        Effect of Collagen Scaffold with Adipose-derived Stromal Vascular Fraction Cells on Diabetic Wound Healing: A Study in a Diabetic Porcine Model

        Diabetes can impair neovascularization important for wound healing, and this study investigated the hypothesis that this effect can be reversed by an active provisional matrix. Here, a 3D collagen scaffold and uncultured adipose-derived SVFs were firstly utilized to construct a collagen-targeting system for wound healing in a diabetic porcine model. The diabetic porcine models were made by injected streptozocin (STZ) intravenously and removed full thickness skin from the dorsum. Each animal received four different samples: Group A (control groups, n=24), Group B (SVFs groups, n=24), Group C (scaffold groups, n=24), Group D (SVFs -scaffold groups, n=24). PBS was applied to the wounds in Group A. 1×105 SVFs suspended in PBS were sprayed on the wounds in Group B. Scaffolds only with PBS were covered on the wounds in Group C. Scaffolds with equal volume of cell suspension containing 1×105 SVFs were covered on the wounds in Group D. The healing rates were calculated and compared among the groups and the tissues of the wound were taken and evaluated for histological analysis. The diabetic wound treated with the SVFs -collagen scaffold showed a significant decrease in wound size, an increase of neovascularization and a maximum VEGF and bFGF expression in comparison with SVFs and scaffold alone. The SVFs-collagen scaffold accelerates wound healing since it stimulates higher capillary formation causes major proangiogenesis factors expression in diabetic pigs suffering from skin defects, suggesting this approach could have utility for diabetic wound healing.

      • Evaluation of digital dental models obtained from dental cone-beam computed tomography scan of alginate impressions

        Objective: To investigate the dimensional accuracy of digital dental models obtained from the dental cone-beam computed tomography (CBCT) scan of alginate impressions according to the time elapse when the impressions are stored under ambient conditions. Methods: Alginate impressions were obtained from 20 adults using 3 different alginate materials, 2 traditional alginate materials (Alginoplast and Cavex Impressional) and 1 extended-pour alginate material (Cavex ColorChange). The impressions were stored under ambient conditions, and scanned by CBCT immediately after the impressions were taken, and then at 1 hour intervals for 6 hours. After reconstructing three-dimensional digital dental models, the models were measured and the data were analyzed to determine dimensional changes according to the elapsed time. The changes within the measurement error were regarded as clinically acceptable in this study. Results: All measurements showed a decreasing tendency with an increase in the elapsed time after the impressions. Although the extended-pour alginate exhibited a less decreasing tendency than the other 2 materials, there were no statistically significant differences between the materials. Changes above the measurement error occurred between the time points of 3 and 4 hours after the impressions. Conclusions: The results of this study indicate that digital dental models can be obtained simply from a CBCT scan of alginate impressions without sending them to a remote laboratory. However, when the impressions are not stored under special conditions, they should be scanned immediately, or at least within 2 to 3 hours after the impressions are taken.

        Biodegradable organosilica-based targeted and redox-responsive delivery system of resveratrol for efficiently alleviating ulcerative colitis

        Ulcerative colitis (UC) is characterized by a low redox potential in the colon. Targeting redox-responsivestrategies are highly desirable due to the capacity of improving the treatment efficiency of drugs againstUC. Resveratrol (Res), a natural ingredient, possesses prominent anti-inflammatory activity. However, thelimited bioavailability and poor water-solubility severely hinder its clinical translation. Herein, we fabricatean oral delivery carrier of Res-encapsulated tetrasulfide-containing organosilica nanoparticles(DSMSNs), functionalized by targeting component chondroitin sulfate (CS). Benefiting from smart‘‘switch” and targeted ‘‘guider”, CS shell enables high internalization and promotes selective accumulationin colon epithelial cells and macrophages through CD44-mediated pathway; meanwhile, glutathione(GSH) response-mediated rapid disassembly exhibits targeted Res release and efficient biodegradation,consequently achieving enhanced intracellular delivery of Res, improved Res-enabled treatment andrecovered gut microbial diversity in the colitis model to mitigate colonic inflammation. These findingsthus bring fresh insights into the potential of MONs in UC treatment as an oral delivery platform.

        NEDD8-activating enzyme inhibitor, MLN4924 (Pevonedistat) induces NOXA-dependent apoptosis through up-regulation of ATF-4

        <P><B>Abstract</B></P> <P>It has been reported that MLN4924 can inhibit cell growth and metastasis in various kinds of cancer. We have reported that MLN4924 is able to inhibit angiogenesis through the induction of cell apoptosis both in vitro and in vivo models. Moreover, Neddylation inhibition using MLN4924 triggered the accumulation of pro-apoptotic protein NOXA in Human umbilical vein endothelial cells (HUVECs). However, the mechanism of MLN4924-induced NOXA up-regulation has not been addressed in HUVECs yet. In this study, we investigated how MLN4924 induced NOXA expression and cellular apoptosis in HUVECs treated with MLN4924 at indicated concentrations. MLN4924-induced apoptosis was evaluated by Annexin V-FITC/PI analysis and expression of genes associated with apoptosis was assessed by Quantitative RT-PCR and western blotting. As a result, MLN4924 triggered NOXA-dependent apoptosis in a dose-dependent manner in HUVECs. Mechanistically, inactivation of Neddylation pathway caused up-regulation of activating transcription factor 4 (ATF-4), a substrate of Cullin-Ring E3 ubiquitin ligases (CRL). NOXA was subsequently transactivated by ATF-4 and further induced apoptosis. More importantly, knockdown of ATF-4 by siRNA significantly decreased NOXA expression and apoptotic induction in HUVECs. In summary, our study reveals a new mechanism underlying MLN4924-induced NOXA accumulation in HUVECs, which may help extend further study of MLN4924 for angiogenesis inhibition treatment.</P> <P><B>Highlights</B></P> <P> <UL> <LI> MLN4924 triggered NOXA-dependent apoptosis in a dose-dependent manner in HUVECs. </LI> <LI> Inactivation of neddylation caused up-regulation of ATF4, a substrate of CRL. </LI> <LI> NOXA was transactivated by ATF4 and further induced apoptosis. </LI> <LI> Knockdown of ATF-4 significantly decreased NOXA expression and apoptotic induction. </LI> <LI> We revealed a new mechanism underlying MLN4924-induced NOXA accumulation in HUVECs. </LI> </UL> </P>

        Influence of heat treatment on complex-shape rotating disk subjected to isothermal precision forging

        Influence of heat treatment on the mechanical properties of the forging was systematically investigated according to tensile test, hardness test and electrical conductivity test. Simultaneously, typical microstructures of the 7A09 aluminum alloy samples subjected to various solution temperatures and aging regimes were characterized by means of Transmission electron microscope (TEM) and Scanning electron microscope (SEM). In the case of the different solution temperatures, the size of precipitations increases with the increase in the solution temperature, which contributes to improving both strength and stress corrosion resistance of the forging. The solution temperature of 465 °C leads to the well comprehensive mechanical properties and the superior stress corrosion resistance of the forging. G.P. zone, η' phase and η phase are able to precipitate in the matrix of the forging in the case of different aging regimes, such as peak aging, two-stage overaging, two-stage aging, and regression and re-aging. The precipitation phases play an important role in enhancing the mechanical properties of the forging as an obstacle against the dislocation movement.

        A Weakly Cationic Temperature Tolerant and Salt Resistant Polymer: Synthesis and Properties

        In the petroleum industry, water-soluble polymers can be used as oil displacement agents. However, the use of water-soluble polymers is limited because of poor temperature and salt resistance. To improve temperature and salt resistance, a weakly cationic polymer with large side groups (PAM/AMPS/VI) was prepared by copolymerizing acrylamide (AM) with 2-acrylamido-2-methylpropane sulfonic acid (AMPS) and 1-vinylimidazole (VI). The viscosity of PAM/AMPS/VI water solutions can be increased more than 20 mPa·s compared with PAM/AMPS. In addition, the viscosity of the solution increased continuously after aging at 80℃, showing good temperature and salt stability. The protonated tertiary amine in the imidazole ring electrostatically interacts with the sulfonic group, increasing the viscosity and salt resistance of the polymer. The five-membered ring of imidazole also enhances the rigidity of the polymer chain and improves the temperature tolerance. As a confirm of the result, a complete spatial network of PAM/ AMPS/VI was observed in scanning electron microscopy (SEM) micrographs. Using weak cationic polymers with large side groups can provide a reference for the design of new temperature tolerance and salt resistant polymer.

        Comparative Study on the Toxic Effects of Red Tide Flagellates <i>Heterocapsa circularisquama</i> and <i>Chattonella marina</i> on the Short-Necked Clam (<i>Ruditapes philippinarum</i>)

        <P><I>Heterocapsa circularisquama</I> showed much higher toxic effects on short-necked clams than <I>Chattonella marina</I>. Clams exposed to <I>H. circularisquama</I> exhibited morphological changes concomitant with an accumulation of mucus-like substances in the gills, a profound reduction in filtration activity, and lysosomal destabilization in hemocytes. <I>Chattonella marina</I> was less effective than <I>H. circularisquama</I>, and <I>Heterocapsa triquetra</I> was almost harmless in all these criteria. These results suggest that <I>H. circularisquama</I> exerted its lethal effect on short-necked clams through gill tissue damage and subsequent induction of physiological stress.</P>

        LincR-PPP2R5C Promotes Th2 Cell Differentiation Through PPP2R5C/PP2A by Forming an RNA–DNA Triplex in Allergic Asthma

        Purpose: The roles and mechanisms of long noncoding RNAs (lncRNAs) in T helper 2 (Th2) differentiation from allergic asthma are poorly understood. We aimed to explore a novel lncRNA, LincR-protein phosphatase 2 regulatory subunit B' gamma (PPP2R5C), in Th2 differentiation in a mouse model of asthma. Methods: LincR-PPP2R5C from RNA-seq data of CD4+ T cells of asthma-like mice were validated and confirmed by quantitative reverse transcription polymerase chain reaction, northern blotting, nuclear and cytoplasmic separation, and fluorescence in situ hybridization (FISH). Lentiviruses encoding LincR-PPP2R5C or shRNA were used to overexpress or silence LincR-PPP2R5C in CD4+ T cells. The interactions between LincR-PPP2R5C and PPP2R5C were explored with western blotting, chromatin isolation by RNA purification assay, and fluorescence resonance energy transfer. An ovalbumin-induced acute asthma model in knockout (KO) mice (LincR-PPP2R5C KO, CD4 conditional LincR-PPP2R5C KO) was established to explore the roles of LincR-PPP2R5C in Th2 differentiation. Results: LncR-PPP2R5C was significantly higher in CD4+ T cells from asthmatic mice ex vivo and Th2 cells in vitro. The lentivirus encoding LincR-PPP2R5C suppressed Th1 differentiation; in contrast, the short hairpin RNA (shRNA) lentivirus decreased LincR-PPP2R5C and Th2 differentiation. Mechanistically, LincR-PPP2R5C deficiency suppressed the phosphatase activity of the protein phosphatase 2A (PP2A) holocomplex, resulting in a decline in Th2 differentiation. The formation of an RNA-DNA triplex between LincR-PPP2R5C and the PPP2R5C promoter enhanced PPP2R5C expression and activated PP2A. LincR-PPP2R5C KO and CD4 conditional KO decreased Th2 differentiation, airway hyperresponsiveness and inflammatory responses. Conclusions: LincR-PPP2R5C regulated PPP2R5C expression and PP2A activity by forming an RNA-DNA triplex with the PPP2R5C promoter, leading to Th2 polarization in a mouse model of acute asthma. Our data presented the first definitive evidence of lncRNAs in the regulation of Th2 cells in asthma.
