        가압가열 및 microwave에 의한 중력분 반죽 gliadin의 항원성 변화

        본 연구에서는 가압가열 및 microwave 처리에 의한 중력분 반죽 추출물 내의 gliadin 단백질의 항원성 변화에 대해 살펴보았다. 중력분 반죽에 가압가열과 microwave를 단독 또는 병행으로 처리하여 ci-ELISA, SDS-PAGE 및 immunoblotting을 실시하였으며, 가압가열 처리에 의해서 anti-gliadin IgG 항체와 gliadin과의 결합력이 다소 감소한 것을 확인하였다. 특히 30 min 이상 처리시 더욱 감소한 것으로 나타났으며, SDS-PAGE와 immunoblotting 결과에서도 gliadin band의 강도가 약해지고 항체와의 반응도 나타나지 않았다. Microwave 처리의 경우, 5 min 이상 처리시 일부 gliadin 단백질의 소실이 관찰되었으나, 항원성에는 큰 변화가 없었다. 또한 가압가열 및 microwave 병행 처리에 의해 항원-항체 결합력이 더욱 감소되었으며, 특히 가압가열 50 min, microwave 10 min 처리시 약 35.0%로 감소되었다. 이상의 결과를 통해 가압가열 처리에 의해 중력분 반죽 추출물 내 gliadin의 항원성이 감소되는 것을 확인하였으며, microwave와 병행 처리하는 경우, 더욱 감소하는 것을 확인하였다. The aim of this study was to determine the optimal physical treatment to reduce the antigenicity of gliadin in wheat dough. Medium wheat dough was treated with an autoclave (5, 10, 30, and 50 min at 121℃, 1 atm), a microwave (1, 5, and 10 min) or both (10, 30, and 50 min/5, 10 min). The proteins in the dough extracts were analyzed by SDS-PAGE and the binding ability of anti-gliadin IgG to gliadin was examined by ci-ELISA and immunoblotting. Results showed that the ability of anti-gliadin IgG to bind to gliadin in wheat dough treated with an autoclave alone or in combination with a microwave was decreased. Especially, it declined to ~77% after autoclaving for 30 min and 35% after both autoclaving for 50 min and microwaving for 5 min. In addition, the intensity of gliadin bands in SDS-PAGE were weakened and anti-gliadin IgG did not recognize gliadin in immunoblotting. However, microwaving alone did not affect the antigenicity of gliadin in wheat dough. These results indicate that autoclaving may affect the reduction of the antigenicity of gliadin in medium wheat dough. Moreover, autoclaving in combination with microwaving is more effective for reducing the antigenicity of wheat dough.

        가압가열 및 Microwave 처리에 의한 중력분 Gliadin의 항원성 변화

        본 연구에서는 가압가열 및 microwave 처리가 gliadin의 항원성에 미치는 영향을 살펴보기 위해 중력분에 가압가열과 microwave를 단독 또는 병행으로 처리하여 Ci-ELISA, SDS-PAGE 및 immunoblotting을 실시하였다. 가압가열 처리의 경우, 처리 시간이 길어질수록 IgG와의 결합력이 감소하였으며, 특히 50분 처리구에서 약 69%로 가장 낮은 결합력을 보였다. 또한 SDS-PAGE와 immunoblotting 결과에서도 무처리구에서 강하게 보였던 gliadin band가 처리에 의해 거의 소실되고 항체와 반응하지 않았다. 가압가열 및 microwave를 병행 처리한 경우도 마찬가지로 gliadin의 결합력이 다소 감소하였으며, 처리구 중에서는 가압가열 50분, microwave 5분 처리구에서 약 73%로 가장 낮은 결합력을 보였다. 반면 microwave를 단독으로 처리하였을 때에는 일부 단백질의 변화는 관찰되었으나 항원성 감소에는 큰 영향을 미치지 않았다. 이상의 결과를 통해 가압가열을 단독 처리에 의해 gliadin의 항원성이 다소 감소되었으며, microwave 병행 처리에 의한 차이는 크게 나타나지 않은 것을 확인하였다. This study was conducted to evaluate the effect of physical treatments on the antigenicity of gliadin in medium wheat flour. The wheat flour was treated with an autoclave (5, 10, 30, 50 min), a microwave (1, 5, 10 min), and both (10, 30, 50 min/ 5, 10 min), and investigated by SDS-PAGE, immunoblotting and Ci-ELISA using anti-gliadin IgG. The results showed that the binding ability of anti-gliadin IgG to gliadin in wheat flour was slightly decreased when autoclaved or when autoclaved and microwaved. Especially, it was reduced to about 69% after autoclaving for 50 min and 73% after autoclaving for 50 min and microwaving for 5 min. In addition, gliadin bands in the 50 min autoclaved group disappeared in both SDS-PAGE and immunoblotting. On the other hand, the antigenicity of gliadin was unaffected by microwaving alone. Consequently, there were no considerable changes in using an autoclave alone or in combination with a microwave. These results suggest that autoclaving may affect the reduction of the antigenicity of gliadin in medium wheat flour.

        인간포배기 배아의 효과적인 유리화 동결법의 개발을 위한 연구

        Objective: The purpose of this study was to evaluate the survival rate of vitrified blastocyst according to the freezing vessels, equilibration time in cryoprotectant and artificial dehydration method. Methods: Human blastocysts were vitrified after loading onto the plastic straw, open-pulled straw (OPS), electron microscopy grid (EM grid) for 1.5 min or 3 min. They also were directly plunged into LN2 within 30sec. For artificial shrinkage of blastocysts, 36 gauge fine needle was pushed at the cellular junction of the trophectoderm into the blstocoele cavity until it shrank without damage of inner cell mass. Results: The survival rate of vitrified blastocysts on plastic straw, OPS, EM grid as freezing vessels were 26.7, 13.0 and 60.5%, respectively. The survival rate of EM grid was significantly higher than that of plastic straw and OPS (p<0.05). For 1.5 min equilibrium, the survival rates of early blastocyst (EB), middle blastocyst (MB) and late blastocyst (LB) were 64.4, 81.0, and 20.0% respectively. For 3 min equilibrium, the survival rates of EB, MB, and LB were 69.9, 50.0 and 57.5% respectively. The survival rates of EB and MB were significantly higher than that of LB in 1.5 min equilibrium group (p<0.05), however, the significance was not observed in 3 min equilibrium groups. In cytoplasmic shrinkage before vitrification, the survival rates of EB, MB and LB were 92.9, 100 and 75.9% respectively. The survival rate of MB was significantly higher than that of LB (p<0.05). The survival rates of vitrified blastocysts by artificial dehydration and slow-frozen blastocysts were not significantly different as 88.9 and 66.7%, respectively. Conclusion: This study showed that the vitrification of human blastocysts using EM grid and artificial dehydration is an effective method. Therefore, these methods would be an useful techniques for blastocyst cryopreservation.

      • Crosslinked anion exchange membranes with primary diamine-based crosslinkers for vanadium redox flow battery application

        <P><B>Abstract</B></P> <P>A series of polysulfone-based crosslinked anion exchange membranes (AEMs) with primary diamine-based crosslinkers has been prepared via simple a crosslinking process as low-cost and durable membranes for vanadium redox flow batteries (VRFBs). Chloromethylated polysulfone is used as a precursor polymer for crosslinked AEMs (CAPSU-x) with different degrees of crosslinking. Among the developed AEMs, CAPSU-2.5 shows outstanding dimensional stability and anion (Cl<SUP>−</SUP>, SO<SUB>4</SUB> <SUP>2−</SUP>, and OH<SUP>−</SUP>) conductivity. Moreover, CAPSU-2.5 exhibits much lower vanadium ion permeability (2.72 × 10<SUP>−8</SUP> cm<SUP>2</SUP> min<SUP>−1</SUP>) than Nafion 115 (2.88 × 10<SUP>−6</SUP> cm<SUP>2</SUP> min<SUP>−1</SUP>), which results in an excellent coulombic efficiency of 100%. The chemical and operational stabilities of the membranes have been investigated via ex situ soaking tests in 0.1 M VO<SUB>2</SUB> <SUP>+</SUP> solution and in situ operation tests for 100 cycles, respectively.         어류 알의 Protease Inhibitor 활성 분포

        To identify and examine the distribution of proteolytic inhibitory activity in crude extracts from fish eggs, and to determine the applicability of these protease inhibitors as anti-degradation agents in surimi-based products and fish meat, we compared the inhibitory activities of various extracts from fish eggs to those of commercial proteases, such as trypsin and papain. We used the optimal conditions for the screening of trypsin activity: 30 ug/uL of 0.1 % trypsin and 0.6 mM Na-benzoyl-L-arginine-p-nitroanilide (BAPNA) with a pH of 8.0 at 40℃ for 60 min. The activities of papain and four commercial proteases were investigated after mixing with 100 ug/uL enzymes and 0.3% casein with a pH of 8.0 at 40℃ for 60 min. We performed a screening assay to detect the inhibitory activity (%) of crude extracts from eight species of fish eggs against the target proteases trypsin and papain. The assay revealed a wide distribution of trypsin and papain inhibitors in fish eggs. The specific inhibitory activities (11.6-28.6 U/mg) of crude extracts from fish eggs against trypsin and BAPNA substrate were higher than that (0.64 U/mg) of egg whites, used as a commercial inhibitor. The inhibitory activities of crude extracts from fish eggs against trypsin, and of egg whites against casein substrate (1.94-4.51 U/mg), were higher than those of papain (0.24-1.57 U/mg) and commercial protease (0.04-0.32 U/mg). The extracts from fish eggs were rich in protease inhibitors that exhibited strong inhibitory activity against trypsin, a serine protease, and papain, a cysteine protease.

      • Ether-free polymeric anion exchange materials with extremely low vanadium ion permeability and outstanding cell performance for vanadium redox flow battery (VRFB) application

        <P><B>Abstract</B></P> <P>Ether-free poly(<I>p</I>-phenylene)-based anion exchange membrane materials with outstanding vanadium ion blocking properties are reported. The synthesized ionomers with a rigid backbone structure show small dimensional variations despite the high ion exchange capacities. Among the synthesized membranes, the QPPP-2 membrane with ion exchange capacity of 2.1 meq·g<SUP>−1</SUP> exhibits high anion (OH<SUP>−</SUP>, SO<SUB>4</SUB> <SUP>2−</SUP>, and Cl<SUP>−</SUP>) conductivity. The QPPP-2 membrane shows an extremely low diffusion coefficient (2.12 × 10<SUP>−9</SUP> cm<SUP>2</SUP> min<SUP>−1</SUP>) for VO<SUP>2+</SUP>, which is much lower than that of Nafion 115 (2.88 × 10<SUP>−6</SUP> cm<SUP>2</SUP> min<SUP>−1</SUP>), indicating the vanadium ion blocking property of QPPP-2. Moreover, the chemical and operational stabilities of the membranes are investigated <I>via</I> ex situ soaking tests in 0.1 M VO<SUB>2</SUB> <SUP>+</SUP> solution and in situ operation tests for 100 cycles, respectively. The vanadium redox flow battery single cell assembled with the QPPP-2 membrane exhibits a coulombic efficiency of 99%, voltage efficiency of 87%, and energy efficiency of 86% during 100 cycles at 80 mA cm<SUP>−2</SUP>.         Lactobacillus pentosus 발효에 의한 담쟁이덩굴 줄기 추출물의 항산화 및 세포보호 효과

        본 연구에서는 담쟁이덩굴 줄기 70% 에탄올 추출물과 발효균주 Lactobacillus pentosus를 이용하여 발효시킨 담쟁이덩굴 줄기 발효추출물에 대하여 항산화 및 세포보호 효과를 측정하였다. 1,1-Diphenyl-2-picrylhydrazyl (DPPH)를 이용한 자유라디칼 소거 활성(FSC<sub>50</sub>)은 담쟁이덩굴 줄기 추출물 및 발효추출물이 각각 42.3 및 34.5 μ g/mL로 발효 후의 라디칼 소거활성이 약 18.4% 더 높게 나타났다. Lumiol-의존성 화학발광법을 이용한 Fe<sup>3+</sup>-EDTA/H<sub>2</sub>O<sub>2</sub>계에서의 총 항산화능(OSC<sub>50</sub>) 평가에서도 담쟁이덩굴 줄기 추출물과 발효추출물은 각각 2.6 및 2.5 μ g/mL로 발효 후가 약 4.2% 정도 더 높은 총 항산화능을 나타냈다. <sup>1</sup>O<sub>2</sub>로 유도된 적혈구 세포손상에 있어서 추출물 및 발효추출물의 세포 보호 효과(τ<sub>50</sub>)는 50 μ g/mL에서 각각 126.4 및 173.0 min을 나타내어 발효 후 세포 보호 효과가 약 34.0% 더 높게 나타났다. 발효추출물은 지용성 항산화제로 알려진 (+)-α-tocopherol (43.4 min)보다도 3.9배 높은 세포 보호 활성을 보여주었다. 사람 섬유아세포인 Hs68을 대상으로 elastase 저해 활성을 조사하였다. Elastase 저해 활성(IC<sub>50</sub>)은 담쟁이덩굴 줄기 추출물과 발효추출물에서 각각 873.6 및 687.8 μ g/mL로 발효 후에 elastase 저해 활성이 약 21.3% 더 높은 것으로 나타났다. 이상의 결과들은 담쟁이덩굴 줄기 발효추출물이 항산화 작용과 더불어 주름개선 효과를 가지는 천연 화장품 소재로써 응용 가능성이 있음을 시사한다. In this study, the antioxidant activities, cellular protective effects, and inhibitory effects on elastase of non-fermented and fermented extracts of Parthenocissus tricuspidata (P. tricuspidata) stem using Lactobacillus pentosus were investigated. The free radical scavenging activities (FSC<sub>50</sub>) of non-fermented and fermented extracts were 42.3 and 34.5 μg/mL, respectively, in which the activity after fermentation was approximately 18.4% higher. Reactive oxygen species (ROS) scavenging activities (OSC<sub>50</sub>) in Fe<sup>3+</sup>-EDTA/H<sub>2</sub>O<sub>2</sub> system of non-fermented and fermented extracts were 2.6 and 2.5 μg/mL, respectively. The activity after fermentation was approximately 4.2% higher. In the <sup>1</sup>O<sub>2</sub>-induced cellular damage of erythrocytes, the cellular protective effects (τ<sub>50</sub>) of non-fermented and fermented extracts were 126.4 and 173.0 min at 50 μg/mL, respectively. The activity after fermentation was approximately 34.0% higher. The effect of fermented extract was 3.9 times higher than (+)-α-tocopherol (τ<sub>50</sub> = 43.4 min), known as a lipophilic antioxidant at 50 μg/mL. The inhibitory effect of elastase was investigated to predict the anti-wrinkle efficacy using Hs68 human fibroblasts cells. The elastase inhibitory activities (IC<sub>50</sub>) of non-fermented and fermented extracts were 873.6 and 687.8 μg/mL, respectively, and the activity after fermentation was approximately 21.3% higher. These results indicated that fermented extract of P. tricuspidata stem has potentials as natural cosmetic ingredients with antioxidant and anti-wrinkle effect.

      • 상대적 교란, 상대가격변동성과 인플레이션율간의 관계에 대한 실증분석

        Analytic contributions in the imperfect information market-clearing framework have shown how uncertainty about inflation can reduce the efficiency of the price system and how relative price variablity is likely to be greater when there are unanticipated changes in the price level. Such analyses, combined with the assumption the high inflation rates are also more uncertain, provide a rationalization for the view that inflation may itself be a factor explaining the poor performance of industrial economies after 1973. In this paper I investigate the empirical realtions between relative shocks, relative price variability, and inflation in Korea for the 1975-1990 perids. Three conclusions are supported by findings of this paper. First, there are positive relations between relative price variability and the rate of inflation in Korea for the periods. Second, the association between relative price variability and inflation is dominated by relative shocks like food and energy shocks. Third, from a welfare economics viewpoint, inflation and relative price variability affect negatively the real GNP.

      • 환율변동성이 수출에 미치는 효과분석 : The Case of Korean Export to the U.S. during the Market Average Exchange Rate System of 1990's

        The purpose of this paper is to show the empirical evidence that the exchange rate volatility risk on export could be positive. GARCH(1,l) process, known to be superior method t o any other measures, is used to generate a measure of volatility risk. Exchange rate volatility risk measured by GARCH(1,l) process must be stationary so that volatility risk cannot be included in export equation with any other I(1) variables. Johansen multivariate cointegration test is applied to investigate if there exists a cointegration vector between export volume index, relative price, and the U.S. industrial production index. We can come to conclude that there is at least 1 cointegration vector between three I(1) variables. So the long-run equilibrium export equation is estimated. In the long-run, the relative price has a inelastic negative effects, while the U.S. industrial production has a elastic positive effects on Korean export to the U.S. At the next step, 1st differenced three variables, which are transformed to I(0) stationary, and volatility risk, which is measured GARCH(1.1) process, make up ECM of short-run adjustment dynamics. In the short-run adjustment, exchange rate volatility risk has a statistically significant positive effect on various period lags. This empirical evidence seems to be explained by the real option model and/or the moral hazard phenomenon of Korean exporting firms.

        Credit rationing is a condition of loan markets in which the lender supply of funds is less than borrower demand at the quoted contract terms. Moral hazard and adverse selection as much more general features of loan markets are the source of credit rationing when there is asymmetrical information. The moral hazard feature arises because individual borrowers choose to operate risker projects at higher loan rates. The adverse selection feature arises become unprofitable at higher loan rates, causing the remaining pool of loan applicants to become risker. Thus, while higher loan rates increase the lender's expected revenue for all borrowers. Given that the risk character of individual borrowers and projects cannot be identified a priori, it may be optimal policy for the lender to set the loan rate below the market-clearing level and to ration credit. For rationing to exist on a continuing basis in loan markets, therefore, the interest rate must be maintained below the market-cleaning level by special factors.
