics of integrons in NTS clinical strains. Materials and Methods : Between 1995-96 and 2000-03, a total 261 NTS clinical strains comprising 39 serotypes were collected from clinical specimens. All strains were serotyped, and the MICs of ampicillin, chloramphenicol, streptomycin, sulfamethoxazole, tetracycline, and trimethoprim were determined by agar dilution method. Integrons were detected by PCR amplification of integrase genes, and gene cassettes were determined by PCR and sequencing, Conjugation experiments were performed using E. coli J53 as a recipient. The clonal relationship was analyzed by pulsed-field gel electrophoresls (PFGE). Results : Of the 261 strains tested, class 1 integrons were present in 21 strains (8.0%). Class 2 and class 3 integrons were not found. The integron-positive rate was higher in S. Typhimurium (24.2% [8/33]) than in S. Enteritidis (2.0% [3/153]). Overall rates of antimicrobial resistance were higher in integron-positive strains. dhfr12-orfF-aadA2gene cassette was detected in 5 strains, aadA2in 4 strains, dhfr17-orfF-aadA5 in 2 strains, and addA1 in 1 strain. Ten integron-positive transconjugants were successfully selected. Among 8 integron-positive strains of S. Typhimurium, 7 had similar PFGE patterns. Conclusion: This study suggests that integrons are already playing a significant role in antimicrobial resistance in NTS. Continuous monitoring is needed to detect the emergence and spread of integron-mediated antimicrobial resistance.

Twenty-three plants of Japanese apricot (Prunus mume) were collected from several sites around Mountains JIRI in Korea. Japanese apricots having the different morphological features were evenly distributed in the groups made from the cluster analysis, indicating no geographic distributions but artificial vegetations in Korea. Japanese apricots were, as based on the PCR-RAPD techniques, clustered into the three groups; a group (prototype) having the five white petals with the five red sepals, a group (green type) having the five white petals with the five green sepals, and a group (hybrid type) having the more than five red petals with various colored sepals. The prototype apricots showed higher toxicities than other type apricot against bacteria and production of less compounds in TLC plates. The polypetal types of Japanes apricot were related to those of p. armebiaca in the characteristics of seed (the ruggedness), but also to be closed to those of p. armebiaca in PCR-RAPD analysis. The cluster analysis of the twenty three apricots and its related species calculated from the two primers were shown to distinguish relationships of cultivars within species, or of individual plants within cultivars, but also to display the two overlapping bands resulted from PCR-RAPD technique.
