        High Expression of KIFC1 in Glioma Correlates with Poor Prognosis

        Objective : Kinesin family member C1 (KIFC1), a non-essential kinesin-like motor protein, has been found to serve a crucial role in supernumerary centrosome clustering and the progression of several human cancer types. However, the role of KIFC1 in glioma has been rarely reported. Thus, the present study aimed to investigate the role of KIFC1 in glioma progression. Methods : Online bioinformatics analysis was performed to determine the association between KIFC1 expression and clinical outcomes in glioma. Immunohistochemical staining was conducted to analyze the expression levels of KIFC1 in glioma and normal brain tissues. Furthermore, KIFC1 expression was knocked in the glioma cell lines, U251 and U87MG, and the functional roles of KIFC1 in cell proliferation, invasion and migration were analyzed using cell multiplication, wound healing and Transwell invasion assays, respectively. The autophagic flux and expression levels matrix metalloproteinase-2 (MMP2) were also determined using imaging flow cytometry, western blotting and a gelation zymography assay. Results : The results revealed that KIFC1 expression levels were significantly upregulated in glioma tissues compared with normal brain tissues, and the expression levels were positively associated with tumor grade. Patients with glioma with low KIFC1 expression levels had a more favorable prognosis compared with patients with high KIFC1 expression levels. In vitro, KIFC1 knockdown not only inhibited the proliferation, migration and invasion of glioma cells, but also increased the autophagic flux and downregulated the expression levels of MMP2. Conclusion : Upregulation of KIFC1 expression may promote glioma progression and KIFC1 may serve as a potential prognostic biomarker and possible therapeutic target for glioma.

      • Study on seismic performance of SRC special-shaped columns with different loading angles

        In order to study the influence of loading angles on seismic performance of steel reinforced concrete (SRC) specialshaped columns, cyclic loading tests and finite element analysis (FEA) were both carried out. Seven SRC special-shaped columns, including two L-shaped columns, three T-shaped columns and two cross-shaped columns, were tested, and the failure patterns of the columns with different loading angles were obtained. Based on the tests, the FEA models of SRC special-shaped columns with different loading angles were established. According to the simulation results, hysteretic curves and seismic performance indexes, including bearing capacity, ductility, stiffness and energy dissipation capacity, were analyzed in detail. The results showed that the failure patterns were different for the columns with the same section and different loading angles. With the increasing of loading angles, the hysteretic curves became fuller and the bearing capacity and initial stiffness appeared increasing tendency, but the energy dissipation capacity changed insignificantly. When the loading angle changed, the ductility got better with the larger area of steel at the failure side for the unsymmetrical section and near the neutral axis for the symmetrical section, respectively.

        Reliability Evaluation and Prediction of Deep Buried Tunnel Based on Similarity Theory and Model Test

        Deep buried tunnel has a complex stress environment and large stress level. At present, it is difficult to dynamic monitoring for such tunnels. In order to make the safety evaluation and prediction of tunnel closer to the engineering practice, this paper deduces the tunnel reliability evaluation algorithm. The time-varying reliability of deep tunnel excavation process is analyzed and predicted. Based on the similarity theory, the actual deep tunnel excavation is simulated by indoor model test, the stress and deformation are monitored. The results show that after the tunnel excavation, the reliability of the structure does not decrease gradually, but increases slowly first and then decreases sharply. With the increase of the buried depth of the tunnel, the structural reliability decreases gradually. When the buried depth reaches a certain depth, the structural reliability will decrease rapidly. The influence of different parameters on the structural reliability has different sensitivity. The sensitivity of friction angle is the highest, and the sensitivity of gravity is the lowest. The sensitivity has little effect with the change of tunnel excavation time and tunnel buried depth. The accuracy of this method is verified by the comparative analysis of model box tests.

        Research on Calculation Method of Suspension Bridge Internal Force under Random Traffic Load

        Suspension bridge is a flexible structure. The weight, location and number of traffic on the bridge will affect the geometric alignment of the bridge. Although the finite element software can analyze the geometric nonlinearity of the bridge, the calculation of random load combination in finite element software is huge. In this paper, based on the deformation coordination, static equilibrium and constitutive model of the bridge, the force expression considering the geometric nonlinearity of the bridge is derived. Finite element software is used to simulate the internal force of components of suspension bridge under train load and vehicle load. The accuracy is verified by comparing the finite element results. Considering the random number of traffics, the random location of traffics, and the random weight of traffics, this paper analyzes the stress state of bridge under the random process of train load and vehicle load respectively. The method in this paper can be well used to analyze the actual stress state of the bridge, and provide a theoretical basis for related calculations.

        Effects of Adding Essential Oil to the Diet of Weaned Pigs on Performance, Nutrient Utilization, Immune Response and Intestinal Health

        The objective of this study was to evaluate the effects of adding essential oils to the diet of weaned pigs on performance, nutrient utilization, immune response and intestinal health. A total of 96 weaning pigs ($8.37{\pm}1.58$ kg) were allotted to one of three dietary treatments. The treatments consisted of an unsupplemented basal diet (negative control, NC) or similar diets supplemented with 0.01% of an essential oil product which contained 18% thymol and cinnamaldehyde (EOD) as well as a diet supplemented with 0.19% of an antibiotic mixture which provided 150 ppm chlortetracycline, 80 ppm colistin sulfate and 50 ppm kitasamycin (positive control, PC). Each treatment was provided to eight pens of pigs with four pigs per pen. Over the entire 35 d experiment, ADG and fecal score were improved (p<0.05) for pigs fed the PC and EOD compared with the NC. Dry matter and crude protein digestibility as well as lymphocyte proliferation for pigs fed the PC and EOD diets were increased significantly compared with NC (p<0.05). IGF-I levels in plasma were significantly increased (p<0.05) in pigs fed the PC diet compared with pigs fed the NC diet. Interleukin-6 concentration was lower (p<0.05) and the tumor necrosis factor-${\alpha}$ level was higher (p<0.05) in the plasma of pigs fed the EOD diet than the NC diet. Plasma total antioxidant capacity level increased (p<0.05) in pigs fed the EOD diet compared with pigs fed the NC. Villus height to crypt depth ratio in the jejunum was greater (p<0.05) in pigs fed the PC and EOD diets than the NC. The numbers of E. coli in the cecum, colon and rectum were reduced (p<0.05) in pigs fed the PC and EOD diets compared with the control. In the colon, the ratio of Lactobacilli to E. coli was increased (p<0.05) in pigs fed the EOD diet compared with NC diet. Total aerobe numbers in the rectum were decreased (p<0.05) in pigs fed the PC and EOD diets compared with the control. Collectively, these results indicate that blends of essential oils could be a candidate for use as an alternative to traditional antibiotics in weaning pig diets.

      • The comparative study of Chinese and Korean sports culture

        PURPOSE: Through cultural exchange and comparison of sports culture between the two countries, this paper aims to explore their common sports values and learn from Korean management experience and successful concepts to further promote the prosperity and development of sports economy and culture in China. METHODS: The author once made an academic visit in Yongin University of South Korea as a sports researcher, and investigated the sports culture in different regions of Korea. By investigating and recording the various forms there, he had a good understanding of Korean sports culture, which provided an important basis for this study. RESULTS: The goal orientation of sports varies from country to country. In the case of school physical education, Chinese schools, influenced by the traditional teaching mode, pay more attention to the teachers" teaching, and all sports activities students engage in are decided and arranged by teachers. Although the current educational reform has improved the students" principal position in the learning process, the status quo has not completely been changed. While Korean school physical education attaches more importance to the students" learning who are allowed to choose their favorite sports and given more consideration in the learning process. Korean educational goal is to cultivate students" awareness of physical exercise, hence their lifelong sports habits. CONCLUSIONS: Korea has been ahead of China in sports management, so we can learn its advanced experience. And we should continue to improve our sports policies and regulations which should be operable and practical on account of the current market economic system, achieving that there are laws to go by and developing sports in accordance with them. In the training of athletes, we should attach great importance to knowledge learning, achieving the common progress of training and learning, hence fundamentally solving the long-standing contradictions between them.

        A Hybrid Double Self-lift Switched-Coupled Inductor Quasi-Z-Source Inverter

        A hybrid double self-lift switch-coupled-inductor quasi-Z-source inverter (HDSCL-qZSI) is proposed in this paper. The voltage boost capability is higher and the voltage and current stress on components are lower than the existing quasi-Z-source topologies. Owing to the strong current handling ability and step-down coupled-inductor, the magnetic elements are smaller than that of mSSCL-qZSI and SCL-qZSI whose structure are similar to that of HDCSL-qZSI. The operation principles, comparisons of voltage and current stress on components and the size of magnetic elements are analyzed theoretically. Finally, the eff ectiveness of the proposed HDSCL-qZSI was validated by a 1 kW prototype fabricated in the laboratory.

        piRNA-1742 promotes renal cell carcinoma malignancy by regulating USP8 stability through binding to hnRNPU and thereby inhibiting MUC12 ubiquitination

        Accumulating studies have confirmed that PIWI-interacting RNAs (piRNAs) are considered epigenetic effectors in cancer. We performed piRNA microarray expression analysis on renal cell carcinoma (RCC) tumor tissues and paired normal tissues and performed a series of in vivo and in vitro experiments to explore piRNAs associated with RCC progression and investigate their functional mechanisms. We found that piR-1742 was highly expressed in RCC tumors and that patients with high piR-1742 expression had a poor prognosis. Inhibition of piR-1742 significantly reduced tumor growth in RCC xenograft and organoid models. Mechanistically, piRNA-1742 regulates the stability of USP8 mRNA by binding directly to hnRNPU, which acts as a deubiquitinating enzyme that inhibits the ubiquitination of MUC12 and promotes the development of malignant RCC. Subsequently, nanotherapeutic systems loaded with piRNA-1742 inhibitors were found to effectively inhibit the metastasis and growth of RCC in vivo. Therefore, this study highlights the functional importance of piRNA-related ubiquitination in RCC and demonstrates the development of a related nanotherapeutic system, possibly contributing to the development of therapeutic approaches for RCC.

        Highly efficient catalytic valorization of biomass-derived furfural in methanol and ethanol

        Catalytic valorization of furfural (FUR) using a simple and low-cost metal catalyst has been considered asa hot topic in sustainable chemistry. In this work, a new Ni@R-Ca catalyst is prepared via the autocombustionand in situ reduction method. When it is employed to promote the catalytic valorization ofFUR in aliphatic alcohol, a nearly complete FUR conversion with excellent product selectivity is obtainedunder mild reaction conditions. Using methanol as reaction medium, the sole product is furfuryl alcohol(FA) in which ca. 99% yield of FA was attained. While, in ethanol, the obtained products mainly contain FAand furan-2-acrolein (FUA) in which a 94% conversion of FUR with a 91% total selectivity of FA and FUAwas acquired. Based on the characterization of catalyst including XRD, SEM, XPS and H2-TPR techniques,it is concluded that the synergy effect of metal Ni and R-Ca crystal lattice plays a crucial role in theefficient conversion of FUR in aliphatic alcohol.
