        울릉도 소의 전염성 병원체 감염률 조사

        Prevalence of infectious agents, including Brucella abortus (BA), Mycobacterium bovis (MB), bovine leukemia virus (BLV), M. avium subsp. paratuberculosis (MP), Neospora caninum (NC) and Toxoplasma gondii (TG), was investigated in all the cattle raised in Ulleung island during 2007∼ 2010. For BA, the prevalences in head and farm were 8.1% (44/545) and 5.5% (4/73) in 2007, all negative in 2008∼2009, and 0.5% (4/774) and 1.7% (1/58) in 2010, respectively. For MB, no sample was positive by PPD or ELISA in 2007∼2010. For BLV and MP, no sample was positive by ELISA in 2007∼2009. For NC, seroprevalences in head and farm were 0.2% (1/545) and 1.4% (1/73), respectively, in 2007 and all negative in 2008∼2009. For TG, seroprevalences in head and farm were 17.6% (97/552) and 54.8% (34/62) by ELISA in 2009. By regions, the seroprevalences of TG in Ulleung-eup, Seo-myeon and Buk-myeon were 26.0%, 9.8% and 16.7%, respectively, which had significant differences (P<0.0001). Tiger cattle were more resistant to TG infection than Hanwoo. The seroprevalence of TG in summer was higher than in autumn. The seroprevalence of TG in cows was higher than in oxen. The seroprevalence of TG in cattle was increased with age. In conclusion, this study indicates that the prevalences of six infectious diseases, except for TG which are widely spread, are relatively low in cattle reared in Ulleung island.

        네모나프라이드의 유효성 및 추체외로 부작용 : Haloperidol 및 Risperidone과의 자연관찰적 개방형 비교연구 A Naturalistic Open Comparative Study with Haloperidol and Risperidone

        Objectives : This open prospective study was performed in order to evaluate the efficacy and extrapyramidal adverse effects of nemonapride in the schizophrenic patients, and was compared wit one of typical antipshchotics. haloperidol and one of atypical antipsychotics, risperidone. Methods : Thirty male and female schizophrenic patients(DSM-Ⅳ) were treated for 12 weeks with haloperidol(n=10), risperidone(n=10) and nemonapride(n=10). The overall efficacy was assessed at baseline, 1st week, 2nd week, 4th week, 8th week, and 12th week by the Positive and Negative Syndrome Scale for Schizophrenia(PANSS). Also the overall safety was assessed in the same time period by the Extrapyramidal Symptom Rating Scale(ESRS). Results : There were no significant differences in PANSS scores(total, positive, negative, and general psycho­pathology subscale) among haloperidol, risperidone, and nemonapride groups, Treatment responders, defined as at least 20% reduction of baseline total PANSS score, were achieved by 8 patients(80%) in haloperidol group, 10 patients(100%) in risperidone group, and 8 patients(80%) in nemonapride group. And there were no significant differences in ESRS total scores among haloperidol, risperidone, and nemonapride group. Inter-group comparison among haloperidol, risperidone and nemonapride group, as assessed by the ESRS, revealed no significant differences in the shifts to the maximum score, 9.4(±9.4), 6.2(±8.4), and 11.3(±8.1) respectively, and also revealed no significant differences in the mean time reaching the maximum score, 4.5(±4.5) week, 7.8(±4.8) week, and 4.6(±4.4) week, respectivily. Conclusion : There wee no significant differences in the efficacy and extrapyramidal adverse effects among haloperidal, rispecridone and nemonapride groups. These results suggest that nemonapride was as efficacious and safe in he treatment of schizophrenia, as well known and widely used antipsychotics, haloperidol and risperidone.

      • 무시멘트성 비구컵 사용시 Polyethylene 마모율 : AML Prosthesis 중심으로

        비구를 metal-back이 있는 polyethylene 컵으로 시멘트를 사용치 않고 치환한 경우에서 비구컵 외전 각도와 인공 골두의 크기가 비구 polyethylene 마모에 미치는 효과를 문헌 고찰과 함께 보고하면서, polyethylene 마모가 고관절 치환술후의 장기적 추적과 연관성이 있는 바 추척 관찰에 유용성을 파악하기 위해 연구를 시행하였다. The amount of linear wear of the polyethlene of eighty four AML acetabular components, implanted without cement during a primary total hip replacement in seventy- two patients, were determined by a comparison of the thickness of the cup as measured on the initial and most recent follow-up radiographs. The measurements were corrected for magnification error by direct measurement of the diamenter of the femoral head on each radiographs. The mean duration of follow-up was seventy-eight months(range, 65 to 125 months). Linear wear averaged 1.94±0.74 millimeter(range, Zero to 9.61 millimeter) over-all, with a mean rate of 0.26±0.15 millimeter(range, zero to 1.10 millimeter) per year. An increase rate of per year correlated with a 32mm femoral head than 28mm femoral head, but there was no relationship with the weight or sex of the patients, the initial thickness of the polyethylene, or the angle of the abduction of the cup.

      • N₂O 가스로 열산화된 게이트 산화막의 전기적 특성

        미래의 ULSI 소자의 게이트 산화막으로 이용하기 위하여 N₂O 가스 분위기에서 기존의 전기로를 이용한 실리콘의 열산화에 의해 N₂O 산화막을 형성하였고, MOS 소자를 제작하여 전기적 특성을 고찰하였다. 900℃에서 90분간 산화한 N₂O 산화막의 경우, 플랫밴드 전압(??), 고정전하밀도(??)와 플랫밴드 전압의 변화량(??)은 각각 0.81 [V], ?? [??]와 80∼95[mV]를 나타내었다. N₂O 산화막의 전기전도기구는 저전계 영역에서는 Fowler-Nordheim 터널링, 고전계영역에서는 Poole-Frenkel 방출이 지배적으로 나타났고, 절연파괴전계는 16 [MV/㎝]로 높게 나타났다. 따라서 N₂O 산화로 형성된 게이트 산화막이 ULSI 소자의 게이트 유전체로 응용이 가능하리라 생각된다. In order to applicate gate insulators in future ULSI devices, electrical properties was investigated in MOS device with N₂O oxide grown by thermal oxidation using conventional furnace in N₂O ambient. For the sample oxidized at 900℃ for 90 min, Flatband voltage(??), fixed charge density(??) and shifts of flatband voltage(??) after BTS was obtained 0.81 [V], ?? [??] and 80∼95[mV], respectively. The dominant conduction mechanism of N₂O oxide appeared to be Fowler-Nordheim tunneling in the low electrical field region(4-6 [MV/㎝]) and to be Poole-Frenkel emission in the high electrical field region( 9 [MV/㎝] or above). Dielectrics breakdown fields of N₂O oxide appeared about 16 [MV/㎝]. These results suggest that the N₂O oxides is a promising candidate for ultrathin gate dielectric for MOS ULSI applications.

        IL-21 ensures TGF-β1-induced IgA isotype expression in mouse Peyer's patches

        <P>It is well established that TGF-beta1 induces IgA and IgG2b class-switching recombination in murine B cells. In the present study, we assessed the activity of IL-21 along with TGF-beta1 in Ig synthesis by murine spleen B cells. IL-21 showed antiproliferative activity on LPS-activated splenic B cells, comparable with that of TGF-beta1. IL-21 alone had little effect on IgA secretion and decreased other isotypes. Likewise, IL-21 also did not alter the TGF-beta1-induced IgA synthesis and concurrently diminished the syntheses of IgM and IgG2a, which were repressed by TGF-beta1. Unexpectedly, IL-21 inhibited the TGF-beta1-induced IgG2b production. This IL-21 effect was examined using B cells from IL-21R knockout mice, where the IgA production profile was paralleled by that seen in wild-type B cells. However, the inhibitory effect of IL-21 on TGF-beta1-induced IgG2b synthesis was not seen in the IL-21R(-/-) mouse, suggesting that IL-21 causes TGF-beta1-stimulated B cells to decrease IgG2b synthesis. Expression patterns of Ig germ-line alpha(GL alpha)/GL gamma 2b transcripts under the influence of TGF-beta1 and IL-21 were paralleled by IgA/IgG2b secretion. This was also observed in the activities of GL(alpha) and GL(gamma 2b) promoters. These results indicate that IL-21 decreases IgG2b secretion mainly through inhibition of GL(gamma 2b) transcription and is ultimately associated with selective IgA secretion induced by TGF-beta1. Our results showed that IL-21 was expressed in greater magnitude in Peyer's patches (PP) than in spleen. These results suggest that IL-21 has an important effect on selective IgA(+) B cell commitment in PP.</P>

      • LIGHT-HVEM Signaling in Innate Lymphoid Cell Subsets Protects Against Enteric Bacterial Infection

        <P><B>Summary</B></P> <P>Innate lymphoid cells (ILCs) are important regulators of early infection at mucosal barriers. ILCs are divided into three groups based on expression profiles, and are activated by cytokines and neuropeptides. Yet, it remains unknown if ILCs integrate other signals in providing protection. We show that signaling through herpes virus entry mediator (HVEM), a member of the tumor necrosis factor (TNF) receptor superfamily, in ILC3 is important for host defense against oral infection with the bacterial pathogen <I>Yersinia enterocolitica.</I> HVEM stimulates protective interferon-γ (IFN-γ) secretion from ILCs, and mice with HVEM-deficient ILC3 exhibit reduced IFN-γ production, higher bacterial burdens and increased mortality. In addition, IFN-γ production is critical as adoptive transfer of wild-type but not IFN-γ-deficient ILC3 can restore protection to mice lacking ILCs. We identify the TNF superfamily member, LIGHT, as the ligand inducing HVEM signals in ILCs.       • Mechanism underlying the suppressor activity of retinoic acid on IL4-induced IgE synthesis and its physiological implication

        <P><B>Abstract</B></P> <P>The present study extends an earlier report that retinoic acid (RA) down-regulates IgE Ab synthesis in vitro. Here, we show the suppressive activity of RA on IgE production in vivo and its underlying mechanisms. We found that RA down-regulated IgE class switching recombination (CSR) mainly through RA receptor α (RARα). Additionally, RA inhibited histone acetylation of germ-line ε (GL ε) promoter, leading to suppression of IgE CSR. Consistently, serum IgE levels were substantially elevated in vitamin A-deficient (VAD) mice and this was more dramatic in VAD-lecithin:retinol acyltransferase deficient (LRAT<SUP>−/−</SUP>) mice. Further, serum mouse mast cell protease-1 (mMCP-1) level was elevated while frequency of intestinal regulatory T cells (Tregs) were diminished in VAD LRAT<SUP>−/−</SUP> mice, reflecting that deprivation of RA leads to allergic immune response.         Analyses of TGF-β1-inducible Ig germ-line γ2b promoter activity: Involvement of Smads and NF-κB

        <P>TGF-β1 directs class switch recombination to IgG2b as well as IgA. We have shown that Smad3/4, Runx3, and p300 mediate TGF-β1-induced germ-line (GL) γ2b transcription and that there is a potential Smad-binding element (SBE, CAGAC, −38/−34) and Runx-binding element (TGTGGGT, +41/+47) in the promoter region. Here, we have characterized more putative transcription factor-binding elements in the promoter. Site-directed mutagenesis revealed that two more putative SBE (GTCTG, −67/−63 and +38/+42) are relevant to TGF-β1-induced GLγ2b promoter activity, a finding that was confirmed by EMSA. However, neither overexpression of Ets (i.e. Elf-1, Fli-1, or Pu.1) nor a mutation deleting a putative Ets-binding element (CAGGAA, −4/+2) affected basal or TGF-β1-induced promoter activity. On the other hand, NF-κB repressed promoter activity without direct binding to two putative NF-κB-binding elements (GGACTCCCC, −63/−55; GGGCCTTTCC,+237/+246). Instead, NF-κB overexpression increased the expression of Smad7 transcripts. Moreover, p300 overexpression failed to rescue the inhibitory effect of NF-κB on GLγ2b promoter activity. These results indicate that there are multiple SBE relevant to GLγ2b promoter activity and that NF-κB acts in cooperation with p300 to downregulate promoter activity through increasing the gene expression of inhibitory Smad7.</P>
