        Increased Non-Rapid Eye Movement Sleep by Cocaine Withdrawal: Possible Involvement of A2A Receptors

        This study attempted to clarify whether cocaine withdrawal altered sleep architecture and the role of adenosine receptors in this process. Cocaine (20 mg/kg) was administered subcutaneously once per day for 7 days to rat implanted with sleep/wake recording electrode. Polygraphic signs of undisturbed sleep/wake activities were recorded for 24 h before cocaine administration (basal recording as control); withdrawal-day 1 (after 1 day of repeated cocaine administration), withdrawal-day 8 (after 8 days of repeated cocaine administration), and withdrawal-day 14 (after 14 days of repeated cocaine administration), respectively. On cocaine withdrawal-day 1, wakefulness was significantly increased, total sleep was decreased, non-rapid eye movement sleep was markedly reduced, and rapid eye movement sleep was enhanced. Sleep/wake cycles were also increased on cocaine withdrawal day 1. However, non-rapid eye movement sleep was increased on withdrawal-day 8 and 14, whereas rapid eye movement sleep was decreased and no significant changes were observed in the total sleep and sleep/wake cycles during these periods. Adenosine A2A receptors expression was increased on withdrawal-day 8 and 14, whereas A1receptors levels were reduced after 14 days of withdrawal and the A2B receptors remained unchanged. Our findings suggest that alterations of sleep and sleep architecture during cocaine subacute and subchronic withdrawals after repeated cocaine administration may be partially involved in A2A receptors over-expression in the rat hypothalamus.

The effects of a low dose (10 mg/kg) and a high dose (200 mg/kg) of RGE were compared in rats. After recovery from a surgical operation enabling electroencephalograms recordings, rats were administered RGE orally. RGE was administered orally for 1 day or once per day for 5 days in either 10 or 200 mg/kg doses. Polygraphic signs were recorded for 12 h after oral administration of RGE. Both treatment with a large dose (200 mg/kg) of RGE for one day and treatment with either a large or a small dose for 5 days reduced the number of sleep.wake cycles. Daily treatment with RGE (either 10 or 200 mg/kg) for 5 days augmented NREM and total sleep, but reduced wakefulness. Delta wave activity recorded during non-REM (NREM) sleep and REM sleep was increased after one treatment with RGE (either 10 or 200 mg/kg). Delta wave activity during NREM was enhanced after daily treatment with RGE (either 10 or 200 mg/kg) for 5 days. Both alpha and beta subunits of the $\gamma$-aminobutyric acid $(GABA)_A$ receptor were significantly over-expressed in the hypothalamus of the RGE-treated groups. Moreover, the expression of glutamic acid decarboxylase was also increased in the hypothalamus. These results demonstrate that RGE may regulate spontaneous sleep via $GABA_A$ergic systems.

        Identification of novel rheumatoid arthritis-associated MiRNA-204-5p from plasma exosomes

        Rheumatoid arthritis (RA) is an autoimmune disease characterized by infiltration of immune cells in the synovium. However, the crosstalk of immune cells and synovial fibroblasts is still largely unknown. Here, global miRNA screening in plasma exosomes was carried out with a custom microarray (RA patients vs. healthy controls = 9:9). A total of 14 exosomal miRNAs were abnormally expressed in the RA patients. Then, downregulated expression of exosomal miR-204-5p was confirmed in both the replication (RA patients vs. healthy controls = 30:30) and validation groups (RA patients vs. healthy controls = 56:60). Similar to the findings obtained in humans, a decreased abundance of exosomal miR-204-5p was observed in mice with collagen-induced arthritis (CIA). Furthermore, Spearman correlation analysis indicated that plasma exosomal miR-204-5p expression was inversely correlated with disease parameters of RA patients, such as rheumatoid factor, erythrocyte sedimentation rate, and C-reactive protein. In vitro, our data showed that human T lymphocytes released exosomes containing large amounts of miR-204-5p, which can be transferred into synovial fibroblasts, inhibiting cell proliferation. Overexpression of miR-204-5p in synovial fibroblasts suppressed synovial fibroblast activation by targeting genes related to cell proliferation and invasion. In vivo assays found that administration of lentiviruses expressing miR-204-5p markedly alleviated the disease progression of the mice with CIA. Collectively, this study identified a novel RA-associated plasma exosomal miRNA-204-5p that mediates the communication between immune cells and synovial fibroblasts and can be used as a potential biomarker for RA diagnosis and treatment.

        Bioactivity of essential oil of Litsea cubeba from China and its main compounds against two stored product insects

        During our screening program for agrochemicals from Chinese medicinal herbs and wild plants, the essential oilof Litsea cubeba fruits was found to possess strong contact toxicity against the cigarette beetle Lasiodermaserricorne adults and the booklouse Liposcelis bostrychophila, with LD50 values of 27.33 μg/adult and71.56 μg/cm2, respectively, and also showed strong fumigant toxicity against the two stored product insectswith LC50 values of 22.97 and 0.73 mg/L, respectively. The essential oil obtained by hydrodistillation wasinvestigated by GC MS. The main components of the essential oil were identified to be E-citral (geranial)(27.49%), Z-citral (neral) (23.57%) and D-limonene (18.82%) followed by β-thujene (3.34%), β-pinene (2.85%), α-pinene (2.57%), 6-methyl-5-hepten-2-one (2.40%) and linalool (2.36%). Citral (Z/E-citral), D-limonene, β-pinene,α-pinene and linalool were separated and purified by silica gel column chromatography and preparative thinlayer chromatography, and further identified by means of physicochemical and spectrometric analysis. Citral andlinalool showed strong contact toxicity against L. serricorne and L. bostrychophila (LD50 = 11.76, 12.74 μg/adultand 20.15, 99.97 μg/cm2, respectively) and fumigant toxicity against L. serricorne and L. bostrychophila (16.54,18.04 mg/L air and 0.14, 0.71 mg/L air, respectively). Otherwise, citral, D-limonene and linalool were strongly repellentagainst the cigarette beetle L. serricorne as the essential oil whereas β-pinene and α-pinene exhibited weakerrepellency against the cigarette beetle compared with the positive control, DEET. Moreover, except α-pinene andlinalool, the other three compounds as well as the essential oil exhibited comparable repellency against thebooklouse relative to DEET.

        Impact performance for high frequency hydraulic rock drill drifter with sleeve valve

        A high frequency hydraulic rock drill drifter with sleeve valve is developed to use on arm of excavator. In order to ensure optimal working parameters of impact system for the new hydraulic rock drill drifter controlled by sleeve valve, the performance test system is built using the arm and the hydraulic source of excavator. The evaluation indexes are gained through measurement of working pressure, supply oil flow and stress wave. The relations of working parameters to impact system performance are analyzed. The result demonstrates that the maximum impact energy of the drill drifter is 98.34J with impact frequency of 71HZ. Optimal pressure of YZ45 rock drill is 12.8 MPa-13.6MPa, in which the energy efficiency reaches above 58.6%, and feature moment of energy distribution is more than 0.650.

        Mixed organic acids improve nutrients digestibility, volatile fatty acids composition and intestinal microbiota in growing-finishing pigs fed high-fiber diet

        Objective: The objective of this study was to investigate effects of mixed organic acids (MOA) on nutrient digestibility, volatile fatty acids composition and intestinal microbiota in growing-finishing pigs fed high wheat bran diet. Methods: Six crossbred barrows (Duroc×Landrace×Yorkshire), with an average body weight 78.8±4.21 kg, fitted with T-cannulas at the distal ileum, were allotted to a double 3×3 Latin square design with 3 periods and 3 diets. Each period consisted of a 5-d adjustment period followed by a 2-d total collection of feces and then a 2-d collection of ileal digesta. The dietary treatments included a corn-soybean-wheat bran basal diet (CTR), mixed organic acid 1 diet (MOA1; CTR+3,000 mg/kg OA1), mixed organic acid 2 diet (MOA2; CTR+2,000 mg/kg OA2). Results: Pigs fed MOA (MOA1 or MOA2) showed improved (p<0.05) apparent total tract digestibility (ATTD) of gross energy, dry matter and organic matter, and pigs fed MOA2 had increased (p<0.05) ATTD of neutral detergent fiber compared to CTR. Dietary MOA supplementation decreased (p<0.05) pH value, and improved (p<0.01) concentrations of lactic acid and total volatile fatty acids (TVFA) in ileum compared to CTR. Pigs fed MOA showed higher (p<0.05) concentration of acetic acid, and lower (p<0.05) content of formic acid in feces compared to CTR. Pigs fed MOA1 had increased (p<0.05) concentration of TVFA and butyric acid in feces. Pigs fed MOA1 showed higher concentration of Lactobacillus and lower concentration of Escherichia in feces compared to CTR. Conclusion: Dietary supplementation of MOA 1 or 2 could improve nutrients digestibility, TVFA concentration and intestinal flora in growing-finishing pigs fed high fiber diet.
