This research with 40 hearing impaired children was performed to find out the characteristics of the phonological processes for each age, hearing loss degree and aided threshold degree through the transcriptions. The phonological processes. of hearing impaired children are similar to those of normal children with a peculiar type of patterns in phonological processes. The results show that: (1) Between 5 and 6 year old groups the phonological processes were significantly different in palatal backing, glottal replacement and frication; between 6 and 7 year old groups the phonological processes were significantly different in velar fronting, labialization, alveolization and labial assimilation; between 7 and 8 year old groups the phonological processes, labialization, alveolization and alveolar assimilation, however 8 year old group showed more phonological processes than 7 year old group. (2) Between moderately-severe and severe hearing impaired groups, phonological processes were significantly different in the omissions of postvocalic, nasal and velar, stopping and stop assimilation. The differences of severe and profound groups were not found at all. (3) Aided hearing thresholds did not show any significant difference.

This paper is a descriptive study that inquires about stress and self-esteem of obese children to lend assistance about understanding emotional care as weight management of children. The data collection was given by the structured questionnaire during five days, from 5th to 10th on march in 2003 at one primary school located in K-city. The subjects were 286 children in 11-12 year old children. The instruments used for this study were an Obesity-Stress Scale(Cho Yang Rae, 1997) and a self-esteem questionnaire including Self-Perception Profile(Harter, 1982). Data were analyzed with frequency, percentages, x²-test, ANOVA, Pearson correlation using the SPSS program. The results were as follows; 1. A stress of obese children was the highest among three group, which was significant of statistical data. On the other hand, self-esteem do be shown the same as the above but was not significant statistically. 2. A relation between stress and self-esteem of obese children was shown to be in inverse proportion ratio so that the higher stress was the lower self-esteem. 3. To research the differences of the stress followed by the general characteristics of children, there are meaningful differences in sex, character, factor of stress, shape of father's body and shape of mother's body. 4. To research the differences of the self-esteem followed by the general character of children, there are meaningful differences in sex, character, shape of mother's body and socioeconomic status.
<i>Salvia miltiorrhiza</i> enhances the survival of mesenchymal stem cells under ischemic conditions

<P><B>Abstract</B></P><P><B>Objectives</B></P><P>To validate the enhanced therapeutic effect of <I>Salvia miltiorrhiza</I> Bunge (SM) for brain ischemic stroke through the anti‐apoptotic and survival ability of mesenchymal stem cells (MSCs).</P><P><B>Methods</B></P><P>The viability and the expression level of cell apoptotic and survival‐related proteins in MSCs by treatment of SM were assessed <I>in vitro</I>. In addition, the infarcted brain region and the behavioural changes after treatment of MSCs with SM were confirmed in rat middle cerebral artery occlusion (MCAo) models.</P><P><B>Key findings</B></P><P>We demonstrated that SM attenuates apoptosis and improves the cell viability of MSCs. In the rat MCAo model, the recovery of the infarcted region and positive changes of behaviour are observed after treatment of MSCs with SM.</P><P><B>Conclusions</B></P><P>The therapy using SM enhances the therapeutic effect for brain ischemic stroke by promoting the survival of MSCs. This synergetic effect thereby proposes a new experimental approach of traditional Chinese medicine and stem cell‐based therapies for patients suffering from a variety of diseases.</P>

Two new guaianolides, 4α,10α-dihydrory-1β(H),5β(H)-guai-11(13)-en-8α,12-olide (2) and 4β,10β-dihydroxy-5α(H)-1,11(13)-guaidien-8α,12-olide (3), from Carpesium macrocephalum were isolated, and their structures were elucidated on the basis of spectroscopic studies.

<P><B>Abstract</B></P> <P>To improve early renal allograft function, it is important to develop a noninvasive diagnostic method for acute T cell-mediated rejection (TCMR). This study aims to explore potential noninvasive urinary biomarkers to screen for acute TCMR in kidney transplant recipients (KTRs) using untargeted metabolomic profiling.</P> <P>Urinary metabolites, collected from KTRs with stable graft function (STA) or acute TCMR episodes, were analyzed using liquid chromatography-mass spectrometry (LC-MS). Multivariate statistical analyses were performed to discriminate differences in urinary metabolites between the two groups. Receiver operating characteristic (ROC) curve analysis was used to evaluate the diagnostic performance of potential urinary biomarkers.</P> <P>Statistical analysis revealed the differences in urinary metabolites between the two groups and indicated several statistically significant metabolic features suitable for potential biomarkers. By comparing the retention times and mass fragmentation patterns of the chemicals in metabolite databases, samples, and standards, six of these features were clearly identified. ROC curve analysis showed the best performance of the training set (area under the curve value, 0.926; sensitivity, 90.0%; specificity, 84.6%) using a panel of five potential biomarkers: guanidoacetic acid, methylimidazoleacetic acid, dopamine, 4-guanidinobutyric acid, and <I>L</I>-tryptophan. The diagnostic accuracy of this model was 62.5% for an independent test dataset.</P> <P>LC-MS-based untargeted metabolomic profiling is a promising method to discriminate between acute TCMR and STA groups. <P>CONTEXT: Polycystic ovary syndrome (PCOS) is a common endocrine disorder characterized by chronic oligo/anovulation, hyperandrogenemia, infertility, and metabolic alterations related to insulin resistance. These abnormalities in PCOS may have complex effects on pathophysiology of the endometrium, contributing to infertility and endometrial disorders. OBJECTIVE: The objective of this study was to examine dysregulated signaling pathways in the endometrium of patients with PCOS (PCOSE) by analyzing expression profiles with a pathway-oriented method. DESIGN: Microarrays, RT-PCR, laser capture microdissection, and immunohistochemistry were performed with endometrial tissues. SETTING: This study was performed at a university hospital laboratory. Patients: This study comprised 12 regularly cycling women and 12 PCOS patients. MAIN OUTCOME MEASURE: Dysregulated signaling pathways in PCOSE were identified as a gene set. RESULTS: Hierarchical clustering revealed distinct expression profiles for PCOSE and the endometrium of normal cycling women. Gene sets associated with androgen signaling were not enriched in PCOSE, although they affect ovarian physiology of PCOS patients. Several biological pathways including cell cycle, apoptosis, glycolysis, and integrin-Rho-cytoskeleton network were aberrantly down-regulated in PCOSE. Expression of genes constituting these gene sets enriched in normal cycling women was systemically down-regulated in PCOSE. Laser capture microdissection-coupled real-time RT-PCR and immunohistochemistry further demonstrated that cell proliferation in the stroma, but not the epithelium, is significantly reduced in PCOSE. CONCLUSIONS: Systemic down-regulation of various signaling pathways in PCOSE with extremely prolonged proliferative phase provides insight into the abnormal phenotypes that reflect pathophysiology of PCOS in the endometrium, possibly leading to increased risks of endometrial disorders.</P>
Identification of Grapevine leafroll-associated virus 3 Ampelovirus from Grapevines in Korea

Grapevine leaf roll-associated virus 3 (GLRaV-3) is one of the most important viral diseases of grapevine in the world. In this study, GLRaV-3 Ampelovirus was identi-fied from grapevines in Korea by analyzing viral coat protein size, nucleotide, and amino acid sequences. The molecular weight of viral coat protein from virus-infected in vitro plantlets was determined by western blot using a commercial GLRaV-3 polyclonal antibody. Western blot analysis showed a coat protein of about 43 kDa. RT-PCR product of about 942 bp which encoded the coat protein (CP) gene was amplified with specific primers. When the viruses existed at low titers in the host plant, the dsRNA had very specific template in RT- PCR amplification of fruit tree viruses. Especially, small-scale dsRNA extraction method was very reliable and rapid. Sequence analysis revealed that the CP of the GLRaV-3 Ko consisted of 942 bp nucleotide, which encoded 314 amino acid residues. The CP gene of GLRaV-3 Ko had 98.9% nucleotide sequence and 98.7% amino acid sequence identities with earlier reported GLRaV-3. This is the first report on molecular assay of GLRaV-3 Ampelovirus identified from Korea. The GLRaV-3 Ko CP clone would be very useful for breeding of virus resistant grapevines.

Ankyrins are a ubiquitously expressed family of intracellular adaptor proteins involved in targeting diverse proteins to specialized membrane domains in both the plasma membrane and the endoplasmic reticulum. We described here that the C-terminal domain of ankyrin-B interact specifically with Z-line portion of titin in yeast two-hybrid analysis, in vitro pull-down assays and localization experiments in COS-7 cells. In this study we provide the first experimental evidence that Z-line portion of titin is necessary for the localization of ankyrin-B and ankyrin-B links between the sarcolemma and the myofibril in costameres.
