        This study was carried out for the purpose of examining the effect of eating on muscular neutral fat and outcome of persistent exercise. Seven men who were taking persistent training had 2 hours of cyclying exercise at an intensity of 65%VO₂max, Thereafter the examinees took meals containing high level of sugars and fats and then carried out 1600kJ by self pace after fasting 12 hours. In the Performance taken 2 hours before starting cycling training (33.0+/- 2.3 vs. 37.0 +/- 2.1mmol/㎏ dry weight ) and in the Performance taken thereafter (30.9+/- 2.4 vs. 32.8+/- 1.6 mmol/㎏ weight) there was no difference in muscular neutral fat. However eating more fatty food showed higher value of muscular neutral fat prior to 2nd self paced exercise in comparison with taking more sugary food. (44.7+/- 2.4 vs. 27.5+/- 2.1 mmol/㎏ dry weight). In terms of outcome of self-paced cycling training Performance where highly fatty food was taken showed significant decline, This outcome did not show any significant change in muscular neutral fat before and after cycling training of medium level intensity but it was observed that absorption of highly fatty food heightened accumulation of muscular neutral fat rather than taking highly sugary food.

        A study was conducted on the effect f creatine loading on persistent exercise and outcome in sprint which was performed immediately after such exercise and subjects of this study were 12 cycle load athletes. Persistent exercise for the purpose of experiment was carried out for 2 hours and 30 minutes. Then it was continued until the condition of tiredness and exhaustion was reaching with showing density of lactate in blood of 4mmol/ℓand then immediately thereafter for 10 seconds 5 sets maximum sprint were carried out. The subject athletes carried out C Performance which featured absorption of placebo liquid during the exercise involving creatine loading prior to experiment exercise, CC Performance featuring absorption of creatine liquid during the exercise involving creatine loading and P Performance featuring absorption of placebo liquid during the exercise without creatine loading-thus totally 3 Performances were carried out. Comparing with P Performance C Performance showed significant improvement in all the 5 sets in terms of peak power and average power for maximum sprint. On the other hand in CC Performance it was same as P Performance. Creatine loading and creatine absorption liquid did not have any effect on duration of persisting exercise until reaching to fatigue and exhaustion. From the result of this study it could be observed that creatine loading improved maximum sprint ability within a short period of time after persistent exercise leading to fatigue and exhaustion However such effect declined when creatine liquid was absorbed during the exercise.

        Rural of Koreas has rapidly changed through the process of industrialization and urbanization. During past 30 years, development differences between urban and rural, metroplitan area and area derived from a highly economic growth, bring about dual specialization that deepens overpopulation and sparsity. Therefore, with a new concern about rural, necessity of rural development policy that developes a settlement and life area for settlement for ensuing stablized settlement and living activity of rural people, has been drived step by step. For rural welfare establishment that is settled and good hereafter, this study aims at attempting a theoretical approach about the way of development for rural settlement as a new rural development strategy through examination of today's rural reality, under assumption that rural people must be able to obtain stable jobs and to be pleasant without leaving rural area, and rural settlement to be able to enjoy urban advantages must be realized. Therefore, items that must be obtained basically rural area to be a human settlement space are equipment connected with a dwelling house, production, consumption, job relaxation, infrastructure connected with transportation or communication network and environment. By the way, the best function of a human settlement space makes a basic pleasant settlement area to be in the middle of that. Then rationalization of land use must be realized as being a best condition for human life around space of a dewelling house. Besides, spacial accessibility about all equipment of inhabitants must be easy. That is, economic distance, social distance and cultural distance as well as spacial distance must be included. Also, those spacial functions must be realized on a basis of suitable enironment for a human settlement life. At the same tiem, the upper area must be connected with the lower area as the system of spacial settlement in settlement space. But truely, as rural people can't satisfy with services of the surrounding rural areas, they desire better city or metroplitan. The these problems bring about inequality between urban and rural. Therefore, here are urgent reasons that must be done rearing of rural central town and rural integrated development of unit area. So, these dys, to correct maldistribution of land use and inequality of settlement space that Korean society faces on, new strategies of rural integrated development for lift area for settlement than gives the meaning. That rural areas become like a place for human settlement life will be able to be chosen in the view of humanism. The say of thinking of rural residents(Jukans-myun, Anseong-gun, Kyeonggi-do) was surveyed in order to develop new rural settlement planning. From the data collected in this survey, rural life-style, view on traditional rural life, agriculture, farming, agri-policy, social attitude, and degree of satifaction of rural life, etc, were analyzed. Firstly, most of rural residents interviewed in this survey have the opinion of not leaving their home village if living conditions improved to a reasonable level in the near future. In decision-making process of village affairs, they want to participate actively and equally. Secondly, in spite of their acceptance of agriculture as a vital sector destined to national security, most of them think that framing should be little of promise. Thirdly, they have the negative attitude to urban-oriented social valued and also to the agricultural policy, while seeking consolation through human and traditional viewing of rural life style. Therefore, in the development of new rural settlement model, idea-making should be directed to the betterment of communal value of village life, stability of rural economy, rationalization of rural life style and enhancement of rural-type happiness.

      • PCR을 이용한 Rat 기관지 세척액에서의 Pneumocystis carinii DNA의 검출 : 기초실험 A pilot study

        Background: Pneumocystis carinii (PC) is a opportunistic pathogen causing serious pneumonia (Pneumocystis carinii pneumonia : PCP) in immunosuppressed patients including AIDS. Laboratory diagnosis of PCP is dependent on microscopic demonstration of the PC by using cytochemical staining or by immunocytochemical staining with monoclonal antibodies. However, these staining methods are not highly sensitive. The development of the polymerase chain reaction (PCR) has made it possible to detect very small numbers of pathogens in clinical specimens. To establish the usefulness of PCR for detection of PC DNA, We performed PCR as a pilot study. Methods: PCP was induced on Spraque-Dawley rats with prednisolone 5mg IM twice a week. Bronchial lavages, impression smears, and permanent sections were performed from the 3rd week to 7th week of cortisonized rats. PC DNA was extracted with bead beater / 10% CTAB method. Results: Impression smears showed PC cyst after the 4th week (100%, 28/28 rats), and permanent sections showed 27/28 rats. Control rats showed some PC cysts (2/7). PCR result was positive only one case among the 6th week rats (1/7), But positive 6 cases at the 7th week rats, (6/7). Among control rats. 2 cases were positive (2/7). ??This data is a pilot study for the PC DNA detection using a PCR. PCP were successfully cortisonized rats. If adequate bronchial washing technique will be used, PCR can be a sensitive for PC DNA detection.

        Antimicrobial Activity of a Halocidin-Derived Peptide Resistant to Attacks by Proteases

        <B>ABSTRACT</B><P>Cationic antimicrobial peptides (AMPs) have attracted a great deal of interest as a promising candidate for a novel class of antibiotics that might effectively treat recalcitrant infections caused by a variety of microbes that are resistant to currently available drugs. However, the AMPs are inherently limited in that they are inevitably susceptible to attacks by proteases generated by human and pathogenic microbes; this vulnerability severely hinders their pharmaceutical use in human therapeutic protocols. In this study, we report that a halocidin-derived AMP, designated HG1, was found to be resistant to proteolytic degradation. As a result of its unique structural features, HG1 proved capable of preserving its antimicrobial activity after incubation with trypsin, chymotrypsin, and human matrix metalloprotease 7 (MMP-7). Additionally, HG1 was observed to exhibit profound antimicrobial activity in the presence of fluid from human skin wounds or proteins extracted from the culture supernatants of <I>Staphylococcus aureus</I> and <I>Pseudomonas aeruginosa</I>. Greater understanding of the structural motifs of HG1 required for its protease resistance might provide feasible ways to solve the problems intrinsic to the development of an AMP-based antibiotic.</P>

        Long-term cardiovascular events in hypertensive patients: full report of the Korean Hypertension Cohort

        Background/Aims: This study evaluated the long-term cardiovascular complications among Korean patients with hypertension and compared them with that of controls without hypertension. Methods: The Korean Hypertension Cohort (KHC) enrolled 11,043 patients with hypertension and followed them for more than 10 years. Age- and sex-matched controls without hypertension were enrolled at a 1:10 ratio. We compared the incidence of cardiovascular events and death among patients and controls without hypertension. Results: The mean age was 59 years, and 34.8% and 16.5% of the patients belonged to the high and moderate cardiovascular risk groups, respectively. During the 10-year follow-up, 1,591 cardiovascular events (14.4%) with 588 deaths (5.3%) occurred among patients with hypertension and 7,635 cardiovascular events (6.9%) with 4,826 deaths (4.4%) occurred among controls. Even the low-risk population with hypertension showed a higher cardiovascular event rate than the population without hypertension. Although blood pressure measurements in the clinic showed remarkable inaccuracy compared with those measured in the national health examinations, systolic blood pressure (SBP) ≥ 150 mmHg was significantly associated with a higher risk of cardiovascular events. Conclusions: This long-term follow-up study confirmed the cardiovascular event rates among Korean hypertensive patients were substantial, reaching 15% in 10 years. SBP levels ≥ 150 mmHg were highly associated with occurrence of cardiovascular event rates.

        Spleen Tyrosine Kinase Participates in Src-Mediated Migration and Proliferation by PDGF-BB in Rat Aortic Smooth Muscle Cells

        Tyrosine kinases, Src and spleen tyrosine kinase (Syk), play crucial roles in cell responses to platelet-derived growth factor (PDGF) and may have their functional interactions. In this study, we focused on investigating the roles of Syk in the regulation of Src signaling in PDGF-mediated vascular cell responses. Migration, proliferation, and activity of kinases were determined in rat aortic smooth muscle cells (RASMCs). PDGF-BB (10ng/mL) induced the migration and proliferation of RASMCs, which were significantly inhibited by PP2 (10 ${\mu}$M) and piceatannol(30 ${\mu}$M), inhibitors of Src and Syk, respectively. The phosphorylation of Syk induced by PDGF-BB was abolished by PP2. PDGF-BB increased the co-association of the PDGF${\beta}$-receptor and the kinases, Src or Syk, and its maximal binding to Src was achieved in a shorter time than that to Syk. PDGF-BB stimulated the phosphorylation of p38 mitogen-activated protein kinase(MAPK) and extracellular signal-regulated kinase (ERK) 1/2, which was inhibited by PP2 and piceatannol. PDGF-BB-induced proliferation and migration were inhibited by SB203580 (30${\mu}$M) and PD98059 (30 ${\mu}$M), inhibitors of p38 MAPK and ERK1/2, respectively. These result simply that Syk is regulated by Src kinase, which participates in migration and proliferation in response to PDGF-BB in RASMCs.

        Quercetin Enhances Human ${\alpha}$7 Nicotinic Acetylcholine Receptor-Mediated Ion Current through Interactions with $Ca^{2+}$ Binding Sites

        The flavonoid quercetin is a low molecular weight substance found in fruits and vegetables. Aside from its antioxidative effect, quercetin, like other flavonoids, has a wide range of neuropharmacological actions. The ${\alpha}$7 nicotinic acetylcholine receptor (${\alpha}$7 nAChR) has a $Ca^{2+}$-binding site, is highly permeable to the $Ca^{2+}$ ion, and plays important roles in $Ca^{2+}$-related normal brain functions. Dysfunctions of ${\alpha}$7 nAChR are associated with a variety of neurological disorders. In the present study, we investigated the effects of quercetin on the ACh-induced inward peak current ($I_{ACh}$) in Xenopus oocytes that heterologously express human ${\alpha}$7 nAChR. $I_{ACh}$ was measured with the two-electrode voltage clamp technique. In oocytes injected with ${\alpha}$7 nAChR cRNA, the effects of the co-application of quercetin on $I_{ACh}$ were concentration-dependent and reversible. The $ED_{50}$ was 36.1 + 6.1 ${\mu}m$. Quercetin-mediated enhancement of $I_{ACh}$ caused more potentiation when quercetin was preapplied. The degree of $I_{ACh}$ potentiation by quercetin preapplication was time-dependent and saturated after 1 min. Quercetin-mediated $I_{ACh}$ enhancement was not affected by ACh concentration and was voltage-independent. However, quercetin-mediated $I_{ACh}$ enhancement was dependent on extracellular $Ca^{2+}$ concentrations and was specific to the $Ca^{2+}$ ion, since the removal of extracellular $Ca^{2+}$ or the addition of $Ba^{2+}$ instead of $Ca^{2+}$ greatly diminished quercetin enhancement of $I_{ACh}$. The mutation of Glu195 to Gln195, in the $Ca^{2+}$-binding site, almost completely diminished quercetin-mediated $I_{ACh}$ enhancement. These results indicate that quercetin-mediated $I_{ACh}$ enhancement human ${\alpha}$7 nAChR heterologously expressed in Xenopus oocytes could be achieved through interactions with the $Ca^{2+}$-binding site of the receptor.
