        Quantitative Assay for the Binding of Jun-Fos Dimer and Activator Protein-1 Site

        The Jun and Fos families of eukaryotic transcription factors form heterodimers capable of binding to their cognate DNA enhancer elements. We are interested in searching for inhibitors or antagonists of the binding of the Jun-Fos heterodimer to the activator protein-1 (AP-1) site. The basic-region leucine zipper (bZIP) domain of c-Fos was expressed as a fusion protein with glutathione S-transferase, and allowed to form a heterodimer with the bZIP domain of c-Jun. The heterodimer was bound to glutathione-agarose, to which were added radiolabeled AP-1 nucleotides. After thorough washing, the gel-bound radioactivity was counted. The assay is faster than the conventional electrophoretic mobility shift assay because the gel electrophoresis step and the autoradiography step are eliminated. Moreover, the assay is very sensitive, allowing the detection of picomolar quantities of nucleotides, and is not affected by up to 50% dimethylsulfoxide, a solvent for hydrophobic inhibitors. Curcumin and dihydroguaiaretic acid, recently known inhibitors of Jun-Fos-DNA complex formation, were applied to this Jun-GST-fused Fos system and revealed to decrease the dimer-DNA binding.

        충남대학교 학술림 산책로 훼손실태 및 관리방안에 관한 연구

        This study concentrates an the trail of the CNU(Chungnam National University) forest. The amount of use and the pattern of damage are considered, as well as the extent of environmental damage. Finally, some managerial strategies are proposed in connection with sustainable maintenance. The trail of the university forest is composed of three sections; one is the neighborhood park and the Institute of Information Technology/Communications, the second is the Institute of Information Technology/Communications and the Dormitory, and the third section is the Dormitory and the Foreign prof's Apt. The total length of the trail is 2.6km. The daily amount of use is 1,269 visitors per day. The amount of using increased three times the amount of that in 1998. This study shows the pattern of damage of the trail in the order of root-exposure, widening, rock-exposure, deepening and the divergence in the frequency of occurrence of all these factors. Environmental damage is also graded thus: grade 3 is 27.4m, grade 4 is 0.8m, grade 5 is 1.5m, and grade 6 is 0.2m. Accordingly, some managerial strategies were proposed in connection with the sustainable trail maintenance.

        Load distribution factors suitable for the analyses of tunnels excavated by bench cut were determined. To achieve this, 3-D analyses were conducted while taking every construction stage into account. Then 2-D analyses were conducted which yield the same results with the 3-D results. The crown settlement normalized by the ultimate value was compared during the process to overcome the discrepancy caused by difference in dimension. When the length of the bench is very short compared to the diameter of the tunnel, in such cases as in short bench cut or in mini-bench cut, the supporting effect of the face does not disappear even after the bench is completely excavated since the face is still too close to the point of interest. The 4th load distribution factor was defined to stand for the advance of the face after the completion of the excavation cycle. The 4th load distribution factor turned out to be very useful in analyzing the behavior of tunnels excavated by bench cut with various bench lengths.

        내용과 형식 스키마가 독해에 미치는 영향

        The purpose of this research was to investigate the effects of content and formal schema on reading comprehension. Five hundred fiftynine subjects from high school were assigned to one of the following levels and treatment conditions : (1) Higher level & Schema Activation, (2) Higher level & Non-schema Activation, (3) Lower level & Schema Activation, and (4) Lower level & Non-schema Activation. To evaluate the effects of schema activation, two experiments were conducted : one was related to the content schema and the other to the formal schema. To evaluate the effects of content schema, three different types of tests were conducted : (1) cloze test, (2) guessing the meanings of nonsense words, and (3) immediate recall test. To evaluate the effects of formal schema instruction, four kinds of tests were conducted : (1) sorting the sentences according to the importance, (2) identifying the signal words, (3) immediate recall test, and (4) identifying the specific information. For content schema condition, results indicated that the subjects given the titles or pictures before reading in "Content Schema Activation" treatment had better grades than those of the other treatment in all types of tests, regardless of their levels. Schema activation helped the subjects to increase the cognitive predictability of missing words and to participate in the tasks more actively with risk-taking. And it was also shown that good readers tend to process the words meaningfully, while poor readers tend to process the words phonetically or morphologically. Formal schema activation through teaching the text organization also had a significant influence on three types of tests : sorting the sentences according to the importance, identifying the signal words, and immediate recall test, but not on identifying the specific information. The implications from this study can be briefly noted as follows : (1) In teaching reading, the student's background knowledge should be activated as a pre-reading activity. (2) In reading, it is more important to emphasize the student's schema than the features of the text. (3) Various educational interventions should be introduced, especially for the lower level students. (4) Teaching text structures can be a powerful method for the top-down processing strategy.

        한국 성인 기억력 평가를 위한 전산화 도구의 개발과 표준화

        연구목적 : 본 연구는 전산화 평가도구를 개발하고, 신뢰도와 타당도를 검증하며, 표준화 자료를 얻기 위하여 수행되었다. 방 법 : 숫자 따라하기검사, 시각 단기기억력검사, 한글언어청각학습검사, 시각도형학습검사를 전산화하였으며, 27명의 정상인을 대상으로 하여 검사-재검사 신뢰도를 검증하였다. 타당도를 검증하기 위하여 48명의 정상인을 대상으로 검사 성적을 기존의 인지기능 검사 성적과 비교하였으며, 또한 본 연구에서 개발된 검사들의 내적 일치도를 평가하였다. 20세부터 50세까지 고졸이상의 학력을 가진 한국 남녀 성인 150명을 대상으로 표준화 자료를 얻었다. 결 과 : 전산화 기억력 검사들의 검사-재검사 성적은 paired t-test 결과 차이가 없었으며, Pearson 상관계수 0.405∼0.873의 높은 상관관계를 보였다. 숫자 따라하기검사, 시각 단기기억력검사, 언어청각학습검사는 기존의 지필 혹은 전산화 검사 결과와 높은 상관관계를 보였다. 시각도형학습검사는 유용한 검사이나, 향후 방법론적 개선이 필요한 것으로 생각된다. 전산화 기억력 검사 성적은 학력이 높을수록 우수하였으며, 성별과 연령도 결과에 영향을 미쳤다. 전산화 기억력 검사의 표준화 자료와 해석기준을 제시하였다. 결 론 : 20세부터 50세까지 고졸 이상의 학력을 가진 한국 성인 남녀에서 신뢰성 있고 타당성 있게 사용할 수 있는 전산화 언어 및 시각 기억력 검사도구가 개발되었으며, 표준화 자료와 해석기준이 마련되었다. Objectives : This study was conducted to develop the computerized memory tests for Korea adults. It also aimed at evaluating the reliability and validity of these tests and providing their normative data. Methods : Computerized versions of digit span, visual span, auditory applications and correction. The test-retest reliabilities of each test was evaluated by the paired t-test and Pearson's correlation coefficient using the data from 27 normal persons. The validity of each test was evaluated by the Pearsons's correlation coefficient between the computerized test scores and the pre-existing cognitive function tests using the data from 48 normal persons. The normative data were obtained from the 150 Korean adults, age 20 to 50, whose educational levels were higher than high school graduate Results : There were no statistical difference between the means of the test and retest scores. High correlations were also observed between the tests and the retest scores(r ranged from 0.405 to 0.873). The computerized digit span, visual span, and auditory verval learning tests scores were highly correlated with the pre-existing memory measures. The computerized memory test scores were greatly influenced by the educational level. The visual learning test seems useful, however, it requires methodological refinement. The normative data and guidelines for interpretation were provided. Conclusions : Clinically applicable computerized memory assessment tools with high reliability and validity were developed. The normative data for the Korean adults aged 20 to 50 were obtained and the guidelines for the interpretation were provided.

      • 전주시의 상수처리에서 하천수와 호소수의 비교분석에 관한 연구

        The study on the water treatment plant of Jikok, Sangkwan, and Daeseong for one year showed that items, such as hydrogen ion concentration, Dissolved Oxygen, Suspended Solid and nitrate, etc., varied respectively, but all items coincide with the standard values. But in Jikok water treatment plant, coliform bacteria increased rapidly, and the value of BOD also increased. The study on treated water seemed that all items except nitrate and sulfonate properly treated and agreed to the standard values. In case of chlorine residues, they were found much during winter, so it needs shortening the input during winter season.

      • Glass-ionomer 시멘트 이장재의 5급와동 치은측 변연폐쇄효과

        The purpose of this study is to evaluate the gingival margin sealing effect of glass-ionomer lining cements in Class V cavities. Freshly extracted 120 human teeth were divided into 4 groups. V-shaped cavity was prepared on the cervical area of facial surface of each tooth. Except the 1 ㎜ inner area from the gingival margin, exposed dentin surface was lined with Dycal in Group 1, Lining cement in Group 2 and Chembond cement in Group 3. No liner was used in Group 4. Prepared enamel surface in Group 1 and Group 4 was etched. In Group 2 and Group 3, prepared enemel surface and lined cement surface were etched simultaneously. After etching, washing and drying, bonding agent was applied and cured, and composite resin was inserted and cured with light in every cavity of teeth of all Groups. Restored teeth were thermocycled altenatively in 2% methlene blue dye solution of 4℃ and 60℃. After that, with diamomd disk under water spray, they were sectioned faciolingually to be divided equally. Dye penetration on the sectioned surface was observed with stereomicroscope. After statistical analysis, the following results were obtained : 1. There was significant difference in gingival margin sealing effect in lined cavities, except those lined with Chembond cement, compared with unlined cavities (p<0.01). 2. There was no significant difference in gingival margin sealing effect of glass-ionomer lining cements such as Lining cement and Chembond cement compared with that of Dycal (p>0.05).

      • 고효율 포토폴리머 회절격자를 이용한 홀로그래픽 역다중화기 분석 및 구현

        우수한 광학적 특성과 저가의 소자 구성이 가능한 포토폴리머 부피형 회절격자를 이용하여 DWDM용 42채널 역다중화기를 설계하고 광학적으로 입증하였다. 실험결과, 50GHz 채널간격에 대해 0.18nm의 3dB 대역폭과 ~20 dB의 누화 억제, 1.6dB의 채널 균일도를 갖는 역다중화 특성을 보였다. A 42 channel demultiplexer for dense wavelength division multiplexing(DWDM) using photopolymer volumetric diffraction grating that has excellent optical properties and low cost capability, has been designed and optically demonstrated. From the experimental results, we have obtained the 3dB bandwidth of 0.18nm, crosstalk suppression of ~20dB and channel uniformity of 1.6dB for a 50GHz channel spacing.
