        CO1 DNA 바코드 염기서열 기반 팽활(蟛螖) 신속 감별용SCAR marker 개발

        Objectives : Paeng-hwal is described as an insect herbal medicine used for digestive diseases in the Dong-ui-bo- gam. The origin of this herbal medicine is limited to several small crabs, such as Helice tridens. These crab species cohabitat in the same environment and share similar morphological characteristics, making it very difficult to distinguish and collect the individual species for use in dietary supplements or herbal medicines. This study was conducted to develop a genetic identification tool for discriminating among these closely related small crab species. Methods : CO1 DNA barcode regions of 15 samples from 6 species of small crabs were analyzed to obtain the individual sequences. To identify the correct species, comparative analyses were carried out using the database of the NCBI GenBank and the NIBR. SCAR primers were designed to develop simple and rapid assay methods using inter-species specific sequences. Optimal SCAR assay conditions were established through gradient PCR, and the limit of detection (LOD) was determined. Results : Six species of small crabs (Helicana tridens, Macrophthalmus abbreviatus, Helicana tientsinensis, Helicana wuana, Chiromantes dehaani, and Hemigrapsus penicillatus), which are distributed as Paeng-hwal, were identified through CO1 sequences analysis. We also developed SCAR markers to distinguish between six small crabs at the species level. Furthermore, we established the optimal PCR assay methods and the LOD of each individual species. Conclusions : The rapid and simple SCAR-PCR assay methods were developed to identify the species and control the quality of herbal medicines for Paeng-hwal based on the genetic analyses of CO1 DNA barcodes.

        Analysis of Genetic Similarity Detected by AFLPand PCoA among Genotypes of Kenaf (Hibiscus cannabinus L.)

        Seventeen kenaf varieties collected from several regions around Asia and Europe were grown in Korea and their genetic diversity was analyzed using morphological characters and AFLP technique. In the morphological analysis, the 17 varieties were divided into two major groups according to stem diameter, plant height, and flowering periods. The late varieties, which could yield more biomass compared with the early-medium varieties, were included in one of two major groups . Nonetheless, it is difficult to identify individual varieties based on morphological characters because of their limited variation. For the AFLP analysis, 34 primer combinations generated a total of 3,193 polymorphic bands (out of 3,914) with a polymorphic rate of 82%. The clusters were divided into two major groups with a similarity coefficient of 0.63 by UPGMA analysis method; but each group did not show a common tendency. Additionally, the results of the AFLP analysis did not show similar tendency compared with morphological data, a result that might be explained in terms of convergent evolution, i.e. the acquisition of morphologically similar traits between distinctly unrelated varieties.

        작약(芍藥) 유전자 감별을 위한 psbA-trnH 부위 염기서열 분석 및 Marker Nucleotide 발굴

        Purpose : The original plant species of Paeoniae Radix is differentially described in the national pharmacopoeia of Korea, China and Japan. To develop the reliable method for the authentication of four Paeonia species, P. lactiflora, P. japonica, Paeonia veitchii and P. suffruticosa, we analyzed psbA-trnH DNA barcode region and identified species-specific marker nucleotides. Materials and Methods : Plant materials were collected from different geographic regions in Korea and China, and psbA-trnH DNA barcode region were amplified using psbA3-f and trnHf-05 primer set. The DNA sequences were analyzed and aligned using the Bio-Edit program. The multiple alignment of each samples were also analyzed with the Bio-Edit program in order to calculate the genetic distance among the samples using unweight pair group method with arithmetic averages (UPGMA) anlysis. Results : PCR amplification rate of 12 samples used in this study were 100% for psbA- trnH region. In comparison of psbA-trnH sequences among four Paeonia species, we identified 1, 2, 2, and 16 species specific nucleotides enough to distinguish each species, respectively. These species-specific sequences could provide useful genetic marker nucleotides to discriminate the precise species among the analyzed four Paeonia species. Conclusion : We analyzed psbA-trnH region to confirm the availability of molecular authentication for Paeoninae Radix on species levels using four Paeonia species. As a result, we amplified PCR products of psbA-trnH region from all of the 12 samples and obtained total 21 marker nucleotides enough to distinguish each four Paeonia species. These marker nucleotides are useful genetic tool for the standardization of Paeonae Radix.

        주거환경만족도가 매입임대주택 입주민의 건강에 미치는 영향 -동네응집력의 매개효과와 거주기간의 조절효과

        본 연구의 목적은 매입임대주택 입주민의 주거환경만족도와 건강의 관계에서 동네응집력의 매개효과와 거주기간의 조절효과를 검증하는 것이다. 분석자료는「2018년 서울시 매입임대주택 공급 및 관리 실태조사」에서 추출하였고, 연구참여자는 서울시 소재 매입임대주택 270동 입주민 540명으로 정하였다. 분석방법으로는 구조방정식모형을 이용하였다. 분석 결과, 주거환경만족도는 건강과 동네응집력에 직접적으로 정적인 영향을 미쳤고, 동네응집력은 건강에 직접적으로 정적인 영향을 미쳤으며, 주거환경만족도는 동네응집력을 매개로 건강에 간접적으로 정적인 영향을 미치는 것으로 나타나 동네응집력의 매개효과를 확인하였다. 또한, 동네응집력이 건강에 미치는 직접적인 정적 영향은 현 주택에 거주한 시간이 짧은 경우에 비하여 긴 경우 유의미하게 커 거주기간의 조절효과도 일부 확인하였다. 연구결과를 토대로 매입임대주택사업의 개선방안을 주거환경 개선, 주택관리 및 운영체계 효율화, 공동체 활성화 사업 추진 등의 측면에서 제시하였다.

        늑간 신경 냉동요법에 의한 개흉술후 흉부 동통 관리

        Post-thoracotomy pain is so severe that lead to postoperative complications, such as sputum retention, segmental or lobar atelectasis, pneumonia, hypoxia, respiratory failure due to the patient`s inability to cough, deep breathing and movement. Many authors have been trying to reduce the post-thoracotomy pain, but there is no method of complete satisfaction. In 1974, Nelson and associates introduced the intercostal nerve block using the cryoprobe. The application of cold directly to the nerves causes localized destruction of the axons while preserving the endoneurium and connective tissue, thereby introducing a temporary pain block and able to complete regeneration of intercostal nerves. One hundred and two patients, who undergoing axillary or posterolateral thoracotomy at the Department of Thoracic and Cardiovascular Surgery in Korea University Medical Center between April 1990 and August 1990, were evaluated the effects of cryoanalgesia for the post-thoracotomy pain reduction. The patients were divided into two groups: Group A, control, the patients without the cryoanalgesia[No.=50], Group B, trial, the patients with cryoanalgesia[No.=52]. Before the thorax closed, in the group A, local anesthetics, 2% lidocaine 3cc, were injected to the intercostal nerves[one level with the thoracotomy, one cranial and caudal intercostal level and level of drainage tube insertion]. In the group B, cryoprobe was directly applied for 1 minute at the same level. Postoperative analgesic effects were evaluated by the scoring system which made arbitrary by author: The pain score 0 to 4, The limitation of motion score 0 to 3, The analgesics consumption score 0 to 3, The total score, the sum of above score, 0 to 10. For the evaluation of immediate analgesic effects, the score were evaluated at the operative day, the first postoperative day, the second postoperative day, and the seventh postoperative day. The effects of incision type, and rib cut to the post-thoracotomy pain were also evaluated. The results were as follows; 1. The intercostal block with cryoanalgesia reduced the immediate postoperative pain significantly compare with control group. 2. The intercostal block with cryoanalgesia improved the motion of the operation side significantly compare with control group. 3. The intercostal block with cryoanalgesia reduced the analgesics requirements at the immediate postoperative periods significantly. 4. The intercostal block with cryoanalgesia lowered the total score significantly compare with control group. 5. The intercostal block with cryoanalgesia were more effective to the mid-axillary incision than to the posterolateral incision 6. The intercostal block with cryoanalgesia were more effective to the patients without rib cut than to the patients with rib cut. 7. No specific complication need to be treated were not occurred during follow-up.

        2002년 농약중독 사망자 통계청 자료 분석 및 제초제 중독으로 인한 사망사고 예방 대책

        Backgrounds: Deaths from pesticides poisoning has increased annually, but no studies have been reported. Especially paraquat is suspected to be a major material in pesticide poisoning deaths. In Korea, simple epidemiology of pesticides poisoning is not well known. So, there is no effective method to prevent pesticides poisoning. We tried to investigate the present status of pesticides poisoning with data from Korea National Statistical Office and suggest an effective counterplan. Methods: We analysed death cause data derived from Korean Statistical association, deaths causes being pesticides poisoning in the year 2002. Also, we investigated data from internet, mainly from Korea National Statistical Office home page. Results: Total 2,875 persons died after pesticides poisoning in 2002 year. Average age is 54.2 year, standard deviation being 17.7 year. Many victims (about $41\%$) died during the herbicides' non-necessary period (JanuaryMarch, October-December) Conclusion: Herbicides sale should be restricted only during herbicides' necessary period, and so called a pesticides safety supervisor system shall effectively prevent herbicides poisoning.

        Phenotyping analysis of p53 knockout mice produced by gene editing and comparison with conventional p53 knockout mice

        Background Knockout (KO) mice developed by homologous recombination (HR) have become useful tools to elucidate gene function. However, HR has low KO efficiency and is time-consuming, labor-intensive, and expensive. ‘Gene editing’ has received much attention for efficient genetic manipulation. Objective As generation of KO mice is simplified, KO mice produced by HR can be feasibly reproduced using gene editing. However, phenotyping analysis and comparison between KO mice produced by these two techniques is necessary. Methods We generated p53 KO mice through gene editing and compared their phenotype with the already reported HRmediated p53 KO mice. Results Tumors occurred in 36 (73%) of 49 homozygous KO mice and the mean age of occurrence was 23 weeks, with lymphoma (64%) and sarcoma (23%) being the most common. Tumors were also developed in 12 heterozygous mice and the mean age of occurrence was 40 weeks, with sarcoma (54%) and lymphoma (46%) in high proportion. Homozygotes had a mean life span of 157 ± 52 days and developmental abnormalities were found in females compared to in males (P < 0.05, P < 0.001). Conclusion We analyzed the basic phenotype of p53 KO mice and observed no significant difference from the conventional HR-mediated p53 KO mice.

        Korean Immigrant Businesses Departing Inner-City African-American Neighborhoods: The Case of Chicago

        This study identifies a determining factor that affects the departure of Korean immigrant businesses from inner-city African-American neighborhoods. Data for this study come from a sample survey of 132 Korean immigrant business owners in the Chicago area. A major finding is that owners’ favorable perception of external economic opportunities, rather than conflicts with African-American customers or residents represents the major factor that contributes to Korean immigrant businesses leaving inner-city African-American neighborhoods. This finding suggests an alternative that invalidates the conflict-centered perspective commonly found in the literature on Korean-American entrepreneurship, instead putting economic factors at the forefront of the analysis.
