We have established a new National Reference Standard(NRS) of filgrastim to be used for the quality management of domestic genetic recombinant drugs. Filgrastim is a recombinant human granulocyte colony stimulating factor( G-CSF) and one of the first-generation biosimilars used as the hematopoietic agent. At first, the candidate substance for the NRS was manufactured on consignment in accordance with the manufacturing process of filgrastim. The identity, purity, potency and protein contents of the candidate substance were assessed using electrophoresis, HPLC systems and a biological assay, which showed it was comparable to filgrastim in quality. In addition the stability test showed it was stable for 12 months. We performed a collaborative study with other institutes to endow it with the labelled protein content and potency values. From the data, it has been assigned with the protein content of 0.240 mg/mL(95% confidence intervals: 0.238~0.242 mg/mL) and the potency value of 2.94 × 107 IU per vial. We had finally enrolled it as the first Korean NRS of filgrastim as approved by Working Committee on Development of Testing and Inspection in Korea Ministry of Food and Drug Safety.
Awareness Survey on Community Water Fluoridation by Region

Background: The purpose of this study is to investigate the pro-con of re-implementation by administrative areas and the difference in perception of community water fluoridation in implemented and non-implemented areas after the community water fluoridation in Korea was suspended. Through this, we intend to provide basic data that can help find ways to increase the support and interest of local residents.Methods: The 601 questionnaires collected through the survey and statistical analysis was conducted using SPSS Statistics 28.0. Results: As a result of analyzing the perception of the community water fluoridation according to the understanding of fluorine, the proportion of people who were not recognized by both fluorine and community water fluoridation was the highest (p<0.05). As a result of the analysis of the pro-con of re-implementation of community water fluoridation, the approval was high. Among those who responded in favor, the place of re-implementation showed that ‘implementation nationwide’ was high. As for the reason for favor, it was found that it was possible to prevent dental caries disease. The reason for the objection was the lack of knowledge about fluoride. Conclusion: The results of the survey for the pro-con of the re-implementation of community water fluoridation showed a higher degree of ‘agree’ and showed that people in the area where community water fluoridation was not implemented showed higher interest in oral health prevention and management. Through this, not only oral education, but also correct information on the implementation method of community water fluoridation, the benefits of community water fluoridation, and the facts that were misunderstood in the past, as well as oral education, can be provided to raise interest in community water fluoridation. It is thought that the expected effect of the re-implementation of community water fluoridation can be obtained if such activities are carried out.

교정복지로 본 학교부적응 청소년 직업훈련과 진로성숙에 관한 연구

본 연구의 목적은 학교부적응청소년의 직업훈련과 진로성숙간의 영향을 분석하는 것에 있다. 이를 통해 직업훈련이 진로성숙에 미치는 영향에 대해 연구하고 이를 통해 효과적인 직업훈련 방안을 모색하여 정책당국과 직업훈련 현장에서 활용하는 데 연구의 의의가 있다. 경기도에 소재하는 직업훈련학교를 중심으로 학교부적응청소년 70명을 대상으로 설문 조사를 한 결과 직업훈련기간과 시간 그리고 훈련만족도는 진로성숙도에 영향을 미치지 못하는 것으로 나타났다. 하지만 훈련내용만족도는 진로성숙도에 영향을 미치는 것으로 나타났다. 그러므로 훈련을 실시할 때 이와 같은 것을 고려하여 훈련내용을 충실히 하거나 훈련교사의 자질을 향상시키는 방법 등을 통해 이러한 문제에 잘 대응할 수 있을 것이다. 관계당국은 이러한 점을 고려하여 훈련프로그램을 작성할 때 훈련과정에 훈련프로그램을 충실하게 운영할 수 있는 방안을 모색하여야 할 것이다. 이러한 연구 결과를 통해 직업훈련에 대한 발전방안을 제언해 보았다. Slow-Binding Inhibition of Soybean Lipoxygenase-1 by Luteolin

Luteolin, isolated from the seeds of Perilla frutescens (perilla seeds), inhibited the peroxidation of linoleic acid catalyzed by soybean lipoxygenase-1 (EC 1.13.11.12, Type 1) with an IC50 of 5.0 μM (1.43 μg/mL)noncompetitively. The progress curves for an enzyme reaction indicate that luteolin shows slow binding kinetics. Both the initial velocity and steady-state rate in the progress curve were decreased with increasing the concentration of luteolin. The kinetic parameters, which described the inhibition by luteolin, were evaluated by nonlinear regression fits.

Colonoscopy is the current standard method for evaluation of the colon. The diagnostic accuracy and therapeutic safety of colonoscopy depend on the quality of colonic cleansing and preparation. Generally, all these preparations have been demon-strated to be safe for use in healthy individuals without significant comorbid conditions. Based on safety and efficacy concerns, polyethylene glycol (PEG) is most commonly utilized as a bowel preparation solution for colonoscopy. Adverse events in pa-tients receiving PEG are mostly clinically non-significant. However, fatal adverse events rarely have been shown to occur in the few individuals who experience vomiting or aspiration. Anaphylactic shock associated with ingestion of PEG electrolyte solu-tion is an extremely rare fatal complication, and reported mainly in Western countries. Here, we report the first case of anaphy-lactic shock following the ingestion of PEG solution in Korea. (Intest Res 2015;13:90-94)

An 82-year-old man diagnosed with supraglottic cancer sought a consultation for percutaneous endoscopic gastrostomy (PEG) placement. Preoperative chest radiography (posterior-anterior [PA] view) revealed no abnormalities, and PEG tube placement was performed using the “pull” method. Chest radiography (PA view) performed 3 days postoperatively showed free air that was not observed immediately after PEG tube placement; therefore, the patient was diagnosed with pneumoperitoneum. Abdominal computed tomography confirmed that the PEG tube was appropriately positioned within the stomach; however, the colon was observed between the abdominal wall and stomach, which indicated that the PEG tube had passed through the colon. Review of preoperative chest radiographs (PA views) confirmed that the colon was visualized in the area wherein usually stomach gas should have been observed. The patient was diagnosed with a gastro-colo-cutaneous fistula that occurred postoperatively, following a procedure that was performed without confirmation of anatomical variations. The PEG tube was removed surgically, and we performed percutaneous gastrostomy.

Objective: Rheumatoid arthritis (RA) is associated with an increased cardiovascular events. These may be related to insulin resistance (IR). We evaluated status of IR and analyzed the relationship between IR and clinical and laboratory characteristics in patients with RA. Methods: We examined 52 RA patients (43 females) and 52 age and sex matched healthy controls. We measured Homeostasis model assessment (HOMA) IR, calculated according to fasting serum glucose and insulin. Results: In patients, age was 50.8±10.2 years old, disease duration was 42.1±30.9 months. In controls, HOMA IR was 0.62±0.30 and in patients, it was 1.28±0.50. This difference was highly significant (p<0.001). Patients with early RA (disease duration is less than 36 months) were 28, and established RA (more than 36 months) were 24. HOMA IR was significantly higher in patients with established RA (1.42±0.45) than those with early RA (1.16±0.45) (p=0.03), and significantly correlated with disease duration (r=0.36, p=0.01), BMI (r=0.36, p<0.001), total cumulative prednisolon dose (r=0.34, p=0.01). Disease duration and BMI were independent predictors for HOMA IR (p<0.01, p=0.03). Conclusion: In patients with RA, IR measured by HOMA IR was more significantly increased than that of healthy control and significantly correlated with disease duration, BMI, and total cumulative prednisolon dose; however, the determinants of HOMA IR in RA patients were disease duration and BMI.
Anthocyanins in Cowpea [Vigna unguiculata (L.) Walp. ssp. unguiculata]

The aim of this study was to isolate and identify the anthocyanins in the black seed coated cowpea [Vigna unguiculata (L.) Walp. ssp. unguiculata] using reverse phase C-18 open column chromatography and high performance liquid chromatography (HPLC) with diode array detection and electro spray ionization/mass spectrometry (DAD-ESI/MS) analysis, respectively. Anthocyanins were extracted from the coat of black cowpea with 1%trifluoroacetic acid (TFA) in methanol, isolated by RP-C-18 column chromatography, and their structures elucidated by 1D and 2D nuclear magnetic resonance (NMR)spectroscopy. The isolated anthocyanins were characterized as delphinidin-3-O-glucoside (2) and cyanidin-3-Oglucoside (4). Furthermore, 5 minor anthocyanins were detected and identified as delphinidin-3-O-galactoside (1),cyanidin-3-O-galactoside (3), petunidin-3-O-glucoside (5),peonidin-3-O-glucoside (7), and malvidin-3-O-glucoside (8) based on the fragmentation patterns of HPLC-DADESI/MS analysis.
