      • 자궁경부암에서 Glutathione S Transferase-π의 분포에 관한 연구

        Uterine cervical carcinoma is the most common malignant tumor of woman in korea. GST-π is multifunctional and is implicated to detoxication of drug and carcinogenesis. This enzyme is expressed in many tumors of human. Relationship between GST-π and drug resistence on uterine cervical carcinoma is not known, but condyloma accuminatum, CIN and invasive carcinoma express this isoenzyme, and it is regarded as a useful marker in preneoplasia and neoplasia. To observe the expression of GST-π in CIN and invasive carcinoma, we analyzed CIN 26 cases and invasive squamous cell carcinoma 61 cases by immunohistochemical stain using monoclonal mouse anti-human GST-π. The results are as followings: 1) The GST-π showed positive reactionin normal and metaplastic epthelium adjacent to CIN and invasive carcinoma in forty-three cases(62.3%) of sixty-nine cases. 2) The GST-π showed positive reactionin CIN in eighteen cases(69.2%) of twenty-six cases. Positive rates of CIN Ⅰ, CIN Ⅱ and CIN Ⅲ are 66.7%, 71.5%, 68.8%, respectively. 3) The GST-π showed positive reaction in invasive squamous cell carcinoma in fifty-one cases(83.6%) of sixty-one cases. Positive rates of microinvasive and invasive squamous cell carcinoma are 84.6% and 83.3%, respectively. According to cell type, keratinizing and nonkeratinizing type are 81.8% and 84.6%, respectively. Above results revealed that GST-π may be significance as a immuohistochemical marker in CIN and invasive SCC, and may be implicated in intrinsic resistance for anticancer drug.

        정신분열병환자의 혈소판막과 ³H-spiperone결합에 관한 연구

        This research was to determine whether changes of dopamine receptors on the platelet membrane of schizophrenics could be used as a biological marker for schizophrenia. ³H-spiperone binding to platelet membranes was carried out in 22 schizophrenic patients who fulfilled DSM-ⅢR criteria and 14 age-matched control subjects. ³H-spiperone specifically binds to dopamine D2 receptor with high affinity. Increased Bmax can suggests increase in the number of dopamine receptors on the platelet membrane. The results are as follows ; 1) Bmax of the patient group was significantly higher than that of control group(p<.05). 2) Kd was not significantly different between the two groups. 3) Bmax of the positive and the negative symptom groups were significantly higher than the control group when the patient group was subdivided into the positive, the negative and the mixed symptom groups(p<0.5). 4) Kd of the positive symptom groups, the negative symptom groups and the control group were not significantly different among each other. 5) Bmax and Kd of the positive and the mixed and the negative symptom groups were not significantly different among each other. This results shows that increase in the number of depamine receptors on the platelet membrane of schizophrenics would be a part of disease process in schizophrenia.

        We show that biased annealing of a-Si:H contacted with metal gives rise to silicide growth and the creation of metastable states that can be to some extent removed by unbiased annealing. This paper gives the first report of a direct correlation of Ⅰ-Ⅴ characteristics to silicide growth in a-Si: H. Variations of Ⅰ-Ⅴ characteristics with annealing biases are analysed by the two series-connected diodes model.

      • 위암에서의 CEA의 분포에 관한 연구

        Carcinoembryonic antigen(CEA) isolated from adenocarcinoma of human digestive system and fetal or embyonic gut tissue is one of tumor associated antigen. In recent years, extensive study in tissue of various condition has been reported but study in gastric cancer, common malignant tumor in korea, was rare. Therefore, to understand positive rate of CEA according to degree of differentiation of gastric cancer, the author collectd serial 95 cases of formalin fixed, paraffin embedded stomach obtained by subtotal resection, performed immunoenzyme histologic reaction by Avidin-Biotin-Complex method using anti-human CEA rabbit serum and analyzed their histologic reation. Results obtained are as followings 1. CEA positive reaction to anti-CEA antibody was noted in 26(77%) of the 95 cases with gastric cancer. 2. CEA postive reaction to anti-CEA antibody was noted in 26(84%) of the 33 cases with well differentiated gastric cancer. 3. CEA positive reaction to anti-CEA abtibody was noted in 20(74%) of the 27 cases with moderately differentiated gastric caner. 4. CEA positive reaction to anti-CEA antibody was noted in 27(73%) of the 37 cases with poorly differentiated gastric cancer.

      • 대장선암에서 p53과 c-erbB-2의 표출 및 상관관계에 관한 연구

        Colorectal adenocarcinoma is relatively common malignant tumor in korea and various oncogenes has found in tumor cells. Colorectal adenocarcinoma show overexpression of p53 and c-erbB-2. The relation between expression of p53 and c-erbB-2 and prognosis of patients in colorectal adenocarcinoma has studied recently, but that is controversy. To evaluate relation between expression of p53 and c-erbB-2 and histopathologic finding prognostic factors, we have performed immunohistochemical staining, using mouse anti-p53 and anti-human c-erbB-2, for 52 cases of colorectal adenocarcinoma. The results were as follows; 1) Expression rate of p53 and c-erbB-2 in colorectal adenocarcinoma were 71.2% and 51.9%, respectively. 2) Expression rate of p53 in left side(70.4%), differentiated(75.6%) and invasion to serosa and surrounding adipose tissue(72.5%) were higher than in right side(37.5%), poorly differnetiated(42.9%) and invasion to muscle(50%). 3) Expression rate of c-erbB-2 in left side(51.9%), differentiated(57.8%) and invasion to serosa and surrounding adipose tissue(54.5%) were higher than in right side(37.5%), poorly differnetiated(14.3%) and invasion to muscle(37.5%). 4) The expression of p53 and c-erbB-2 showed coexpression in colorectal adenocarcinoma. 5) The expression of p53 and c-erbB-2 showed no differenece according to presence or absence of lymphatic metastasis. Above result revealed that p53 and c-erbB-2 are coexpression in colorectal adenocarcinoma. The expression of each p53 and c-erbB-2 and the simultaneous expression of p53 and c-erbB-2 may be related to left side and differentiated cases. Our studies for relation with prognosis of patients in colorectal adenocarcinoma showed different results according to depth of invation and metastasis to lymph node and we should be followed for them.

        노인 외상 환자의 중증도 분류 시 Triage-revised Trauma Score의 의의

        Backgrouds: The mortality of geriatric trauma patients is higher than that of other age groups. However, little research has been done the methods or criteria of triage for geriatric trauma patients. This study evaluated a clinical significance of the triage-revised trauma score(t-RTS) for triage of geriatric trauma patients. Methods: We retrospectively analyzed clinical data on 528 trauma patients over 65 years of age who were treated from Jan 1999 to Dec 2000. The t-RTS was calculated utilizing the RR(respiratory rate), SBP(systolic BP), and GCS scores and the ISS was abstracted from the final diagnosis. The obtained t-RTS and ISS were evaluated using the measures of sensitivity, specificity, accuracy, and AUC curve. Results: The overall mortality rate was 9%, and there was no significant differences between the survival group and the mortality group according to age and sex. The mean scores of SBP, RR and GCS of the survival group were significantly higher than those of mortality group(p=0.001). The mean of t-RTS and RTS of the survival group were also significantly higher(p=0.001), but the ISS was significantly higher in the mortality group(p=0.001). The t-RTS, RTS, and ISS showed good prediction rates on the ROC curve(p=0.001), and the AUC value was higher in the ISS than in the t-RTS and the RTS. The sensitivity and the accuracy were high in the t-RTS and the RTS, and the specificity was high in the ISS. The t-RTS is less than 10 for a survival probability of 50% or less Conclusions: Implementation of the t-RTS in the triage of geriatric trauma patients in the field and in emergency room would be very useful. The probability of death in geriatric trauma patients is high when the t-RTS is less than 10.

      • 대장선암에서 Metallothionein과 p53의 표출

        Colonic adnocarcinoma is common malignant tumor in Korea. Many oncogenes have been found in malignant tumor cells. Among these oncogenes, MT has high affinity for metals such as copper and zinc. The fuction of MT is metabolism and detoxification of copper and zinc. The relation between the expression of MT and prognosis of patients in various cancers are reported but is controversial. p53 is a regulator of cellular proliferation, and the over expression of p53 relates to malignant trasformation and prognosis. To evaluate the expression of MT and p53 according to histopathologic features in colonic adenocarcinoma, we have performed immune staining, using mouse anti-MT and mouse anti-p53. The results were as follow; 1) The expression rates of MT in hyperplastic polyp, adenoma, and adenocarcinoma of colon were 100%, 64.1%, and 46.6%, p53 were 0%, 64.1%, and 48.9%, respectively. 2) MT and p53 in colonic adnoma showed increased espressionrat according to severity of dysplasia 3) MT incolonic adenoarcinoma showed statisfically significant higher expressionrates in female9P=0.04), in cases showing lymph node metastasis(P=0.012), and in stage C(P=0.0001), but there is no significant difference according to age, site, differentiation, and invasion depth. 4) p53 in colinic adenocarcinoma showed statisfically significant higher expression rates rates in cases showing lymph node metastasis(P=0.047), but there is no statisfically significant difference in age, sex, site, invasion depth, stage. 5) In colonic adenocarcinomas expressing MT(+)p53(+) there is more many cases in carcinomas within the wall, in carcinoma showing lymph node metastasis, and stage C, but no statisfically significant difference. 6) Correlation between MT and p53 in colonic adenocarcinoma were not seen.

        PC12를 이용한 신경세포 허혈모델에서 irp94 유전자의 발현

        Background: The ischemia responsive protein 94 kDa(irp94) gene belongs to the heat shock protein 110 family and was isolated in 1999 from rat brain by transiently induced forebrain ischemia. The PC12 cell is the pheochromocytoma cell line of rat, which is differentiated to a sympathetic neuron-like cell by the stimulation of a nerve growth factor. This study is to determine whether irp94 is expressed when an ischemia-like condition is induced by ATP depletion in cultured PC12 cells in vitro. Methods: PC12 cells were maintained as monolayer cultures in RPMI-1640 medium(Sigma) supplemented with 10% horse serum, 5% fetal bovine serum, 5 mg/ml transferrin, and 1 mg/ml insulin in a humidified 5% CO2 incubator at 37℃. The ATP depleting agent antimycin A was added at concentrations of 1, 2.5, and 5 μM to simulate ischemia, and 10 μg/ml of tunicamycin, which is expected to express heat shock protein maximally, was used as a positive control. The cells were harvested after a 60-minute incubation, and the total RNA was extracted. The reverse transcription polymerase chain reaction(RT-PCR) was performed to use 501 bp irp94 cDNA as a molecular probe, and the expression of irp94 mRNA was analyzed by northern blotting. Results: The irp94 mRNA expression was enhanced, compared to the negative control group, as the concentration of antimycin A was increased. Conclusion: This study suggests that irp94 mRNA expression is enhanced as the severity of ischemia is increased. Thus, it is possible to investigate the mechanism of ischemic neuronal injury indirectly by using this in-vitro model of neuronal ischemia.

        CNC 공작기계에서 상호결합제어기를 위한 새로운 윤곽오차모델

        In the control of CNC machine tools, it is significant for precise machining to reduce the contour error. The object of servo-control is reduction of contour error and necking error. In past studies, there were two approaches to control a servo-system. One was to eliminate axial tracking errors, and the other was to control contour errors. The Cross-coupled con-troller(CCC) was introduced from a viewpoint of contour errors. For constructing CCC, it is necessary to make the contour error model. Recently, for machining part with free form surfaces, we propose a new contour error model based on curve interpolator. It is presented here that performance of CCC using proposed model is enhanced. Therefore, we can make more precise parts with the curve interpolator and the new contour error model.
