틸리피아에서 분리한 Edwardsie tarda 의 성상과 항원성에 대하여

1987年 3月 釜山水産 大學 魚病學 實驗室에서 飼育 中이던 틸라피아 (Oreochromis niloticus)에 細菌性 疾病이 發生하여 慢性的으로 몇마리씩 斃死하였다. 本 硏究는 이러한 疾病의 原因을 밝히기 위하여 原因菌을 分離하였으며, 型態的, 生化學的 性狀을 觀察하고 病原性, 藥劑感受性 實驗을 하였으며 試驗抗原을 制作하여 그 抗原性을 조사하였는바, 그 結果를 要約하면 다음과 같다. 1. 病魚에서 分離된 原因菌은 Edwardsiella tarda로 同定되었다. 2. 分離菌을 틸라피아에 접종하여 再現性實驗을 한 結果 전형적인 E. tarda의 감염증상을 나타내고 死亡했으며, 魚類로부터 同一菌을 얻을 수 있었다. 3. 分離菌은 oxytetracycline에서 感受性이 높았고, MIC는 6.9로 나타났다. 4. 分離菌은 病原性이 강한 菌이었으며, 48-LD5O은 10^(6.4)cell/fish 이었다. 5. 試驗抗原으로, 포르말린사균항원, 초음파처리항원, 균여과액항원을 사용하였으며, 이 세가지를 사용한 抗原性試驗 결과 포르말린사균항원의 시험구에서 높은 응집가와 방어력을 나타내었다. In March 1987, a bacterial disease occured among cultured tilapia (Oreochromis niloticus). in the aquarium of Fish Pathology Laboratory of National Fisheries University of Pusan. The diseased fish showed abdominal inflamation of ascites. The causative organisms isolated from the kidney were Gram negative rods. On SS agar colonies developed within 48 hours at 25℃. On the basis of the morphological and biochemical characteristics. the organisms were identified as Edwardsiella tarda. They were sensitive to chloramphenicol, ampicilline and oxytetracycline. The pathogenicity was proved positive by intraperitoneal injection. Three kinds of antigen were prepared with E. tarda for the immune response in tilapia : i.e., by inactivating the strain with 0.4% formalin for 24 hrs at 25℃ ultrasonicating it 3 times with 5 watts for 30 seconds each time and filtrating it through millipore filter. The formalin killed antigen showed good efficacy at the injection concentration of over 10^7 cells per fish of 50g. All the immunized tilapia showed only a little agglutination titer to the three antigens. The highest survival rate was recorded in challenged tilapia immunized with formalin killed cells.
실험적 녹농균성 각막염에 대한 Cefotetan의 치료 효과

This study was carried out for the purpose of evaluation of effect of cefotetan against the pseudomonas induced keratitis. The corneal infection caused by pseudomonas organisms have a poor prognosis and their increased prevalence and general resistance to therapy make them a serious problem for to ophthalmologists. So, the author introduced experimental keratitis in the rabbit cornea by Ps. aeruginosa and Compared effects of the cefotetan and tobramycin. The cefotetan is a new antibiotic of cephalosporin system which is effective in vivo and vitro against not only gram positive but also gram negative organism and is stable against the β-lactamase. The results obtained were as follows: 1. Clinically, cefotetan and gentamicin were both effective against experimentally induced .pseudomonas keratitis and there was no significant difference between the efficacy of cefotetan and that of tobramycin. 2. In culture study, the test after treatment showed significantly fewer colonies on the platesfrom the cefotetan, tobramycin treated groups than those from the saline treated group, and there was no significant difference between cefotetan treated, and tobramycin treated group.
중합효소연쇄반응법(Polymerase Chain Reaction, PCR)에 의한 남해안 양식산 어류로부터 Birnavirus의 검출

To detect birnavirus from cultured marine fish, RT-PCR assay was developed. This method was specific for aquatic birnaviruses that include IPNV Sp., IPNV Ab, IPNV VR-299 and MBV Y6. The birnavirus gene was detected (birnavirus positive samples detected 46/50) from clinical samples signed with abdominal distension and overall darkening even though the samples gave negative results in virus isolation (birnavirus isolate with CHSE-214 cell showed 12/50).
