        Reduction of d-lactate content in sauerkraut using starter cultures of recombinant Leuconostoc mesenteroides expressing the ldhL gene

        <P>The n-form of lactate, which causes metabolic stress upon excessive dietary intake, is mainly produced by Leuconostoc sp., the predominant species in sauerkraut. To shift the metabolic flux of D-lactate from pyruvate to L-lactate, we expressed the L-lactate dehydrogenase (ldhL) gene in Leuconostoc mesenteroides ATCC 8293. The IdhL gene from Lactobacillus plantarum was introduced into L. mesenteroides using the shuttle vectors pLeuCM and pLeuCM42. To elevate the expression level of IdhL in L. mesenteroides, the nucleotides for pyruvate kinase promoter were fused to IdhL and cloned into above vectors to construct pLC18pkL and pLC42pkL. As results, introduction of pLC42pkL in L. mesenteroides significantly improved both L-LDH activity and L-lactate productivity during fermentation, decreasing the D-/L-lactate ratio. When used as a starter culture for sauerkraut fermentation, recombinant L. mesenteroides harboring pLC42pkL increased L-lactate concentration and decreased D-lactate concentration compared to the wild type strain. We newly developed a recombinant L. mesenteroides which has high L-lactate dehydrogenase activity and applied this strain to minimize the harmful effect of D-lactate during the sauerkraut fermentation. To the best of our knowledge, we demonstrate for the first time the effective use of recombinant Leuconostoc sp. for quality improvement of fermented foods.       • Ultrasound-assisted chiral derivatization of etodolac with (1R)-(-)-menthyl chloroformate for the determination of etodolac enantiomers

        <P>This study presents the first report of an ultrasound-assisted derivatization reaction between a carboxylic acid of etodolac (ETO) and a chiral derivatization reagent, (1R)-(-)-menthyl chloroformate (R-MCF). Fifty mu L of deproteinated mouse serum containing ETO enantiomers was derivatized with 125 mu L of 200 mM R-MCF and 17 mu L of pyridine (a catalyst), with the reaction facilitated by ultrasonic radiation for 13 min, which were the optimal conditions as determined by response surface methodology. After quenching the reaction by adding an aqueous L-proline solution, the mixture was subjected to salting-out assisted liquid-liquid extraction (SA-LLE), which provided phase separation for sample concentration as well as cleanup. The ETO diastereomers were separated on a Phenomenex Gemini C-18 column (150 x 4.6 mm, 5 mu m) under a simple gradient elution of a mobile phase containing a mixture of methanol: acetonitrile (10: 1, V/V) and 10 mM acetic acid at a flow rate of 1.0 mL min(-1), followed by fluorescence detection with excitation and fluorescence emission wavelengths of 235 nm and 345 nm, respectively. The developed method was validated for specificity, sensitivity, linearity, accuracy, precision, stability, and matrix effect. A good linearity in the range of 0.5-50.0 mu g mL(-1) for each ETO enantiomer with r(2) > 0.998 and acceptable values for the intra-day and inter-day accuracy and precision as well as negligible matrix effects supported the suitability and reliability of the method.         Studies on the Intestinal Microflora of Chicken Under Tropical Condition

        Three media, i. e., MOD-SD, M98-5 and M98-5 supplemented with chicken fecal extract were tested as isolation media for anaerobic bacteria present in the duodenum, jeju-ileum and cecum of chicken. The results showed that the mean colony counts of medium M98-5 were similar with those of MOD-SD medium in all intestinal samples at the incubation periods of 2, 6 and 10 days. Supplementation with chicken fecal extract of M98-5 medium significantly increased (p < 0.05) the colony counts of bacteria from the duodenum, jeju-ileum and cecum. The colony counts at 6-day incubation were similar with those at 10-day incubation, but were much higher than the counts at 2-day incubation. The major types of bacteria found in the duodenum and jeju-ileum of chicken were tentatively identified as Lactobacillus, Streptococcus and E. coli. In the cecum, ten tentatively identified groups of bacteria, namely, Streptococcus, Staphylococcus, Lactobacillus, E. coli, anaerobic coccus, Eubacterium, Propionibacterium, Clostridium, Fusobacterium and Bacteroides were isolated. Anaerobes were found to comprise nearly the entire microbial population of the cecum. Predominating in all sections of the intestine were homofermentative lactobacilli. The main Lactotacillus species in chicken intestine were L. acidophilus, L. fermentum and L. brevis.

      • High concentrations of single-walled carbon nanotubes lower soil enzyme activity and microbial biomass

        Nanomaterials such as single-walled carbon nanotubes (SWCNTs) may enter the soil environment with unknown consequences resulting from the development of nanotechnology for a variety of applications. We determined the effects of SWCNTs on soil enzyme activity and microbial biomass through a 3-week incubation of urban soils treated with different concentrations of SWCNTs ranging from 0 to 1000μgg<SUP>-1</SUP> soil. The activities of cellobiohydrolase, β-1,4-glucosidase, β-1,4-xylosidase, β-1,4-N-acetylglucosaminidase, l-leucine aminopeptidase, and acid phosphatase and microbial biomass were measured in soils treated with powder and suspended forms of SWCNTs. SWCNTs of concentrations at 300-1000μgg<SUP>-1</SUP> soil significantly lowered activities of most enzymes and microbial biomass. It is noteworthy that the SWCNTs showed similar effects to that of multi-walled carbon nanotubes (MWCNTs), but at a concentration approximately 5 times lower; we suggest that this is mainly due to the higher surface area of SWCNTs than that of MWCNTs. Indeed, our results show that surface area of CNTs has significant negative relationship with relative enzyme activity and biomass, which suggests that greater microorganism-CNT interactions could increase the negative effect of CNTs on microorganisms. Current work may contribute to the preparation of a regulatory guideline for the release of CNTs to the soil environment.

        Elimination of the cryptic plasmid in Leuconostoc citreum by CRISPR/Cas9 system

        <P>Leuconostoc spp. are important lactic acid bacteria for the fermentation of foods. In particular, L. citreum strains isolated from various foods have been used as host strains for genetic and metabolic engineering studies. In order to develop a food-grade genetic engineering system, L. citreum CB2567 was isolated from Kimchi. However, the isolated bacterium contained a cryptic plasmid which was difficult to eliminate. As the existence of the plasmid might hinder strain engineering, we eliminated the plasmid using an RNA-guided DNA endonuclease CRISPR/Cas9 system. We demonstrated that a plasmid-free L. citreum CB2567 host strain could be efficiently constructed through a two-step procedure: 1) transformation of the 'killer' plasmid expressing Cas9 endonuclease and a guide RNA (gRNA) targeting for a specific sequence in the cryptic plasmid, and 2) serial subculture without antibiotics for curing the killer plasmid. When the crude extract of L. citreum expressing Cas9 and the guide RNA was incubated with a PCR fragment containing the specific sequence recognized by the guide RNA, the PCR fragment was cleaved. Also, the cryptic plasmid pCB42 was successfully eliminated from the host strain after transforming the plasmid harboring Cas9 and the guide RNA. The Cas9 and gRNA expression plasmid used in this study can be applied for genome engineering purposes by additionally introducing an editing DNA template to repair the double strand DNA breakage caused by Cas9 in the genome of L. citreum. This study demonstrates the feasibility of developing CRISPR/Cas9-based genetic engineering tools to develop a safe host strain and construct food-grade lactic acid bacteria without residual antibiotic markers.</P>

        Potato (Solanum tuberosum L. cv. Gogu valley) protein as an antimicrobial agent in the diets of broilers.

        <P>Two experiments were conducted to evaluate potato protein (PP, experiment 1) and refined PP (RPP, experiment 2) obtained from Gogu valley tubers as an antimicrobial agent in broiler diets. In both the experiments, 1-d-old male Ross 308 chicks were allotted to 5 treatments and performance, nutrient retention, and microbial populations in excreta and cecum were studied. Dietary treatments were as follows: basal diet (negative control, NC), basal diet with antibiotic (positive control, PC, 10 mg/kg of avilamycin), and low, medium, or high levels of PP (0.25, 0.50, and 0.75%, respectively, in experiment 1) or RPP (200, 400, and 600 mg/kg, respectively, in experiment 2). The overall gain and retention of DM (d 20 to 21) and CP (d 20 to 21 and d 41 to 42) were greater in birds fed PC and high PP diets than birds fed the NC diet. Population of total aerobic bacteria and coliforms was lowest in the cecum and excreta of birds fed the PC diet and highest in birds fed the NC diet. An increase in dietary PP linearly improved BW gain, feed intake, and feed conversion ratio during starter phase and overall BW gain. Also, there was linear improvement in retention of DM (d 20 to 21) and CP (d 20 to 21 and d 41 to 42) and reduced populations of total aerobic bacteria and coliforms in the cecum (d 42) and excreta (d 28 and 42) due to an increase in dietary PP. In the second experiment, the PC diet and diets with increasing levels of RPP had no effect on performance and nutrient retention. Birds fed the PC diet had the lowest microbial population in excreta and cecum, whereas the population of total aerobic bacteria and coliforms in excreta and cecum decreased (linear, P < 0.05) as the level of RPP was increased in the diet. These results suggest that both PP and RPP obtained from Gogu valley potato tubers have in vivo antimicrobial activity.</P>

        Purpose: The study aimed to investigate the factors influencing consciousness of sexual violence in female high school students, focusing on sexual attitude and parental attitude. ethods: Data were collected from 248 female high school students in South Korea. A structured questionnaire was used for data collection through online survey. The collected data were analyzed by descriptive statistics, t-test, ANOVA, Tukey’s test, Pearson correlation coefficient, and multiple linear regression using the SPSS 25.0 program. Results: The mean score of consciousness of sexual violence in female high school students was 43.54 out of 48. The findings indicate that the mean score of consciousness of sexual violence differed y number of years in high school and academic achievement. Among sexual attitude, parental ttitude, and consciousness of sexual violence, there were meaningful correlations between onsciousness of sexual violence and sexual attitude (r=.132, p=.038,), love-hostility (r=.172, p=.006),and autonomous-control (r=.132, p=.038). Love-hostility aspect of parental attitude (=.159, p=.049) and academic achievement (=.208, p=.001) were found to be the important influencing factors on female high school student’s consciousness of sexual violence (F=5.720, p<.001). Conclusion: The findings of the study suggest that the love-hostility aspect of parental attitude and academic achievement influence consciousness of sexual violence of female high school students. Therefore, in order to improve consciousness of sexual violence in female high school students, sex education program and parent-education program should be developed with further research.

        Magnetic and Magneto-Optical Properties of Co-Sputtering FePt Films

        FePt thin .lms have been prepared at dierent substrate temperatures by DC magnetron co-sputtering. The .lms change from a disordered fcc structure to a L10 fct structure with temperature. The magnetic and the magneto-optical properties of those FePt .lms have been studied. The dependence of spectral changes on the formation of a chemically-ordered L10structure is observed in the polar magneto-optical Kerr rotation. In the spectra, the ultraviolet peak located in the high-energy range (4.0 { 4.5 eV), which is related to electronic transitions on Pt atomic sites, is observed for all samples. On the contrary, the peak related to transitions on Fe atomic sites, which is located in the low-energy range (1.5 { 2.0 eV), is only observed for highly chemically-ordered samples. Compared with the spectra for the L10 structure of FePt .lms pre- pared by using molecular beam epitaxy, the infrared peak related to Fe sites is shifted to low energy.

        강탄성 BiVO₄분말은 고상반응시켜서 제작하였으며, 원료 분말의 화학 반응 상태는 X-ray 회절 방법으로 확인했다. Czochralski 방법에 의하여 이 물질의 단결정을 육성하였으며 상전이점은 유전 상수의 온도 변화에 의하여 조사되었다. Ferroelastic BiVO₄has been synthesized through the conventional solid state reaction technique. Its phase and reaction mechanism are investigated by the X-ray diffraction method. The single crystals of BiVO₄have been grown by Czochralski method along with the optimum conditions. The phase transition of BiVO₄was observed through the temperature dependent dielectric constant measurements.
