      • 박 種子油中의 C30Δ5,24-Triterpene Alcohol 및 C29Δ7,22-Sterol의 構造

        Two kinds of sterol isolated from unsaponifiables of gourd seed oil were identified, the structure as cucurbitadienol(10α-cucurbita-5.24-dien-3β-o1, C30??5,24-triterpene alcohol) and chondrillasterol[(24R)-24ethy1-5α-cholesta-7, E-22-dien-3β-o1; C29??7,22-setrol], by infrared, mass and proton nuclear magnetic resonance spectrometry using lanthanide shift reagent. The former has not been detected in higher plants so far although the compound has been synthesized from similar compound. The latter is unusual in the plant kingdom and has hitherto been found only in several species of green algae(Chlorophyta).

        米糠油, 大豆油, 참깨油, 들깨油, 산초 및 菜種油의 불감화물중에 存在하는 free sterol을 column chromatography法과 T.L.C法에 의해 分溜 定量하였고 total tocopherol에 對해서는 Emmeri-Engel法에 의해 定量하였다. 1. Free steol을 용출할 때는 n-hexane-20% benzenedyddor에서만 용출됨을 확인하였다. 2. 이들 植物油의 不??化物中에 存在한 free sterol와 total tocopherol의 含量은 다음과 같다. Free Sterol(㎍/g) Total Tocopherol(㎍/g) 米糠油 1300 490 大豆油 1500 565 참깨油 773 425 들깨油 516 635 산초油 520 740 菜種油 640 470 Free sterols contained in unsaponifiable matters of vegetable oils such as, rice bran oil, soybean oil, sesame oil, japanes pepe oil, rape seed oil. and perilla oil are determined by the column chromatographic and thin layer chromato raphic methods. And also total tocopherols are investigated by Emmerie-Engel Procedure 1. For the elution of free sterol, it is only eluted when the n-hexane-20% benzene solution were added on the column paked silica gel. 2.       • Prochiral 탄소를 갖는 화합물에서 광학활성체 합성(Ⅱ)

        The chiral heterocycle groups as an auxiliary use for asymmetric synthesis, Especially C-4chiral thiazolidine -2-thione has been shown to most effective for the regioselectivity of asymmetric synthesis. In order to increase the differentiation of regioselectivity between to idential groups in prochiral symmetric dicarboxylic acids. I selected 4(S)-isopropy1-1, 3-thiazolidine-2-thione [4(S)-IPTT which was synthesized from L-valine as an auxiliary. The 3-methy[ glutaric acid was reacted with 4(S)-IPTT in presence of DCC in pyridne, to give 1,5-di[4(S)-isoprony1-2-thione-1,3-thiazolidinyl]-3-methyl-1,5-pentanedione. This diamide was subjected to aminolysis with 1 molar equivalent of piperidine in CH₂Cl₂at low temperature. As resutl, very nigh regioselectivity of the reaction was observed in case.

      • 실험적 녹농균성 각막염에 대한 Cefotetan의 치료 효과

        This study was carried out for the purpose of evaluation of effect of cefotetan against the pseudomonas induced keratitis. The corneal infection caused by pseudomonas organisms have a poor prognosis and their increased prevalence and general resistance to therapy make them a serious problem for to ophthalmologists. So, the author introduced experimental keratitis in the rabbit cornea by Ps. aeruginosa and Compared effects of the cefotetan and tobramycin. The cefotetan is a new antibiotic of cephalosporin system which is effective in vivo and vitro against not only gram positive but also gram negative organism and is stable against the β-lactamase. The results obtained were as follows: 1. Clinically, cefotetan and gentamicin were both effective against experimentally induced .pseudomonas keratitis and there was no significant difference between the efficacy of cefotetan and that of tobramycin. 2. In culture study, the test after treatment showed significantly fewer colonies on the platesfrom the cefotetan, tobramycin treated groups than those from the saline treated group, and there was no significant difference between cefotetan treated, and tobramycin treated group.

        The incidence of double primary cancer is about 10% in all kinds of cancers. Double primary cancer is usually accompanied by gastrointestinal cancer. Esophageal cancer is supposed to be frequently accompanied by squamous cell carcinoma of the head and neck or the upper respiratory tract, because of such common carcinogens as smoking and alcohol ingestion. Esophageal cancer and hypopharyngeal cancer are usually diagnosed by esophagogastric endoscopy with biopsy. We performed endoscopic ultrasound-guided Trucut biopsy(EUS-TCB) to diagnose a mediastinal mass in a 49 year-old patient, who visited our hospital because of voice change. During procedure, we accidently found enlarged celiac lymph node, and obtained tissue from the mediastinal lymph node and the celiac lymph node. We confirmed double primary cancer of squamous cell carcinoma in the esophagus and the hypopharynx via esophagogastric endoscopy.

      • 결핵균 30-kDa와 32-kDa 단백항원의 분리정제와 마우스 비장세포 증식에 미치는 영향

        Identification and characterization of individual components of M.tuberculosis have long been a focus of research on tuberculosis. The 30-kDa and 32-kDa antigens are major constituents of M bovis BCG and M. tuberculosis culture fluids. Because 30-kDa and 32-kDa antigens are partially identical, these antigens are difficult to purify in large amounts by biochemical techniques. This study was performed to purify the 30-kDa and 32-kDa antigens complexes primarily purified by 50% ammonium sulfate precipitation, hydroxylapatite chromatography and Sephadex G-75 gel filtration. And then further purification for separation of the two antigens was accomplished on preparative isoelectric focusing. Recovery of 30-kDa and 32-kDa antigens during above the purification procedures were 28% and 14%, respectively, and 147.0 and 59.8-fold purification were showed, respectively. On silver stained SDS-PAGE gels, the purified 32-kDa antigen gave a single band at 32-kDa molecule, while 30-kDa antigen gave one major band at 30-kDa molecule and faint additional band at 32-kDa. The pI of 30kDa-and 32-kDa antigens were 4.3 and 4.6, respectively. The partial identity between these two antigens was observed through the same pattern of reactivity of antigens in the ELISA and precipitation pattern of immunodiffusion. We also examined the immunological activities of both purified antigens by splenocyte proliferation of BCG-infected mice. Proliferative response to the 30-kDa and 32-kDa antigens did not show significant difference. However, macrophage-like cells existed in spleen cells from BCG-infected mice were involved in the suppression of the proliferative response to mycobacterial antigen. These results suggest that the 30-kDa and 32-kDa antigens could be effectively purified by the IEF and reactions of partial identity between the two antigens were found. However, 30-kDa antigen was the more immunogenic antigen than 32-kilodalton.

        The hydroindanedione and the hydronaphthalenedione intermediates have been synthesized for the purpose to obtain important CD-rings for total synthesis of steroids. The starting materials, 2-methylcyclopentane-l,3-dione was obtained through the reaction of succinic acid with propionyl chloride and also 2-methylcyclohexane-l,3-dione newly prepared by the treatment of glutaric acid with propionyl chloride in AIC_3/CH_3N0_2. In the synthesis of 2-methyl-2-(3'-oxobutyl)-cyclopentane-l,3-dione by Michael addition of methyl vinyl ketone to 2-methylcyclopentane-l,3-dione the yield and purity of the product could be appreciably increased by working in an aqueous medium without a basic catalyst, but in the synthesis of 2-methyl-2-(3'-oxobutyl)-cyclohexane-l,3-dione by Michael addition of methyl vinyl ketone to 2-methylcyclohexane-l,3-dione the reaction time of the product could be shortened. with a basic catalyst. (+)-(7aS)-7, 7a,-Dihydro-7a-methyl-l, 5(6H)-indanedione and (+)-(8aS)-3, 4, 8, 8a-tetrahydro -8a-methyl-I, 6(2H,7H)-naphthalenedione were obtained by the stereos elective cyclization of 2methyl-2-(3'-oxobutyl)-cyclopentane-l, 3-dione and 2-methyl-2-(3'-oxobutyl)-cyclohexane-I, 3-dione with optically active catalyzers.       • 3-아실-1.3-티아졸리딘-2-티온을 이용한 아미드 및 아미드유도체 합성

        3-아실 티아졸리딘-2-티온(ATT)의 가아민 분해반응은 아미드 및 아미드 유도체 합성에 효과적으로 이용 되었다. ATT와 아미노 알콜이 반응할때 화학 선택적인 반응이 관찰되었다. ATT를 이용하여 몇종의 아미드 알카리오디 화합물과 Li에 민감한 ionophore를 합성 하였다. 아울러 ATT유도체가 페니실린에 민감한 Bacillus Substilus에 생물학적 활성을 나타내었었다.

        純粹 메탄올과 메탄올-물 混合溶液에 空氣를 飽和시켜 Co??-γ線을 照射하여 H₂O₂, HCHO을 測定하였다. 本 實驗에서 얻어진 G값과 眞空中의 값을 比較검토하고 이를 토대로 Scavenger의 影響을 照射하였다. Yields of gydrogen peroxide and formaldehyde by Co??-gamma radiolysis of pure methanol-water mixture solution saturated with air have been investigated at room temperature. The G-values obtained by the present study and degassed mixture solution is compared and the effect of scavenger is discussed, on the basis of the observed date.

      • 산딸나무 열매로부터 저밀도 지질 단백질(LDL) 산화 역제물질의 분리

        산딸나무 열매를 100% MeOH로 추출하고, 얻어진 추출물을 EtOAc, n-BuOH 및 물로 용매 분획 하였다. 이 중 EtOAc 분획으로부터 silica gel과 octadecyl silica gel(ODS) column chromatography를 반복하여 1종의 lignan 화합물을 분리, 정제하였다. 화합물의 화학구조는 NMR, Ms 및 IR 등의 스펙트럼 데이터를 해석하여, (+)-pinoresinol (1) 로 동정하였다. 이 화합물은 산딸나무에서 처음 분리되었다. LDL-항산화활성 억제활성이 20㎍/mL 처리농도에서 85.1±1.3%로 나타났다. (+)-Pinoresinol was isolated from the EtOAc fraction of methanol extracts of the fruits of Cornus kousa Burg. The chemical structures of (+)-pinoresinol was determined by spectroscopy method, as well as 1D- and 2D-NMR (^(1)H-^(1)H COSY, HSQC and HMBC) spectroscopy. This compound was the first to be isolated from C. kousa Burg. Also, (+)-pinoresinol inhibited LDL-oxidation with the inhibitory activity of 85.1±1.3% at a concentration of 20 ㎍/mL.
