        FSCB phosphorylation in mouse spermatozoa capacitation

        It is generally accepted that spermatozoa capacitation is associated with protein kinase A-mediated tyrosine phosphorylation. In our previous study, we identified the fibrous sheath CABYR binding protein (FSCB), which was phosphorylated by PKA. However, the phosphorylation status of FSCB protein during spermatozoa capacitation should be further investigated. To this aim, in this study, we found that phosphorylation of this 270-kDa protein occurred as early as 1 min after mouse spermatozoa capacitation, which increased over time and remained stable after 60 min. Immunoprecipitation assays demonstrated that the tyrosine and Ser/Thr phosphorylation of FSCB occurred during spermatozoa capacitation. The extent of phosphorylation and was closely associated with the PKA activity and spermatozoa motility characteristics. FSCB phosphorylation could be induced by PKA agonist DB-cAMP, but was blocked by PKA antagonist H-89.Therefore, FSCB contributes to spermatozoa capacitation in a tyrosine-phosphorylated format, which may help in further elucidating the molecular mechanism of spermatozoa capacitation.         Development of Nested PCR, Multiplex PCR, and Loop-Mediated Isothermal Amplification Assays for Rapid Detection of Cylindrocladium scoparium on Eucalyptus

        Eucalyptus dieback disease, caused by Cylindrocladiumscoparium, has occurred in last few years in largeEucalyptus planting areas in China and other countries. Rapid, simple, and reliable diagnostic techniquesare desired for the early detection of Eucalyptus diebackof C. scoparium prior to formulation of efficientcontrol plan. For this purpose, three PCR-based methodsof nested PCR, multiplex PCR, loop-mediatedisothermal amplification (LAMP) were developed fordetection of C. scoparium based on factor 1-alpha (tef1)and beta-tubulin gene in this study. All of the threemethods showed highly specific to C. scoparium. Thesensitivities of the nested PCR and LAMP were muchhigher than the multiplex PCR. The sensitivity of multiplexPCR was also higher than regular PCR. C. scopariumcould be detected within 60 min from infectedEucalyptus plants by LAMP, while at least 2 h wasneeded by the rest two methods. Using different Eucalyptustissues as samples for C. scoparium detection,all of the three PCR-based methods showed muchbetter detection results than regular PCR. Base on theresults from this study, we concluded that any of thethree PCR-based methods could be used as diagnostictechnology for the development of efficient strategiesof Eucalyptus dieback disease control. Particularly,LAMP was the most practical method in field applicationbecause of its one-step and rapid reaction, simpleoperation, single-tube utilization, and simple visualizationof amplification products.

        Raman Lidar for the Measurement of Temperature, Water Vapor, and Aerosol in Beijing in the Winter of 2014

        To measure atmospheric temperature, water vapor, and aerosol simultaneously, an efficient multi-function Raman lidar using an ultraviolet-wavelength laser has been developed. A high-performance spectroscopic box that utilizes multicavity interference filters, mounted sequentially at small angles of incidence, is used to separate the lidar return signals at different wavelengths, and to extract the signals with high efficiency. The external experiments are carried out for simultaneous detection of atmospheric temperature, water vapor, and aerosol extinction coefficient in Beijing, under clear and hazy weather conditions. The vertical profiles of temperature, water vapor, and aerosol extinction coefficient are analyzed. The results show that for an integration time of 5 min and laser energy of 200 mJ, the mean deviation between measurements obtained by lidar and radiosonde is small, and the overall trend is similar. The statistical temperature error for nighttime is below 1 K up to a height of 6.2 km under clear weather conditions, and up to a height of 2.5 km under slightly hazy weather conditions, with 5 min of observation time. An effective range for simultaneous detection of temperature and water vapor of up to 10 km is achieved. The temperature-inversion layer is found in the low troposphere. Continuous observations verify the reliability of Raman lidar to achieve real-time measurement of atmospheric parameters in the troposphere.

        The preservation effect of Metschnikowia pulcherrima yeast on anthracnose of postharvest mango fruits and the possible mechanism

        This study aimed to determine the effects of Metschnikowia pulcherrima yeast on storage quality of 'Tainong' mango, and elucidate it's possible anti-disease mechanism. The results showed that M. pulcherrima could inhibit the changes in peel colour, fruit firmness, the contents of total soluble solids, total acid and vitamin C, and maintain the storage quality of mango fruits. An investigation of the mechanism showed that M. pulcherrima competed not only for the primary carbon source, but also for living space with Colletotrichum gloeosporioides. In addition, M. pulcherrima promoted the activities of defence-related enzymes, including ${\beta}$-1,3-glucanase(GLU) and chitinase (CHT), and secreted a small amount of antimicrobial substances composed of volatile and nonvolatile anti-fungal compounds. The results strongly demonstrated that antagonistic yeast M. pulcherrima could be applied as a biocontrol agent for deducing the spoilage and decay of mango fruit.

      • Simultaneous quantitation and validation of method for the quality evaluation of Eucommiae cortex by HPLC/UV

        A new HPLC/UV method has been developed for the simultaneous quantitative determination of four major components in Eucommiae cortex, namely geniposidic acid (1), geniposide (2), pinoresinol di-O-β-D-glucopyranoside (3), and liriodendrin (4). Simultaneous separations of these four components were achieved on a J``sphere ODS CI8 column (250 x 4.6 mm, 4 mm), The elution was done using water with 0.1 % phosphoric acid (A) and acetonitrile with 0.1 % phosphoric acid (B) in a two-step elution of the mobile phase at a flow rate of 1.0 mL/min and a wavelength of 230 nm. The method was validated for linearity, recovery, precision, accuracy, stability and robustness. All calibration curves showed good linear regression (r<sup>2</sup> > 0.999) within the test ranges. This method showed good recovery and repro-ducibility for the quantification of these four components in 85 species of Eucommiae cortex. The intra-day and inter-day precisions were lower than 0.53 % (as a relative standard deviation, RSD) and accuracies between 93.00 and 106.28 % for all standards. The results indicate that the established HPLC/UV method is suitable for quantitation and quality evaluation of Eucommiae cortex.

        Dynamic analysis method for the progressive collapse of long-span spatial grid structures

        In the past, the progressive collapse resulting from local failures during accidents has caused many tragedies and loss of life. Although long-span spatial grid structures are characterised by a high degree of static indeterminacy, the sudden failure of key members may lead to a catastrophic progressive collapse. For this reason, it is especially necessary to research the progressive collapse resistance capacity of long-span spatial grid structures. This paper presents an evaluation method of important members and a novel dynamic analysis method for simulating the progressive collapse of long-span spatial grid structures. Engineering cases were analysed to validate these proposed method. These proposed methods were eventually implemented in the progressive collapse analysis of the main stadium for the Universiade Sports Center. The roof of the structure was concluded to have good resistance against progressive collapse. The novel methods provide results close to practice and are especially suitable for the progressive collapse analysis of long-span spatial grid structures.

        Simultaneous analysis of seven marker compounds from Saposhnikoviae Radix, Glehniae Radix and Peucedani Japonici Radix by HPLC/PDA

        A new combination of high performance liquidchromatography (HPLC) method coupled with photodiodearray (PDA) analysis has been developed for the simultaneousquantitative determination of seven major components inSaposhnikoviae Radix (SR), Glehniae Radix (GR) and PeucedaniJaponici Radix (PR), namely peucedanol 7-O-b-Dglucopyranoside(1), prim-O-glucosylcimifugin (2), cimifugin(3), 40-O-b-D-glucosyl-5-O-methylvisamminol (4), bergapten(5), sec-O-glucosylhamaudol (6), and imperatorin (7). Clearseparation of these seven components were achieved on aPhenomenex Kinetex C18 (250 9 4.6 mm, 5 lm) column bygradient elution of water (A) and methanol (B) as mobilephase. The flow rate was 1.0 mL/min and the UV detectorwavelength was set at 254 nm. The method was successfullyused in the analysis of SR, GR, and PR with relatively simpleconditions and procedures, and the results were satisfactory forlinearity, recovery, precision, accuracy, stability and robustness. The results indicate that the established HPLC/PDAmethod is suitable for the classification of SR, GR, and PR.

        Knockdown of Chloride Channel-3 Inhibits Breast Cancer Growth In Vitro and In Vivo

        Purpose: Chloride channel-3 (ClC-3) is a member of the chloride channel family and plays a critical role in a variety of cellular activities. The aim of the present study is to explore the molecular mechanisms underlying the antitumor effect of silencing ClC-3 in breast cancer. Methods: Human breast cancer cell lines MDAMB- 231 and MCF-7 were used in the experiments. Messenger RNA and protein expression were examined by quantitative realtime polymerase chain reaction and western blot analysis. Cell proliferation was measured by the bromodeoxyuridine method, and the cell cycle was evaluated using fluorescence-activated cell sorting. Protein interaction in cells was analyzed by co-immunoprecipitation. Tumor tissues were stained with hematoxylin- eosin and tumor burden was measured using the Metamorph software. Results: Breast cancer tissues collected from patients showed an increase in ClC-3 expression. Knockdown of ClC-3 inhibited the secretion of insulin-like growth factor (IGF)-1, cell proliferation, and G1/S transition in breast cancer cells. In the mouse xenograft model of human breast carcinoma, tumor growth was significantly slower in animals injected with ClC- 3-deficient cells compared with the growth of normal human breast cancer cells. In addition, silencing of ClC-3 attenuated the expression of proliferating cell nuclear antigen, Ki-67, cyclin D1, and cyclin E, as well as the activation of extracellular signalregulated protein kinases (ERK) 1/2, both in vitro and in vivo. Conclusion: Together, our data suggest that upregulation of ClC- 3 by IGF-1 contributes to cell proliferation and tumor growth in breast cancer, and ClC-3 deficiency suppresses cell proliferation and tumor growth via the IGF/IGF receptor/ERK pathway.
