        HPLC-ELSD와 LC/MS를 활용한 건강기능식품 중 글루콘산철 최적 분석법 확립 및 검증

        본 연구는 국내에서 식품첨가물로 사용되는 산도조절제 글루콘산철(ferrous gluconate)의 고성능 액체 크로마토그래피(High Performance Liquid Chromatography, HPLC)를 이용한 분석법 개발 및 이에 대한 타당성을 검증하기 위해 진행되었다. 글루콘산철의 정량을 위해 증기화 광산란 검출기(Evaporative Light Scattering Detector, ELSD), 역상분배형 컬럼(HyperClone<SUP>TM</SUP> ODS column)을 사용하였으며, 이동상은 1.3% 트리플루오로아세트산과 아세토니트릴을 이용하였다. 개발된 분석법의 직선성은 결정계수(R²)가 0.999로 우수하였고, 검출한계는 12.8 μg/mL, 정량한계는 38.9 μg/mL로 나타났다. 정확도는 일내분석에서 95.6~101.5%와 일간분석에서 98.6~103.1%로 나타났으며, 정밀도는 일내분석과 일간분석에서 각각 1.5~2.6%와 0.7~2.9%로 나타났다. 또한, 건강기능식품을 시료로 분석한 측정불확도는 954.7 μg/mL 농도에 대해 확장불확도가 27.73μg/mL로 산출되어 약 3%의 측정불확도 값이 산출되었다. 이 분석법 검증 결과는 유럽연합 집행위원회(European Commission, EC) 및 국제표준화기구(International Organization for Standardization, ISO)의 기준을 모두 충족시키는 결과였다. Ferrous gluconate has been used in Korea, the United States, Japan, and the European Union to control the acidity in foods. We optimized an analytical method to determine the levels of ferrous gluconate in foods by a high-performance liquid chromatography-evaporative light scattering detector (HPLC-ELSD) and an ultra-performance liquid chromatography-mass spectrometry (UPLC-MS). The linearity of the developed analysis method using HPLC was excellent with a coefficient of determination (R²) of 0.999 obtained from calibration curve, and limits of detection and quantitation were 12.8 μg/mL and 38.9 μg/mL, respectively. In intra-day and inter-day analysis, the accuracy was 95.6∼101.5% and 98.6∼103.1%, respectively, and the precision was 1.5∼2.6% and 0.7∼2.9%, respectively. The measurement uncertainty of analyzing the health functional food samples was within 3%, and the verification results of all analytical methods met the standards of the European Commission (EC). UPLC-MS was also used to confirm ferrous gluconate. As a result, the field applicability of the ferrous gluconate analytical method developed in this study was confirmed.

        CONCEPTUAL STRUCTURAL DESIGN AND COMPARATIVE POWER SYSTEM ANALYSIS OF OZONE DYNAMICS INVESTIGATION NANO-SATELLITE (ODIN)

        The Ozone Dynamics Investigation Nano-Satellite (ODIN) is a CubeSat design proposed by Chungnam National University as contribution to the CubeSat Competition 2019 sponsored by the Korean Aerospace Research Institute (KARI). The main objectives of ODIN are (1) to observe the polar ozone column density (latitude range of 60° to 80° in both hemispheres) and (2) to investigate the chemical dynamics between stratospheric ozone and ozone depleting substances (ODSs) through spectroscopy of the terrestrial atmosphere. For the operation of ODIN, a highly efficient power system designed for the specific orbit is required. We present the conceptual structural design of ODIN and an analysis of power generation in a sun synchronous orbit (SSO) using two different configurations of 3U solar panels (a deployed model and a non-deployed model). The deployed solar panel model generates 189.7 W through one day which consists of 14 orbit cycles, while the non-deployed solar panel model generates 152.6 W. Both models generate enough power for ODIN and the calculation suggests that the deployed solar panel model can generate slightly more power than the non-deployed solar panel model in a single orbit cycle. We eventually selected the non-deployed solar panel model for our design because of its robustness against vibration during the launch sequence and the capability of stable power generation through a whole day cycle.

      • Digestive protease activity and antibacterial activity against spoilage-relate bacteria of the black soldier fly (Diptera: Stratiomyidae)

        Larvae black soldier fly, Hermetia illucens, is beneficial because its larvae feed on organic materials derived from plants, animals and humans and promote the recycling of food waste and organic materials. Chymotrypsin serine protease is one of the main digestive proteases in the midgut of and is involved in various essential processes. In a previous study, a gene encoding a chymotrypsin-like protease, Hi-SP1, was cloned from the larvae of Hermetia illucens and characterized. The objective of this study was to compare the digestive enzyme activity with various enzymes such as papain, protease and α-chymotrypsin. And also, we investigated the antimicrobial activity of the Hi-SP1 against the spoilage relate bacteria. The growth of the bacteria was inhibited in nutrient broth containing the Hi-SP1.

        미야자와 켄지의 동화와 『본생경』(本生經) : 동물우화를 중심으로 動物寓話を中心に About animal fables

        今まで宮澤賢治の童話の佛敎からの影響を論ずる時、法華文學の創作という側面からの硏究が强調されてきたわけである。勿論、それは看過できない重要な部分であるが、佛敎說話の母體でなる『本生經』との關係についても一層關心を注ぐべき時期ではないかと思われる。『本生經』は、世界の說話の寶庫である。幼い頃から佛敎に親しんでいたこと、彼の童話が動物及び植物。また人間に及ぶ異彩な登場物を持つことを考えると、一層『本生經』との關連性が高まる。本稿では、その中でも樣??な動物の登場する動物寓話に焦點を合わせ、賢治の童話と『本生經』との比校を通した考察を行った。佛敎國家で崇拜の對象である象、想像の動物ながら東洋の民話によく登場する龍、食用の對象として描かれている豚、跛を引く馬、業を惱む鳥などの話が代表的な例である。ここで言及した『本生經』と賢治の童話は、動物の設定からスト-リ-に至るまで殆んど、致していることが分かった。その類似性が必然的であれ偶然的であれ、影響關係の究明が求めらむるし、一層細分化された分類と正確な分析が必要になる。 兩側の思想の種になっているのは、佛敎の輪廻思想であり、人間のこの世での行爲、卽ち、業の重要性が强調されている。敎訓的な性格は同じであるが、賢治の童話には時代の要求に應じて、近代の物質文明に對する辛辣な批判まで含まれている。この批判精神は、賢治童話のファンタ-ジ-とともに尤も評價される特性でもある。文明が發達すればするほど、そこに住んでいる人間の心の暖かさは次??失われるようになったからである。とにかく、兩側の重要な共通點は、直接行動として善行を勤めるということであろう。 『本生經』と賢冶童話については、ここで扱われた動物以外にも植物と人間の話など、檢討すべきところが多く殘っている。又、賢治童話の正確な影響關係の究明のため、西洋とか日本での樣??な話の詳しい調べも今後の課題にする。

        다문화가정 청소년들의 이중문화 수용태도와 심리적 적응 및 학교적응

        본 연구는 청소년정책연구원의 다문화청소년패널 자료 중에서 2015년 중학교 2학년 시점의 학생들 1348명을 대상으로 다문화가정 청소년들의 이중문화 수용태도에 따른 문화적응 유형 및 특징을 파악해보고, 각 유형에 따른 심리적·학교적응의 차이를 분석하였다. 우선 첫 번째, 군집분석을 통해 문화적응 유형은 크게 한국문화 친밀형, 외국문화 친밀형, 이중문화 소외형의 세 유형으로 구분되었으며, 이중문화 소외형이 가장 높은 비율을 차지하였고, 한국문화 친밀형과 외국문화 친밀형이 비슷한 비율을 차지하는 것으로 나타났다. 다음으로 다문화청소년의 문화적응 유형별 심리적·학교적응의 차이를 분석한 결과 한국문화 친밀형을 보이는 청소년들이 적응을 가장 잘 하고 있었으며, 이중문화 소외형의 아이들은 심리적·학교적응에 어려움을 겪는 것으로 나타났다. 연구결과를 통해 이중문화 소외형을 보이는 청소년들의 문화적응을 도울 수 있는 방법이 마련되어야 함을 추론해 볼 수 있으며, 이를 위한 구체적인 방안을 논의하였다. This study aims to examine acculturation type, difference of psychological·school adaption according to bicultural acceptance attitude. The research subjects were 1348 adolescents of 2015(the forth year survey) multi-cultural family panel by National Youth Policy Institute. First, as the results of cluster analysis, the acculturation type were identified as friendly type toward Korean culture, friendly type toward Foreign culture, and bicultural alienated type. The proportion of bicultural alienated type was highest, and the rate of friendly type toward Korean culture, toward Foreign culture were similar. Second, as the results of MANOVA analysis, adolescents of friendly type toward Korean culture adapted better than other two types, and adolescents of bicultural alienated type had difficulty in psychological·school adaption. This study shows that the programs had to be made to help acculturation of bicultural alienated type, so specific methods for that students were discussed.

        의과대학 수업수준에서의 준거설정 사례 연구

        역량중심교육은 학생교육의 질적 강화 및 구체적인 교육효과를 요구하고 있다. 이를 위해서는 적절한 성취수준 설정 및 학생의 도달 여부를 확인하는 준거참조평가(절대평가)가 보다 적절할 것이다. 준거참조평가를 위해서는 준거설정방법을 통해 각 성취수준을 구분하는 분할점수를 산출해야 한다. 기존 준거설정방법들은 대부분 고부담시험에 적용되었으나, 대학의 수업수준에서 이뤄진 연구는 거의 없었다. 본 연구는 준거참조평가를 시행하는 의과대학의 실제 수업에서 활용하는 준거설정방법을 소개하였다. 이를 위해 준거설정방법 중 Modified Angoff 방법과 Modified Ebel 방법을 이용하여 성취수준별 분할점수를 산출했다. 산출결과 Modified Ebel 방법이 Modified Angoff 방법보다 엄격하게 평정되어 분할점수가 높았다. 또한 Modified Ebel 방법은 예상정답률과 실제정답률을 사용했을 때 분할점수에 차이가 있었으며, 실제정답률을 반영하는 것이 보다 현실적이었다. 본 연구는 고부담시험에서 주로 이뤄지는 준거설정을 실제 의과대학에서 사용하는 방법으로 의과대학 성적에 적용해 보았다는데 의의가 있다. 창의성 기반의 역량중심교육을 보다 활성화하려면, 경쟁에 관심을 두는 규준참조평가보다는 협력학습을 가능하게 하는 준거참 조평가가 더욱 강조될 것이고, 이때 다양한 준거설정방법이 수업수준에 적용될 것으로 예측된다. Competency-centered education and the Fourth Industrial Revolution require qualitative strengthening of student education and concrete educational effects. To do this, it is appropriate to establish appropriate levels of achievement and to assess the criterionreferenced evaluation (so-called absolute evaluation). For the criterion-referenced evaluation, the division score between each grade should be calculated through the standard setting method. The existing standard setting methods were carried out in high-stakes examinations, but there were few studies conducted at the class level of universities. This study compares the standard setting method of the medical school which performs the criterion referenced evaluation in the actual class level. Among the standard setting methods, the modified score was calculated using Modified Angoff method and Modified Ebel method. As a result, Modified Ebel method was more strict than Modified Angoff method, and the Modified Ebel method had a higher cutoff score than the Modified Angoff method. It was more realistic to reflect actual difficulty. In this study, it is meaningful to apply the standard setting method, which is mainly used in the high stakes test, to the results of the medical college as a method used in actual medical college. In the future, competency-based evaluation will be more emphasized in order to activate competence-based education and creativity through cooperative learning rather than competition.

      • Antioxidant activity and quality characteristics of enzymatic hydrolysis and alcohol-treated mealworms for the development of aging-friendly formulae

        The purpose of this study is to develop an aging-friendly food material using mealworms (Tenebrio Molitor), considered as future food resources. To improve the antioxidant activity of mealworms, Alcalase and Flavozyme were added to mealworms in 0.5% and 1%, and then treated at 60°C for 4 hours. In addition, in order to improve the storage properties, the mealworms were treated with 50% alcohol for 24 hours to remove fat, and the quality characteristics of mealworms before and after degreasing were evaluated. Alcalase-treated mealworms showed superior in total polyphenol content, DPPH radical scavenging activity and ABTS scavenging activity to control or Flavozyme treatment groups. In addition, as a result of measuring acid value and peroxide value while storing mealworms at 70°C for 21 days, mealworms undergone the degreasing process showed lower results than those of the control group, indicating that the mealworms had better storage stability. In conclusion, our study confirmed that storage stability and antioxidant effect could be further improved by treatment with alcalase after removing the fat from mealworm.
